The availability of zinc in endosperm, whole grain and bran-enriched wheat crispbreads fed to rats on a Zn-deficient diet.

The hypothesis that factors such as dietary fibre and phytate in wheat bran limit the availability of Zn was tested in growing rats fed on low-Zn diets with different wheat crispbreads as the major source of Zn. Six groups of six weanling male rats each were fed on 5 parts semi-synthetic Zn-deficient diet and 1 part wheat-endosperm crispbread for 1 week. At the beginning of the second week, the crispbread in the diet of five groups was exchanged for crispbread made using one of the following wheat flours: (1) whole grain, (2) bran-enriched whole grain, (3) endosperm with Zn added to the whole-grain level, (4) endosperm with Zn added to the bran-enriched level, (5) whole grain with Zn added to the bran-enriched level. These diets were given ad lib. together with deionized water for 2.5 weeks. The relative absorption of Zn was lowest from the three non-supplemented diets (75-82%). All the added Zn was absorbed. As appetite, body-weight increase, Zn absorption, Zn retention and the Zn concentrations in serum and bone differed only slightly among groups fed on diets with similar Zn concentrations, it is concluded that factors such as dietary fibre or phytate in wheat bran limit the availability of Zn in wheat crispbreads very little when all the Zn is needed for growth and development in rats.

Gastrointestinal implications in pigs of wheat and oat fractions. 2. Microbial activity in the gastrointestinal tract.

The present work was undertaken to study the microbial activity in various segments of the gastrointestinal (GI) tract of pigs as influenced by the source and level of wheat and oat dietary fibre (DF). Eight experimental diets were prepared from wheat and oat fractions and studied in a series of two experiments using wheat flour as the DF-depleted control. The diets in Expt 1 were based on wheat flour and three iso-DF enriched diets comprising fractions rich in wheat aleurone, pericarp/testa or bran. In Expt 2, oat bran was added to wheat flour to achieve the same DF intake level as in Expt 1. This series included further diets based on rolled oats and rolled oats plus oat bran. The eight diets were given to thirty-two ileal-cannulated pigs, with sixteen pigs in each experiment. After a total period of 34 d (Expt 1) and 42 d (Expt 2), the pigs were slaughtered 4 h post-feeding and samples taken for adenine nucleotides (adenosine 5'-triphosphate (ATP); adenylate energy charge (AEC)), organic acids (lactic acid (LA); short chain fatty acids (SCFA)) and pH at twelve sites of the GI tract. The microbial activity as measured by the ATP concentration was low in the stomach and the cranial two-thirds of the small intestine, but tended to increase in the distal third. In the caecum a sharp rise in microbial activity was observed; the highest level was found for the diet providing most fermentable substrates. In all the diets but the rolled oats + oat bran diets, microbial activity showed a descending pattern as the digesta moved through the colon. In the large intestine source and level of residues had a marked influence on microbial activity. LA was the chief organic acid in the stomach and small intestine (10-40 mmol/l) while LA relative to SCFA was a minor component in the caecum and colon (10-20 mmol/l). The contribution of SCFA to total organic acids was reciprocal to LA, i.e. low in the stomach and small intestine (less than 20 mmol/l) and high in the caecum and colon. In the large intestine the concentration of SCFA decreased from 100-140 mmol/l in the caecum and proximal colon to 40-80 mmol/l in the distal colon. The acetic: propionic acid ratio increased from the caecum to the distal colon. With the diets based on oat alone (rolled oats; rolled oats + oat bran) the increase was less significant.(ABSTRACT TRUNCATED AT 400 WORDS)

Effect of ammonia treatment of wheat straw with or without supplementation of potato protein on intake, digestion and kinetics of comminution, rumen degradation and passage in steers.

Untreated wheat straw (UWS) or ammoniated wheat straw without (AWS) or with (AWSP) a supplement of potato protein of a low rumen degradability was fed to three steers according to a 3 x 3 Latin square design. All rations were supplemented with sugar-beet pulp and minerals. Voluntary organic matter intake (OMI, g/kg0.75 per d) was 67.8, 76.0 and 80.1 for whole rations (51.1, 59.7 and 59.2 for straw) for UWS, AWS and AWSP respectively, which was significantly higher for AWS and AWSP than for UWS. Organic matter digestibility (OMD, g/kg) was 561, 596 and 625 for the respective rations UWS, AWS and AWSP, also significantly higher for AWS and AWSP than for UWS. The increased voluntary intake and digestion of ammoniated wheat-straw-based rations were associated with a significantly higher potentially degradable fraction (D) of neutral detergent fibre (NDF) in offered straw (556 and 661 g/kg for untreated and ammoniated wheat straw respectively) and in the rumen pool (469, 555 and 554 g/kg for UWS, AWS and AWSP respectively). Isolated small rumen particles (retained on sieves with a pore size < 1.25 and > 0.041 mm) had a significantly lower D of NDF (average 588 g/kg) than isolated large rumen particles (average 663 g/kg). Fractional rates of degradation of NDF did not differ significantly either between untreated and ammonia-treated wheat straw offered (2.9 and 2.6%/h respectively) or between rumen pools (1.8, 1.7 and 2.1%/h for UWS, AWS and AWSP respectively). Rations based on ammoniated wheat straw had a significantly higher rumen NH3-N concentration than UWS. Although the rumen pool size of total contents differed significantly between treatments, those of dry and organic matter and of cell wall constituents were not significantly different. The proportion of rumen dry matter passing through a sieve with a pore size of 1.25 mm averaged 0.684 over rations (not significantly different between rations). Daily rumination (96 min) and eating (52 min) times/kg NDF ingested did not differ between rations. The rate of comminution of large particles estimated from the disappearance of indigestible NDF in large rumen particles from the rumen of animals without access to feed was 4.1, 6.3 and 7.1%/h for UWS, AWS and AWSP respectively. These values were not significantly different. The fractional rate of passage estimated from the faecal excretion of Cr-NDF was 5.4, 6.1 and 6.3%/h for UWS, AWS and AWSP respectively (significantly higher for AWS and AWSP than for UWS) but the turnover rate of indigestible NDF did not differ between treatments.

Effects of whole wheat dilution v. substitution on coccidiosis in broiler chickens.

To test the hypothesis that inclusion of whole cereals in the diet of broiler chickens reduces the severity of a coccidial infection, the effects were investigated, in birds infected with Eimeria acervulina, of feeding a complete pelleted control food, the control food diluted with whole wheat (400 g/kg), or a diet in which ground wheat in the pellets was substituted with whole wheat (400 g/kg) so as to achieve the same composition as the control diet. In the weeks prior to and after infection, (days 14--20 and days 21--27), no significant differences in performance were observed between birds fed the complete pellet and substituted feeds. Birds fed the diluted feeds had significantly lower gains per unit of feed and numerically lower weight gains than the birds fed the control and substituted feeds during this period. No significant differences in performance were observed between treatments from days 28--34. Diluting feeds with whole wheat (400 g/kg) significantly increased ileal digesta viscosity levels, compared with feeding the control and substituted feeds in birds dissected on day 21, although no significant differences in digesta viscosity levels were observed in birds dissected on day 35. Whole wheat feeding, either by dilution or substitution, significantly increased gizzard sizes in birds dissected on day 21 and day 35. Neither dilution nor substitution of feeds with whole wheat (400 g/kg) significantly affected the level of an Eimeria acervulina infection, as measured by daily and total faecal oocyst yields.

Cadmium absorption and retention by rats fed durum wheat (Triticum turgidum L. var. durum) grain.

A whole-body radioassay procedure was used to assess the retention and apparent absorption by rats of Cd in kernels of durum wheat (Triticum turgidum L. var. durum) harvested from plants grown hydroponically in 109Cd-labelled nutrient solution. Wholegrain wheat, containing 5 micromol Cd (570 microg)/kg dry weight labelled intrinsically with 109Cd, was incorporated into test meals fed to rats that had been maintained on diets containing marginally adequate, adequate or surplus levels of Zn (0.12 mmol (8 mg), 0.43 mmol (28 mg) or 1.55 mmol (101 mg) Zn/kg respectively), and either 0 or 50 g durum wheat/kg. Regardless of diet, all rats consumed about 99 % of the test meal offered. In rats fed diets without wheat, initial Cd absorption averaged 7.7, 4.6 and 2.4 % of the dose when the diet contained 0.12 mmol (8 mg), 0.43 mmol (28 mg) or 1.55 mmol (101 mg) Zn/kg diet respectively. In rats fed wheat-containing diets, initial Cd absorption averaged 3.8 and 2.6 % of the dose when dietary Zn concentration was 0.12 mmol (8 mg) and 0.43 mmol (28 mg)/kg diet respectively. The amount of Cd retained in the body at 15 d postprandial was <2 % of the dose in all rats, and decreased as Zn in the diet increased. Even at 15 d postprandial, 32 to 44 % of the Cd retained in the body was still in the gastrointestinal tract. The results show that: (1) the bioavailability to rats of Cd in wholegrain durum wheat was depressed when wholegrain wheat was part of the regular diet; (2) increased intake of dietary Zn lowered Cd absorption and retention; (3) retention of Cd in the body at 15 d postprandial from diets containing adequate Zn was <1.3 %.

Variation of cadmium concentration in Swedish wheat and barley. An indicator of changes in daily cadmium intake during the 20th century.

In order to evaluate whether or not an increase in daily cadmium intake may be taking place, 75 wheat samples (spring wheat and fall wheat) harvested from the same "nonpolluted'' farm fields between the years 1916 and 1972 were analyzed. The individual scatter was large but a significant (Psmaller than.05) tendency toward increasing concentration with time was evident for fall wheat. A similar tendency was found for spring wheat, through not a statistical significance. This increase may be explained by general air contamination or the cumulative effect of fertilizers. There was a clear covariation in cadmium concentrations of differenc genotypes of wheat and barley harvested in the same year despite yearly variations in average concentration.

Differences in the digestive tract characteristics of broiler chickens fed on complete pelleted diet or on whole wheat added to pelleted protein concentrate.

1. The effects of whole grains of wheat on the digestive tract of broiler chickens was studied. A complete pelleted feed was compared with free choice feeding of whole wheat and a pelleted protein concentrate, given from 7 to 29 d of age. 2. Pepsin activity in proventriculus tissue was lower in whole wheat-fed birds than in complete diet-fed birds. The weight (g/kg body weight) of the gizzard was higher in whole wheat-fed birds and its contents had a lower pH. 3. In the intestine, there were no differences in deoxyribonucleic acid (DNA) concentration, protein/DNA, ribonucleic acid (RNA)/DNA, RNA/protein ratios or alkaline phosphatase activity expressed per tissue weight. The weight (g/kg body weight) of the duodenum was lower in whole wheat-fed birds and its contents had a higher pH. Also the activities of alkaline phosphatase and leucine aminopeptidase in the duodenum, and maltase in the ileum, expressed per unit of bird weight, were lower in whole wheat-fed birds. 4. These results suggest that whole grain feeding increases the chemical (pepsin in proventriculus) and physical (gizzard muscle) functionality of the upper part of the digestive tract but decreases the digestive capacity of the intestine. Higher gizzard functionality may play a positive role in the control of bacterial populations. The lower digestive enzyme activities in the intestine may be detrimental in situations of mucosal deterioration caused by intestinal disease.

Effect of ambient storage of wheat samples on their nutritive value for chickens.

1. Two wheat cultivars (Abbott and Equinox) were grown using the same crop husbandry conditions. Batches of each cultivar were stored at ambient temperature (AT) for 0, 6, 12, 18 or 24 weeks. Those stored at AT for less than 24 weeks were then stored at -20 degrees C up to the end of the storage period. The 24 week (AT) samples were divided into two lots and one lot was frozen to -20 degrees C for 24 h. 2. Each of the 12 wheat samples was included (650 g/kg) in a nutritionally complete diet and fed to broiler chickens from 7 to 21 d of age. The apparent metabolisable energy (AME) of each sample was determined using a rapid bioassay with adult cockerels. 3. The Equinox cultivar sample had a high proportion (92/1000) of visibly sprouted grains and had a low Hagberg falling number (high amylase activity) and high content of free sugars compared to the Abbott sample. There were no major changes in dry matter, free sugar content, water extract viscosity or Hagberg falling number due to the different times of ambient storage of either of the two wheat cultivar samples. 4. The chickens fed on the Abbott cultivar wheat had a greater weight gain, although there were no differences in the determined AME of the two wheat cultivars. 5. The Abbott sample stored at ambient temperature for 6 weeks had a greater AME than the sample given no ambient storage and the samples given 18 and 24 weeks ambient storage. There were no significant differences between the Equinox cultivar samples. There were no effects of storage time on the growth performance of the broiler chickens. 6. The AME of the unfrozen Abbott cultivar was greater than the frozen (for 24 h) sample but there was no effect on broiler growth performance. This study suggests that, in some wheat samples, there may be a benefit in avoiding freezing during storage.

Wheat grass juice in the treatment of active distal ulcerative colitis: a randomized double-blind placebo-controlled trial.

BACKGROUND: The use of wheat grass (Triticum aestivum) juice for treatment of various gastrointestinal and other conditions had been suggested by its proponents for more than 30 years, but was never clinically assessed in a controlled trial. A preliminary unpublished pilot study suggested efficacy of wheat grass juice in the treatment of ulcerative colitis (UC). METHODS: A randomized, double-blind, placebo-controlled study. One gastroenterology unit in a tertiary hospital and three study coordinating centers in three major cities in Israel. Twenty-three patients diagnosed clinically and sigmoidoscopically with active distal UC were randomly allocated to receive either 100 cc of wheat grass juice, or a matching placebo, daily for 1 month. Efficacy of treatment was assessed by a 4-fold disease activity index that included rectal bleeding and number of bowel movements as determined from patient diary records, a sigmoidoscopic evaluation, and global assessment by a physician. RESULTS: Twenty-one patients completed the study, and full information was available on 19 of them. Treatment with wheat grass juice was associated with significant reductions in the overall disease activity index (P=0.031) and in the severity of rectal bleeding (P = 0.025). No serious side effects were found. Fresh extract of wheat grass demonstrated a prominent tracing in cyclic voltammetry methodology, presumably corresponding to four groups of compounds that exhibit anti-oxidative properties. CONCLUSION: Wheat grass juice appeared effective and safe as a single or adjuvant treatment of active distal UC.

Wheat starch-containing gluten-free flour products in the treatment of coeliac disease and dermatitis herpetiformis. A long-term follow-up study.

BACKGROUND: We investigated whether wheat starch-based gluten-free products are safe in the treatment of gluten intolerance. METHODS: The study involved 41 children and adults with coeliac disease and 11 adults with dermatitis herpetiformis adhering to a gluten-free diet for 8 years on average. Thirty-five newly diagnosed coeliac patients at diagnosis and 6 to 24 months after the start of a gluten-free diet and 27 non-coeliac patients with dyspepsia were investigated for comparison. Daily dietary gluten and wheat starch intake were calculated. Small-bowel mucosal villous architecture, CD3+, alphabeta+, and gammadelta+ intraepithelial lymphocytes, mucosal HLA-DR expression, and serum endomysial, reticulin, and gliadin antibodies were investigated. RESULTS: Forty of 52 long-term-treated patients adhered to a strict wheat starch-based diet and 6 to a strict naturally gluten-free diet; 6 patients had dietary lapses. In the 46 patients on a strict diet the villous architecture, enterocyte height, and density of alphabeta+ intraepithelial lymphocytes were similar to those in non-coeliac subjects and better than in short-term-treated coeliac patients. The density of gammadelta(+)cells was higher, but they seemed to decrease over time with the gluten-free diet. Wheat starch-based gluten-free flour products did not cause aberrant upregulation of mucosal HLA-DR. The mucosal integrity was not dependent on the daily intake of wheat starch in all patients on a strict diet, whereas two of the six patients with dietary lapses had villous atrophy and positive serology. CONCLUSION: Wheat starch-based gluten-free flour products were not harmful in the treatment of coeliac disease and dermatitis herpetiformis.

Dietary wheat bran inhibits DNA synthesis and enhances 5-fluorouracil toxicity in rat pancreas.

The effect of a 20% wheat bran dietary supplement on pancreatic growth and on the content and synthesis of pancreatic RNA and DNA was examined in male Sprague-Dawley rats fed defined diets for 4-5 weeks. Nutrient intake and body weight gain were similar in the control group (fed a fiber-free diet) and the groups fed the wheat bran supplement. The wheat bran diet produced no significant change in pancreatic weight, protein, RNA content, or DNA content, but was associated with a 28.8% fall in pancreatic DNA synthesis, measured by [3H]thymidine incorporation into DNA, when compared with the controls (p less than 0.05). When rats were injected with 50 mg/kg of 5-fluorouracil (two injections, one week apart), those fed wheat bran exhibited a 40.1% increase in pancreatic wet weight (p less than 0.005) and a 24.5% decrease in RNA content (p less than 0.001), when compared to controls fed a fiber-free diet. RNA synthesis, measured by the incorporation of 32P into RNA, increased 3.6-fold in rats fed wheat bran, when compared with controls (p less than 0.01). These results suggest that wheat bran should be tested further for any inhibitory action on pancreatic carcinogenesis and for its modifying effect on 5-fluorouracil toxicity.

Estrogen profiles in postmenopausal African-American women in a wheat bran fiber intervention study.

High dietary fiber intake has been hypothesized to lower blood estrogen concentrations, an effect thought to be beneficial for decreasing breast cancer risk. This study investigated the association between dietary supplementation of wheat bran and circulating estrogen levels in postmenopausal African-American women participating in a community intervention trial. Seventeen postmenopausal women (aged 63 +/- 1.6 yr) participated in the study. Nutritional status was assessed and blood and 24-hour urine samples were collected before and after five to six weeks of daily supplementation of the diet with 35 g of wheat bran cereal (11.6 g insoluble dietary fiber) marked with 28 mg of riboflavin. Riboflavin confirmed that all postmenopausal participants adhered to the intervention protocol. Nine of the 17 postmenopausal women were taking some form of estrogen replacement therapy (PM-ERT). Baseline hormone levels in the PM-ERT group did not significantly change after the dietary intervention. Estradiol (96.8 +/- 20.3 vs. 113.8 +/- 23.3 pg/ml), androstenedione (0.47 +/- 0.06 vs. 0.45 +/- 0.06 ng/ml), and sex hormone-binding globulin (SHBG, 107 +/- 13.5 vs. 106.6 +/- 13.3 nmol/l) levels remained constant. In the eight postmenopausal women who were not receiving exogenous hormones (PM), wheat bran consumption was not associated with predicted decreased levels of estradiol (25.7 +/- 2.7 vs. 31.0 +/- 1.9 pg/ml), estrone (38.3 +/- 10.1 vs. 39.3 +/- 10.6 pg/ml), and androstenedione (0.78 +/- 0.08 vs. 0.68 +/- 0.11 ng/ml) or with increased concentrations of SHBG (35.2 +/- 6.4 vs. 34.8 +/- 6.5 nmol/l). Participants receiving ERT had baseline and postintervention levels of estradiol and SHBG significantly higher and androstenedione significantly lower than those not receiving ERT. No association between wheat bran supplementation and hormone levels was found in PM or PM-ERT African-American participants. These results in postmenopausal women are in contrast to findings of earlier studies in premenopausal women indicating that wheat bran fiber decreases serum sex hormones. Estrogen levels in postmenopausal women are only 5-10% of those in premenopausal women; therefore, a high wheat bran fiber diet alone may not be sufficient to depress these low levels even further.

Fusarium toxins in wheat from an area in Henan Province, PR China, with a previous human red mould intoxication episode.

Wheat samples of the 1998 and 1999 crops from Puyang, an area in Henan Province, PR China with a previous human red mould intoxication episode, were analysed for trichothecenes and zearalenone (ZEA). For the 1998 Puyang crop, deoxynivalenol (DON) was the predominant toxin detected abundantly and frequently at a level of up to 14,000 microg kg(-1) (mean 2850 microgkg(-1)) in 30 of 31 (97%) wheat samples. Among these were 21 (70%) with a DON level that exceeded the Chinese regulation of 1,000 microg kg(-1). Nivalenol (NIV) and 15-acetyl-DON (15-ADON) were also found at 578 microg kg(-1) (one sample) and 59-1,800 microg kg(-1) (mean 365 microg kg(-1), 20 samples), respectively. ZEA co-occurred in 21 samples at 9-1,400 microg kg(-1) (mean 209 microg kg(-1)). Twenty-five (89%) wheat samples from Zhumadian, a region without a history of human red mould intoxication in the same province, contained low levels of DON (53-1240, mean 223 microg kg(-1)) with seven (25%) co-contaminated with ZEA (10-217, mean 108 microg kg(-1)). All were free from 15-ADON and NIV. Significant differences in DON, 15-ADON and ZEA concentrations between both areas were found. DON (<1000 microg kg(-1)) and ZEA (5-111 microg kg(-1)) were also detected in the 1999 Puyang wheat. Proper environmental conditions for Fusarium species surviving winter combined with unusual high precipitation during wheat flowering were responsible for a high concentration of Fusarium mycotoxins in the 1998 Puyang wheat.

Distribution of toxigenic Fusarium spp. and mycotoxin production in milling fractions of durum wheat.

A reliable and sensitive PCR assay to specifically detect trichothecene-producing Fusarium spp. in milling fractions and kernel tissue of naturally infected durum wheat is reported. Assays were based on a combination of primers derived from the trichodiene synthase and the beta-tubulin genes. The occurrence of toxigenic Fusarium spp. in semolina and wheat tissue (grain ends, crease, pericarp, aleurone layer, germ and albumen) was detected, even for a weakly contaminated wheat sample. Penetration of toxigenic Fusarium spp. into the interior of durum wheat kernel was demonstrated for the Nefer variety, indicating that none of the tissue structures within the wheat kernel acted as an effective barrier to fungal invasion. Moreover, after inoculation by toxigenic Fusarium strains, semolina was shown to allow high yields of trichothecenes, while bran was demonstrated to contain biochemical inhibitors able to significantly reduce trichothecene production. These results will be useful in improving breeding strategies to control trichothecene contamination of durum wheat kernels.

Distribution of the trichothecene mycotoxin deoxynivalenol (vomitoxin) during the milling of naturally contaminated hard red spring wheat and its fate in baked products.

In order to assess human intake of deoxynivalenol (DON, vomitoxin) from wheat sources for regulatory purposes, the effects of food processing on DON retention and distribution must be known. Two hard red spring wheats (1 and 2) naturally contaminated with DON at 7.5 micrograms/g and 1.4 micrograms/g were subjected to cleaning, tempering and scouring processes then milled in the Grain Research Laboratory (GRL) pilot mill; the low-level wheat was also milled in an experimental mill (Allis-Chalmers mill) and commercial-scale pilot mill (Canadian International Grains Institute). Flours were baked into bread and, in some cases, cookies and doughnuts. No appreciable losses of DON occurred during the cleaning, milling or baking processes. Some fractionation took place during milling: up to two-fold increases in DON concentrations were observed in shorts and feed flour fractions, while lesser increases were found in the bran from wheat 2 (none for bran from wheat 1); there was some correlation of ash concentration with DON levels in the flour streams from GRL pilot milling of the two wheats.

Application of capillary gas chromatography to a survey of wheat for five trichothecenes.

A previously published method for the determination of deoxynivalenol (DON) and nivalenol (NIV) in grains by capillary gas chromatography (GC) with electron-capture detection (ECD) has been modified to include HT-2 toxin (HT-2), T-2 toxin (T-2) and diacetoxyscirpenol (DAS). Recoveries of the five trichothecenes from wheat averaged 71-92% in the range 0.8-4 micrograms/g. Detection or quantitation limits found in two laboratories were from 0.02 to 0.4 micrograms/g, with those for T-2 and DAS at the high end of this range. The method proved of practical use in survey work for the screening and determination of these trichothecenes in wheat from the 1987 western Canadian crop. There were no false positives for HT-2, T-2 and DON in durum and HY-320 wheat. However, interferences precluded the determination of 4- and 15-monoacetoxyscirpenol (MAS) by GC-ECD. The natural occurrence of HT-2 toxin (0.06-0.59 micrograms/g) was demonstrated by GC-ECD and confirmed by GC-mass spectrometry (MS) in 23 samples of durum and HY-320 wheats (1986 and 1987 crops); it was almost always accompanied by DON and in one sample a low concentration of NIV (0-05 micrograms/g). False positives for HT-2 and DAS in red spring wheat detected in 1987 by GC-ECD were 6% (nearly all identified as variety Katepwa) and 8%, respectively. Hence confirmation of suspected HT-2 and DAS by GC-MS is necessary, particularly with red spring wheat.

Necrotic enteritis: effect of barley, wheat and corn diets on proliferation of Clostridium perfringens type A.

Necrotic enteritis, caused by Clostridium perfringens type A, is more prevalent in broilers fed wheat or barley diets than in those fed a corn diet. We compared the effects of wheat, barley and corn diets on in vitro proliferation of C. perfringens type A. Bacteria were inoculated into the supernatants delivered from either digested or non-digested barley, wheat and corn diets mixed with thioglycollate medium (1:3). Colony forming units were counted following incubation for 6 h at 40 degrees C. There were no significant differences in clostridial proliferation among non-digested diets. Bacterial proliferation in the digested wheat and barley diets was significantly higher than in the digested corn diet. These findings suggest that the increased incidence of necrotic enteritis in broilers fed barley and wheat diets compared with those fed a corn diet may be due in part to increased clostridial proliferation associated with the wheat and barley diets, or to decreased proliferation associated with the corn diet.

Breath hydrogen and methane responses of men and women to breads made with white flour or whole wheat flours of different particle sizes.

OBJECTIVE: While it is well known that consumption of whole grain foods results in beneficial health effects, the great majority of Americans prefer bread made with white flour. Consumption of whole grain foods in high fiber menus may cause undesirable intestinal responses. The purpose of this study was to determine whether consumption of bread made with ultra fine ground whole wheat flour retained beneficial effects while reducing undesirable effects. METHODS: Twenty-six men and women (31 to 55 years of age) consumed glucose solutions or bread made with white, whole wheat or fine ground whole wheat flour (1 g carbohydrate/kg body weight) in a Latin square design after two days of controlled diet. Breath methane and hydrogen were determined over the 24 hours after consumption of test foods. RESULTS: Hydrogen and methane responses of men and women were similar. There were no significant differences in methane responses to the different treatments, but hydrogen response was increased by all breads (p<0.0001). Although the overall mean response values were similar for all three breads, the patterns of hydrogen response differed (treatment*time, p<0.003). Gastrointestinal symptoms were not associated with fiber content or particle size of bread; however, subjects reporting symptoms tended to have higher methane responses (0.05<p<0.10). CONCLUSION: Neither fiber content of bread nor particle size of whole wheat flour substantially affected breath hydrogen or methane responses or gastrointestinal symptoms. Fine ground whole wheat breads may provide a more acceptable food choice than standard whole wheat bread without sacrificing the beneficial health effects of higher fiber.
