Forensic applications of chemical imaging: latent fingerprint detection using visible absorption and luminescence.

Chemical imaging technology is a rapid examination technique that combines molecular spectroscopy and digital imaging, providing information on morphology, composition, structure, and concentration of a material. Among many other applications, chemical imaging offers an array of novel analytical testing methods, which limits sample preparation and provides high-quality imaging data essential in the detection of latent fingerprints. Luminescence chemical imaging and visible absorbance chemical imaging have been successfully applied to ninhydrin, DFO, cyanoacrylate, and luminescent dye-treated latent fingerprints, demonstrating the potential of this technology to aid forensic investigations. In addition, visible absorption chemical imaging has been applied successfully to visualize untreated latent fingerprints.

[Genetic fingerprinting (PFGE) of bacterial isolates for their molecular epidemiology].

The determination of fragment patterns of bacterial genomic DNA after digestion with long-range-cutting restriction enzymes and their separation in pulsed-field-gelelectrophoresis is currently regarded as reliable fingerprinting tool for surveillance and identification of infectious sources and routes of bacterial pathogens. The PFGE based molecular fingerprinting has been applied and used for a broad range of bacterial species. As a prerequisite for its application in national and international network studies (German PulseNet, PulseNet USA, Salmgene) a common protocol for rapid production (1-2 days) of standardised and electronically readable fragment patterns has been developed. National and international outbreaks, diffuse outbreaks and long term surveillance of the ups and downs of pathogens have been successfully carried out by means of PFGE studies. The international outbreak of Salmonella enterica serovar Oranienburg (2001) due to contaminated chocolate is described here underlining the usefulness of PFGE as genetic fingerprinting for epidemiological purposes.

A rapid gas chromatographic method for the fingerprinting of illicit cocaine samples.

A gas chromatographic (GC) fingerprint method, based on the presence or absence of six congeners, was developed for illicit cocaine samples. The fingerprint utilizes the relative abundances of these congeners towards each other, disregarding cocaine as the main constituent, and can be expressed numerically or graphically in the form of pictograms for rapid visual comparison. The method can be applied directly to a solution of the sample in chloroform, without previous workup procedures. More than 70 unrelated samples were analyzed and a great variation was observed in the parameter composition. On the other hand, a remarkable similarity could be seen between related samples. The GC fingerprint method may be considered an important contribution for sample comparison, as is exemplified by a subdivision of the analyzed samples in different categories, based on the number and types of congeners found.

[Pattern recognition applied to the fingerprint of traditional Chinese medicine characterized by two-dimension information data].

AIM: To establish the two-dimension information fingerprints chromatograph of traditional Chinese medicine and investigate via pattern recognition. METHODS: Hierarchical cluster was applied to fingerprints described by one-dimension information data and by two-dimension information data respectively and corresponding results was compared. RESULTS: The gross classes, in which the samples denoted by two-dimension information data, classified by all kinds of hierarchical cluster methods had less differences and more robustness than the methods containing the samples expressed by one-dimension information data. The classes to which the unknown samples would be belonged were determined by hierarchical cluster analysis and artificial neural network (ANN), and the same results were obtained. CONCLUSION: The fingerprints characterized by two-dimension information data could provide more overall and special information as compared with the methods indicated by one-dimension information data.

The analysis was performed on a Zorbax SB-C18 column (5 microm, 4.6 mm i.d. x 250 mm) with acetonitrile-water (containing 0.4% (v/v) acetic acid ) (the volume fraction of acetonitrile from 0 to 40% within 70 min) as mobile phase at a flow rate of 1.0 mL/min, and at a column temperature of 25 degrees C. The detection wavelength was 254 nm. The relative standard deviations of relative retention values and relative peak areas were less than 3%. The mutual fingerprint of the extract of Salvia miltiorrhiza Bge. collected from Danshen Base in Tongren City of Guizhou Province was established by using the similarity calculation software of Chinese herbal fingerprint. The method is reliable and can be helpful in effectively controlling the quality of Salvia miltiorrhiza Bge.

Genipin, a novel fingerprint reagent with colorimetric and fluorogenic activity, part II: optimization, scope and limitations.

Genipin, a hydrolytic product of geniposide extracted from gardenia fruit, was thoroughly studied as a potential fingerprint reagent, and optimal conditions for fingerprint development have been determined. Latent fingerprints on paper items that have been treated with a non-ink running formulation containing 0.17% of the reagent, showed up as both colored and fluorescent images. On brown wrapping paper and on papers with highly luminescent backgrounds, genipin developed more visible and clearer prints than did classical reagents such as ninhydrin or DFO. Another potential advantage of genipin is that it is totally harmless and an environmentally friendly reagent.

Monitoring bovine viral diarrhea virus vaccines for adventitious virus, using T1 ribonuclease viral RNA oligonucleotide fingerprinting.

Viral RNA oligonucleotide fingerprinting was used to discriminate 3 cytopathic vaccine bovine viral diarrhea viruses (BVDV) grown in medium supplemented with serum contaminated with noncytopathic BVDV from the same 3 viruses grown in cell culture free of BVDV. Oligonucleotide fingerprinting also effectively discriminated between reference Singer BVDV, NADL BVDV, and New York-1 BVDV grown in BVDV-free noncontaminated or BVDV-contaminated cell cultures. Oligonucleotide fingerprint mapping of viral RNA maybe used to determine the purity of virus stocks, as well as that of BVDV vaccines.

[The effects of different clinicopathologic variables on serum protein fingerprint in hepatocellular carcinoma patients].

OBJECTIVE: To investigate the effects of different clinicopathologic variables on serum protein fingerprint in hepatocellular carcinoma (HCC) patients. METHODS: Serum samples were collected from 112 HCC patients, Special serum protein or peptide spectra was determined by surface enhanced laser desorption/ionization-time of flight-mass spectrometry (SELDI-TOF-MS) measurement after treating the sample onto weak cation exchange (WCX2) protein chip for each case. The serum protein profiles were compared by BioMarker Wizard Software among the patients stratified according to gender, AFP, presence of portal vein tumor thrombus (PVTT), tumor size, tumor number, presence of cirrhosis, respectively. RESULTS: According to serum protein fingerprints of 112 HCC patients, a total of 100 protein peaks were identified at the m/z value ranging from 1100 to 30,000. (1) Sixteen significant differential proteins were found between the groups of HCC with single tumor and those with multiple tumors (P < 0.01). (2) Only one significant differential protein was found between the groups of HCC with tumor size > 3 cm and those with tumor size 5 cm and those with tumor size 10 cm and those with tumor size <or= 10 cm (P < 0.01). (3) Sixteen significant differential proteins were found between the groups of macroscopic portal vein tumor thrombus (Ma-PVTT) and those without PVTT (N-PVTT); Only 2 significant differential protein were found between the groups of microscopic portal vein tumor thrombus (Mi-PVTT) and N-PVTT (P < 0.01); eight significant differential proteins were found between the groups of Ma-PVTT and Mi-PVTT (P < 0.01). (4) No significant differential protein was found when patients stratified according to gender, presence of cirrhosis and AFP. CONCLUSIONS: PVTT, tumor number and tumor size had significant effects on serum protein fingerprint, while no significant effect on serum protein from gender, presence of cirrhosis and AFP. The most profound impact on the serum protein was attained when cutoff was chosen to be presence of Ma-PVTT compared to less effect from Mi-PVTT and 5cm for tumor size.
