Effect of administrating oxytocin or prostaglandin F2alpha on characteristics of the canine ejaculate.

To test the hypothesis that male dogs treated with smooth muscle contracting drugs have an increase in the total number of spermatozoa in the ejaculate but no change in all other ejaculate characteristics, such as progressive motility of spermatozoa or percentage morphologically normal spermatozoa, dogs were treated with oxytocin or prostaglandin F2alpha (PGF2alpha) and compared to saline treatments. Semen was collected from each of the 3 dogs once every 3 to 4 d for a total of 6 collections per dog. Ten minutes before each collection, 1 of 3 injections (oxytocin 10 IU [0.5 mL], IM; PGF2alpha 2.5 mg [0.5 mL], IM; or saline 0.5 mL, IM) was administered. Compared to the saline controls, neither treatment had any significant effect on any measured variable when collected in this manner with an estrus bitch present. Therefore, the use of these drugs does not appear to be a viable treatment to increase the number of spermatozoa.

Premature ejaculation. 1. Definition and etiology.

There is no general agreement on the definition of premature ejaculation, therefore scientific studies often reach discordant results depending on whether they assess the increase in ejaculatory latency or the couple's sexual satisfaction. Etiological theories can be divided into psycho-sexual (anxiety-related, behavioral) and biological (pelvic floor alteration, hypersensitivity of the glans penis, accelerated conduction and cortical amplification of the genital stimuli), both sharing the neurobiological assumption of serotonergic mediation. Premature ejaculation can be iatrogenic (amphetamine, cocaine, dopaminergic drugs) or secondary to urological diseases (prostatovesiculitis, frenulum breve) or to neurological diseases (multiple sclerosis, peripheral neuropathies, medullary expansion processes).

[Effect of experimental classical swine fever (CSF) infection on libido and ejaculate parameters in boars].

The objective of this study was to characterize the effect of an experimental infection with the classical swine fever (CSF) virus on libido and ejaculate parameters of adult boars. Four boars 10 month old were infected with a CSF field isolate (Visbek/Han95). Semen was collected every second day after infection and daily during the pyrexic phase. The only clinical signs in the boars were an increase in body temperature, but never above 39.9 degrees C and a temporally reduction of food intake. The libido was always good, so semen collection was performed in three boars without difficulty and the semen quality was always in the range of the minimum requirements for sperm that is used for artificial insemination. Although one boar had a good libido only a sperm free ejaculate could be collected on one day. The results show that a CSF virus infection of adult boars hardly causes any clinical symptoms and that sperm can be obtained despite fever. Insemination boars may thus be of special epidemiological relevance for the dissemination of the CSF virus.

[Characterization of the mRNA profile in ejaculated spermatozoa from healthy fertile men].

OBJECTIVE: To explore the complexity of mRNA in the ejaculated sperm from healthy fertile men. METHODS: Semen samples were collected from 10 healthy fathers. The swim-up method was adopted to purify the sperm from possible contamination of somatic cells and the spermatozoal total RNA extracted by Trizol was used for SAGE library analysis. RESULTS: A totle of 21 052 SAGE raw tags were sequenced from 877 clones and 2 712 unique tags that occurred at least twice in the library were given further analysis. 19.7% of the unique tags had no match in the existing SAGE map, representing novel genes. Molecular function analysis revealed 67% of unique tags related to protein binding or nucleic acid binding categories, 41% to catalytic activity, 13% to message transducer activity, and 10% to transporter, structural and transcription regulator activity, respectively. CONCLUSION: There exists a complex repertoire of mRNAs in the ejaculated spermatozoa from fertile men.

Levels of cholesterol and phospholipids in freshly ejaculated sperm and Percoll-gradient-pelletted sperm from fertile and unexplained infertile men.

Cholesterol and phospholipid levels were determined in individual sperm samples obtained from 20 fertile and 20 unexplained infertile men. The determination was performed on both washed freshly ejaculated sperm and Percoll-gradient-pelletted sperm. Although sperm cholesterol levels in unexplained infertile patients were significantly lower, i.e., 10.6 +/- 1.3 (mean +/- SD) nmol/10(7) freshly ejaculated sperm and 5.4 +/- 0.7 nmol/10(7) Percoll-gradient-pelletted sperm as compared with 19.9 +/- 1.9 nmol/10(7) and 12.6 +/- 1.5 nmol/10(7) for corresponding sperm populations in fertile donors. Motility parameters measured in 10 sperm samples of the two groups of fertile and unexplained infertile men revealed increases in the amplitude of lateral head displacement and decreases in percent of straightness in sperm tracks from unexplained infertile men.

Successful pregnancy despite retrograde ejaculation. A case report.

In retrograde ejaculation sperm is passed into the bladder causing infertility. Among patients seen at the Reproductive Biology Research Unit of the University of Pretoria retrograde ejaculation is relatively uncommon (0.004%), but is a potentially treatable cause of infertility. The technique for collecting sperm, sperm processing and recovery described was successful and resulted in a pregnancy after transfer of 1 four-cell embryo.

An investigation into the origin of the first fraction of the canine ejaculate.

Histological examination of the reproductive tract of the male dog failed to identify glandular tissue other than the prostate gland and its disseminated portion. A biochemical study of the three fractions of the ejaculate demonstrates similarities between the first and the third fractions. The composition of three ejaculates collected over 12 hours indicated that storage of prostatic secretion was a possible explanation for the biochemical differences observed. It is suggested that both the first and third fractions originate from the prostate gland.

[The function of the closure apparatus of the bladder in retrograde ejaculation in diabetics].

The impairment of sympathetic innervation of the urinary bladder neck was supposed to be a cause of the syndrome of diabetic retrograde ejaculation (DRE). The performance of the sphincter system had not been studied in this category of patients. The liquid profilometric technique was used to examine 3 groups of males: 8 patients with the DRE syndrome; 5 patients with diabetes mellitus (DM) without ejaculation disorders; and 7 apparently healthy subjects. All the examinees showed no organic changes in the prostate and urethra. The groups were matched by age, type, duration and severity of DM. The DRE syndrome patients had no elevation of intraureteral pressure in the area the inner sphincter of the urinary bladder, which evidenced its atony. In health the elevation of vesical pressure is usually accompanied by an increase in ureteral resistance (r = 0.96; p less than 0.001), thus maintaining the stability of the positive pressure gradient and preventing the escape of urine. Correlation analysis revealed alterations of interrelations between intravesical and sphincter pressures in diabetes mellitus patients. This phenomenon evidenced the disorders of somatic innervation of the outer ureteral sphincter more pronounced in patients with the DRE syndrome. No significant differences were revealed in the length of the anatomical and functional ureter as well as in the length of the posterior ureter of all the patients involved in the study.(ABSTRACT TRUNCATED AT 250 WORDS)

[Retrograde ejaculation following a Y-V plasty of the bladder neck carried out during childhood. Treatment using the Abrahams procedure].

Retrograde ejaculation of Y-V plasty of the bladder neck performed during childhood appears to be a rare complication. Based on a case treated with success, the authors stress the need for non-aggressive treatment of bladder neck disease in children (endoscopic radial incisions). The procedure proposed by J.J. Abrahams for the treatment of retrograde ejaculation (simple calibration of the bladder neck and posterior urethra) proves to be reliable, although recurrences may occur.

[Retrograde ejaculation, a little known complication of ectopic ureterocele with ureteral duplication].

A 9 year old boy with an ectopic ureterocele on a left ureteral duplication was treated by radical surgery: nephro-uretero-ureterocelectomy. At 22 years of age he presented with total absence of ejaculation, shown to be due to retrograde ejaculation from permanent gaping of bladder neck. Urinary affections were absent and urologic results of operation were good. The prevention of this disturbing genital complication, and the manner to treat it if it does develop, are discussed.

Effect of flushing the vasa deferentia at the time of vasectomy on the rate of clearance of spermatozoa from the ejaculates of dogs and cats.

Flushing the vasa deferentia (ductus deferentes) at the time of vasectomy reduced to zero the number of intact spermatozoa by postvasectomy day 6 in the dog and by postvasectomy day 7 in the cat and shortened the time from vasectomy to azoospermia in the dog, but not in the cat. The fluid used to flush the vasa deferentia was not eliminated through the penile urethra, but flowed into the urinary bladder, indicating that the least resistant pathway for the exit of vasal content in the anesthetized dog and cat is toward the urinary bladder. Both control and treated dogs and cats had spermatozoa in the urine obtained by cystocentesis immediately after ejaculation or ejaculation and flushing of the vasa deferentia. Flushing the vasa deferentia at the time of vasectomy is easy to do, safe, and can be used in clinical practice to decrease the time from vasectomy to the safe utilization of dogs and cats as teasers. The procedure has potential application to males of other species.

Immunoreactive relaxin-like substance in human split ejaculates.

Immunoreactive relaxin-like substance concentrations were estimated in whole seminal plasma of normozoospermic patients, in seminal fluid of both fractions of split ejaculates, as well as in the seminal plasma of patients affected by excretory azoospermia of various causes. The relaxin concentration was significantly higher in the first fraction of partitioned ejaculates (P less than 0.001). In cases of azoospermia after vasectomy or after epididymal obstruction, the relaxin levels were similar to the values observed in normozoospermia. Relaxin concentrations, however, were high in cases of congenital agenesis of vasa deferentia and seminal vesicles. Relaxin traced in human male seminal plasma seems thus to have an important extragonadic source, probably localized in the prostate.

Studies of human seminal parameters with frequent ejaculation. I. Clinical characteristics.

The intra-subject variation in seminal parameters (e.g. volume; spermatozoal density, motility and normal morphology, and total spermatozoal count) have been examined in 172 men following a set ejaculatory pattern designed to exhaust their extra-gonadal spermatozoal reserve. The results have demonstrated that the motility and normal morphology of spermatozoa do not change significantly with ejaculation frequency. Also, while the volume of the ejaculate and the total number of spermatozoa decreased during a period of frequent coitus, an individual's depletion and recovery of his spermatozoal reserve could not be accurately predicted. It is suggested that coital frequency during the woman's periovulatory period could be modified by the partner's response to this test. Furthermore, while it is acknowledged that the variation in results from a 'routine' semen analysis is large, data obtained after depletion of the extra-gonadal reserve and recovery over a set time may have more value and could be the method of choice for the future.

An assessment of clomipramine (Anafranil) in the treatment of premature ejaculation.

A trial of clomipramine in the treatment of premature ejaculation is described. For the first 4 weeks the drug was administered in a double-blind manner and compared with matching placebo, the two medications being randomly assigned to suitable patients. During the second month patients 'crossed over' to the alternative therapy. Following the completion of the double-blind period all patients received clomipramine for a further 3 months. The drug was administered initially in a dose of 10 mg daily with an option to increase in individual cases to 40 mg. Twenty patients entered the trial, sixteen completed it. During the double-blind study no difference could be shown between clomipramine and placebo in their effect on premature ejaculation. But during the follow-up period nine patients were found to have derived benefit from combined treatment.
