Diagnosis and treatment of primary malignant tumors of the seminal vesicles.

The author, after suggesting that malignant tumors of the seminal vesicles are not as rare as usually believed, considers the best method of early diagnosis to be deferentovesiculography carried out in all suspected cases. Surgery of these tumors includes vesiculectomy by the transvesical route which permits simultaneous treatment of vesicular, ureteral, vesical, and prostatic lesions. A place of choice is reserved for high-energy radiotherapy.

Extraskeletal Ewing's sarcoma contiguous with the seminal vesicle.

A 25-year-old man presented with acute pain in the right lower quadrant of the abdomen and a locally palpable mass. The tumour was extirpated together with the right seminal vesicle, adjacent ureter and bladder wall. The histologic diagnosis was extraskeletal Ewing's sarcoma. No tumour tissue was seen within the seminal vesicle. Intensive postoperative chemotherapy was given and at 18-months follow-up the patient was apparently tumour-free.

Effects of Azadirachta indica leaves on the seminal vesicles and ventral prostate in albino rats.

Oral administration of 20, 40, 60, mg of dry Azadirachta indica leaf powder for 24 days resulted in decrease in the weights of seminal vesicles and ventral prostate, reduction in epithelial height, nuclear diameter and the secretory material in the lumen. Biochemically, there was a decrease in total protein, acid phosphatase activities. Seminal vesicles and ventral prostate being androgen dependent, the regressive changes histologically as well as biochemically, suggests the antiandrogenic property of the neem leaves.

Prenatal exposure of male mice to diethylstilbestrol alter the expression of the lactotransferrin gene in seminal vesicles.

We have previously isolated an estrogen-inducible secretory protein, lactotransferrin (LTF), and a cDNA to its messenger RNA from the uterus of mice. In this report we determined that the level of LTF mRNA is minimal in the seminal vesicles of normal mice. In contrast, expression of LTF mRNA in the seminal vesicles of developmentally estrogenized males was both constitutive and estrogen inducible. The results suggested that this alteration may be an example of atypical gene expression after hormonal manipulation early in development.

Patterns of motor and seminal vesicle activities during copulation in the male rat.

The interaction between behavioral and visceral events during copulation was studied in nine intact adult male Wistar rats. The motor copulatory pattern was analyzed by an accelerometric technique and the seminal vesicle pressure (SVP) was recorded through a catheter chronically inserted into the right seminal vesicle. Mounting was associated with a small broad rise in SVP that often preceded the initiation of pelvic thrusting. Penile insertion resulted in a second sharp rise in SVP superimposed on the primary wave related to mounting. A steep rise in SVP occurred during the performance of the motor ejaculatory pattern after a variable period of intravaginal thrusting. This final rise was associated with ejaculation. Analysis of the temporal relationships ships between the SVP changes and copulatory behavior revealed that each of the three rises of SVP was related to distinct patterns of stimulation, suggesting that they were mediated by separate reflex arcs.

Metabolism of ATP during anoxia in guinea pig seminal vesicle mucosa.

The content of adenine nucleotides in guinea pig seminal vesicles was determined enzymatically. Inosine monophosphate (IMP) was isolated from other tissue nucleotides by two-dimensional thin-layer chromatography on cellulose PEI plates, eluted, and quantitated spectrophotometrically. The intracellular concentration of IMP increased 10-fold over control values (aerobic tissue) concomitantly with the strong ATP drain caused by anoxia. This contrasts markedly with other tissues in which ATP depletion is usually associated with an increase in AMP. Tissue inorganic phosphate also accumulated in an amount corresponding to the apparent ATP depletion. Uric acid did not accumulate. Our data suggest the possibility of a special role for IMP in metabolic control in this tissue.

Ultrastructure of apyrene and eupyrene spermatozoa from the seminal vesicle of Euptoieta hegesia (Lepidoptera: Nymphalidae).

The ultrastructure of the seminal vesicle's spermatozoa of the butterfly Euptoieta hegesia was analyzed. The apyrene spermatozoa measure about 300 microm in length and swim freely in a secretion. The anterior end consists in a cap with a cylindrical extension and a globular structure. The flagellum has a 9+9+2 axoneme, two mitochondrial derivatives with paracrystalline matrices and an external coat formed by concentric layers. The eupyrene spermatozoa measure about 550 microm in length and are grouped into bundles. The anterior end consists in an amorphous globule. Posterior to this globule, a coat with a dense material covers the spermatozoon where an acrosome and a nucleus appear. The flagellum has a 9+9+2 axoneme and two mitochondrial derivatives. External to the coat and attached to the dense material, there is a reticular appendage, which has a paracrystalline core and extends to the distal tip of the spermatozoon.

Castration-resistant secretion in the hamster seminal vesicle does not depend on androgens.

The secretory activity of seminal vesicles (SV) in the castrated hamster was studied by stereological analysis and biochemical approaches following treatment with cyproterone acetate (CPA) and adrenalectomy in order to investigate whether extra-testicular androgens are responsible for castration-resistant protein secretion. Treatment of castrated animals with CPA decreased the size of secretory granules and increased the number of apical granules, though neither the absolute nor the relative volume of all the components analysed was changed. In addition, CPA-treatment increased the amount of protein exocytosed by SV in castrated animals, though total protein synthesis remained unchanged. Adrenalectomy neither suppressed secretion nor induced any further ultrastructural changes in the SV epithelium. Our results demonstrate that secretory activity of the hamster SV following castration is not controlled by extra-testicular androgens and suggest that SV secretory proteins, which are heterogeneous with regard to their sensitivity to androgen withdrawal, might be regulated differentially by androgens.

Characterization of a genomic clone for rat seminal vesicle secretory protein IV.

The entire coding region for rat seminal vesicle secretory protein IV was obtained on a 3.5 kb Eco RI fragment isolated from a genomic library in lambda Charon 4A. The coding sequence for SVS IV message is interrupted twice by introns. The first lies just downstream from the juncture of the 21 amino acid secretory signal peptide with the start of the mature protein, and the second lies in the 3'-nontranslated region. The major transcriptional start site was mapped by primer extention and is 22 nucleotides upstream from the translational initiation codon. S1 protection experiments indicated additional minor transcriptional starts about 27 and 50 nucleotides further upstream from the major cap site. The entire transcriptional unit comprises about 1740 nucleotides. The SVS IV gene does not belong to an obvious gene family, and it is conserved in mice and guinea pigs.

Zinc content in the epididymis, vas deferens, prostate, and seminal vesicles of juvenile rhesus monkey (Macaca mulatta): effect of androgen and estrogen.

The zinc content in the three segments of the epididymis (caput, corpus, and cauda), vas deferens, seminal vesicles, and prostate of juvenile monkeys was determined by atomic absorption spectrophotometry. Zinc content (micrograms/gm wet weight) was found to be maximum (328) in the vas deferens; in the other organs it measured in the following order: caput 191, corpus 238, cauda 193, prostate 133 and seminal vesicles 85. In order to investigate the endocrine control of the zinc in these organs, two groups of animals were treated with testosterone propionate (2 mg) or estradiol dipropionate (10 micrograms) once daily for 30 days. In response to androgen, a rise in both concentration and content of zinc was evident only in the prostate. The results further suggested that the prostatic zinc may be under dual hormonal control, but in the epididymis and vas deferens it may be under the influence of estrogen. It is concluded that the hormonal effects on zinc content and growth stimulation in accessory sex organs are quite separate and may be under different hormonal control.

The androgen-dependent mouse seminal vesicle secretory protein IV: characterization and complementary deoxyribonucleic acid cloning.

Seminal vesicle secretory protein IV of a mouse has been isolated, and the cDNA coding for its mRNA has been cloned and sequenced. The 556-nucleotides encode 16 amino acid signal peptides and 92 residues of mature protein. Considerable homology between mouse and rat SVS IV cDNA was found. In the leader peptide and 3'-noncoding region there is 92% and 85% homology, respectively. The other regional homologies are 86% for the first 12, 68.5% for the last 35, and 40% for the middle 44 amino acids. The expression of mouse SVS IV mRNA is under the control of androgen. Administration of testosterone to castrated mice resulted in induction of the mRNA level to 50% of the mature male in 96 h of hormone treatment. Secretion of the protein after testosterone injection follows a similar pattern.

Cellular autophagocytosis in mouse seminal vesicle cells in vitro.

Cellular autophagocytosis was observed in mouse seminal vesicle cells incubated in vitro up to 8 h in medium 199 or Krebs-Ringer bicarbonate buffer. During the first 2 h of incubation, early forms of autophagic vacuoles were seen in the cells, advanced forms containing degraded material began to cumulate later. After 6--8 h, early vacuoles occurred sparsely, while advanced forms were detected in a great number. During the first 2 h of incubation, we often observed smooth surfaced membrane pairs between the cisternae of rough surfaced endoplasmic reticulum resembling isolating membranes of autophagic vacuoles. They varied in size and shape from short, straight cisternae to long, curved ones, almost completely encircling areas of the cytoplasm. Based on these observations, we propose a tentative scheme of the formation of autophagic vacuoles, viz., the short, straight cisternae would represent the first stage in the development of an autophagic vacuole, while the curved sack-like forms are interpreted as successive steps leading to the complete sequestration of an area of the cytoplasm.

Purification and characterization of a trypsin inhibitor from mouse seminal vesicle secretion.

A Kazal-type trypsin inhibitor in mouse seminal vesicle secretion was purified to homogeneity via a series of purification steps including ammonium sulfate fractionation, affinity chromatography on a trypsin Affi-Gel 10 column, and HPLC on a reverse phase C4 column. It was shown to be a weak basic protein with an isoelectric point of 8.7 and to contain no carbohydrate. The protein had a specific activity of 184 U/mg protein in the inhibitory effect on the trypsin digestion of N-benzoyl-Pro-Phe-Arg-p-nitroanilide. Analysis of the kinetic data for the trypsin digestion of N-benzoyl-Phe-Val-Arg 7-amido-4-methylcoumarin revealed that the protein was a competitive inhibitor with an inhibitory constant (Ki) of 0.15 nM. The molecular mass of the protein was determined to be 7 kDa by both gel chromatography and electrophoresis. Results of direct amino acid determinations indicated that this protein corresponded to the reading frame of MP12 cDNA identified from mouse prostate. We found that cleavage only at the reactive site of this protein (Arg19-Ile20) resulted in its denaturation.

An ex vivo evaluation of regulatory role of biogenic amines in rat seminal vesicle after pharmacological manipulation.

We studied the neural regulation of seminal vesicles (SV) by determining the changes of intraluminal pressure of rat SV in response to an electric stimulation of the lesser splanchnic nerve (LSN). After pharmacological manipulation with neurotoxin, the contents of monoamines and their metabolites in SV were estimated. In rats receiving electric stimulation of the LSN, an increase of intraluminal pressure was obtained with a reduction of the serotonergic turnover rate in SV. An intraperitoneal injection of DSP-4 (100 mg/kg), the noradrenergic neurotoxin, into rats decreased the level of norepinephrine (NE) in SV significantly but did not influence this functional response. Also, the intraluminal pressure was lowered by an intrathecal injection of 6-OHDA (20 microg/rat) to denervate spinal monoaminergic nerves in rats although the contents of monoamines in SV were not changed. This indicates that noradrenergic neurotransmission appears unimportant in this regulation. The lowering of intraluminal pressure in rats by 6-OHDA is mainly related to an attenuation of dopaminergic neuroregulation from the decrease of turnover rate of DA. Otherwise, an intrathecal injection of 5,7-DHT (60 microg/rat) to abolish spinal serotonergic nerves did not influence the level of monoamines in SV but increased the intraluminal pressure indicating an involvement of inhibitory regulation from spinal serotonergic pathway. These results suggest that contraction by electric stimulation of the LSN in Wistar rat SV is mainly regulated by the dopaminergic nervous pathway and an inhibitory regulation of the serotonergic nervous pathway from spinal cord while the noradrenergic nervous system seems unimportant for this regulation.

Recurrence of a non-seminomatous germ cell tumor in the seminal vesicle 20 years after initial diagnosis and treatment.

We present a case of a pathologic stage 1, right sided, non-seminomatous germ cell tumor recurrence in the left seminal vesicle, 20 years after initial diagnosis and treatment. The patient was treated with three salvage cycles of bleomycin, etoposide, and cisplatinum. At 24 months of follow-up after completion of chemotherapy, digital rectal and TRUS examinations revealed complete resolution of the lesion. We believe that this tumor is a late metastasis to the contralateral seminal vesicle.

Carcinoma in situ of the bladder extending into the seminal vesicles.

Involvement of the urethra or distal ureter in 30 to 50 per cent of the patients with carcinoma in situ of the bladder is well known. Carcinoma in situ of the bladder extending to the seminal vesicles rarely has been mentioned. We discuss 2 patients with extension of carcinoma in situ into the seminal vesicles and replacement of the normally present cylindroid mucosal lining. Carcinoma in situ of the ureter, bladder and prostatic ducts also was present in both patients.

Changes in enzyme activities of thymidine kinase and 5'-nucleotidase for dTMP during hormonal regeneration of seminal vesicles of mice.

An increase of thymidine kinase [EC 2.7.1.21] activity and decrease of 5'-nucleotidase [EC 3.1.3.5] activity for dTMP were found during hormonal regeneration of the seminal vesicles by daily or single administration of testosterone propionate into mice castrated 2 weeks previously. Actinomycin D injected on day 0 of testosterone treatment completely inhibited both the increase of thymidine kinase and the decrease of 5'-nucleotidase. When injected on day 2, actinomycin D decreased thymidine kinase activity below the control level and 5'nucleotidase activity was not restored to the normal level. The activity of 5'-nucelotidase in a mixed sample, in which seminal vesicles of castrated mice and those of testosterone-treated mice were homogenized together, was intermediate between the activities determined separately. This indicates the absence of any inhibitor of 5'nucleotidase in the regenerating vesicles. Changes in total activity of 5'nucleotidase and total protein content in extracts during various treatments showed that the decrease in specific activity of 5'-nucleotidase in the first 2 days of testosterone treatment was not due to inhibition of enzyme activity but to dilution of the enzyme with other proteins which increased in content more rapidly than 5'-nucleotidase.
