Kinetic regularities of the reaction of superprecipitation of myometrium actomyosin, as well as the effect of different concentrations of EGTA, EDTA and diphosphonic acids on this process have been studied. Results obtained are of interest from the viewpoint of possible practical use of diphosphonates as factors modifying interaction of the contractile proteins of the uterus smooth muscles under the pathology of contractile response.

AIM: To construct a recombinant vector containing fused gene of heat shock protein A(HspA) and outer membrane protein(OMP) with M(r) 18,000, from human Helicobacter pylori(Hp) and express the fusion protein in E.coli BL21. METHODS: The gene encoding HspA was amplified from Hp chromosome by PCR. After digestion with kpn I and BamH I, the HspA gene was inserted into the prokaryotic expression vector pET32a(+). After recombinant vectors pET32a(+)/HspA and pET32a(+)/Omp (18) were digested with Hind III and BamH I, the pET32a(+)/HspA and 18,000 OMP gene segments were recovered through agarose electrophoresis, and connected by T4 ligase. The recombinant vector pET32a(+)/HspA-Omp(18) was transformed into E.coli BL21(DE3). The antigenicity of recombinant fusion protein was analysed by Western blot. RESULTS: Enzyme digestion analysis and sequencing showed that the fused gene had 891 base pairs. As compared with gene reported in GenBank, there were 1.15% and 1.26% differences in cloned nucleotide sequence and amino acid sequence respectively. SDS-PAGE analysis showed that recombinant vector could be expressed in E.coli BL21. The relative molecule mass (M(r)) of expressed fusion protein was 51x10(3). And soluble expression product accounted for 18.96% of total bacterial protein. After purification via Ni-NTA agarose resin, the purity of recombinant fusion protein was about 95%. The Western blot analysis showed that recombinant fusion protein could be recognized by anti-Hp positive serum and anti-18,000 OMP mAb, suggesting that this protein had good antigenicity. CONCLUSION: The fused gene of HspA and OMP is cloned and expressed successfully, which lays the foundation for development of protein and DNA vaccines and a diagnostic kit of Hp infection.

UNLABELLED: Safety, targeting, and antitumor efficacy of pretargeted radioimmunotherapy using anti-carcinoembryonic antigen (CEA) hMN-14 x m734 bispecific antibody (BsmAb) and 131I-di-diethylenetriamine pentaacetic acid (DTPA)-indium hapten were evaluated in a phase I study performed on patients with CEA-expressing tumors. METHODS: Twenty-two patients with nonmedullary thyroid carcinoma (non-MTC) (group I, 13 patients) or medullary thyroid carcinoma (MTC) (group II, 9 patients) were enrolled. These patients received a 75 mg/m2 (11 patients) or 40 mg/m2 (11 patients) dose of BsmAb and escalating activities of (131)I-di-DTPA-indium 5 d later. Toxicity and tumor response were assessed in 20 patients who received a therapeutic (>2.2 GBq) hapten dose of radioactivity. RESULTS: The percentage of lesions detected by immunoscintigraphy after injection of the therapeutic dose of hapten was 70% on an anatomic-site basis. High bone uptake was relatively frequent. A transient grade I or II hepatic toxicity was observed in 5 patients (45%) injected with 75 mg/m2 of BsmAb and in 1 patient (11%) injected with 40 mg/m2. No other nonhematologic toxicity was observed. With 75 mg/m2 of BsmAb, hematologic toxicity was high: 5 cases of grade III or IV leukopenia (45%) and 5 cases of grade III or IV thrombopenia (45%). With a 40 mg/m2 dose of BsmAb, hematologic toxicity was reduced significantly: 3 cases of grade III or IV leukopenia (33%) and 1 case of grade III or IV thrombopenia (11%) (P = 0.02). Toxicity was significantly higher in MTC patients than in non-MTC patients (P = 0.019). Nine cases of tumor stabilization of 3 mo to more than 12 mo were observed (45%), 6 in the MTC group and 3 in the non-MTC group. The rate of disease stabilization was significantly higher with 75 mg/m2 of BsmAb (64%) than with 40 mg/m2 (22%) (P = 0.04). Human antimouse antibody elevation was observed in 1 patient (8%) and human antihuman antibody in 4 (33%). CONCLUSION: A BsmAb dose of 40 mg/m2 and a 5-d interval appeared to be a better dose/schedule regimen, with acceptable toxicity. Under these conditions, the maximal tolerated activity was 3 GBq of 131I-di-DTPA-indium in MTC patients. In non-MTC patients, dose escalation should continue.
A combination of the 69 kDa outer membrane protein and filamentous hemagglutinin (FHA), both isolated from pertussis toxin (ptx) minus mutants of B. pertussis, is protective in the mouse intracerebral challenge potency test ("Kendrick test"). A combination of the same 69 kDa protein and lymphocytosis promoting factor (LPF, pertussis toxin, PTX) is approximately 15 times less effective. The data show that the 69 kDa protein in tandem with FHA is the most relevant combination for mouse protection, and may be a more suitable acellular pertussis vaccine candidate than the LPF/FHA combination which has never been satisfactorily protective in the mouse test. However, the preparation of the 69 kDa protein of acceptable quality is still difficult. Attempts were made at screening the most suitable batches of the preparation by exploiting some recently discovered properties of the 69 kDa protein, i.e. its chromatofocusing pattern and lymphocyte affinity. Although both these properties may differentiate 69 kDa preparations, none of them was able to replace the mouse intracerebral challenge potency test for the final quality assessment.

276 volunteers aged 19 years and over were placed under observation in the course of the trial of oral cholera vaccine in tablets, containing choleragen toxoid, O-antigens of serovars Inaba and Ogawa and a number of Vibrio cholerae exoenzymes, for safety, reactogenic properties and immunological effectiveness. The vaccine was found to produce no reactions in a dose of 1-4 tablets; the administration of 3 tablets (300,000 binding units of the toxoid and 10,000 units of O-antigens, serovars Inaba and Ogawa) was shown to induce the most intensive synthesis of both antitoxins and vibriocidal antibodies in the blood sera of volunteers, as well as IgA coproantibodies. The oral vaccine was found to have an advantage over parenteral vaccines due to the absence of reactogenic properties and the formation of local immunity: coproantibodies appeared in 80% and 9% of the vaccinees respectively.
