[Aseptic osseous necrosis after renal transplantation].

Of 165 renal Transplantated patients, 12 developed aseptic bone necrosis in the femoral head (6 patients), in the femoral condyle (5 patients), in the astragalus (1 patient). The onset of symptoms was 6 to 23 months after transplantation. 99mTc-O4-MDP bone scintigraphy and radiological examination associated with clinical signs confirmed the diagnosis. Unresolved hyperparathyroidism, phosforus depletion, ponderal increase, total i.v. prednisolone-boluses and trauma represented conditions which might predispose to the development of lesion. 8 patients were managed with conservative treatment. 4 patients required a total of 8 operations: head replacement arthroplasty and articular cartilage reimplant in two patients with disease involving femoral head; articular cartilage reimplant and condyle replacement arthroplasty in two patients with disease involving femoral condyle.

[Computerized tomography of the talo-crural joint].

CT provides an additional method of investigating the ankle joint in cases of trauma and posttraumatic lesions. Concerning fractures CT displays preoperatively the true extent of the bony lesion. The ligaments of the tibiofibular syndesmosis are directly visible in CT. Posttraumatic states (reparatively ossified avulsions of ligaments, partial osteonecrosis of the astragalus) are recognizable in CT.

Limb deformities in foals associated with ingestion of locoweed by mares.

Five of 26 pregnant mares observed ingesting locoweed (Astragalus mollisimus) subsequently aborted and another 10 produced foals with various limb deformities. Seven of the foals had deformities of a flexion-extension type and 3 had angular deformities. Four foals were normal. The limb deformities either resolved spontaneously (5 cases) or were treated successfully (in 4 of the other 5 cases). The problems of abortion and limb deformities were attributed to locoweed ingestion on the basis of the mares having eaten the plants, and the similarity of the syndrome to that previously reported in sheep and cattle.

[Pathology of injuries due to snow surfing].

The amount of injured snowboarders is increasing each year. The reason is due to the success of snowboarding for teenagers. Accidents occur more during the first hours of practice. Upper limb is the first location: fore-arm breaks and wrist fractures are very common. Elbow and shoulder dislocations also occur often for snowboarders. The location of most injuries for lower leg is the ankle; we found many ankle sprains and astragalus's breaks. The knee sprain is rare unlike alpine ski. Prevention has to be developed to give a better image of this sport, in fact the attitude of the participants is the main reason for the accidents.

[Clinical and experimental study on effects of man-shen-ling oral liquid in the treatment of 100 cases of chronic nephritis].

A new Chinese herbal preparation, Man-Shen-Ling (MSL, consisted of medicinal herbs such as Astragalus and Rehmannia) in treating 100 cases of chronic nephritis. The effective rate was 91% in comparing to 66.7% in the control group, P < 0.001. It was markedly effective for proteinuria, hematuria, improvement and recovery of renal functions, edema, anemia, anorexia etc in comparing with the control group. It showed no adverse effects on functions of liver, kidney, heart and GI tract. Animal model of chronic nephritis was established and the effects of MSL were observed. The laboratory findings and histopathological investigation on kidney revealed and confirmed that MSL has therapeutic effects on chronic nephritis. Pharmacodynamically, MSL exhibited effects of anti-allergy, its immuno-suppressive effect corresponded to that of cyclophosphamide, with diuretic, hypotensive, proteinuria eliminating, anti-inflammatory, anti-coagulatory, renal blood flow and glomerular filtration enhancing, the excretion of urea-nitrogen, potassium and sodium promoting function; in addition, it also could promote and modulate the immunity. Acute and chronic toxicity tests on animal models neither showed toxic, mutagenic, teratogenic nor carcinogenic effects. It is a new preparation of Chinese medicinal herbs in treating chronic nephritis, it is safe and effective.

[Experimental study on effects of 18 kinds of Chinese herbal medicine for synthesis of thromboxane A2 and PGI2].

The effect of 18 kinds of Chinese herbal medicine for the synthesis of TXA2 and PGI2 was studied. The porcine lung microsoma was used as donor of enzymes such as cyclo-oxygenase, thromboxane A2 synthetase and prostacyclin synthetase, etc. It was found that Glehnia littoralis could inhibit the synthesis of TXA2 and increase the formation of PGI2. Rheum palmatum (300 mg group) and Erigeron breviscapus significantly inhibited the synthesis of TXA2, but no apparent effection on the synthesis of PGI2. When Codonopsis pilosulae, Astragalus membracaceus, Angelica sinensis, Ginsenosides and Baicalin, etc. markedly inhibited the formation of TXA2 and mildly affected the formation of PGI2. Some Chinese herbs for promoting blood circulation play an important role on the inhibition of TXA2 synthesis. Some tonic herbs could either inhibit the synthesis of TXA2 and increase the synthesis of PGI2 or inhibit the formation of PGI2. Therefore these tonic herbs had the characteristics of both strengthening the body resistance and eliminating pathogenic factors. In this aspect they are better than the control drug (Aspirin) and other herbs of promoting blood circulation.

[Phytotherapeutic aspects of diseases of the cardiovascular system. 5. Saponins and possibilities of their use in prevention and therapy].

The summarizing paper deals with the structure and biological effects of saponins generally and in relation with the treatment and prevention of diseases of the heart and circulatory system. In this field, mainly the saponins from the plants of the genera Panax, Gynostemma or Bupleurum are of use. Also soya saponins and saponins of the genera Astragalus, Salvia, Boussigaultia and Litchi can be employed. Saponins exert a positive effect on the function of the heart direct, or they help treat related diseases. For instance, they inhibit the formation of lipid peroxides in the cardiac muscle or in the liver, they influence the function of enzymes contained in them, they decrease blood coagulation, cholesterol and sugar levels in blood, they stimulate the immunity system. They act either direct, blocking the transfer of Ca2+ ions or modulating the function of Na(+)-K(+)-ATPase, or they help resorb other active principles. It can be concluded that saponins are a prospective group of drugs of natural origin for the prevention and treatment of diseases of the heart and circulatory system.

Evaluation of clinicopathological parameters in cattle grazing timber milkvetch.

The purpose of this study was to determine whether hematological and serum biochemical parameters could be used to diagnose poisoning in cattle grazing timber milkvetch (Astragalus miser var. serotinus), a nitropropanol-containing legume. Blood samples were obtained from 20 Hereford cattle on timber milkvetch range in southern British Columbia during 1992 and 1993. Relevant changes in blood parameters were not detected and the parameters could not be used to predict nitropropanol ingestion or intoxication. The same parameters were measured in dosing trials with cattle with rumen fistulas, but relevant changes were not detected. Succinate dehydrogenase is the key target in nitropropanol intoxication and an assay for this enzyme may provide a probe for diagnosis.

Free radical generation by selenium compounds and their prooxidant toxicity.

Selenium (Se) and many of its compounds are among the most toxic of nutrients. Selenium toxicity was first described in range animals in the western United States in the 1930's which consumed "selenium accumulator" plants of the genus Astragalus, Xylorrhiza, Oonopsis, and Stanleya. Selenites and selenates from the soil accumulate in these plants primarily as methylated selenium compounds and plants evolve dimethyldiselenide and dimethylselenide. Dietary selenium, primarily as selenomethionine and selenocysteine for humans fulfill the dietary requirement for selenoenzymes and proteins. In humans and animals excessive dietary selenium may be toxic. In vitro, selenium compounds such as selenite, selenium dioxide and diselenides react with thiols, such as glutathione, producing superoxide and other reactive oxygen species. This catalytic reaction of selenium compounds with thiols likely accounts for selenium toxicity to cells ex vivo and in vivo where the major glutathione producing organ, the liver, is also the major target organ of selenium toxicity. Selenium enzymes and selenoethers that do not readily form a selenide (RSe-) anion and compounds such as Ebselen where selenium is sequestered, are not toxic. Methylation of selenium by both plants and animals serves to detoxify selenium by generating methylselenides. Alternatively, full reduction of Se to elemental selenium (Se0) as done by some bacteria and the formation of heavy metal selenides such as Ag2Se or Hg2Se, results in a non-catalytic non-toxic form of selenium. This catalytic prooxidant attribute of some selenium compounds appears to account for its toxicity when such activity exceeds plant and animal methylation reactions and antioxidant defenses. This prooxidant activity may also account for cellular apoptosis and may provide a useful pharmaceutical application for selenium compounds as antibacterial, antiviral, antifungal and anticancer agents.

Protective effects of radix astragali against anoxic damages to in vitro cultured neurons.

To study the protective effects of radix astragali against anoxic damages to in vitro cultured neurons in rats, NaCN was used to develop a hypoxic model of in vitro cultured neurons from newborn rat cerebral cortex. The cellular morphology, A value (cell survival number) and effluxes of lactate dehydrogenase (LDH) and K+ from cells were measured in the radix astragali group and the control group respectively. After 48 h of anoxia, A value was decreased from 0.325 +/- 0.031 before anoxia to 0.145 +/- 0.011, the effluxes of LDH and K+ were increased from 65.80 +/- 2.90 U/L and 5. 23 +/- 0.11 mmol/L before anoxia to 148.80 +/- 8.40 U/L and 7.31 +/- 0.18 mmol/L, respectively. It was found that in the anoxic circumstance in the Radix astragali group, the mophological changes were mild, the effluxes of LDH and K+ were decreased and A value increased as compared with those in the control group. It was suggested that Radix astragali could protect the cultured rat neurons against anoxic damages in the anoxic circumstance.

Immuno-enhancement effects of Huang Qi Liu Yi Tang in a murine model of cyclophosphamide-induced leucopenia.

Tonic Chinese prescriptions are widely used in daily foods to improve health in Taiwan. Huang Qi Liu Yi Tang (HQLYT) is one such tonic Chinese prescription. In this study, the immuno-enhancement effects of HQLYT tonic were explored using an in vitro splenocyte proliferation assay and by evaluating improvements in cyclophosphamide-induced leucopenia in a BaLb/c mice model. HQLYT is composed of Radix Astragali, licorice, and jujube, and extracted in boiling water. The contents of glycyrrhizin and formononetin were used as bioactive markers to control the quality of the HQLYT water extract, at 7.26+/-0.04mg/g and 112.07+/-0.08 micro g/g, respectively. In the in vitro assay, HQLYT (12.5 micro g/ml) showed better immuno-enhancing effects than Radix Astragali, licorice, or jujube alone, and enhanced the survival rate of splenocytes by 88%. Moreover, HQLYT significantly enhanced the cytotoxic effects of natural killer cells of splenocytes against YAC-1 cells. The BaLb/c mice were fed HQLYT once a day for 14 days. After oral treatment with HQLYT 400mg/kg body weight (BW), the IL-2 release in BaLb/c mice and the natural killer cell activity of the splenocytes were both enhanced, and the WBC level of cyclophosphamide-induced leucopenic mice was improved by treatment with 400mg HQLYT/kgBW. HQLYT significantly delayed the decline in the WBC level and affected a faster recovery of the WBC level back to a normal status. In summary, HQLYT enhanced immunologic functions in both in vitro and in vivo studies. These results offer scientific evidence to prove the efficacy of this tonic Chinese prescription.

[Preventive effect of radix Astragali on insulin resistance caused by tumor necrosis factor-alpha].

OBJECTIVE: To investigate the preventive effect of Radix Astragali (RA) on insulin resistance caused by tumor necrosis factor-alpha (TNF-alpha). METHODS: Normal rats were pretreated with RA or distilled water by intragastric infusion for one week. Glucose-insulin tolerance test was conducted in the rats 4 h after low dose TNF-alpha injection by caudal vein to estimate the change in insulin sensitivity. Meanwhile, the plasma glucagon, ACTH, blood lipid, and glycogen, triglyceride in tissue were also observed. RESULTS: Exogenous TNF-alpha could induce hyperinsulinemia in normal rats. K value in glucose-insulin tolerance test decreased, serum ACTH, glucagon, blood lipid increased, glycogen content in liver and red quadriceps muscles decreased, the liver triglyceride level increased in the TNF-alpha treated rats. RA could improve all the above-mentioned changes significantly except the blood lipid and triglyceride depots in liver. CONCLUSION: RA has obvious preventive effect on insulin resistance caused by TNF-alpha, it may be due to its action in decreasing insulin antagonistic hormones and increasing glycogen synthesis in tissue.

The regulatory action of Radix Astragali on M-cholinergic receptor of the brain of senile rats.

The changes in density of M-cholinergic receptors in different areas of senile rats and the regulatory action of Huang Qi ([symbol: see text] Radix Astragali, a drug for warming yang and replenishing qi) were observed by autoradiography. The results showed that the gray scale displayed in brain sections was clear and mainly distributed in the cortex, hippocampus and striate body, while that due to nonspecific combination was negligible. The gray scale in the cortex, hippocampus and striate body of the experimental group was markedly lower than that in the young control rats, decreased respectively by 24.87%, 14.12% and 12.76% (all P < 0.05); but it was obviously higher than those in the senile control rats, increased respectively by 24.15%, 14.38% and 13.47% (P < 0.05). The data indicate that Huang Qi ([symbol: see text]Radix Astragali) may up-regulate the decreased density of M-cholinergic receptors in the brain of senile rats.

[Effect of po li gan capsule on immunofunction in mice].

This study aimed at the immune response of mice to Po Li Gan Capsule (PLGC). The effects of PLGC on phagocytosis of phagocyte, on serum hemolysin, and on IL-1, IL-2 in mice were investigated. The results showed that the coefficient and index of phagocytosis obviously increased in the experiment group, compared with that in the control group (P < 0.01); the serum hemolysin in the experiment group increased too (P < 0.01). The results also showed that PLGC could restore the capacity of macrophage for secreting IL-1 that had been inhibited by cyclophosphamide and PLGC could enhance the release of IL-2 from spleen cell in the experiment group (P < 0.05-0.01). These suggest that PLGC enhances both cellular and humoral immunity function in mice.
