Morphometric analysis of differentiation in human breast carcinoma. Tumor grading.

Subjective grading systems suffer from limited reproducibility and do not assign a sufficiently large proportion of patients to the most predictive grades (1 and 3). In 50 cases of stage 1 or stage 2 mammary ductal carcinoma having at least five-year follow-up, we quantitated the proportion of neoplastic cells showing glandular differentiation (DI) or remaining within ducts (IN). Patients who experienced recurrence had significantly less of both DI and IN, and the point at which 10% of neoplastic cells was composed of DI + IN appeared to be predictive for recurrence; ie, 24 recurrences appeared among the 33 patients who had tumors with DI + IN less than 10%, whereas none developed among the 14 patients having tumors with DI + IN greater than 10%. Only three patients could not be classified with this marker. Tumor stage and grade were independent variables. Reproducibility among three observers averaged 95%. The morphometric evaluation can be performed by a technician under the direction of a pathologist.

Morphometric analysis of differentiation in human breast carcinoma. Tumor heterogeneity.

The assumption that tumor grading is best performed in the region having the least differentiation was tested in a series of mammary ductal carcinomas by quantitating the relative amounts of neoplastic cells that remain within ducts (intraductal tumor [IN]) or participate in glandular formations (differentiated infiltrating tumor [DI]). Field-to-field variation in these two elements displayed a linear correlation with their overall level in each tumor. Since having less than 10% DI + IN is a marker for recurrence among breast cancer patients, this marker was applied to find variant regions of potential biological importance. Such regions were common, but their distribution was random, and they varied so much within themselves that they were not significantly different from the remainder of the tumor. Furthermore, the existence of such poorly differentiated regions did not correlate with a poor prognosis. Grading should be performed on a sample that is representative of the entire tumor, rather than its least differentiated part.

[Use of monoclonal antibodies against the light-chain immunoglobulins of the rat in the radioimmune analysis of differentiated lymphocyte antigens of the mouse].

Spleen cells from SJL mice immunized with rat immunoglobulin light chains were fused with X-63 myeloma. Seven hybridoma lines were obtained. Monoclonal antibodies (McAB) secreted by lines L1G9, L2B2 and L3E8 were purified and labeled by 125I. McAB L1G9 did not bind directly to murine lymphocytes but reacted in a different manner with rat splenocytes. McAb L2B2 and L3E8 but not L1G9 reacted with murine lymphocytes treated with rat McAb against Lyt-1, Lyt-2, C1-9, C4-20 antigens whatever the class of heavy chains of rat McAb used. The binding levels obtained with 125I-McAb reflect the quantitative differences in the expression of respective antigens.

[Morphometric analysis of differentiation of skeletal muscle fibers of the maxillofacial region during the process of embryonic development].

The skeletal muscle tissue of the lip, tongue, soft palate and masticatory muscles have been morphometrically studied in 70 human fetuses (4-10-month-old). In the muscles mentioned decrease in the nuclear volume and in the nuclear-cytoplasmic ratio is observed. Increase of the muscle fibres in the area of transversal sections is especially intensive during last three months. The labial and glossal fibres reach the greatest thickness. On the 6th month and further, there is an essential difference in number of myons with nuclei in the transversal section of the tongue and lips, as compared with the masticatory muscle and soft palate. In 9-10-month-old fetuses the index on saturation of the glossal and labial muscle fasciculi with capillaries is much greater than that of the soft palate and the masticatory muscle. There exists a statistically important and rather high positive correlation between the area of transversal sections of the muscle fibres, the number of nuclei in them and the density of capillaries in the muscle fasciculi. The relation between histochemical profile of the muscles under investigation and the number of capillaries in them is discussed.

Localized bowel distension in the newborn: a review of the plain film analysis and differential diagnosis.

Neonatal abdominal plain films were prospectively analyzed over a 41-month period in order to generate a differential diagnosis for focal bowel distension during the first 6 weeks of life. A total of 31 examples of locally distended bowel were collected. The study discredits the concept that asymmetrically distended bowel in the newborn is usually caused by necrotizing enterocolitis (only 9/31 cases) and indicates that distended bowel in the lower abdomen or left upper quadrant is often caused by air trapping in normal but redundant rectosigmoid (11 cases) or in distal transverse colon (three cases). Five other pathologic conditions not normally specified in differential diagnoses of focally distended bowel included inguinal hernia (three cases), small bowel atresia (two cases), midgut volvulus (one case), Hirschsprung's disease (one case), and small left colon (one case). The study emphasizes the value of sequential supine and prone filming for distinguishing normal from pathologic dilated loops of bowel.

Analysis of differentiation of mouse hemopoietic stem cells in culture by sequential replating of paired progenitors.

Blast cell colonies seen in cultures of spleen cells from 5-fluorouracil-treated mice provide a highly enriched population of primitive hemopoietic progenitors. Our recent studies of the differentiation potentials of the paired daughter cells of these progenitors showed different patterns of differentiation in the colonies produced by the separated daughter cells. In this study, we carried out sequential micromanipulation of paired progenitors followed by cytologic examinations of the colonies derived from these progenitors. Of the total 94 evaluable cultures, consisting of three or more colonies, 52 consisted of macrophage colonies and one consisted of megakaryocyte colonies. In the remaining 41 cultures, diverse combinations of colonies revealing heterogeneous compositions of cell lineages were identified. Presumptive genealogic trees of the differentiation of hemopoietic progenitors constructed for the latter group of cultures suggested that monopotent progenitors may be derived from pluripotent progenitors in two ways: (1) directly during one cell division of pluripotent cells or (2) as a result of progressive lineage restriction during successive division of the pluripotent progenitors. The results also suggested that some of the oligopotent progenitors are capable of limited self-renewal.

Analysis of differentiated T-cell subpopulations: difference in radiosensitivity of immune responses to allogeneic antigen.

The radiosensitivity of immune responses to allogeneic tumor cells was compared. Delayed hypersensitivity as demonstrated by macrophage migration inhibition and antibody production as detected by immune adherence test were extremely radiosensitive. The radiosensitive nature of macrophage migration inhibition was in marked contrast to he radioresistance of the delayed footpad reaction. The generation of cytotoxicity was relatively radioresistant.
