Fate of Irgarol 1051, diuron and their main metabolites in two UK marine systems after restrictions in antifouling paints.

Two major antifouling biocides used worldwide, Irgarol 1051 and diuron, and their degradation products in Shoreham Harbour and Brighton Marina, UK were studied during 2003-2004. The highest concentrations of Irgarol 1051 were 136 and 102 ng L(-1) in water and 40 and 49 ng g(-1) dry weight in sediments for Shoreham Harbour and Brighton Marina, respectively. As the degradation product of Irgarol 1051, M1 was also widespread, with the highest concentration of 59 ng L(-1) in water and 23 ng g(-1) in sediments in Shoreham Harbour, and 37 ng L(-1) in water and 5.6 ng g(-1) in sediments in Brighton Marina. The target compounds showed enhanced concentrations during the boating season (May-July), when boats were being re-painted (January-February), and where the density of pleasure crafts was high. Overall, the concentration of Irgarol 1051 decreased significantly from late 2000 to early 2004, indicating the effectiveness of controlling its concentrations in the marine environment following restricted use. Diuron was only detected in 14% of water samples, and mostly absent from sediment samples.

Copper emissions from antifouling paint on recreational vessels.

Trace metals, especially copper, are commonly occurring contaminants in harbors and marinas. One source of copper to these environs is copper-based antifouling coatings used on vessel hulls. The objective of this study was to measure dissolved copper contributions from recreational vessel antifouling coatings for both passive leaching and hull cleaning activities. To accomplish this goal, three coating formulations, including hard vinyl, modified epoxy and a biocide free bottom paint were applied on fiberglass panels and placed in a harbor environment. In situ measurements of passive leaching were made using a recirculating dome system. Monthly average flux rates of dissolved copper for the hard vinyl and modified epoxy coatings were 3.7 and 4.3 microg/cm(2)/day, respectively, while flux rates for the biocide free coating was 0.2 microg/cm(2)/day. The highest passive flux rates were measured initially after cleaning activities, rapidly decreasing to a baseline rate within three days, regardless of copper-based coating formulation. Hull cleaning activities generated between 8.6 and 3.8 microg dissolved copper/cm(2)/event for the modified epoxy and hard vinyl coatings, respectively. Aggressive cleaning using an abrasive product doubled the copper emissions from the modified epoxy coating, but produced virtually no change in the much tougher hard vinyl coating. When compared on a mass basis, roughly 95% of copper is emitted during passive leaching compared to hull cleaning activities over a monthly time period for a typical 9.1 m power boat.

A study on the treatment of antifouling paint waste from shipyard.

A study on the treatment of antifouling paint waste from shipyards, including sandblast waste and ship hull washing wastewater, was performed. The sandblast waste could be effectively detoxified by heat treatment, and the efficiency was affected by the temperature of the heating vessel and treatment time. The removal efficiency of total organotin compounds from the sandblast waste was over 99% at 1000 degrees C and treatment for 1 h. For the treatment of ship hull washing wastewater by the solvent extraction, ship diesel was a good solvent for the tributyltin (TBT) extraction, and the proper amount of solvent was about 10 mL for TBT extraction from 1L of wastewater. The extraction efficiency of TBT was significantly affected by the agitation intensity. The TBT in the wash wastewater was rapidly extracted within 1 h. The level of the TBT residual in the wastewater extracted for 1h was 2.8 microg L(-1), and this was further decreased to 0.8 after 5 h extraction.

The effects of exposure to different clastogens on the pattern of chromosomal aberrations detected by FISH whole chromosome painting in occupationally exposed individuals.

The pattern of chromosomal aberrations (CA) was studied by fluorescence in situ hybridization (FISH) technique (whole chromosomes #1 and #4 painting) in workers occupationally exposed to any of the four following conditions: acrylonitrile (ACN), ethyl benzene (EB), carcinogenic polycyclic aromatic hydrocarbons (c-PAHs), and irradiation in nuclear power plants (NPP), respectively. Decrease in the relative frequency of translocations was observed in EB group, and an increase in reciprocal translocations in ACN and NPP-exposed groups. An increase in a relative number of insertions was registered under all four conditions (significant at ACN, EB, c-PAHs, quasisignificant at NPP-exposed groups). Significant differences in the percentage of lymphocytes with aberrations on chromosome #1 (58.8+/-32.7%, versus 73.8+/-33.6% in the controls, P < 0.05), and chromosome #4 (47.0+/-34.1%, versus 29.4+/-32.2%, P < 0.01) were found in workers exposed to ACN. Similarly, a decrease in the proportion of cells with aberration on chromosome #1 (61.0+/-24.0%, versus 73.8+/-33.6%, P < 0.05) and an increase on chromosome #4 (45.6+/-24.6%, versus 29.4+/-32.2%, P < 0.05) were observed in workers exposed to EB. Frequency of aberrant cells (%AB.C.) as well as genomic frequency of translocations (F(G)/100) increased with age (P < 0.001). Aging also increased the percentage of translocations and reciprocal translocations (P < 0.05), but decreased the relative number of acentric fragments (P < 0.01). Smoking led to significantly increased F(G)/100 (P < 0.05), but did not affect the pattern of chromosomal aberrations. Our results seem to indicate that different carcinogens may induce a different pattern of chromosomal aberrations.

Microdissection and reverse painting reveals a microdeletion 6(q26qter) in a de novo r(6) chromosome.

Ring chromosomes 6 are rare constitutional abnormalities with inconsistent phenotypic and clinical features. One of the reasons for this variability is the cytogenetically undetectable loss of chromosomal material from the telomeric segments at 6p or 6q. We have therefore used fluorescence in situ hybridization (FISH) to analyse a ring chromosome 6 that was detected in a newborn boy with dysmorphic features. Reverse painting of the microdissected ring chromosome onto normal metaphase spreads revealed a small deletion of the terminal region of the long arm, 6(q26qter). Moreover, the simple all-telomeric sequence (TTAGG)n was lost, whereas the p-specific subtelomeric sequence was still present. Our findings confirm that microdeletions occur during the formation of r(6) chromosomes and, therefore, are an important determinator of the associated phenotype.

On the use of computational fluid dynamics in the prediction and control of exposure to airborne contaminants-an illustration using spray painting.

Computational fluid dynamics (CFD) is employed to simulate breathing-zone concentration for a simple representation of spray painting a flat plate in a cross-flow ventilated booth. The results demonstrate the capability of CFD to track correctly changes in breathing-zone concentration associated with work practices shown previously to be significant in determining exposure. Empirical data, and models verified through field studies, are used to examine the predictive capability of these simulations and to identify important issues in the conduct of such comparisons. A commercially available CFD package is used to solve a three-dimensional turbulent flow problem for the velocity field, and to subsequently generate particle trajectories for polydisperse aerosols. An in-house algorithm is developed to convert the trajectory data to breathing-zone concentrations, transfer efficiencies and aerosol size distributions. The mesh size, time step, duration of the simulation, and number of particles per size interval are all important variables in achieving convergent results.

Hemispheric memory for surrealistic versus realistic paintings.

The issue of hemispheric processing of art works, either alone or in relation to a certain aspect of language, was investigated in normal subjects. Three experiments were performed. In the first, memory for surrealistic versus realistic pictures was investigated. In the second, memory for metaphoric versus literal titles of these pictures was measured. In the third, memory for the paintings was determined as a function of the same titles. The results of the first experiment showed a right visual field (RVF) advantage for the surrealistic pictures. No field difference emerged for the realistic pictures. The results of the second experiment indicated a RVF advantage in memory for metaphoric titles. Moreover, in the RVF, there was an advantage for titles from surrealistic-metaphoric pairs over all other pairings. Results of experiment three showed a RVF advantage in remembering pictures from surrealistic-metaphoric pairs and, in the left visual field (LVF) there was advantage for pictures with literal titles. Taken together, the results suggest left hemisphere advantage in processing meaningful, yet incongruous arrays, both pictorial and linguistic. The results are discussed in terms of hemispheric memory for art works, metaphors, and the relationship between the two in the brain.

Determination of some aromatic amines in finger-paints for children's use by supercritical fluid extraction combined with gas chromatography.

A test method based on supercritical fluid extraction (SFE) and gas chromatography has been developed for some aromatic amines, such as 4-chloro-o-toluidine, beta-naphthylamine and 4-aminobiphenyl. A two-level factor design was used as the optimization procedure. Four main variables were considered: CO2 pressure, extraction temperature, static extraction time and volume of modifier (methanol). Results obtained for 4-chloro-o-toluidine, indicated that the volume of modifier was the variable with the most important influence on extraction, CO2 pressure had a negative effect and temperature and time were less significant. For the other amines, static time was the most important variable in both cases, followed by CO2 pressure and volume of modifier, with no influence of temperature. SFE was compared with Soxhlet extraction, and was found to give higher recoveries in all cases. Other commercial finger-paints were tested for the presence of aromatic amines.

Translocation analysis by the FISH-painting method for retrospective dose reconstruction in individuals exposed to ionizing radiation 10 years after exposure.

Fluorescence in situ hybridization (FISH) is a powerful method largely used for detecting chromosomal rearrangements, translocations in particular, which are important biomarkers for dose assessment in case of human exposure to ionizing radiation. To test the possibility of using the translocation analysis by FISH-painting method in retrospective dose assessment, we carried out in vitro experiments in irradiated human lymphocytes, in parallel with the analysis of translocations in lymphocytes from 10 individuals, who were exposed to 137cesium in the Goiânia (Brazil) accident (samples collected 10 years after exposure). The in vitro dose-response curve for the genomic translocation frequencies (FGs) fits a linear quadratic model, according to the equation: Y=0.0243X(2)+0.0556X. The FG values were also calculated for the individuals exposed to 137cesium, ranging from 0.58 to 5.91 per 100 cells, and the doses were estimated and compared with the results obtained by dicentric analysis soon after the accident, taking the opportunity to test the validity of translocation analysis in retrospective biodosimetry. A tentative of retrospective dosimetry was performed, indicating that the method is feasible only for low level exposure (below 0.5Gy), while for higher doses there is a need to apply appropriate correction factors, which take into consideration mainly the persistence of chromosomal translocations along with time, and the influence of endogenous and exogenous factors determining the inter-individual variability in the cellular responses to radiation.

Antifouling paint booster biocide contamination in Greek marine sediments.

Organic booster biocides were recently introduced as alternatives to organotin compounds in antifouling products, after restrictions imposed on the use of tributyltin in 1987. In this study, the concentrations of three biocides commonly used as antifoulants, Irgarol 1051 (2-methylthio-4-tertiary-butylamino-6-cyclopropylamino-s-triazine), dichlofluanid (N-dichlorofluoromethylthio-N',N'-dimethyl-N-phenyl sulphamide) and chlorothalonil (2,4,5,6-tetrachloro isophthalonitrile) were determined in sediments from ports and marinas of Greece. Piraeus (Central port, Mikrolimano and Pasalimani marinas), Thessaloniki (Central port and marina), Patras (Central port and marina), Elefsina, Igoumenitsa, Aktio and Chalkida marinas were chosen as representative study sites for comparison with previous monitoring surveys of biocides in coastal sediments from other European countries. Samples were collected at the end of one boating season (October 1999), as well before and during the 2000 boating season. All the compounds monitored were detected at most of sites and seasonal dependence of biocide concentrations were found, with maxima during the period June-September, while the winter period (December-February) lower values were encountered. The concentrations levels ranged from 3 to 690 ng/g dw (dry weight). Highest levels of the biocides were found in marinas (690, 195 and 165 ng/g dw, for Irgarol, dichlofluanid and chlorothalonil respectively) while in ports lower concentrations were observed. Antifouling paints are implicated as the likely sources of biocides since agricultural applications possibly contributed for chlorothalonil and dichlofluanid inputs in a few sampling sites.

Cytogenetic characterization of two colon cell lines by using conventional G-banding, comparative genomic hybridization, and whole chromosome painting.

The heterogeneous nature of genetic alterations in cancer cells handicaps the full characterization of its occurrence and the analysis of their molecular bases and relation to biological processes. Although many cancer cells are highly aneuploid, in other cases, as in a subset of colorectal carcinomas displaying microsatellite instability, chromosomal aberrations are scarce. The aim of this study was to fully characterize both qualitatively and quantitatively, the karyotypes of two established colon carcinoma cell lines (LoVo and HCT 116) previously reported as being near diploid. An array of complementary cytogenetic techniques were used: G-banding, comparative genome hybridization (CGH), and whole-chromosome painting (WCP). Combinations of these techniques provided an accurate karyotype for the two cell lines: LoVo cells showed 49,XY,t(2;12)(q13;p11.2),+5,+7,+12,i(15)(q10) and HCT 116 cells showed 45,X,-Y,dup(10)(q24q26),der(16)t(8;16)(q13;p13), der(18)t(17;18)(q21;p11.3). Heterogeneity was also observed in both cell lines as shown by G-banding. Chromosomal unbalances determined by CGH (many of them related to structural reorganizations) were characterized by WCP, allowing the reliable identification of those chromosome markers that could not be completely identified by G-banding. We show that combined analysis with classical and molecular cytogenetic techniques provides an accurate map of chromosomal aberrations in these two cell lines not identified in previous investigations.

A comprehensive karyotypic analysis on a newly established sarcomatoid hepatocellular carcinoma cell line SH-J1 by comparative genomic hybridization and chromosome painting.

We first established a sarcomatoid hepatocellular carcinoma cell line, designated as SH-J1, and applied comparative genomic hybridization and fluorescence in situ hybridization (FISH) with chromosome painting probes for the characterization of the chromosomal rearrangements. In the SH-J1 cell line, the pleomorphic spindle cells were arranged in bundles of interlacing patterns and were positive in immunohistochemical staining with hepatocyte-related markers. By G-banding and FISH, the chromosomal gains were detected at 6p and 17, whereas losses were observed at 3p21-pter, 3q27-qter, 4, 6q, 13pter-q11, 16, 18, 19p13, and Y.

Detection of chromosome 6 abnormalities in melanoma cell lines by chromosome arm painting probes.

Chromosome 6 abnormalities, particularly the deletion of 6q, are among the most frequent chromosomal changes in human malignant melanoma. In this study, chromosome 6 rearrangements in 21 melanoma cell lines were identified by fluorescence in situ hybridization (FISH) with the use of chromosome 6 arm specific painting probes (CAPs). Structural abnormalities of chromosome 6 were detectable in 18 of 21 (86%) cases, including the identification of structural abnormalities that were not detectable by routine G-banding analysis. The results of this study correlate the frequency of 6q alterations and demonstrate that FISH with CAPs can be of considerable use for rapid screening of specific chromosome abnormalities in melanoma.

Posterior capsule rupture after a paint-pellet injury.

We report a case in which a high-velocity paint pellet injury to the eye ruptured the posterior lens capsule. In the setting of blunt trauma, posterior capsule rupture has rarely been reported. Recognizing such a rupture allows the surgeon to plan the best surgical approach.

Reciprocal chromosome painting shows that squirrels, unlike murid rodents, have a highly conserved genome organization.

We present the first report of reciprocal chromosome painting between humans and a rodent. Gene mapping and sequencing data lead to the generalization that rodent genomes are highly rearranged. In contrast, our results show a surprising conservation of genome structure between humans and squirrels. The synteny of 12 human chromosomes was entirely conserved (5, 6, 9, 11, 13-15, 17, 18, 20, 21, and X). Of the 12 syntenic associations of human chromosomes present in the squirrel, six are well-known ancestral eutherian associations (3/21, 4/8, 7/16, 12/22, 14/15, 16/19). Apparently, few derived translocations characterize the evolutionary origin of the rodents. One association (10p/1qter) may be a cladistic marker for the cohort Glires, linking rodents and lagomorphs.

Chromosome painting for cytogenetic monitoring of occupationally exposed and non-exposed groups of human individuals.

The suitability of a three-color fluorescence in situ suppression hybridization technique was examined for monitoring five different groups of individuals: 30 occupied in radiology, 26 occupied in nuclear medicine or radiation physics, 32 patients with breast cancer, 26 occupied with military waste disposal, all presumably exposed to low doses of radiation or chemical mutagens and a non-exposed control group (N=29). The average frequency of breaks constituting the various aberrations did not significantly differ between the groups of medical radiation appliers and the control group. However, breast tumor patients and military waste disposers, as groups, showed a higher aberration rate than did healthy controls. Stable rearrangements mainly characterized the groups of controls, tumor patients, and radiation appliers, while a higher proportion of unstable aberrations was found in the chemically exposed individuals. Individuals with an increased frequency of aberrations could be detected within each examined group, which clearly determined the average values of the whole group. With respect to interchromosomal distribution of the breakpoints constituting the found aberrations and the involvement of the labeled chromosomes in rearrangements, the observed values were very close to the expected ones in the controls. A rather similar trend of deviations from expectation was observed in all other groups. Chromosome 4 was slightly over-affected, while chromosome 2 was slightly underrepresented in all analyzed groups (except tumor patients). Rearrangements of the labeled chromosomes with the unlabeled ones exceeded expectation. In conclusion, chromosome painting if included in further attempts of human population monitoring will broaden the basis of argumentation with respect to health risks introduced by mutagen exposure.

Construction of chromosome-specific paints for meta- and submetacentric autosomes and the sex chromosomes in the horse and their use to detect homologous chromosomal segments in the donkey.

A pilot study comparing horse and donkey karyotypes on a molecular basis was initiated using the chromosomal microdissection approach. All equine meta- and submetacentric chromosomes, viz. ECA1 to ECA13 and the X and Y chromosomes, were microdissected. The DNA was PCR amplified, non-radioactively labelled and used as probes on equine metaphase chromosomes to confirm their origin. Once tested, the paints were used as probes on donkey metaphase chromosomes to detect homologous chromosomal segments between the two species. The results not only detected conservation of whole chromosome and/or arm synteny between the two karyotypes, but also highlighted varying degrees of rearrangements. The findings also enable deduction of homology between parts of donkey and human karyotypes. In light of the molecular evidence, this study examines the accuracy of the available comparative cytogenetic data between horse and donkey.

Karyotype relationships between four distantly related marsupials revealed by reciprocal chromosome painting.

Marsupial karyotypes have shown extensive conservation even between distantly related groups with a high diversity of life forms and reproductive biology. Banding analysis has been the main test for assessing their homologies and chromosome rearrangements. More recently, cross-species reciprocal chromosome painting has been developed and applied to several mammalian species and has shown homologies and rearrangements not revealed by banding analysis. Karyotype relationships between four marsupial species, Sminthopsis crassicaudata, Potorous tridactylus, Trichosurus vulpecula and Macropus eugenii, which are from different families in two orders, were investigated and presented in the form of comparative chromosome maps. These show that only a limited number of chromosomal rearrangements have occurred during their evolution. A karyotype phylogeny of the four marsupials was derived from these maps. A comparison between published gene location and the comparative chromosome maps for these species is presented and inconsistencies with previous gene mapping data indicated.
