Imaging liposomes at electron microscopic level: encapsulated colloidal iron as an electrondense marker for liposome-cell interactions.

Colloidal iron was encapsulated into liposomes prepared by different methods to provide an electrondense marker for easy identification of liposomes in cell and tissue culture. Stable colloidal iron solution can be prepared at virtually any concentration. The diameters of more than 75 per cent of the iron particles measured between 1 and 5 nm. Liposomes with a distinct electrondense core were evident at a colloidal iron solution concentration of 0.6 g/l. The colloidal iron labelled liposomes were easily identified in cells after incubation by routine electron microscopic procedures. Liposomes could be found in lysosomes or endosomes of human M21 melanoma cells. Intact, as well as partially degraded liposomes were present after two hours of incubation.

Colloidal gold: a pluripotent receptor probe.

Colloidal gold is an electron-dense, lyophobic colloid that readily forms a stable electrostatic interaction with a variety of macromolecules. Monodispersed colloids ranging from 3-150 nm in diameter can be produced to provide the researcher with flexibility in selecting the optimally sized probe. Gold labeling of antibodies and lectins has been extensively used to study surface antigens and cell components. Recently, the use of gold labeling has been extended to study receptor-ligand binding, enzyme-substrate reactions, and transcellular pathways. Published applications include gold labeling of metabolites (low-density lipoproteins), enzymes (DNAase and RNAase, RNA polymerase, thrombin, collagenase, elastase), hormones (insulin, epidermal growth factor, glucagon), circulating plasma proteins (asialoglycoprotein, alpha 2-macroglobulin, factor VIII-von Willebrand factor), and endotoxins (tetanus toxin, cholera toxin). This broad spectrum of applications emphasizes the versatility and usefulness of colloidal gold as a probe in areas of cell biology related to receptors, endocytosis, transport, and functions of proteins.

[An evaluation of radioactive colloid for measuring the volume of effective liver].

A methods for liver scintigraphy for finding the effective liver volume were developed. Matrix ROI (region of interest) was set on the anterior liver scintigram with 99mTc tin colloid and 198Au colloid, respectively. The various depths of liver under matrix ROI were calculated from the count rate noting the absorption coefficient. The effective liver volume was the sum total of the areas of matrix ROI to the various depths. In order to check the accuracy of this method, the effective liver volume was calculated using the liver phantom and the patient's liver scintigraphy. The volume obtained for the liver phantom was nearly equal to the real volume. The liver volume in 19 patients using 99mTc tin colloid and 198Au colloid showed good correlation with the volume as measured by computed tomography (CT) scan (r = 0.841, r = 0.749).

Correlation between increased colloid osmotic pressure and the resolution of refractory ascites after transjugular intrahepatic portosystemic shunt.

BACKGROUND: This study compared the changes in serum albumin, globulin, and colloid osmotic pressure (COP) before and after transjugular intrahepatic portosystemic shunt (TIPS) or large volume paracentesis (LVP) in patients with ascites. METHODS: Of 23 patients with refractory ascites, 17 had TIPS and 6 had LVP with infusion of albumin. Colloid osmotic pressure measurements were calculated, using the formula previously proposed by Hoefs: COP = A (1.058G + 0.163A + 3.11) where A = serum albumin and G = serum globulin. RESULTS: After 1 month, ascites resolved in 9 of the 17 patients who had TIPS and in none of the 6 who had LVP. Colloid osmotic pressure increased significantly in patients whose ascites resolved after TIPS. Colloid osmotic pressure did not change in the patients whose ascites did not resolve after TIPS, and COP decreased significantly in the LVP group. A statistically significant difference was found in the pre-TIPS COP measurements between those patients who had resolution of ascites and those who did not. A pre-TIPS COP of < or =20 mm Hg predicted resolution of ascites with an 88% sensitivity and a 78% specificity. CONCLUSIONS: Serum COP increased significantly in patients with resolution of ascites but remained unchanged in patients with persistent ascites after TIPS. Serum COP decreased after LVP. A statistically significant difference in the pre-TIPS COP was found between patients whose ascites resolved and patients having persistent ascites.

Colloid cyst of the third ventricle: imaging-pathologic correlation.

Colloid cysts are relatively rare intracranial lesions located in the rostral aspect of the third ventricle. They may produce acute hydrocephalus, brain herniation, and lead to death. Although the clinical and imaging features of colloid cysts are well known, their etiology and the factors responsible for their imaging features continue to be a subject of debate. We present the imaging-pathologic correlation of a patient with a colloid cyst as well as data supporting the fact that the presence of cholesterol is probably responsible for the MR imaging features exhibited by some colloid cysts.

[Diurnal rhythms of thyroid hormones in rooster chicks (Gallus domesticus). 1. Studies on the colloid epithelium portion, the radioiodine accumulation in the thyroid gland and the plasma radioiodide concentration].

Four and five week old White Leghorn cockerels were investigated concerning diurnal changes of thyroidal activity. They were kept under normal conditions, including the changes in day light (light from 5.00 am to 8.00 pm). In the thyroid maxima of the per cent amount of colloid were found at 3.00 am and at noon. The values differed significantly from the intermediate ones (2P less than 0.001) which reached only 22 per cent of the maxima. The thyroidal uptake of radioiodine had also a maximum at 3.00 am (51 percent and 78 percent resp., 90 min. or 24 hours p.i.). The 24-h uptake at 3.00 am was significant different from the corresponding uptakes between 6.00 am and 6.00 pm (53-61 percent; 2P less than 0.05). The plasma radioiodide increased at afternoon and in the evening. The 9.00 pm concentration differed significantly from the 9.00 am concentration (2P less than 0.001). Since in the afternoon most of the food is ingested the thyroid can take up a greater amount of untracered iodide and therefore the radioiodide concentration of plasma may remain high. The above observations may be explained in the following way: (1) The thyroidal iodine uptake is increased in the afternoon corresponding to the increase in colloid; however these increase is not detectable by measuring the radioiodine uptake because of dilution effects (during the night the increase in colloid coincidences with the increase in iodine uptake); (2) looking at the daily changes of colloid and radioiodine uptake the thyroid may be stimulated in the early morning and in the evening.

Increased erythropoietin production in anephric rats with hyperplasia of the reticuloendothelial system induced by colloidal carbon or zymosan.

Adult rats were subjected sequentially to (1) administration of colloidal carbon, zymosan, or gadolinium; (2) subtotal hepatectomy, bilateral nephrectomy, or sham operation; and (3) a 6-hr bout of hypoxia, starting 1 hr or 24 hr after the operation. Control animals received the respective vehicles. The erythropoietin (Ep) activity was assayed in exhypoxic polycythemic mice on the basis of 48-hr per cent RBC(-59) Fe incorporation values. Ep levels in serum of anephric rats primed with either colloidal carbon or zymosan were considerably more elevated than in control animals. This potentiating effect was observed in rats subjected to hypoxia starting either 1 or 24 hr after nephrectomy. On the other hand, gadolinium did not enhance the extrarenal Ep response to hypoxia. It is emphasized that both colloidal carbon and zymosan induced marked hyperplasia of the hepatic and splenic reticuloendothelial system (RES), while gadolinium did not induce this effect. A strict correlation was thus established between potentiation of extrarenal Ep production and hyperplasia of the RES. It is therefore tentatively concluded that the RES is a source of extrarenal Ep. Additionally, since the liver plays a prominent role in extrarenal Ep production, Kupffer cells may represent a major source for Ep in the anephric rat. Finally, it is of interest that both colloidal carbon and zymosan did not potentiate the Ep response to hypoxia in sham-operated or subtotally hepatectomized rats.

[Mechanism of inhibition of tumor angiogenesis by Bletilla colloid: an experimental study].

OBJECTIVE: To study the mechanism of inhibition of tumor angiogenesis by Bletilla colloid. METHODS: Human Hep-G2 hepatocellular carcinoma cells were cultured and treated with Bletilla colloid of different concentrations. Pure culture of Hep-G2 cells was used as control and pure culture medium without Hep-G2 cell was used as blank control. The concentration of vascular endothelial growth factor (VEGF) in the supernatant was detected by ELISA. The apoptosis and proliferation of the Hep-G2 cells were examined by flow cytometry. Cells of human endothelial cell line ECV-304 were cultured in the supernatant of culture media of Hep-G2 treated with Bletilla colloid of different concentrations. MTT method was used to observe the growth of the ECV-304 endothelial cells. Eighty rats were made animal model of transplanted Walker-256 hepatoma and then randomly divided into 4 groups of 20 rats 12 - 14 days after to undergo transarterial chemoembolization (TACE) treated with normal saline, 5-fluouracil (5-Fu), iodized poppy seed oil, and 5-Fu-Blettila microsphere respectively. Two weeks after, the rats were killed and the tumors were taken out. Immunohistological staining was conducted to detect the expression and localization of factor VIII, VEGF, and basic fibroblast growth factor (b-FGF). The microvcascular density (MVD) was calculated by counting the factor VIII positive endothelial cells. RESULTS: There was no statistically significant difference in the apoptosis rate, proliferation rate and supernatant VEGF level between the control group and the Bletilla groups. The inhibition rates of ECV-304 endothelial cell growth were 57.6%, 66.7%, 86.4%, 87.5%, and 94.8% in the groups of Bletilla of the concentrations of 0.5, 1.0, 2.0, 4.0, and 8.0 micro g/ml respectively in a dose-dependent manner. The MVD of tumor was 59 +/- 34 per field in the 5-Fu-Blatilla group,significantly lower than those in the other 3 groups (F = 5.177, P = 0.003). The expression of VEGF and the expression of b-FGF were not significantly different among the 4 TACE groups. CONCLUSION: Bletilla colloid inhibits angiogenesis after TACE, potentially, through inhibition of the binding of vascular endothelial growth factor to its receptor.

[Effect of hypotonic liquid containing colloid on hemorrhagic dogs with celiac seawater immersion wound].

OBJECTIVE: To determine hypotonic liquid containing colloid on hemorrhagic dogs with celiac seawater immersion wound. METHODS: Thirty-five dogs were randomly divided into five groups: control group (group A), 0.9% NaCl therapy group (group B) and 0.45% NaCl therapy group (group C), 5% glucose therapy group(group D) and hypotonic liquid containing colloid therapy group (group E). Changes of mean artery pressure (MAP), cardiac output (CO), urine volume, plasma osmotic pressure and pathologic changes of lung and brain were observed. RESULTS: hypotonic liquid containing colloid could effectively ameliorate MAP and CO, improve urine volume, decrease plasma osmotic pressure and prevent occurrence of lung and brain edema. CONCLUSION: Hypotonic liquid containing colloid can exert satisfactory therapeutic effects on hemorrhagic dogs with celiac seawater immersion wound.

[Effect of colloid gold on physiologic-biochemical processes in Escherichia coli 1257].

The influence of colloid gold on the growth processes, ATP-ase activity and extrusion of protons in Escherichia coli 1257 was studied. The particles of colloid gold exert nonmonotonous influence on these processes with different direction is such a way that small concentration of this metal (5 x 10(-7)-5 x 10(-6) mg/ml) exert stimulative effect, while higher concentrations of colloid gold result in the suppression of biological activity of the bacterial cells. The discovered peculiarities of colloid gold influence of E. coli strain may be determined by specificity of contact interaction of metal particles with the surface of bacterial cells.

99mTc-stannous colloid white cell scintigraphy in childhood inflammatory bowel disease.

UNLABELLED: 99mTc-Labeled white cell scintigraphy (WCS) has been used for the investigation of inflammatory bowel disease (IBD) in adults, but data on children are limited. The most common agent used is (99m) Tc-hexamethylpropyleneamine oxime (HMPAO); however, this agent has limitations. In a retrospective study, we assessed the use of (99m)Tc-stannous colloid WCS for the initial evaluation of children with suspected IBD. METHODS: Diagnostic, endoscopic, and contrast radiography results were retrospectively collected from the medical records. Two experienced nuclear physicians unaware of the patient data interpreted the WCS results, with agreement reached by consensus. Statistical analysis was performed on the ability of WCS to detect active disease and localize it topographically and on a comparison of diagnostic methods, using a combination of clinical features and endoscopy as the reference standard. RESULTS: Between 1996 and 1999, 64 patients (35 male and 29 female; mean age, 12.5 y; age range, 2-19 y) had WCS performed, with IBD subsequently diagnosed in 34 patients. (99m)Tc-Stannous colloid WCS had an 88% sensitivity, 90% specificity, and 8.8 likelihood ratio for initial investigation of IBD. Agreement was poor for topographic localization of disease. Small-bowel series had a 75% sensitivity, 50% specificity, and 1.5 likelihood ratio for detecting endoscopic disease of the terminal ileum and proximal colon. CONCLUSION: Our results confirm that WCS is a useful imaging technique for the initial evaluation of patients with suspected IBD. (99m)Tc-Stannous colloid had results at least comparable to those of other WCS agents, and in children, (99m)Tc-stannous colloid WCS should be preferred in view of lower cost, shorter preparation time, and the smaller blood volumes required.

[Effect of low molecular weight organic acids on Pb2+ adsorption and desorption by constant charge soil colloids].

The isothermal curves of Pb2+ adsorption-desorption by soil colloids followed Freundlich and Langmuir equation. The adsorption constant Ka showed the trend of old manured loessal soil > yellow loessal soil > dark cultivated loessal soil > yellow cinamon soil, which was consistent with surface charge density sigma 0, an indicator of the strength of Pb2+ adsorption by each soil colloid. Low molecular-weight organic acids (LMWOAs) made Pb2+ adsorbed amount decreased and adsorption affinity increased, and the effect of citric acid was larger than that of oxalic acid. The constant Kd of residual Pb2+ adsorption after desorption showed the trend of old manured loessal soil > yellow cinamon soil > dark cultivated loessal soil > yellow loessal soil, which reflected the adsorption strength of residual Pb2+ after desorption, and was related to the contents of organic matter and free ferric oxide. The isothermal curves of Pb2+ desorption from soil colloids were obvious higher than those of Pb2+ adsorption. In NaNO3 and oxalic acid background solution, the isothermal curves of Pb2+ adsorption were far away from those of Pb2+ desorption, showing that the hysteretic degree between adsorption and desorption was very large. The isothermal curves of Pb2+ desorption from soil colloids were closed to those of Pb2+ adsorption in citric acid background solution, which showed that the Pb adsorption and desorption was reversible.

[Criteria for the safety of transfusion of colloidal solutions in patients with acute lung lesions].

The prospective study explored the hemodynamic effects of colloidal solution replacement therapy and the criteria for its safety in patients with acute lung parenchymatous lesions (ALPL) attended by hypoalbuminemia and coagulopathy. There were 68 observations of the effects of colloidal solutions: 20% albumin solution (n=25), freshly frozen plasma (FFP) (n=20), 6% hydroxyethylated starch (HES) 130/0.4 9:1 (n=23). The colloidal solutions were infused at a constant rate; the infusion was stopped until pulmonary wedge pressure (PWP) was 25% greater than its baseline value. Before and after infusion, the parameters of central hemodynamics and oxygen transport, extravascular lung water index (ELWI), pulmonary vascular permeability index (PVPI), and colloid-osmotic pressure (COP) were measured. The infusion volumes were 3.8 +/- 0.4, 13.7 +/- 1.4, and 13.4 +/- 1.3 ml/kg for 20% albumin solution, 6% HES 130/0.4, and FFP, respectively. The PWP-COP gradient increased in all groups. After FFP infusion, there was an increase in ELWI and lung shunt. After 20% albumin solution, there was a delayed increase in ELWI. There was no rise in ELWI after 6% HES administration. In the 20% albumin solution group, the increased ELWI was recorded in patients who had positive baseline PWP-COP gradients (p < 0.05). A combination of higher PVPI and a positive PWP-COP value causes a greater increase in ELWI after 20% albumin solution infusion than in the normal PVPI group. In patients with ALPL, FFP infusion may lead to an increase in the accumulation of extravascular lung water. A negative preinfusion PWP-COP gradient is a safety criterion for the infusion of 20% albumin solution in patients with ALPL. The increased PVPI in combination with a positive PWP-COP gradient is an aggravating factor.

Intraglandular colloid induced nuclear proliferation of murine thymic cells as determined by flow cytometry.

The ability of intraglandular colloid, the holocrine secretion of cells of the marginal half of the bovine pituitary intermediate lobe, to enhance or suppress the proliferation of immature thymic cells of CH3/6JH mice, 4 to 6 weeks of age, was studied. The mice were given subcutaneous injections of colloid for 3 consecutive days at a dose of 20 mg/kg body weight in 0.2 ml of saline. The analysis of propidium iodide stained thymic cell preparations by means of flow cytometry showed that there was a significant increase in the percentage of cells in the S phase (proliferative phase) of the cell cycle of mice injected with colloid (33%) compared with saline injected (15%) and non-injected controls (7%). These findings are the first to suggest that various combinations and concentrations of intraglandular colloid neuropeptides play a role in the immune system and shed light on additional functions of the intermediate lobe.

[Effects of prenatal factors on the colloid-osmotic status of the newborn].

Studies of prenatal factor effects on the newborns' colloid-osmotic status, a pressing problem in present-day obstetrics, were carried out in 2 groups of women: Group 1 consisted of 50 women with spontaneous delivery and Group 2, also consisting of 50 women, who delivered via cesarean section, 25 of them because of a cicatrix on the uterus and the rest because of severe gestoses characterized by nephropathy and Stage III pre-eclampsia. Blood samples were collected directly after delivery from the mother's vein and from the newborn's umbilical vein, and the colloid-osmotic status of the plasma was analyzed. The results permitted singling out a number of prenatal factors influencing the newborns' colloid-osmotic status. These factors are as follows: obstetrical abnormalities, gestosis among other things; intensive care of the mother during delivery or after surgery, administration of diuretics, delivery mode, etc. These results have brought the authors to a conclusion that standardized infusion therapy without due consideration for the colloid-osmotic status is inadmissible, for it may result in iatrogenic complications in the newborns.

[A new method for immunological detection of fecal blood, using colloidal gold labeled with anti-human hemoglobin monoclonal antibody].

Application of colloidal gold agglutination to detection of fecal blood was done. Colloidal gold labeled with monoclonal antibody recognized human hemoglobin was agglutinated with hemoglobin and changed the color from red to gray. The color that diminished rapidly with an antigen was unchangeable from 30 min to 4 hr. Reaction was single step, and positive tests were easily distinguished from negative tests. Detectable hemoglobin concentration was 0.5 micrograms/ml. Comparison of colloidal gold agglutination with other methods, as low as latex agglutination and RPHA, showed good agreements in 1409 fecal samples. Colloidal gold agglutination method is superior in easiness of judgement and unnecessity of keeping time, and will be suitable for mass screening for colorectal cancer using 96 well microplate.

Colloidal bismuth subcitrate evokes adaptive cytoprotection in rat gastric mucosa.

The 6-keto-prostaglandin-F1 alpha content (as the stable break-down product of prostacyclin) of rat antral and fundic gastric, as well as of duodenal mucosa, significantly increases after 1, 5 and 10 mg/kg orally administered colloidal bismuth subcitrate treatment. The results indicate that (i) colloidal bismuth subcitrate-induced stimulation of endogenous prostacyclin content ("adaptive cytoprotection") of rat gastroduodenal mucosa may contribute to its therapeutic effect; (ii) the effect of colloidal bismuth subcitrate is not due exclusively to the bismuth content of the molecule, but seems to be connected with the structure of colloidal bismuth subcitrate itself; (iii) the effect seems to be dose-dependent, showing a dose-response relationship.

Colloids versus crystalloids in fluid resuscitation: an analysis of randomized controlled trials.

BACKGROUND: Controversy about fluid therapy in resuscitation has existed since the 1960s. The difficulty could be that fluid behavior at the lung capillary membrane level may vary depending on the patient's particular pathology. METHODS: Mortality rates taken from randomized controlled trials were analyzed to compare colloidal and crystalloidal fluid for resuscitation efforts. We controlled for the underlying pathological process by categorizing subjects into three groups: (1) surgical stress, (2) hypovolemia, and (3) severe pulmonary failure. A cost-effectiveness analysis also was performed. RESULTS: No statistically significant differences in mortality rates were found. The cost of each life saved using crystalloids is $45.13, and the cost of each life saved using colloidal solutions is $1493.60. CONCLUSIONS: Because there is no significant mortality-rate advantage to using colloids, and because the cost-effectiveness ratio for crystalloids is much lower than for colloids, it is concluded that crystalloids should always be used in resuscitation efforts.
