[Sanitary chemical research on the atmosphere in wards for cardiac patients].

Man passes a greater part of the day-and-night in closed premises. The pollutants of the inside air, in quantity and quality are more various than that of the atmospheric air, which is due to the additional sources of pollution. The investigation performed is part of a complex study and hygienic assessment of the environment in the hospital rooms of two cardiological wards in view of the prevailing contingent of patients with heart diseases and their specific sensitivity to its qualities. The chemical state of the aerial environment was traced according to several basic indices: concentration of CO2, ammonia, total content of organic substances (oxidizable) and some pollutants, migrating from the polymeric materials, used for furnishing of the hospital rooms. The results received show the necessity for strictly following the regime of ventilation, ensuring the indispensable area for each patient and high light for the rooms. In view of economy of time, labour and means it is recommended the determination of oxidizability and content of ammonia in the air to be carried out one daily. It is also recommended to use synthetic polymer materials in the hospital premises subject to enforced sanitary protection, as little as possible.

Measurement of emissions from air pollution sources. 5. C1-C32 organic compounds from gasoline-powered motor vehicles.

Gas- and particle-phase organic compounds present in the tailpipe emissions from an in-use fleet of gasoline-powered automobiles and light-duty trucks were quantified using a two-stage dilution source sampling system. The vehicles were driven through the cold-start Federal Test Procedure (FTP) urban driving cycle on a transient dynamometer. Emission rates of 66 volatile hydrocarbons, 96 semi-volatile and particle-phase organic compounds, 27 carbonyls, and fine particle mass and chemical composition were quantified. Six isoprenoids and two tricyclic terpanes, which are quantified using new source sampling techniques for semi-volatile organic compounds, have been identified as potential tracers for gasoline-powered motor vehicle emissions. A composite of the commercially distributed California Phase II Reformulated Gasoline used in these tests was analyzed by several analytical methods to quantify the gasoline composition, including some organic compounds that are found in the atmosphere as semi-volatile and particle-phase organic compounds. These results allow a direct comparison of the semi-volatile and particle-phase organic compound emissions from gasoline-powered motor vehicles to the gasoline burned by these vehicles. The distribution of n-alkanes and isoprenoids emitted from the catalyst-equipped gasoline-powered vehicles is the same as the distribution of these compounds found in the gasoline used, whereas the distribution of these compounds in the emissions from the noncatalyst vehicles is very different from the distribution in the fuel. In contrast, the distribution of the polycyclic aromatic hydrocarbons and their methylated homologues in the gasoline is significantly different from the distribution of the PAH in the tailpipe emissions from both types of vehicles.

Assessment of new vehicles emissions certification standards in the metropolitan area of Mexico City.

Light duty gasoline vehicles account for most of CO hydrocarbons and NOx emissions at the Metropolitan Area of Mexico City (MAMC). In order to ameliorate air pollution from the beginning of 2001, Tier 1 emission standards became mandatory for all new model year sold in the country. Car manufacturers in Mexico do not guarantee the performance of their exhaust emissions systems for a given mileage. The purpose of this study was to assess whether the Tier 1 vehicles will stand the certification values for at least 162000 km with the regular fuel available at the MAMC. Mileage accumulation and deterioration show that certified carbon monoxide emissions will stand for the useful life of the vehicles but in the case of non-methane hydrocarbons will be shorter by 40%, and nitrogen oxides emissions above the standard will be reached at one third of the accumulated kilometers. The effect of gasoline sulfur content, on the current in use Tier 1 vehicles of the MAMC and the impact on the emissions inventory in year 2010 showed that 31000 extra tons of NOx could be added to the inventory caused by the failure of the vehicles to control this pollutant at the useful life of vehicles.

A novel marine algal toxicity bioassay based on sporulation inhibition in the green macroalga Ulva pertusa (Chlorophyta).

A 5-day aquatic toxicity test based on sporulation inhibition of the green macroalga Ulva pertusa Kjellman has been developed. Optimal test conditions determined for photon irradiance, salinity and temperature were 60-200 micromol photons m(-2)s(-1), 25-35 per thousand and 15-20 degrees C, respectively. Tests were conducted by exposing U. pertusa thallus disks to a reference toxicant (sodium dodecyl sulfate; SDS), metals (Cd2+, Cu2+, Zn2+, Pb2+) and elutriates of sludge collected from nine different locations. The EC(50) values for SDS was 5.35 mg L(-1). When four heavy metals were assayed, the NOECs were highest for lead (0.625 mg L(-1)) and lowest for copper (0.031 mg L(-1)). The EC50 values showed the following toxicity rankings: Cu(2+) (0.061 mg L(-1))>Cd2+ (0.326 mg L(-1))>Zn2+ (0.738 mg L(-1))>Pb2+ (0.877 mg L(-1)). The bioassay indicated also that the sporulation endpoint could be a sensitive indicator of toxicity effects of elutriates of sludge as reflected from the NOEC values equal to or lower than the lowest concentration employed (6.25%). Sporulation was significantly inhibitied in all elutriates with the greatest and least effects observed in elutriates of sludge from industrial waste (EC50 6.78%) and filtration bed (EC50 15.0%), respectively. The results of the Spearman rank correlation analysis for EC50 data versus the concentrations of toxicants in the sludge presented a significant correlation between toxicity and four heavy metals (Cd2+, Cu2+, Pb2+, Zn2+). Introduction of the concept of toxicity unit (TU) showed that these metals were the main cause of toxicity in elutriates of at least four out of nine sludge samples. Members of the order Ulvales show a wide geographic distribution and have similar reproductive characteristics, thus making it possible to apply the present test method to other algae of this taxa, elsewhere. This novel method will be a useful tool for assessing the aquatic toxicity of a wide range of toxicants, once the respective sensitivities are demonstrated.

Up-regulation of the alligator CYP3A77 gene by toxaphene and dexamethasone and its short term effect on plasma testosterone concentrations.

In this study we describe an alligator hepatic CYP3A gene, CYP3A77, which is inducible by dexamethasone and toxaphene. CYP3A plays a broad role in biotransforming both exogenous compounds and endogenous hormones such as testosterone and estradiol. Alligators collected from sites in Florida that are contaminated with organochlorine compounds exhibit differences in sex steroid concentrations. Many organochlorine compounds induce CYP3A expression in other vertebrates; hence, CYP3A induction by organochlorine contaminants could increase biotransformation and clearance of sex steroids by CYP3A and provide a plausible mechanism for the lowering of endogenous sex steroid concentrations in alligator plasma. We used real time PCR to examine whether known and suspected CYP3A inducers (dexamethasone, metyrapone, rifampicin, and toxaphene) up-regulate steady state levels of hepatic CYP3A77 transcript to determine if induction patterns in female juvenile alligators are similar to those reported in other vertebrates and whether toxaphene, an organochlorine compound found in high concentrations in Lake Apopka alligators, induces this gene. Estrogen receptor alpha (ERalpha), estrogen receptor beta (ERbeta), androgen receptor (AR), glucocorticoid receptor (GR), progesterone receptor (PR), and steroid-xenobiotic receptor (SXR) transcripts were also measured to determine whether any of these nuclear receptors are also regulated by these compounds in alligators. Dexamethasone (4.2-fold) and toxaphene (3.5-fold) significantly induced CYP3A77 gene transcript, whereas rifampicin (2.8-fold) and metyrapone (2.1-fold) up-regulated ERbeta after 24h. None of the compounds significantly up-regulated AR, ERalpha, GR, PR, or SXR over this time period. Plasma testosterone (T) did not change significantly after 24h in alligators from any of the treatment groups. Dexamethasone treated animals exhibited a strong relationship between the 24h plasma T concentrations and CYP3A77 (R(2)=0.9, positive) and SXR (R(2)=0.77, negative) transcripts, which suggests that the expression of these genes is related to plasma T in alligators. In light of our findings, we hypothesized that higher steady state CYP3A77 (and possibly SXR) gene expression would be observed in alligators collected from Lake Apopka, a polluted lake containing organochlorine compounds known to induce CYP3A isoforms in other taxa. Therefore, we measured basal levels of CYP3A77 and SXR gene transcripts in wild juvenile alligators collected from Orange Lake (reference lake), Lake Woodruff (reference lake), and Lake Apopka (contaminated lake). We found that no differences existed in CYP3A77 or SXR gene expression among animals from the lakes sampled suggesting that exposure to organochlorine compounds at concentrations present in Lake Apopka does not lead to variation in the expression of these genes, although capture stress could be interfering with these results since the glucocorticoid dexamethasone induces CYP3A77 transcript in alligators.

Quantitative structure-property relationships on photolysis of PCDD/Fs adsorbed to spruce (Picea abies (L.) Karst.) needle surfaces under sunlight irradiation.

By partial least squares (PLS) regression, quantitative structure-property relationship (QSPR) models were developed for photolysis half-life (t1/2) of PCDD/Fs and PAHs sorbed to spruce (Picea abies (L.) Karst.) needle surfaces and irradiated by sunlight. Quantum chemical descriptors computed by PM3 Hamiltonian were used predictor variables. PLS analysis for the PCDDs and PAHs respectively resulted in no correlation by our statistical methods. The cross-validated Qcum2 value for the optimal QSPR model of PCDFs is 0.722, indicating a good predictive ability for logt1/2 of PCDFs adsorbed to spruce needle surfaces. The QSPR results show that the number of chlorine atoms bonded to the parent structure and (E(LUMO)-E(HOMO))2 has a dominant effect on t1/2 values of PCDFs. Increasing the number of chlorine atoms and (E(LUMO)-E(HOMO))2 values leads to increase of logt1/2 values of PCDFs.

Effect of sediment turbidity and color on light output measurement for Microtox Basic Solid-Phase Test.

In this work, sediment samples collected from several Spanish harbours were tested with two toxicity procedures, designed for solid samples: the Microtox Basic Solid-Phase Test (BSPT) and a modified procedure of the previous test protocol (mBSPT). According to the BSPT procedure, after initial light readings, pure bacteria were exposed to sediment suspension dilutions and light production was directly measured on suspended sediments without any further manipulation. As measurements are likely to be affected by sediment turbidity and color, a variation in initial light measurement has been here suggested, in order to consider the sample effect at all time readings during the test. Firstly, when sediment suspensions at different concentrations were added to bacteria suspension, immediately the initial light output drastically decayed by more than 50% in signal difference, resulting in a false inhibition, as effect of sample turbidity/color. This effect was more evident at high EC50 values, when slightly or not toxic samples were assessed. Secondly, the comparison of the EC50 obtained with both procedures, demonstrated that the mBSPT produced higher EC50 values (less toxic) than those obtained with the standard procedure. The mBSPT procedure resulted rapid and effective and it could be applied simultaneously with BSPT, in order to better evaluate the toxicity.

Lichen and moss bags as monitoring devices in urban areas. Part I: influence of exposure on sample vitality.

Samples of the lichen Pseudevernia furfuracea (L.) Zopf and the moss Hypnum cupressiforme Hedw. were exposed for 6 weeks in nylon bags in two air pollution monitoring stations in Trieste and Naples (Italy) with different climates and pollution loads to evaluate influence of environmental conditions on sample vitality. This was assessed before and after exposure by transmission electron microscopy observations, K cellular location, and measurements of C, N, S and photosynthetic pigments content, CO2 gas exchange, and chlorophyll fluorescence. Almost all data sets indicate that exposures caused some damage to the species, considerably heavier in the moss, especially in Naples. The two cryptogams differed significantly in accumulation and retention of C, N, and S, the lichen clearly reflecting NO2 availability. The difference in vitality loss was related to the different ecophysiology of the species, because concentrations of phytotoxic pollutants were low during exposure. Critical notes on the analytical techniques are also given.

Synthesis and characterization of mesoporous silica films encapsulating titanium dioxide particles; Photodegradation of 2,4-dichlorophenol.

Degussa P-25 TiO2 encapsulated in a SBA-15 mesoporous thin film silica matrix prepared using a block copolymer templating method. The film structure was characterized using small angle X-ray diffraction (XRD), variable angle spectroscopic ellipsometry (VASE), and polarized light microscopy. The photoactivity of the films was assessed using HPLC and UV-vis spectroscopy to study the photodegradation of 2,4-dichlorophenol, an industrial pollutant, in water under 350 nm irradiation. The unoptimized film was found to be approximately half as photoactive as an equivalent dispersion of TiO2.

The effects of variability in the inherent optical properties on estimations of chlorophyll a by remote sensing in Swedish freshwaters.

During late summer of 1997 measurements were made of the inherent optical properties in Lake Mälaren, Sweden. These included measurements of the total particulate absorption, phytoplankton absorption, yellow substance absorption and total backscattering. Measurements were also made of concentrations known to affect the inherent optical properties; chlorophyll a and suspended particulate organic and inorganic matter. At the same time measurements of underwater radiance reflectance were made. The above measurements were used to better define the relationships between the inherent optical properties and the concentrations affecting them. Correlation analyses were also made in order to study covariation of the concentrations of suspended and dissolved substances in the data set. These relationships were then used to better parameterize a semi-analytical model predicting remote sensing reflectance. Multivariate sensitivity analyses showed that large variations on the Rr(700-710)/Rr(678-685) ratio vs. chlorophyll concentrations could be expected in a heterogeneous environment such as Lake Mälaren. This could lead to large errors in subsequent estimates of chlorophyll concentrations.

Selective photocatalytic oxidation of NH3 to N2 on platinized TiO2 in water.

Selective photocatalytic oxidation of NH3 to N2 is proposed as a new treatment method for controlling the levels of ammonia in water. The photocatalytic oxidation of ammonia on naked and metallized TiO2 in water saturated with air, nitrogen, or NO2 gas was investigated. While the slow photocatalytic oxidation of NH3 to NO2-/NO3- is the only pathway for decomposition of NH3 on naked TiO2 and Au/TiO2, a new pathway, that of selective oxidation of ammonia to dinitrogen, opens up on Pt/TiO2. The formation of dinitrogen from the oxidation of 15NH3 was confirmed by mass spectrometric detection of 15N2. The photocatalytic conversion of NH3 to N2 greatly increases when the Pt/TiO2 suspension is saturated with NO2 gas, whereas NO2 itself shows little reactivity with naked TiO2 and Au/TiO2. Over 80% of the total nitrogen available in ammonia (0.1 mM) is converted into N2 within 40 min illumination of the N2O-saturated Pt/TiO2 suspension. The ability of N2O to accept the conduction band electrons of Pt/TiO2 was verified by photoelectrochemical measurements. NO2 reductively decomposes to generate OH radicals on Pt/TiO2; the rate of ammonia degradation in the NO2-saturated Pt/TiO2 suspension significantly decreases in the presence of excess tert-butyl alcohol, an OH radical scavenger. The presence of Pt deposits on the TiO2 particles changes the photocatalytic pathway of ammonia conversion by both enhancing OH radical production from NO2 and stabilizing intermediate NHx (x=0, 1, 2) species to facilitate their recombination into N2.

Photo-irradiated titanium dioxide catalyzes site specific DNA damage via generation of hydrogen peroxide.

Titanium dioxide (TiO2) is a potential photosensitizer for photodynamic therapy. In this study, the mechanism of DNA damage catalyzed by photo-irradiated TiO2 was examined using [32P]-5'-end-labeled DNA fragments obtained from human genes. Photo-irradiated TiO2 (anatase and rutile) caused DNA cleavage frequently at the guanine residue in the presence of Cu(II) after E. coli formamidopyrimidine-DNA glycosylase treatment, and the thymine residue was also cleaved after piperidine treatment. Catalase, SOD and bathocuproine, a chelator of Cu(I), inhibited the DNA damage, suggesting the involvement of hydrogen peroxide, superoxide and Cu(I). The photocatalytic generation of Cu(I) from Cu(II) was decreased by the addition of SOD. These findings suggest that the inhibitory effect of SOD on DNA damage is due to the inhibition of the reduction of Cu(II) by superoxide. We also measured the formation of 8-oxo-7,8-dihydro-2'-deoxyguanosine, an indicator of oxidative DNA damage, and showed that anatase is more active than rutile. On the other hand, high concentration of anatase caused DNA damage in the absence of Cu(II). Typical free hydroxyl radical scavengers, such as ethanol, mannnitol, sodium formate and DMSO, inhibited the copper-independent DNA photodamage by anatase. In conclusion, photo-irradiated TiO2 particles catalyze the copper-mediated site-specific DNA damage via the formation of hydrogen peroxide rather than that of a free hydroxyl radical. This DNA-damaging mechanism may participate in the phototoxicity of TiO2.

Membrane filter-fluorescent-antibody method for detection and enumeration of bacteria in water.

A technique which employs nonfluorescing membrane filters and specific fluoresceinisothiocynate-labeled antiserum has been successfully used in the identification and enumeration of known species of Escherichia coli which have been added to natural populations of bacteria found in water. The quantitative results compared favorably with those of standard tests. The use of a dissecting microscope with an external lighting arrangement provided a simple requirement for equipment. This method may be useful in monitoring specific bacterial types from waters which were being monitored for specific pollution.

Photosynthetic photon flux, photoperiod, and temperature effects on emissions of (Z)-3-hexenal, (Z)-3-hexenol, and (Z)-3-hexenyl acetate from lettuce.

To investigate the effects of environment on plant volatile emissions, 'Waldmann's Green' leaf lettuce was cultivated under different levels of photosynthetic photon flux (PPF), photoperiod, and temperature. A modified growth chamber was used to sample plant volatile emissions nondestructively, over time, and under controlled conditions. Total volatile emission rates were significantly higher from lettuce cultivated under PPF of 360 or 200 micromoles m-2 s-1 compared to 105 micromoles m-2 s-1, and significantly higher under a 16-h photoperiod than an 8-h photoperiod. No differences were detected among emission rates from different temperature treatments. In controlled environments, emissions could be regulated by adjusting environmental conditions accordingly.

Photocatalytic degradation of Chicago Sky Blue 6B and Benzopurpurin 4B using titanium dioxide thin film.

Aqueous solutions of azo dyes undergo degradation to form harmless intermediates and colorless products following irradiation by visible light in the presence of titanium dioxide thin films. The dyes that were studied in this work are: Chicago Sky Blue 6B and Benzopurpurin 4B. Results obtained indicated that complete mineralization of the dyes took place under the experimental conditions. There was an increase in conductivity after the complete mineralization experiments possibly indicating the formation of ions such as NO3- and SO4(2-). Chemical oxygen demand (COD) measurements show a decrease in organic matter for both dyes following complete degradation. The effect of how changing experimental conditions such as pH, temperature and starting concentrations of dyes affected the rate of dye degradation was measured. There was an increase in the rate of disappearance of the dye color at lower pH. High concentrations of dye solutions required long degradation time.

[Urban asthma and habitat].

This study examines in urban area the housing conditions in asthma patients who called a first-aid association of doctors (SOS Médecins) and their relationships to symptoms. The patient population is compared with the non asthmatic population who called SOS Medecins during the same period. During the first six months of 1989, among 102,791 calls, 701 were related to acute asthma symptoms, and 100 non asthma patients were sorted at random as a control group. A telephonic questionnary has been carried out to asthmatic patients and to control group. A particular aspect of asthma in Paris is given in this study: the homogeneous distribution of asthma attacks in the different districts of Paris, (with a light prevalence in three districts of the city). Sex, age, occupation were non discriminant and no significant difference was established in housing conditions and indoor air pollution sources, between the two groups of asthma and non-asthma patients.
