Tandem mass spectrometric analysis of aspergillus niger pectin methylesterase: mode of action on fully methyl-esterified oligogalacturonates.

The substrate specificity and the mode of action of Aspergillus niger pectin methylesterase (PME) was determined using both fully methyl-esterified oligogalacturonates with degrees of polymerization (DP) 2-6 and chemically synthesized monomethyl trigalacturonates. The enzymic activity on the different substrates and a preliminary characterization of the reaction products were performed by using high-performance anion-exchange chromatography at neutral pH. Electrospray ionization tandem MS (ESI-MS/MS) was used to localize the methyl esters on the (18)O-labelled reaction products during the course of the enzymic reaction. A. niger PME is able to hydrolyse the methyl esters of fully methyl-esterified oligogalacturonates with DP 2, and preferentially hydrolyses the methyl esters located on the internal galacturonate residues, followed by hydrolysis of the methyl esters towards the reducing end. This PME is unable to hydrolyse the methyl ester of the galacturonate moiety at the non-reducing end.

[Structure and physiological activity of lemnan, Lemna minor L. pectin].

A pectic polysaccharide, lemnan, was isolated from freshly collected duckweed Lemna minor L. Its sugar chain was shown to be mainly composed of the residues of D-galacturonic acid (64%), galactose, arabinose, xylose, and D-apiose, a branched chain sugar. The high content of D-apiose (25%) indicated that lemnan is an apiogalacturonan type pectin similar to zosteran, a pectic polysaccharide from a sea phanerogam of the Zosteraceae family. The results of partial acidic hydrolysis, pectinase digestion, and NMR studies of lemnan demonstrated that its macromolecule contains regions of the linear alpha-1,4-D-galacturonan and branched apiogalacturonan. The side chains of apiogalacturonan were found to be formed of single and 1,5-linked residues of D-apiofuranose attached to 2- and 3-positions of the D-galacturonic acid residues of the apiogalacturonan backbone. Lemnan was shown to exhibit an immunomodulatory effect by activating the system of phagocytosis.

[The difference of the bismuth absorption from a single colloidal bismuth pectin therapy and quadruple therapy for eradicating Helicobacter pylori infection].

OBJECTIVE: To investigate whether the quadruple therapy influences the absorption of bismuth from Colloidal Bismuth Pectin (CBP). METHODS: 24 male Wistar rats weighing from 400 g to 500 g were randomly divided into two groups: CBP group (group A(1)) and CBP+AMO+MTR+ LAN group (group A(2)). The serum bismuth concentration was observed at 0,1,2,3,4,5,6 hour after these medicines were fed. Other 24 male Wistar rats weighing from 200 g to 250 g were randomly divided into two groups: CBP group (group B( 1)), CBP+AMO+MTR+LAN group (group B(2)). These medicines had been taken every day for 14 days. The accumulation of bismuth in the heart, brain, kidney and liver of rats were detected eight weeks later. RESULTS: The serum peak value of bismuth is not significantly different between group A(1) and group A(2) (P > 0.05), but the peak time of bismuth of the group A(2) is one hour earlier than that of group A(1). The amounts of the accumulation of bismuth in the kidney are 154 +/- 15 ng/g and 212 +/- 20 ng/g in the group B(1) and B(2) respectively at the eighth week after the medicines were fed in rats with a significant difference (P < 0.05). CONCLUSION: The quadruple therapy causes an increase of the absorption and the accumulation of bismuth from CBP in the kidney in rats.

[Effectiveness of natural pectin-containing fruit pastes in patients with chronic enteritis].

As many as 56 patients with chronic enteritis in different stages aged 19-55 years were studied for myoelectrical activity of the intestines before treatment, following a single ingestion of pastes, and at the end of the course, intestinal absorptive function, and condition of the microflora. Combined treatment involving the use of pectin-containing apple paste in patients with chronic enteritis has a positive effect on the basic pathogenetic links of the disorder, which fact permits avoiding polypragmasy, undesirable medicinal preparations-associated complications, allergic reactions. It can come to be widely used in medical practice for treating patients with intestinal disorders.

Effect of guar, pectin, psyllium, soy polysaccharide, and cellulose on breath hydrogen and methane in healthy subjects.

Different types of dietary fiber are fermented to various extents in vitro, but little is known about the effects of fiber on breath hydrogen and methane levels in vivo. Therefore, we studied the effects on breath hydrogen and methane of 15 g of guar, pectin, psyllium, soy polysaccharide, or cellulose in eight healthy subjects over a 12-h period. None of the fibers had a significant effect on breath hydrogen or methane concentrations, compared with the control (fasting). The four methane producers had lower breath hydrogen levels than the nonproducers 1 h after 15 g of lactulose (3 +/- 1 vs. 42 +/- 9, p less than 0.005) and 5-12 h after the different fibers (3.3 vs. 4.8 ppm; pooled SEM = 0.8; p less than 0.025). When the methane responses of the methane producers were expressed as increments relative to the control, there were small differences between treatments, with guar producing a larger response, 8.2 +/- 3.3 ppm, than cellulose, -2.9 +/- 2.3 ppm (p less than 0.05). The incremental methane responses of the different fibers in vivo were related to the previously reported production of propionic acid (r = 0.94, n = 5, p less than 0.02) and methane (r = 0.93, n = 4, NS) from in vitro fermentation of the same fibers. We conclude that methane producers have lower breath hydrogen levels than nonproducers. Purified fermentable and nonfermentable dietary fibers have no effect on breath hydrogen levels over 12 h in subjects previously consuming a normal diet. However, fermentable fibers may produce small increases in breath methane in methane-producing subjects.

[Lowering cholesterol with Anticholest--a high fiber guar-apple pectin drink].

In order to determine the efficacy of Anticholest, an apple-pectin-guar soft-drink in reducing elevated cholesterol (c) levels, 33 participants (aged from 8 to 73 years) were divided into three groups of comparable age, body-mass index, total c, LDL-c, HDL-c, and triglycerides. They received this combined fiber product either (group 1) at dosages of 1 cup (17 g) every second day, or (group 2) of 1 cup a day or (group 3) of 2 cups a day. Anticholest significantly reduced total c, LDL-c and the total c/HDL-c ratio. In group 3 HDL-c was increased significantly (p less than 0.01). The average percentage decrease in total c was 10.0% for group 1, 10.7% for group 2, and 15.7% for group 3. LDL-c was lowered 14.4% in group 1, 13.8% in group 2 and 19.1% in group 3. The highest individual reduction amounted to more than 30% for total-c and LDL-c. A significant decrease was also detected in the triglycerides (tg) in group 3 (p less than 0.05). 7 patients were non-responders. Anticholest appears to be an appropriate treatment for patients at risk of coronary heart disease with insufficient or inadequate response to dietary measures, but where long-term pharmacotherapy is not yet indicated. The lack of side effects and its form of administration as a viscous drink are important determinants for long-term compliance, which is essential in the management of a chronic progressive disease such as atherosclerosis.

Combined enzymatic hydrolysis and HPAEC method for simultaneous analysis of galacturonic acid and neutral sugars of pectin.

Pectic substances are heteropolysaccharides from plant cell walls, which mainly consist of a homogalacturonan backbone of predominantly alpha-(1-->4) linked galacturonic acid (GalA) residues. This chain is interrupted by ramified rhamnogalacturonan regions with a certain amount of neutral sugars (rhamnose, arabinose, galactose, glucose, xylose and mannose) present as side-chains. The GalA residues can be methyl-esterified at the carboxyl group. Usually uronic acids of pectin are determined as anhydrogalacturonic acid by spectrophotometry, using the metahydrodiphenyl method (Thibault, 1979), while the monosaccharides are determined by High-Performance Liquid Chromatography (HPLC) after a methanolysis (Quemener and Thibault, 1990) or by Gaz Liquid Chromatography (GLC) as alditol acetates according to Blakeney et al. (1983). In this paper, a combined enzymatic hydrolysis and HPLC method is validated for simultaneous analysis of galacturonic acid and neutral sugars without any derivatisation.

[Maintenance of bactericidal activity of pectin stabilized aqueous solutions].

Maintenance of the bactericidal activity and organoleptic properties of 3% and 5% stabilized aqueous solutions of pectin (Pepidol PEG, 3% and Pepidol PEG, 5%) stored for prolonged periods and at various temperatures was studied. Pepidol PEG was shown to preserve its bactericidal activity and organoleptic properties for 28 months when stored at rather wide ranges of the environmental temperature. The drug did not lose its properties after freezing and subsequent melting. The results of the study allowed to increase the period of Pepidol PEG use up to 18 months and the storage temperature ranges up to -30 degrees to +30 degrees C.

[Effect of acid suppression therapy for eradicating Helicobacter pylori infection on bismuth absorption from colloidal bismuth pectin].

OBJECTIVE: To investigate whether acid suppression therapy influences the absorption of bismuth from colloidal bismuth pectin (CBP). METHODS: 48 male SD rats were randomly divided into five groups to be administer with different medicines once a day for 14 days: group A1 (administered with CBP only and killed on the cessation day of administration), group B1 (administered with CBP only and killed 8 weeks after the cessation of administration), group A2 [administered with CBP + amoxicillin (AMO) + metronidazole (MTR) + losec and killed on the cessation day of administration], group B2 (administered with CBP + AMO + MTR + losec and killed 8 weeks after the cessation of administration), and control group (administered with distilled water). The kidney issue sections were counterstained after AMG development. The bismuth deposited in tissues was observed by microscopy. The gray level of kidney tissue sections were measured and compared through image processing program. The deposition of bismuth and the degrees of cell organ's impairment were observed by electron microscopy. By using electron probe microanalysis bismuth was identified from the chemical elements in the specimens. RESULTS: Under the light microscopy, black-brown granules were discovered in the cell bodies of the proximal convoluted renal tubule. The amounts of bismuth accumulated in kidney of the 2 quadruple therapy groups were much more than those of the 2 single compound therapy groups (all P < 0.05). The amount of bismuth accumulated in kidney on the cessation day of administration was more than that eight weeks later (both P < 0.01). Under electron microscopy, black-brown granules were observed exclusively in the lysosomes of the proximal convoluted renal tubule cell. Electron microscopy found cell impairment in the quadruple therapy groups. Impairment of these cells could be recovered 8 weeks after the cessation of administration. CONCLUSION: Acid suppression therapy causes an increase of absorption and accumulation of bismuth from CBP in the kidney. Bismuth can be accumulated in the cell bodies of proximal convoluted renal tubule after its absorption. The absorbed bismuth can be discharged out of the body via kidney. Large amounts of bismuth accumulation in kidney can impair the functions of proximal convoluted renal tubule cells.

[Enterosorption and nutritious support with pectin-containing preparation in the treatment of stress immunodeficiency in peritonitis].

Fifty-eight patients aged from 18 to 65 years with peritonitis of different etiology in toxic and terminal phases were treated. Nasoenteral intubation, enterosorption and early nutritious support with pectin-containing preparation (PCP) were carried out in all the patients just after surgery with standard course of 5 days. Baseline data was compared with that after PCP-supported treatment. Powder products of red beet were used as PCP. Immunodeficiency in peritonitis is characterized by imbalance of stress-realizing and stress-limiting mechanisms of immunocompetent cells. Enterosorption and early nutritious support with PCP in peritonitis decrease the level of plasmic cortisol and enhance stress-limiting reception that reduces a harmful effect of cortisol and reestablishes immunity.

[Influence of the food products, enriched with pectin, on properties of potentially patogenic representatives of the microflora of the large intenstine].

The treatment method of disbios suffered by patients with syndrome of irritated intestines with constipations was under research. The method was based on introduction of sour-milk beverage with addition of beet pectin in a dietary intake. The comparison product was kefir. The following developments were detected against the positive dynamics of clinical symptoms: improvement of microbiocenosis structure, reduction of potentially pathogenic germs, inhibition of pathogenic properties of enterotoxigenic Staphylococcus aureus. Comparison group results showed lower clinical efficiency and poorer effect of dietary treatment on large gut microorganisms, contents of enterotoxigenic S. aureus remained unchanged. Introduction of sour-milk beverage with addition of alimentary fibre in a dietary intake of patients suffering the syndrome of irritated intestines with constipations was found expedient.

[Immunomodulating properties of pectin from seawater grass Zostera].

Immunomodulating activity of zosterin was studied. Zosterin is a pectin from sea grass belonging to Zostera with pronounced antibacterial activity and therapeutic efficacy in experimental infections. It was shown that parenteral administration of the drug 24 hours before the antigenic irritation stimulated the humoral and cellular immunity in F1 (CBA X C 57 BL/6) mice: an increase in the number of the antibody cells in the spleen, the DTH and the index of the splenocyte spontaneous proliferation. In the animals treated with injections of zosterin and infected intraperitoneally with a virulent strain of S. enteritidis there was observed a marked increase in migration of polymorphonuclear leukocytes to the abdominal cavity accompanied by an increase in their phagocytic activity. The results suggested the possible use of zosterin as a drug with associated antibacterial and immunomodulating activities.

[Antibacterial and therapeutic effectiveness of a pectin from sea grass Zostera].

The kinetics of the antibacterial activity of zosterin, a polysaccharide preparation of a sea grass belonging to Zoster, was studied. By its chemical structure zosterin is a low ++methoxylated pectin. In vitro the preparation markedly inhibited the growth of ++Gram-negative and ++Gram-positive organisms: S. aureus, E. coli, Y. pseudotuberculosis, S. typhimurium and Ps. aeruginosa. On a model of experimental pseudotuberculosis++ infection caused by oral contamination of mice F1 (CBA X C57B1) with a suspension of Y. pseudotuberculosis zosterin was shown to have a therapeutic effect. It protected 30 to 40 per cent of the animals when administered per os simultaneously with or 24 hours after the contamination. The results are in favour of the zosterin further investigation as a preparation useful in prevention of intestinal infections in persons being in contact with the patients.

[Thermoanalytical studies of sodium salt of carboxymethylcellulose (CMC-Na) and pectin].

The influence of physical and chemical properties of electrolytes on thermal degradation of CMC-Na and pectiny salt were studied. A differential thermal analysis showed that thermal decomposition of CMC-Na and pectin salt proceeds in three stages. The heat of conversion of the studied systems was shown to increase according to the concentration of the applied electrolytes. Significant changes in the structure of CMC-Na at pH = 3.0 were shown by IR analysis. Confirmation of transition of the above system to the acidic form were obtained by DTA and TG curves.
