Diacylglycerol kinase epsilon regulates seizure susceptibility and long-term potentiation through arachidonoyl- inositol lipid signaling.

Arachidonoyldiacylglycerol (20:4-DAG) is a second messenger derived from phosphatidylinositol 4,5-bisphosphate and generated by stimulation of glutamate metabotropic receptors linked to G proteins and activation of phospholipase C. 20:4-DAG signaling is terminated by its phosphorylation to phosphatidic acid, catalyzed by diacylglycerol kinase (DGK). We have cloned the murine DGKepsilon gene that showed, when expressed in COS-7 cells, selectivity for 20:4-DAG. The significance of DGKepsilon in synaptic function was investigated in mice with targeted disruption of the DGKepsilon. DGKepsilon(-/-) mice showed a higher resistance to electroconvulsive shock with shorter tonic seizures and faster recovery than DGKepsilon(+/+) mice. The phosphatidylinositol 4,5-bisphosphate-signaling pathway in cerebral cortex was greatly affected, leading to lower accumulation of 20:4-DAG and free 20:4. Also, long-term potentiation was attenuated in perforant path-dentate granular cell synapses. We propose that DGKepsilon contributes to modulate neuronal signaling pathways linked to synaptic activity, neuronal plasticity, and epileptogenesis.

Partial limbic kindling--brain, behavior, and the benzodiazepine receptor.

Partial kindling (PK) of the left perforant path (PP) lastingly increased feline defensiveness. Perforant path PK produced long-term potentiation (LTP) in the amygdalo-ventromedial hypothalamic (AM-VMH) pathways in both hemispheres, and in the ventroamygdalofugal (VAF)-VMH efferents of the amygdala of the left hemisphere. Long-term potentiation paralleled behavioral changes. Perforant path PK did not affect recurrent inhibition in area CA3 of the ventral hippocampus. Long-term potentiation of CA3 EPSP and population spikes appeared, but before behavioral changes. Changes in excitability of the periaqueductal grey also accompanied behavioral changes. After kindling, the benzodiazepine receptor antagonist, flumazenil, reduced defensive response to rats in a drug-dependent manner. Flumazenil also reduced LTP in the AM-VMH pathway, but did not affect LTP in the VAF-VMH pathway. Therefore, flumazenil was acting in the amygdala, and not at the VAF-VMH synapse. Kindling caused flumazenil to act like an agonist on behavior, and in the amygdala, and as an agonist or an inverse agonist in area CA3 depending upon the physiological measure taken.

Perforation of the gastro-intestinal tract by a foreign body. A case report.

Most ingested foreign bodies pass through the gastro-intestinal tract without giving rise to complications. If they become lodged in a narrow segment of the gastro-intestinal tract, perforation may occur. The resulting morbidity depends on the further route of the penetrating object and whether septic sequelae ensue. This article describes an unusual case of foreign body perforation with a protracted clinical course. It emphasises that there are no insurmountable barriers in the pathway of foreign bodies.

Endogenous activation of mu and delta-1 opioid receptors is required for long-term potentiation induction in the lateral perforant path: dependence on GABAergic inhibition.

Opioid peptides costored with glutamate have emerged as powerful regulators of long-term potentiation (LTP) induction in several hippocampal pathways. The objectives of the present study were twofold: (1) to identify which opioid receptor types (mu, delta, or kappa) regulate LTP induction at lateral perforant path-granule cell synapses and (2) to test the hypothesis that endogenous opioids regulate LTP induction via modulation of GABAergic inhibition. LTP of lateral perforant path-evoked field EPSPs was induced selectively by high-frequency stimulation applied to the outer third of the molecular layer of the dentate gyrus of rat hippocampal slices. No changes in medial perforant path responses occurred. LTP was blocked when high-frequency stimulation was applied in the presence of the mu receptor antagonist CTAP, the selective delta-1 receptor antagonist BNTX, or the delta-1 and delta-2 receptor antagonist naltrindole. By contrast, the kappa-1 opioid receptor antagonist NBNI had no effect on LTP induction. The role of GABAergic inhibition was investigated by comparing the effect of naloxone on LTP induction in slices maintained in standard buffer and picrotoxin-containing buffer. Naloxone blocked LTP in standard buffer, whereas normal LTP was induced in picrotoxin-treated, disinhibited slices. Finally, NMDA receptor blockade completely inhibited LTP in both standard and disinhibited slices. The results show that mu and delta-1 opioid receptors regulate LTP induction and that this mechanism critically depends on GABAergic inhibition. A key issue then becomes how endogenous opioids fine-tune the activity of intact inhibitory networks in the dentate gyrus, effectively gating synaptic plasticity in specific dendritic strata.

[Fulguration of atrial flutter in man. A pathological case].

A 63 year old man with ischemic heart disease underwent two sessions of catheter ablation in the inferior right atrium for poorly tolerated resistant and recurrent atrial flutter. After endocavitary mapping and comparison with surface recordings of the f waves, a shock of 50 joules was delivered to the zone situated anteriorly to the inferior vena cava, under the orifice of the coronary sinus and behind the tricuspid valve. Early recurrence of the arrhythmia led to a second attempt and another 50 joules shock was administered to the same area. Another short term recurrence led to definitive nodohisian interruption with a 270 joules shock. Thirty months later, the patient died suddenly during an episode of cardiac failure. Macroscopic examination of the right atrium showed a zone of parietal congestion measuring 4 x 3 cm with a very thin, translucid, central zone measuring 3 x 1.5 cm, just anterior to the inferior vena cava in the right atrial free wall. Histological examination of this zone showed an intense, mutilating fibrosis dissociating the muscular fibres, of the pectinate muscle and even replacing the myocardium in certain regions. In the Eustachian valve, there were muscular fibres, probably representing the posterior internodal pathway, which were also fibrosed. These observations suggest that: 1) in view of the extreme thinness of the atrial wall at the site of ablation there is a high risk of perforation even when right endoatrial catheter ablation is performed with low energy shocks; 2) the posterior internodal pathway does not seem to be an essential component for atrial flutter.

Facilitative effect of carbamazepine on previously induced hippocampal long-term potentiation.

The effects of carbamazepine (CBZ) on previously induced hippocampal long-term potentiation (LTP) were examined. Acute experiments were performed on 33 adult, male rabbits. Field potentials in the dentate gyrus were elicited by single shocks to the perforant path, and LTP was induced by tetanic stimulation to the pathway without induction of seizure discharge. At a CBZ serum level of about 5 micrograms/ml (value +/- SD = 5.40 +/- 1.28 micrograms/ml), the previously induced LTP in population spikes (PSs) and population excitatory postsynaptic potentials (EPSPs) was facilitated. At a CBZ serum level of about 15 micrograms/ml (value +/- SD = 14.28 +/- 1.29 micrograms/ml), the LTP in PS alone was decreased. The effects of carbamazepine on synaptic inhibition were examined by the paired-pulse test. The inhibition was enhanced with induction of LTP. After administration of CBZ, at a CBZ serum level of about 5 micrograms/ml the inhibition was further enhanced, while it was attenuated at a CBZ serum level of about 15 micrograms/ml. These results suggest that CBZ has a facilitative effect on previously induced LTP.

Modulation of Ca2+ channels by cyclic nucleotide cross activation of opposing protein kinases in rabbit portal vein.

Cyclic nucleotides are known to modify voltage-gated (L-type) Ca2+ channel activity in vascular smooth muscle cells, but the exact mechanism(s) underlying these effects is not well defined. The purpose of the present study was to investigate the modulatory role of the cAMP- and cGMP-dependent protein kinase (PKA and PKG, respectively) pathways in Ca2+ channel function by using both conventional and perforated-patch-clamp techniques in rabbit portal vein myocytes. The membrane-permeable cAMP derivative, 8-bromo cAMP (0.1 to 10 micromol/L), significantly increased (14% to 16%) peak Ba2+ currents, whereas higher concentrations (0.05 to 0.1 mmol/L) decreased Ba2+ currents (23% to 31%). In contrast, 8-bromo cGMP inhibited Ba2+ currents at all concentrations tested (0.01 to 1 mmol/L). Basal Ca2+ channel currents were significantly inhibited by the PKA blocker 8-Bromo-2'-O-monobutyryladenosine-3',5'-monophosphorothioate, Rp-isomer (Rp 8-Br-MP cAMPS, 30 micromol/L) and enhanced by the PKG inhibitor beta-Phenyl-1,N2-etheno-8-bromoguanosine-3',5'-monophosphorothioate, Rp-isomer (Rp-8-Br PET cGMPS, 10 nmol/L). In the presence of Rp 8-bromo PET cGMPS (10 to 100 nmol/L), both 8-bromo cAMP (0.1 mmol/L) and 8-bromo cGMP (0.1 mmol/L) enhanced Ba2+ currents (13% to 39%). The excitatory effect of 8-bromo cGMP was blocked by Rp 8-bromo MB-cAMPS. Both 8-bromo cAMP (0.05 mmol/L) and forskolin (10 micromol/L) elicited time-dependent effects, including an initial enhancement followed by suppression of Ba2+ currents. Ba2+ currents were also enhanced when cells were dialyzed with the catalytic subunit of PKA. This effect was reversed by the PKA blocker KT 5720 (200 nmol/L). Our results suggest that cAMP/PKA stimulation enhances and cGMP/PKG stimulation inhibits L-type Ca2+ channel activity in rabbit portal vein myocytes. Our results further suggest that both cAMP and cGMP have a primary action mediated by their own kinase as well as a secondary action mediated by the opposing kinase.

[Infarcts in the region of the middle cerebral artery. Study of 501 cases in 484 patients].

We report a study of 501 infarcts in the middle cerebral artery (MCA) territory, in 484 patients. All cases had an appropriate low-density area on computed tomography. The basal (perforators) territory was involved in half the cases and the motor pathways either in the pre-rolandic area or in the internal capsule in 90%. In all locations, except for watershed infarcts, the main cause was cardiac embolism. Atrial fibrillation accounted for 59%, the two other main causes were myocardial infarction and paradoxical embolism. Atherosclerosis accounted for less than one third of the cases. Among 102 internal carotid artery occlusions less than one half were due to atherosclerosis, cardiac embolism and dissecting aneurysms accounted for 22% each. None of 34 MCA occlusions were due to atherosclerotic thrombotic occlusion. Transient ischemic attacks were recorded in 22% of the cases. Stroke-in-progression with a mean duration of 6-8 hours in hospital, was noted in nearly half the cases. In a small group of MCA infarcts paralysis began and predominated on the lower limb, fifteen contralateral old MCA infarcts were silent. Ten of these were on the right side. The 15 patients with silent infarcts were all right-handed.

Antagonist activity of methyl-substituted analogues of 2-amino-4-phosphonobutanoic acid in the hippocampal slice.

Four monomethyl-substituted analogues of 2-amino-4-phosphonobutanoic acid (APB), an antagonist of excitatory pathways in the central nervous system, were prepared in order to investigate the steric requirements of the APB receptor. Methyl groups were incorporated at the amino, alpha-, beta-, and gamma-positions. The beta- and gamma-methyl-substituted analogues of APB were found to be moderately potent antagonists in excitatory synapses of the hippocampal perforant path, as judged by extracellular recording techniques, while the N- and alpha-methyl-substituted analogues had much lower potencies. All of these APB analogues had very low potencies in the Schaffer collateral pathway. The APB receptors in the perforant path displayed more tolerance of methyl-substitution at the beta- and gamma-positions of APB than at the amino or alpha-positions in this system.

Substance P-induced calcium entry in endothelial cells is secondary to depletion of intracellular stores.

Substance P (SP) induces an elevation in cytosolic Ca2+ concentration ([Ca2+]i) in porcine coronary artery endothelial cells by way of Ca2+ influx and release from intracellular stores. We tested the hypothesis that SP-induced Ca2+ influx occurs due to activation of a Ca(2+)-permeable influx pathway coupled to depletion of intracellular stores. With the use of the perforated patch technique and fura 2 microfluorimetry, a fivefold greater increase in [Ca2+]i per unit decrease in membrane potential was obtained in the presence of SP (10 nM) compared with resting state, implying that SP increased Ca2+ conductance. When K+ channels were blocked, SP activated a net inward current with a reversal potential (2.5 +/- 1 mV) not significantly different from that (2 +/- 1 mV) for inward current recorded in response to store depletion by (2,5-di-tert-butylhydroquinone) (BHQ, 10 microM). Increasing bath [Ca2+] induced a similar shift in reversal potential for SP- and BHQ-induced currents. Inositol 1,4,5-trisphosphate (20 microM), applied through the patch pipette, activated an inward current with Ca2+ selectivity similar to SP- and BHQ-activated currents. Dialysis of cells with heparin (5 mg/ml) completely blocked SP-induced inward current but not BHQ-induced current. These results suggest that the SP-induced increase in Ca2+ conductance can be completely explained by activation of a Ca(2+)-permeable influx pathway coupled to depletion of intracellular stores.

Histamine activates Cl- and K+ currents in guinea-pig tracheal myocytes: convergence with muscarinic signalling pathway.

1. We investigated the effects of histamine on membrane currents and contractile state of isolated guinea-pig tracheal myocytes using perforated patch and whole-cell recording techniques. The effects of histamine were compared to those of acetylcholine (ACh) and caffeine. 2. During voltage clamp (Vhold = -60 mV), histamine elicited contraction and an inward current (Ihist) which was often followed by current oscillations. Ihist had a reversal potential (Vrev) of -9 +/- 3 mV. 3. Ihist was dependent on the Cl- gradient and was antagonized by the Cl- channel blocker niflumic acid. Vrev was more positive (+2 +/- 1 mV) when K(+)-selective currents were blocked by Cs+ and TEA. When all external Na+ was replaced with N-methyl-D-glucamine, there was a small reduction in the amplitude of Ihist. 4. The histamine-induced current was similar to that elicited by ACh and by caffeine with respect to time course, amplitude, and current-voltage relationship. Responses to histamine and to ACh were non-additive, consistent with a convergence of histaminergic and cholinergic signalling pathways. Ihist was antagonized by the H1 histaminergic receptor antagonist astemizole, but not by atropine. 5. When recorded using the perforated patch configuration, Ihist could be elicited repeatedly for more than 30 min. When cells were studied in the whole-cell configuration using a pipette solution containing 0.025 mM EGTA, the amplitude of Ihist was initially the same as that obtained using perforated patch but then decreased; the time required for the responses to decrease to 50% (t1/2) was 8.2 +/- 1.0 min. When 1 mM EGTA was included in the pipette solution (whole-cell configuration), the initial response to histamine was significantly decreased in size and t1/2 was reduced to 3.3 +/- 0.7 min. 6. The characteristics of the signalling pathway were examined in cells studied using the whole-cell configuration with 0.025 mM EGTA in the recording pipette. Heparin significantly reduced t1/2 to 4.3 +/- 0.8 min. GTP gamma S elicited inward current and oscillations; both effects were enhanced by histamine. GTP gamma S also reduced t1/2 to 1.4 +/- 0.1 min. Pertussis toxin did not alter the amplitude or time course of Ihist. 7. We conclude that in guinea-pig tracheal myocytes, binding of histamine to H1 receptors leads to release of Ca2+ from intracellular stores and subsequent activation of Cl- and K+ conductances as well as contraction. Furthermore, we demonstrate that ACh elicits similar physiological responses due to a convergence of the histaminergic and muscarinic signalling pathways.

[Standards in diagnosis of diverticulitis].

The diagnostic procedure is determined by the severity of the diverticulitis. In complicated cases of diverticulitis, it is necessary to detect those patients with obstructive ileus, perforation, and peritonitis who require instant emergency surgery. In all other cases, diagnostic procedures serve as a tool to determine the best therapeutic options. The CT scan of the abdomen seems to have the highest reliability for determining therapy. Other investigations such as barium enema and coloscopy are not indicated in emergency cases or cannot exactly describe the stage of the diverticular disease. Many surgeons still prefer an enema with water-soluble contrast medium in emergency cases to visualize a perforation. CT scans have the same ability to answer this question, but they are more expensive and are not available everywhere at any given time.

Growth hormone-releasing peptide-2 reduces inward rectifying K+ currents via a PKA-cAMP-mediated signalling pathway in ovine somatotropes.

Inward-rectifying potassium (Kir) channels are essential for maintaining the resting membrane potential near the K(+) equilibrium and they are responsible for hyperpolarisation-induced K(+) influx. We characterised the Kir current in primary cultured ovine somatotropes and examined the effect of growth hormone-releasing peptide-2 (GHRP-2) on this current and its related intracellular signalling pathways. The Kir current was, in most cases, isolated using nystatin-perforated patch-clamp techniques. In bath solution containing 5 mM K(+), the Kir current was composed of both transient (fast activated) and delayed (slowly activated) components. An increase in the external K(+) concentration from 5 to 25 mM induced an augmentation of approximately 4-fold in the delayed part of the Kir current and both BaCl(2) and CsCl dose-dependently inhibited this current, confirming the presence of the Kir current in ovine somatotropes. Moreover, this specific effect of high K(+) on the Kir current was only observed in the cells that showed positive staining with anti-growth hormone (GH) antibodies, or in GC cells that belong to a rat somatotrope cell line. Application of GHRP-2 (100 nM) reversibly and significantly reduced the Kir current in bath solutions with 5 or 25 mM K(+) in ovine somatotropes. In addition, we found that the reduction in the Kir current mediated by GHRP-2 was totally abolished by the pretreatments with H89 (1 microM) or Rp-cAMP (100 microM) or by intracellular dialysis of a specific protein kinase A (PKA) inhibitory peptide PKI (10 microM). The specific PKC blocker chelerythrine (1 microM) or inhibitory peptide PKC(19-36) (10 microM) did not show any effects on the GHRP-2-induced decrease in the Kir current. These results suggest that the inhibition of Kir current through PKA-cAMP pathways may play an integral role in GHRP-2-induced depolarisation and GH release in ovine somatotropes.

Trafficking of sulfated glycoprotein-1 (prosaposin) to lysosomes or to the extracellular space in rat Sertoli cells.

Sulfated glycoprotein-1 (prosaposin) exists in 2 forms: a 65kDa form targeted to lysosomes and a 70kDa form secreted extracellularly. In order to understand the sorting and targeting mechanisms of the two forms of SGP-1, we have compared their maturation, processing, and secretion in rat Sertoli cells in vivo. Metabolic labeling experiments in vivo demonstrated that the 65kDa form is synthesized first, then post-translationally modified to the 70kDa form of SGP-1. Subcellular fractionation of testicular homogenate was used to obtain Golgi fractions containing up to 50-fold enrichment in galactosyltransferase. Permeabilization of enriched Golgi fractions with saponin released the 70kDa form, but did not affect the 65kDa protein. While excess free mannose 6-phosphate did not release lysosomal SGP-1, it released the 35kDa cathepsin L from Golgi membranes. Using quantitative electron-microscopic immunocytochemistry, the lysosomal contents of SGP-1 were shown to increase significantly after the administration of tunicamycin in vivo. Therefore, the trafficking of the 65kDa form of SGP-1 to the lysosomes appears to be independent of the M6P-receptor pathway. The 70kDa form of SGP-1 was found to aggregate within perforated Golgi fractions in a process which depends on low pH and calcium ions. We conclude that the targeting of the 65kDa form of SGP-1 to the lysosomes involves an early association with Golgi membrane that is independent of mannose 6-phosphate receptors.

Orotate improves memory and enhances synaptic long-term potentiation in active avoidance behaviour in rats with perforant path stimulation as the conditioned stimulus.

Male Wistar rats were trained in an active avoidance task with stimulation of the perforant path with impulse trains of 15 Hz as the conditioning stimulus. Immediately after the first training session, methylglucamine orotate (225 micrograms), a memory improving drug, was injected intraventricularly. The retention of the learned behaviour was determined on the following day in a relearning session. Field potentials evoked in the dentate area by test stimuli from the perforant path electrode were recorded at different times after learning and relearning sessions to determine whether there were functional changes in the perforant path-granular cell synapses, which are involved in the conditioning pathway. Untreated control animals exhibited a so-called 'postconditioning potentiation', expressed as a long-lasting increase of both the excitatory postsynaptic potential (EPSP) and the population spike of the granular cells of the evoked test potentials. This finding reproduces previously published results. Methylglucamine orotate-treated rats showed significantly more conditioned reactions in the relearning sessions compared with untreated controls and a significantly more pronounced potentiation of the population spike, whereas the postconditioning potentiation of the field EPSP remained unaffected by the treatment. When both the control animals and the methylglucamine orotate-treated rats were divided into subgroups of good and poor learners according to their learning scores from the first training session, differences between the effect of the drug became evident. In good learners, the treatment with methylglucamine orotate after the learning session slightly, but significantly, improved retention compared with that of untreated good learners.(ABSTRACT TRUNCATED AT 250 WORDS)

Leukocyte glycolysis and lactate output in animal sepsis and ex vivo human blood.

Lactate is released in large quantity from sites of sepsis and inflammation. We asked whether the increased lactate production found in sepsis can be explained by the augmented glycolysis of inflammatory cells. The glycolytic metabolism of rat peritoneal leukocytes was measured following cecal ligation and perforation (CLP) or sham laparotomy. CLP augmented glucose uptake, the pentose phosphate pathway, and glucose oxidation. Lactate output increased from 1.03 +/- 0.05 to 1.20 +/- 0.05 fmol x cell(-1) x min(-1) (P < .001). Total lactate output of peritoneal lavage fluid increased from 7.94 +/- 2.59 to 28.12 +/- 5.60 nmol L x min(-1) (P < .005). The effect of lipopolysaccharide (LPS) on the lactate output of whole blood from 31 critically ill patients was measured. Leukocyte lactate production was calculated by multiple linear regression analysis. Following exposure to LPS, human leukocyte lactate output increased from 0.20 +/- 0.09 to 1.22 +/- 0.14 fmol x cell(-1) x min(-1) (P < .001). This rate of production is so high that it suggests that the lactate output of different tissue beds in sepsis may be affected by their different cell populations and state of activation. This study supports the hypothesis that lactate may be more a product of inflammation than a marker of tissue hypoxia in sepsis.

Extracellular matrix molecules and synaptic plasticity: immunomapping of intracellular and secreted Reelin in the adult rat brain.

Reelin, a large extracellular matrix glycoprotein, is secreted by several neuron populations in the developing and adult rodent brain. Secreted Reelin triggers a complex signaling pathway by binding lipoprotein and integrin membrane receptors in target cells. Reelin signaling regulates migration and dendritic growth in developing neurons, while it can modulate synaptic plasticity in adult neurons. To identify which adult neural circuits can be modulated by Reelin-mediated signaling, we systematically mapped the distribution of Reelin in adult rat brain using sensitive immunolabeling techniques. Results show that the distribution of intracellular and secreted Reelin is both very widespread and specific. Some interneuron and projection neuron populations in the cerebral cortex contain Reelin. Numerous striatal neurons are weakly immunoreactive for Reelin and these cells are preferentially located in striosomes. Some thalamic nuclei contain Reelin-immunoreactive cells. Double-immunolabeling for GABA and Reelin reveals that the Reelin-immunoreactive cells in the visual thalamus are the intrinsic thalamic interneurons. High local concentrations of extracellular Reelin selectively outline several dendrite spine-rich neuropils. Together with previous mRNA data, our observations suggest abundant axoplasmic transport and secretion in pathways such as the retino-collicular tract, the entorhino-hippocampal ('perforant') path, the lateral olfactory tract or the parallel fiber system of the cerebellum. A preferential secretion of Reelin in these neuropils is consistent with reports of rapid, activity-induced structural changes in adult brain circuits.

Calcium and long-term transmission damage following anoxia in dentate gyrus and CA1 regions of the rat hippocampal slice.

The mechanism of long-term anoxic damage in brain tissue is investigated using the rat hippocampal slice as a model system. The effects of short durations of anoxia on subsequent transmission through two neural pathways are studied. 10 min of anoxia irreversibly abolishes transmission between the perforant path and the dentate granule cells while only 7 min of anoxia irreversibly abolishes transmission between the Schaeffer collaterals and the CA1 pyramidal cells. We examine the involvement of Ca2+ in this irreversible transmission damage and, also, the differential sensitivities of the dentate gyrus and CA1 regions. Substitution of a buffer containing 0 Ca2+ and 10 mM-Mg2+ during the anoxic period substantially improves the recovery of synaptic transmission in both regions of the slice. Dentate gyrus transmission recovers completely after 20 min of anoxia and CA1 transmission survives 10 min of anoxia. These results suggest that Ca2+ influx during anoxia may be an important cause of the long-term damage. The uptake of 45Ca2+ into the intracellular space of the slice is increased during anoxia. This effect is approximately twice as large in CA1 as in the dentate gyrus. Thus, in the dentate gyrus the calculated exchangeable pool of Ca2+ is increased 30% by anoxia and in the CA1 it is increased by 70%. Two incubating conditions which decrease the amount of 45Ca2+ uptake during anoxia protect transmission against long-term damage. (a) Pre-incubation of the slices with 25 mM-creatine elevates tissue phosphocreatine and attenuates the fall in adenosine 5'-triphosphate (ATP) during anoxia. This is associated with partial protection against transmission damage and an approximate 50% attenuation of the anoxic uptake of 45Ca2+. (b) Inclusion of 2 mM-cobalt in the buffer reduces the normoxic uptake of 45Ca2+ so that the uptake during anoxia is no greater than normoxic uptake in the absence of cobalt. This is associated with a complete protection against long-term transmission damage following 10 min of anoxia in the dentate gyrus. A kinetic analysis of the 45Ca2+ uptake shows that the anoxic uptake results primarily from inhibition of the unidirectional efflux of Ca2+ from the cells; there is no calculable increase in the undirectional influx. This suggests that anoxia increases Ca2+ uptake by inhibiting one or more Ca2+-extrusion processes and not by opening depolarization-sensitive Ca2+ channels.(ABSTRACT TRUNCATED AT 400 WORDS)
