Characterization of salicylate uptake across the basolateral membrane of the malpighian tubules of Drosophila melanogaster.

The organic anion salicylate is a plant secondary metabolite that can protect plants against herbivores. Transport of salicylate across the basolateral membrane of the Malpighian tubules of Drosophila melanogaster was studied using a radioisotope tracer technique. The uptake of [(14)C]salicylate by the Malpighian tubules was active, saturable and Na(+)-dependent; the maximum uptake rate (J(max)) and the half saturation concentration (K(t)) were 12.6 pmoltubule(-1)min(-1) and 30.7micromoll(-1), respectively. In contrast to organic anion transport by vertebrate renal tissues, salicylate uptake was not trans-stimulated by glutarate (0.01-1.0 mmoll(-1)) or cis-inhibited by high concentrations (5 mmoll(-1)) of various alpha-keto acids (glutaric acid, alpha-ketoglutaric acid, succinic acid, and citric acid). Changes in basolateral membrane potential or physiologically relevant changes in bathing saline pH did not affect the rate of [(14)C]salicylate uptake. Ring-structure monocarboxylic acids (benzoic acid, nicotinic acid, gentisic acid, unlabelled salicylic acid, alpha-cyano-4-hydroxycinnamic acid, probenecid, fluorescein, and P-aminohippuric acid) strongly inhibited [(14)C]salicylate uptake rate. In contrast, short-chain monocarboxylic acids had little (butyric acid) or no effect (lactic acid, pyruvic acid, and propionic acid). Our results suggest that salicylate uptake across the basolateral membrane of D. melanogaster Malpighian tubules is mediated by a non-electrogenic, alpha-cyano-4-hydroxycinnamic acid-sensitive, Na(+):salicylate cotransport system.

Specific method for serum creatinine determination based on ion exchange chromatography and an automated alkaline picrate reaction -- a proposed reference method.

A proposed reference method for serum creatinine has been developed under the auspices of the Committee on Reference Methods and Reference Materials of the Canadian Society of Clinical Chemists. A serum ultrafiltrate at pH 2.0 is applied through a closed sample loop injection system to a short column containing cationic resin of high resolving power. Elution with sodium citrate buffer by means of minipump at constant rate passes the eluate into alkaline picrate reagents in a continuous flow system (Technicon AAIII pump, AAII colorimeter 50 mm x 1.5 mm flow cell, narrow band width filter). The colour reaction peak is monitored visually to verify specificity and the area is calculated electronically. Specificity has been demonstrated by use of Jaffé-reactive substances such as glucose, sodium acetoacetate, L-ascorbic acid, pyruvic acid, L-dopa and glycocyamidine and also by use of an alternate colour reaction, sodium 3,5-dinitrobenzoate in place of alkaline picrate in the analysis of serum pools. Routine methods in common use, i.e., manual and automated alkaline picrate procedures, demonstrated a statistically significant high bias in interlaboratory studies in which this procedure was used for reference.

Gas chromatographic-mass spectrometric analysis of acids and phenols in distilled alcohol beverages. Application of anion-exchange disk extraction combined with in-vial elution and silylation.

A GC-MS protocol for profiling spirits, based on 19 acids and phenolic compounds, has been proposed and evaluated. The method combined a simple preconcentration procedure based on solid-phase (anion-exchange) disk extraction, and in-vial elution and silylation of the analytes. The derivatized extract was directly injected into the GC-MS system. These analytes were: C6, C8, C10, C12 acids, pyruvic acid, 2-furoic acid, succinic acid, fumaric acid, glutaric acid, lactic acid, glycolic acid, malic acid, tartaric acid, citric acid, vanillin, syringaldehyde, coniferaldehyde, vanillic acid and gallic acid. The profiles of six different spirits were found reproducible from day-to-day with <20% RSD for measurements of most of the analytes at different concentrations. Recoveries of individual analytes appear to be affected by the level of tannins in the spirits, and they varied from sample to sample. The method of standard addition was used to quantify age-related analytes. Good linearity of response with correlation coefficients in the range of 0.992-0.999 was obtained. The results of the study indicate that for spirits of the same brand but of different ages, the amounts of these analytes appear to increase with the ageing period.

Thermaerobacter subterraneus sp. nov., a novel aerobic bacterium from the Great Artesian Basin of Australia, and emendation of the genus Thermaerobacter.

A strictly aerobic, thermophilic, gram-positive, spore-producing, rod-shaped bacterium (2.0-10.0 x 0.3 microm), designated isolate C21T, was isolated from a sample collected from an open drain run-off channel of a bore in the Great Artesian Basin of Australia (New Lorne Bore, registered number 17263). Isolate C21T grew optimally at 70 degrees C (temperature range for growth was 55-80 degrees C) and pH 8.5 (pH range for growth was 6.0-10.5), with a generation time of 90 min. The isolate was strictly heterotrophic and grew on yeast extract and/or tryptone as carbon and energy sources. An increase in growth was not observed with carbohydrates (sucrose, cellobiose, glucose, dextrin, amylopectin, chitin, carboxymethylcellulose, xylan, inositol, arabinose, mannose, fructose, gelatin, starch, amylose, galactose, dextrose, xylose, maltose, L-sorbose or raffinose), organic acids (lactic acid, pyruvic acid or benzoic acid) or Casamino acids as sole carbon sources or in the presence of yeast extract and/or tryptone. The G+C content of the chromosomal DNA, as measured by the thermal denaturation method, was 71 mol%. Phylogenetic analysis of the 16S rRNA gene of isolate C21T placed it as a member of the phylum Firmicutes, with Thermaerobacter marianensis as the closest relative (similarity value of 98%). However, isolate C21T and T. marianensis differed in a number of key physiological and phenotypic properties and also had a DNA-DNA hybridization value of less than 5%. Based on this evidence, it is proposed that strain C21T be designated Thermaerobacter subterraneus sp. nov. (type strain C21T = ATCC BAA-137T = DSM 13965T).

The characteristics of a new non-spore-forming cellulolytic mesophilic anaerobe strain CM126 isolated from municipal sewage sludge.

A new mesophilic anaerobic cellulolytic bacterium, CM126, was isolated from an anaerobic sewage sludge digester. The organism was non-spore-forming, rod-shaped, Gram-negative and motile with peritrichous flagella. It fermented microcrystalline Avicel cellulose, xylan, Solka floc cellulose, filter paper, L-arabinose, D-xylose, beta-methyl xyloside, D-glucose, cellobiose and xylitol and produced indole. The % G + C content was 36. Acetic acid, ethanol, lactic acid, pyruvic acid, carbon dioxide and hydrogen were produced as metabolic products. This strain could grow at 20-44.5 degrees C and at pH values 5.2-7.4 with optimal growth at 37-41.5 degrees C and pH 7. Both endoglucanase and xylanase were detected in the supernatant fluid of a culture grown on medium containing Avicel cellulose and cellobiose. Exoglucanase could not be found in either supernatant fluid or the cell lysate. When cellulose and cellobiose fermentation were compared, the enzyme production rate in cellobiose fermentation was higher than in cellulose fermentation. The optimum pH for both enzyme activities was 5.0, the optimum temperature was 40 degrees C for the endoglucanase and 50 degrees C for the xylanase. Both enzyme activities were inhibited at 70 degrees C Co-culture of this organism with a Methanosarcina sp. (A145) had no effect on cellulose degradation and both endoglucanase and xylanase were stable in the co-culture.

Superficial chemical peels.

Superficial chemical peeling (SCP) involves the application of a peeling agent to the skin, resulting in destruction of part or all of the epidermis. SCP is mainly recommended for facial rejuvenation, photoaging and superficial rhytides, pigmentary dyschromias and acne. It can be used on all Fitzpatrick skin types, no sedation is needed, and the desquamation is usually well accepted. Overpeel and complications are very rare. The most commonly used SCP agents are glycolic acid 20-70%, trichloroacetic acid 10-35%, Jessner's solution, salicylic acid, pyruvic acid, resorcinol 30-50% preparations, and solid carbon dioxide. The careful selection of patients is critical for the outcome of a SCP and contraindications must be seriously considered. The peel procedure is generally common for all SCP agents but a good knowledge of the specific characters of each agent is of great importance in order to decide which to use for each individual patient.

Characterization of low molecular weight organic acids from beech wood treated in supercritical water.

Japanese beech (Fagus crenata Blume), its cell wall components, and model compounds were treated by supercritical water (380 degrees C, 100 MPa) for 5 s using a batch-type reactor to investigate the production behavior of low molecular weight organic acids. It was found that cellulose and hemicellulose were decomposed to formic acid, pyruvic acid, glycolic acid, acetic acid, and lactic acid, whereas lignin was barely decomposed to such organic acids under the given conditions. However, after prolonged treatment (380 degrees C, 100 MPa, 4 min) of lignin, some organic acids were recovered owing perhaps to the decomposition of the propyl side chain of lignin. It was additionally revealed that the predominant organic acid recovered was acetic acid, which might be derived from the acetyl group of hemicellulose in Japanese beech.

Improved somatic growth following adenoidectomy and tonsillectomy in young children. Possible pathogenetic mechanisms.

The effect of Adenotonsillectomy on somatic growth was studied prospectively in 57, randomly selected children (31 boys, 26 girls), aged 5.03+/-1.32 (mean+/-1SD) years. The indication for surgery was adenotonsillar hypertrophy with or without recurrent infections. Weight, height, triceps skinfold thickness, and Body Mass Index were measured prior to the operation and 6-13 months afterwards. Weight was significantly improved following T&A in all children. The improvement in height was significantly only for children under 5 years. In an attempt to uncover the pathogenetic mechanisms, lactic acid, pyruvic acid, somatomedin-C (IGF-I), growth hormone(GH), insulin, glucose, pH, hemoglobin (Hbeta). and white cell count (WBC) were also determined in the last 18 children, prior to and 6-8 months post operatively. For the comparison of pre and post operative values the paired t test was applied. Although the values of GH and IGF1 did not significantly increase post-op the IGF-1/GH ratio increased, possibly indicating improved IGF1 generation. There was also a rise in Hbeta values and a lowering of WBC, probably reflecting the lower frequency of infections. All other metabolic indices did not change. In conclusion, linear grow post-Adenotonsillectomy improved in children aged <5 years and was associated with improved with IGF-1/GH ratio, increased Hbeta values and decreased in WBC.

Effects of artificial circulation by pulsatile and non-pulsatile flow on brain tissues.

We examined the effects of artificial circulation by pulsatile and non-pulsatile flow on microcirculation in the brain from the viewpoints of circulation and metabolism in the brain. A centrifugal pump was fixed in the bypass in the right heart of 10 pigs. In 5 of the 10 pigs, a pulsatile flow pump was fixed in the bypass in the left heart (P group), and in the remaining 5 pigs, a centrifugal pump was fixed in the bypass in the left heart (NP group). Hemodynamics were periodically monitored for 3 hours while maintaining about 100 mmHg of the mean aortic pressure. Intracranial pressure (ICP), cerebral tissue blood flow and cerebral blood flow (CBF) were measured and compared with the initial values. As the parameters of metabolism in the brain, the cerebral oxygen consumption and lactic acid - pyruvic acid ratio were evaluated. If the cerebral blood flow was reduced by cardiogenetic shock, we suggest that blood circulation and metabolism in the brain were maintained by artificial circulation. It also indicated that there was no significant difference in blood circulation and metabolism in the brain between artificial pulsatile and non-pulsatile flow circulation.

Comparison of ion-pair chromatography and capillary zone electrophoresis for the assay of organic acids as markers of abnormal metabolism.

The abnormal organic acids in urine are closely related with physiological metabolism. To determinate the low-molecular-mass metabolites in human biological fluids, although there were some previous reports by both of capillary electrophoresis and ion-exchange high-performance liquid chromatography, but it was rarely found by reverse phase of liquid chromatography using ion pair reagent. The objective of this study was aimed to suggest and compare two methods, an additional chromatographic method-ion-pair chromatography (IPC) and a sharp capillary zone electrophoresis (CZE), to determinate organic acids, acting as the abnormal metabolic markers, namely uric acid, orotic acid, pyruvic acid, alpha-ketoglutaric acid, fumaric acid, and hippuric acid. The proposed method of IPC possessed both the extreme stability for column and the good results of reproducibility, linearity and detection limit. The optimum mobile phase was 22% methanol and 10 mM tetra-n-butyl ammonium hydrogen sulfate (pH 4) by gradient elution. As well as the optimum condition of CZE was 5% acetonitrile and 0.5 mM CTAB in phosphate buffer. From the results, CZE showed better recovery and sharp lucid electropherogram. Finally, the two proposed analytical methods were applied to assay human urine with direct and spiked analysis. CZE showed good potency to overcome the sample-to sample variation with standard deviation less than 10%. By comparison results of urinary spiked analysis between IPC and CZE by statistical paired t-test, the results were evaluated no significant difference under P < 0.05. The quantitative linearity of both methods was fitted in application of clinical biological analysis even with 50-fold dilution.

[Effects of anesthesia on the metabolic response during pelvic surgery].

Dividing anesthesia for gynecological laparotomy into nitrous oxide-oxygen-halothane (GOF) anesthesia (group A), nitrous oxide-oxygen (GO) plus ketamine anesthesia (group B) and GOF plus epidural anesthesia (group C), we studied effects of each anesthesia on circulatory and metabolic systems. During the operation, influence on the heart rate was slight in three groups, while systolic pressure decreased significantly in group C. The lactic acid in the blood was elevated in three groups, but the elevation was least in group C. The elevation of lactic acid pyruvic acid ratio (L/P) was less in group C. The blood glucose level was elevated significantly in three groups, and it was highest in group A. On the other hand NEFA was elevated significantly in group A, while no change was noted in group B, and decreasing was noted in group C. The results indicate that no large influence of each anesthesia on the metabolic system was noted in group A and B, and its influence was the least in group C.

[A 60-year-old man with intention tremor as an initial symptom followed by cerebellar ataxia, peripheral neuropathy and dementia].

We report a 60-year-old man with a 6-year history of tremor in his hands. He noted the onset of short of breath and gait disturbance in 1994; both of these symptoms were slowly progressive. Then recently he developed fever two months prior to the present admission. He was admitted to the rheumatology department of our hospital and neurological consultation was asked on December 13, 2000. On neurologic examination, he showed Gottron sign and fine crackle in both lungs. Pertinent neurological findings were bilateral dysmetria in finger-to-nose and heel-to-knee tests and a broad-based gait. In addition, he showed intention tremor in upper extremities more on the left. Romberg sign was positive. Deep tendon reflexes were decreased. Vibratory sensation was reduced at the wrists. The patient's hemoglobin was 11.1 g/dl, with a mean corpuscular volume of 92.0 fl. Vitamin B12 level was 190 (reference range, >230 pg/ml). Serum lactic acid, pyruvic acid and ceruloplasmin were slightly elevated. Chest X-ray showed interstitial pneumonia. Muscle biopsy showed grouping of small angular fiber. Brain MRI showed diffuse atrophy of the cerebral cortex and the cerebellum hemisphere. Thalamotomy did not improve his tremor. He was admitted again in November 2001. General worsening of his neurological findings was observed. IL2-receptor was markedly elevated. Serum anti-Hu, Yo and Ri antibodies were negative. An anaplastic carcinoma was found in his jejunum. He died from respiratory failure in February 2002. He was discussed in a neurological CPC and the chief discussant arrived at a conclusion that the patient had paraneoplastic syndrome. Other diagnosis entertained included MERRF, GSS, Ramsay Hunt syndrome, subacute combined degeneration, spinocerebellar degeneration. Majority of the participants thought that paraneoplastic syndrome was most likely. Post-mortem examination revealed poorly differentiated carcinoma in the small intestine. Myeline pallor was noted in the posterior and the lateral columns in the thoracic spine. Neuronal cell loss was observed in the Purkinje cell and granular cell layer in the cerebellum. Sural nerve demonstrated loss of myelinated fibers and grouping of small fibers. Neuropathological findings were consistent with Friedreich ataxia; nevertheless, no mutation was reported in frataxin in Japan. The neuropathologist concluded that neuropathological diagnosis was a spinocerebellar ataxia with neuropathological similarities to Friedreich ataxia.

[Changes in respiratory, circulatory, endocrine, and metabolic systems under induced total spinal block].

Total spinal block was induced by 2% lidocaine in adult mongrel dogs, and its influence on the respiratory, circulatory, endocrine and metabolic systems was studied. HR, MAP, CI and LV dp/dt max during total spinal block decreased significantly compared with those during the control period. The blood gas, Qs/Qt and ETVI were hardly influenced by total spinal block. Regarding the endocrine and metabolic systems, plasma renin activity (PRA) was significantly elevated by total spinal block, while plasma epinephrine and norepinephrine levels showed no changes. Blood levels of lactic acid, pyruvic acid and blood sugar were not influenced by total spinal block.

[The effect of galbanic acid on the course of experimental hepatitis].

Administration of galbanic acid isolated from the roots of Ferula kopetdaghensis Eug. Kor. to rats orally in a dose of 50 mg/kg was found to improve the course of toxic hepatitis induced by fourfold subcutaneous injections of a 50% oil solution of CCl4. Galbanic acid produced much more earlier than in control normalization of the activity of the enzymes alanine- and aspartataminotransferase in blood serum, increased glycogen content and improved the parameters of the redox potential of lactic acid-pyruvic acid system in the liver. Galbanic acid exerted the antioxidant effect. In the animals with a developed hepatitis receiving galbanic acid there was a more rapid restoration of intensity of bile secretion, synthesis of bile acids and bilirubin, cholesterol excretion as compared with control.

[Biological and biochemical characteristics of the oral spirochetes isolated from the focus of marginal periodontitis].

Since they are found to be increased in lesions of acute necrotic ulcerative gingivitis or marginal periodontitis, agents for these diseases. In the present study, 38 pure cultured strains were obtained as a result of isolation and culture of samples collected from lesions of marginal periodontitis (periodontal pokets), and the biological and biochemical characteristics of these strains were investigated. 1) Light microscopy (including dark-field microscopy) and transmission electron microscopy (negative staining) were used for observation of the morphology and cellular structure of the strains. The cells had a spiral shape, and showed active movement. Based on the above findings the cultured strains were all confirmed to be spirochetes of small to medium size, being 0.08-0.24 micron in width. 2) Growth and motility of the strains were investigated on various types of culture medium. Intense growth and movement were noted in strains cultured in bovine liver exudate medium containing horse serum (pH 7.2) at 37 degrees C under anaerobic conditions produced by the evacuation-replacement method (95% N2, 5% CO2) for 3-7 days after inoculation. 3) Thirty-five strains were positive for indole production and decomposition of urea, mucin, hippuric acid and esculin. Production of hydrogen sulfied was observed in 31 strains. In decomposition tests for 17 carbohydrates, 17 strains were positive for galactose and 14 strains were positive for glucose, while 11 strains were positive for dextrin and 10 strains for fructose upon decomposition of soluble starch. Other carbohydrates were also decomposed by a few strains. 4) In an investigation of the production of alcohol and lower fatty acids, among the metabolic products detected by gas chromatography, a large amount of acetic acid and small amounts of ethanol, lactic acid, propionic acid, pyruvic acid were observed. 5) The results of enzyme activity tests using an API ZYM system indicated relatively high activities of esterase, esterase-lipase, alpha-glucosidase, alkaline phosphatase, trypsin and acid phosphatase.

Morphological and biochemical studies on liver, kidney and gill of fishes affected by pesticides.

Effect of a herbicide, paraquat (1,1'-dimethyl-4,4'-bipyridilium-dichloride), the fungicide copper sulphate, and zinc chloride was studied on the histological structure of liver, kidney and gill of three fish species with different feeding habits, viz.: a herbivorous, silver carp (Hypophthalmichthys molitrix); an omnivorous, common carp (Cyprinus carpio L.) and a carnivorous, sheatfish (Silurus glanis L.). The organs were studied electron microscopically after fixation according to Karnovsky. The toxic effect manifested itself characteristically on the respective species, regardless of the type of the chemical applied and the species specificity. Upon the effect of the treatments applied the cytoplasm of the respiratory cells of the gill became electron transparent and the cytoplasmic organelles disappeared almost totally. In the chloride cells showing focal necrosis, residuals of nuclear, mitochondrial and endoplasmic origin were seen. Pillar cells and the pericytes remained intact. In the nucleus of the liver cells, electron dense heterochromatin was not present. The degree of the damage in the liver cells was indicated by swollen mitochondria with electron transparent matrix and by dilatation and vacuolation of the endoplasmic reticulum system. Epithelial cells decreased in electron density, the endoplasmic reticulum was vesiculated, mitochondria were swollen. Leucocytes increased in number, and empty vacuoles and vacuoles filled with dense granules appeared in them during toxicosis. Copper sulphate or paraquat increased serum transaminase enzyme activities (glutamic acid-oxalacetic acid transaminase, glutamic acid-pyruvic acid transaminase) in all the three fish species. These damages can cause serious disturbances in energy uptake and secretion processes of fish.

[Use of clofelin as an anesthetic and component of neurovegetative defense in ENT oncologic surgery].

Metabolism indices (blood sugar, lactic acid, pyruvic acid) and hemodynamics (mean arterial pressure, 1-hour diuresis) were measured in 41 hypertensive subjects undergoing resection or removal of the larynx for cancer under endotracheal anesthesia through tracheostoma. 18 patients of group 1 received calypsol anesthesia, whereas 23 patients of group 2 received combination of calypsol with clofelin. Croup 2 patients benefited more because clofelin potentiated the analgetic effect of calypsol, intensified neurovegetative block, promoted more rapid recovery of normal respiration, prolonged analgetic effect in postoperative period.

[Effect of hemodilutional autotransfusion on coagulative-fibrinolytic dynamics and metabolic response].

We studied the effect of hemodilutional autotransfusion on coagulative-fibrinolytic dynamics and metabolic response. Hemodilutional solutions used were Dextran-40 (group A) and Salin-HES (group B). The activated partial thromboplastin time (APTT) immediately after hemodilution was prolonged in both groups. Prothrombin time (PT) and hepaplastin decreased significantly, and a remarkable variation was observed particularly in group A. The results suggest that effect on fibrinolytic dynamics is not exerted in either group because antithrombin-III (AT-III) and fibrinogen decreased significantly, while fibrinogen degradation products (FDP) are within normal ranges, and plasminogen as well as alpha 2-plasmin inhibitor (alpha 2-PI) decreased significantly. On the other hand, all other parameters such as lactic acid level, pyruvic acid level in blood, lactic acid pyruvic acid ratio, and blood glucose level were elevated during surgery, but no difference was observed regarding these parameters between the two groups.
