Gas/particle partitioning of seven volatile polycyclic aromatic hydrocarbons in a heavy traffic urban area.

Air samples (vapor- and particle-phase) were taken for 19 sampling events during the period from December 1997 to July 1998 in an urban site in the center of Athens. The urban site is densely populated and characterized by heavy traffic circulation and elevated concentrations of VOCs, NO(x), CO and smoke. Seven volatile polycyclic aromatic hydrocarbons (PAHs) were determined in samples. The temperature dependence of gas-phase atmospheric concentration of PAHs, C(g), was investigated using diagrams of natural logarithm of partial pressures (lnP) vs. reciprocal mid-point temperatures. For the six of seven volatile PAHs, the temperature dependence of lnP was statistically significant (at least at the 90% confidence level) and the temperature accounted for 21-67% of the variability in gas-phase concentrations. The gas-phase concentration C(g) of the very volatile PAHs was affected more significantly by changes in temperature, but the variation of the less-volatile PAHs fluoranthene and pyrene C(g), was better explained by changes in temperature. The temperature dependence of gas/particle partitioning constant K(P) was also examined. Regressions of log(K(P))(-1) vs. T(-1) for fluorene, fluoranthene and pyrene were classified into two different temperature ranges. The gas/particle partitioning of PAHs was studied by correlating the partition constant to the sub-cooled liquid saturation vapor pressure (P(L)(o)). The Junge adsorption model underestimated the particle fraction of volatile PAHs probably due to the presence of non-exchangeable fraction. Slopes (m(r)) of the regressions logK(P) vs. logP(L)(o) were different from the value -1 as Pankow's theory predicts. The short distance between the sampling point and the emission sources is also estimated to be a factor that causes deviations from the theoretical value. Evidence that atmospheric conditions favorable for secondary aerosol formation coincide with higher value of m(r), was provided by limited sampling events. An interrelation was found to exist between the m(r) values, allowing the prediction of the gas/particle partitioning of a series of seven PAHs by the measurement of a single PAH partitioning.

A combined chemical and bioassay analysis of traffic-emitted polycyclic aromatic hydrocarbons.

The objective of this study was to determine the concentrations of an extended series of polycyclic aromatic hydrocarbons (PAHs) and nitrated PAHs in outdoor air samples collected in low-contaminated urban areas, affected mainly by traffic emissions, and to estimate in vitro mutagenic and dioxin-like toxicity of extracts from these samples. Data on concentrations of PAHs and toxic in vitro potencies were compared in extracts obtained by different sampling methods. PAHs and their derivatives were analysed by high performance liquid chromatography with diode array and fluorescence detection, as well as gas chromatography with mass spectrometry. The total sum of 39 PAHs under study ranged from 6.7 to 62.7 ng.m(-3); of this, the sum of 16 U.S. EPA priority PAHs in urban air samples ranged from 3.2 to 6.2 ng.m(-3). Phenanthrene was the prevalent PAH in all air samples tested, with concentrations up to 17.6 ng.m(-3), followed by fluorene, fluoranthene and pyrene present mostly in the gaseous phase. Also, other low molecular weight PAHs (with MW up to 228) were distributed mostly in gaseous phase. The particulate phase contained mostly carcinogenic PAHs, among which, benzo[a]pyrene, indeno[1,2,3-cd]pyrene and benzofluoranthenes were predominant compounds (with benzo[a]pyrene reaching levels up to 1.57 ng.m(-3)). Traffic emissions were confirmed as the major source of PAHs in the airborne samples due to the presence of elevated concentrations of benzo[ghi]perylene and coronene. The most abundant nitrated PAH derivatives were nitronaphthalenes, which were present exclusively in the vapor phase; 9-nitroanthracene, 9-nitrophenantrene and 3-nitrofluoranthene were associated mostly with particulate matter (PM(10)). Bioassays for detection of the Ah receptor-mediated activity and mutagenicity in vitro were used as a screen of potential adverse effects of air pollutants emitted from traffic. The major part of mutagenic and aryl hydrocarbon receptor (AhR)-mediated activities was found to be present in the PM(10) fraction. Although the PM(10) sampling technique was found to be a suitable method regarding the subsequent determination of mutagenic and AhR-mediated activities in vitro, relative toxic potencies, associated with low molecular weight PAHs (such as tumor promotion and other adverse effects), could be underestimated.

Polycyclic aromatic hydrocarbons in dustfall in Tianjin, China.

Atmospheric dustfall samples from 23 locations in Tianjin, China, were collected and analyzed for 16 polycyclic aromatic hydrocarbons (PAHs) classified by the Environmental Protection Agency as priority pollutants from March 2002 to March 2003. SigmaPAH16 (sum of 16 PAH compounds) concentrations in the dustfall collected during heating season ranged from 2.5 to 85.5 mug/g, while that during the non-heating season varied from 1.0 to 48.2 microg/g dry weight. The dominant components in the heating season included naphthalene, phenanthrene, fluoranthene, and chrysene, while naphthalene, fluorene, phenanthrene, and fluoranthene were dominant during the non-heating season. Compared with the non-heating season, the heating season was characterized by a higher fraction of high-molecular-weight PAHs with four to six rings with exception of the samples from the east industrial area. The east industrial area had more significant correlations between individual PAH compounds, and more discrete triangular components of three-, four-, five- and six-ring PAHs. No significant correlations were observed between the PAHs concentrations and total organic carbon (TOC) in the dustfall samples. The deposition fluxes of sigmaPAH15 (sum of 15 PAHs except naphthalene), sigmaPAH6 (sum of 6 carcinogenic PAHs recommended by IARC) and benzo[a]pyrene (BaP) from atmospheric deposition to the whole area were estimated as 1911, 196, and 53 microg/m2/year, respectively. The deposition rates for PAH compounds in the east industrial area were higher than those in the urban and rural areas. Furthermore, the deposition contribution of PAHs during domestic heating season in winter was not significant relative to the annual inputs.

Gradient distribution of persistent organic contaminants along northern slope of central-Himalayas, China.

High mountains may serve as condenser for persistent organic pollutants (POPs) and the vegetation in remote areas has been used as a means to characterized atmospheric concentrations of air pollutants. In this study, organochlorine pesticides (OCPs) and polycyclic aromatic hydrocarbons (PAHs) in Himalayan spruce needle samples from Zhangmu-Nyalam region (central-Himalayas) were analyzed and the altitudinal gradient of these pollutants was investigated. Total HCHs and DDTs concentration in needles were in the range of 1.3-2.9 ng g(-1) dry weight and 1.7-11 ng g(-1) dry weight, which were lower than concentrations reported in spruce needles from Alps, however higher than concentrations in conifer needles from mountain areas of Alberta. Total Himalayan spruce needle PAHs was below 600 ng g(-1) and fluorene, phenanthrene and acenaphthene were abundant individual compounds measured. The ratios of alpha-HCH/gamma-HCH in pine needles were similar with the usual values for technical HCH, implying technical HCHs might be used in this region. The high ratios of o-p'-DDT/p-p'-DDT and no p-p'-DDE measured in this study led to the suspicion that a new source of o-p'-DDT and/or p-p'-DDT existed in this region. In addition, higher ratios of low molecular weight-/high molecular weight-PAHs in this region indicated that petroleum combustion, vehicle emission and low-temperature combustion might be the major contributions of PAH source. To examine the POPs distillation, the analyte concentrations were correlated with altitude. The more volatile OCPs, alpha-HCH, gamma-HCH, aldrin and alpha-endosulfan positively correlated with altitude, however, less volatile OCPs (DDT and DDD) inversely related with elevation. Almost all PAHs detected in this area showed positive correlations with altitude. It is worthy to note that heavy PAHs (Benzo[k] fluoranthene and Benzo[a]anthracene) displayed positive correlation, which implied the sources of PAHs were near the sampling sites. The distillation of POPs was strongly affected by the proximity between sampling sites and contaminant sources. If the contaminant sources are close to the mountains, it may be the dominant factor that controls the concentration gradient.

Hardwood smoke alters murine splenic T cell responses to mitogens following a 6-month whole body inhalation exposure.

The purpose of these studies was to assess the effects of hardwood smoke (HWS) inhalation (30-1000 microg/m3) on the systemic immune responses of A/J mice evaluated after 6 months of daily exposures. Spleen cells obtained from mice were assessed for changes in cell number, cell surface marker expression [B, T, macrophage, and natural killer (NK) cells], and responses to B cell (LPS, endotoxin) and T cell (Con A) mitogens. Results showed that HWS smoke increased T cell proliferation in the 100 microg/m3 exposure group and produced a concentration-dependent suppression of T cell proliferation at concentrations >300 microg/m3. There were no effects on B cell proliferation or in spleen cell surface marker expression. Analyses of the exposure atmospheres revealed the presence of significant levels of naphthalene and methylated napthalenes, fluorene, phenanthrene, and anthracene in the exposure chambers, as well as low concentrations of several metals (K, Ca, and Fe). Our results demonstrate that environmentally relevant concentrations of HWS may be immunosuppressive to the immune system of mice exposed during a 6-month period.

Activation and up-regulation of nitric oxide synthase in human umbilical vein endothelial cells by polycyclic aromatic hydrocarbons.

PAHs, including naphthalene, fluoranthene and fluorene rapidly induced extracellular Ca(2+) influx and hence elevation of intracellular Ca(2+) concentration and NO production. The effect can be inhibited by Ca(2+) channel blocker but not by P450 inhibitor. In addition to the rapid effect, we have also found that the eNOS mRNA and protein expression were augmented in HUVECs treated with PAHs at concentration as low as 0.1 microM for 24 h. These effects were abolished when the HUVECs were pretreated with the BAPTA, NiCl(2) and SKF96365, as well as SKF525A. Our results revealed that, for the first time, PAHs induce the activation of eNOS and enhance eNOS protein expression in HUVECs both in a Ca(2+)-dependent manner.

Biological monitoring of exposure to polycyclic aromatic hydrocarbons by determination of unmetabolized compounds in urine.

In this paper we evaluated the possibility to assess occupational exposure to polycyclic aromatic hydrocarbons (PAHs) measuring unmetabolized PAHs in urine. With this aim, 24 road paving (RP) workers, exposed to bitumen fumes, and 6 road construction workers (CW), exposed to diesel exhausts, were investigated. Median personal exposure to low boiling PAHs (from naphthalene to pyrene) during the work shift ranged from 0.5 to 369 ng/m(3), with naphthalene as the most abundant compound. Three urine samples were collected for each worker: baseline (after 2 days of vacation), before- and end-shift samples (in the second part of the work week). The following urinary compounds were measured by headspace-solid phase microextraction GC/MS: naphthalene (U-NAP), acenaphthylene (U-ACY), acenaphthene (U-ACE), fluorene (U-FLE), phenanthrene (U-PHE), anthracene (U-ANT), fluoranthene (U-FLU), pyrene (U-PYR). Urinary PAHs were detected in almost all samples. Median levels for U-NAP, U-PHE, U-PYR and U-FLE in end-shift samples were 82, 48, 54 and 21 ng/L in RP and 69, 14, 24 and 15 ng/L in CW, respectively. Significant differences in the levels of U-PHE, U-FLU and U-PYR were found between RP and CW (p<0.05). Moreover in RP samples the urinary excretion of most analytes increased during the work shift (p<0.05). These results suggest that urinary PAHs may be useful biomarkers of occupational exposure.

Prediction of percutaneous absorption from physicochemical data: a model based on data of in vitro experiments.

Correlations between in vitro percutaneous absorption data and physicochemical properties of industrial chemicals are evaluated in order to develop predictive mathematical models based on said properties. Percutaneous diffusion of 16 pounds of occupational interest, eight of which were polycyclic aromatic hydrocarbons (acenaphthene, anthracene, benzo(a)anthracene, chrysene, phenanthrene, fluorene, naphthalene, pyrene), six organophosphorus insecticides (acephate, chlorpyrifos, dimethoate, fenitrothion, methamidophos, omethoate) and two phenoxycarboxylic herbicides (2,4-D, MCPA), were tested in vitro using monkey (Cercopithecus aetiops) skin. The test apparatus consisted of nine static diffusion cells with normal saline, gentamycin sulphate and 4% bovine serum albumin as receiving solution. Test compounds were applied at various concentrations in 30 microliters of acetone solution and determined, in the receiving phase, by chemical analysis. Values for ln Kow (octanol/water partition coefficient) were correlated with experimentally determined values of the permeability constant Kp (r = 0.90, P < 0.001) and lag time (r = 0.81, P < 0.01). Analysis of variance in a model of multiple linear regression between Kp, ln Kow and water solubility [water] of the compounds, showed that the data had a highly significant fit (P < 0.0001). A more general model which also included molecular weight (MW) and vapour pressure was evaluated as well, but the two variables made no substantial difference. Multiple regression analysis between lag time, ln Kow and [water] was significant (P < 0.0001), whereas introduction of vapour pressure and MW as independent variables did not significantly improve the predictive effect on lag time. Our experimental system, therefore, enables the values of Kp and lag time to be predicted with reasonable precision on the basis of ln Kow and [water] values, using the algorithm derived from the multiple linear regression equation.

DNA repair in human fibroblasts treated with a combination of chemicals.

Excision repair of DNA damage was measured by the photolysis of bromodeoxy-uridine incorporated during repair in normal human and xeroderma pigmentosum group C fibroblasts (XP C) treated with a combination of the carcinogens N-acetoxy-2-acetylamino-fluorene (AAAF), and 4-nitroquinoline 1-oxide (4NQO). Repair was additive in normal and XP C cells treated with AAAF plus 4NQO, indicating that there are different rate limiting steps for removal of 4NQO and AAAF lesions.

Membrane interaction of a beta-structure-forming synthetic peptide comprising the 116-139th sequence region of the cytotoxic protein alpha-sarcin.

alpha-Sarcin is a cytotoxic protein that strongly interacts with acid phospholipid vesicles. This interaction exhibits a hydrophobic component although alpha-sarcin is a highly polar protein. A peptide comprising the amino acid sequence corresponding to the 116-139th segment of the alpha-sarcin cytotoxin has been synthesized by a standard fluoren-9-yl-methoxycarbonyl-based solid phase method. Its primary structure is: (116)-NPGPARVIYTYPNKVFCGIIAHTK-(139). Two beta-strands have been predicted in this region of alpha-sarcin, where the less polar stretches of the protein are found. The synthetic peptide interacts with negatively charged large unilamellar vesicles of either natural or synthetic phospholipids. An apparent fragmentation of the vesicles is produced by the peptide based on electron microscopy studies. The peptide promotes leakage of the intravesicular aqueous contents and lipid mixing of bilayers. The packing of the phospholipid molecules is greatly perturbed by the peptide, as deduced from the drastic changes induced by the peptide in cooperative properties associated with the phase transition of the bilayers. At saturating peptide/phospholipid ratios, the phase transition of dimyristoylphosphatidylglycerol vesicles is abolished. All of these effects are saturated at about 0.3 peptide/lipid molar ratio. The peptide adopts a mostly random structure in aqueous solution. A conformation composed of a high proportion of antiparallel beta-sheet is induced as a consequence of the interaction with the phospholipid vesicles in opposition to trifluoroethanol that promotes alpha-helical peptide structures, as deduced from circular dichroism measurements. The obtained results are discussed in terms of the potential involvement of the region comprising residues 116-139 of alpha-sarcin in the hydrophobic interactions of this cytotoxic protein with membranes.

Post-treatment with a novel PARG inhibitor reduces infarct in cerebral ischemia in the rat.

Poly(ADP-ribose) is synthesized from nicotinamide adenine dinucleotide (NAD(+)) by poly(ADP-ribose) polymerase (PARP) and degraded by poly(ADP-ribose) glycohydrolase (PARG). Overactivation of the poly(ADP-ribose) pathway increases nicotinamide and decreases cellular NAD(+)/ATP, which leads to cell death. Blocking poly(ADP-ribose) metabolism by inactivating PARP has been shown to reduce ischemia injury. We investigated whether disrupting the poly(ADP-ribose) cycle by PARG inhibition could achieve similar protection. We demonstrate that either pre- or post-ischemia treatment with 40 mg/kg of N-bis-(3-phenyl-propyl)9-oxo-fluorene-2,7-diamide, a novel PARG inhibitor, significantly reduces brain infarct volumes by 40-53% in a rat model of focal cerebral ischemia. Our result provides the first evidence that PARG inhibitors can ameliorate ischemic brain damage in vivo, in support of PARG as a new therapeutic target for treating ischemia injury.

Investigation of the nematic-isotropic transition of a liquid crystalline polymer and determination of molecular diffusion coefficients using gas chromatography.

Inverse gas chromatography has been used to study the nematic-isotropic transition of a side chain liquid crystalline polymer (LCP). The mesogenic side groups are laterally attached to a polysiloxane backbone through a flexible spacer. The nematic-isotropic transition of this LCP coated onto a glass capillary column is detected by considering the variation with temperature of the retention volume and of the theoretical plate number for the several probes. The molecular diffusion coefficients, D, of naphthalene, fluorene, pyrene and o-terphenyl have been determined at different temperatures in the nematic phase of the LCP as well as in the isotropic melt. The values ranged between 10(-14) and 10(-12) m2 s(-1) for the polynuclear aromatic hydrocarbon probes tested.

Gas chromatographic-mass spectrometric determination of aromatic hydrocarbon metabolites from livers of fish exposed to fuel oil.

Metabolites of several two- and three-ring aromatic hydrocarbons (AHs) have been found in livers of English sole exposed to No. 2 fuel oil. Four metabolites of the C2H5-naphthalenes, six of the C3H7-naphthalenes and one each of fluorene, phenanthrene and anthracene have been partially characterized and their concentrations, which ranged from 50 to 1100 ng/g, were determined. Metabolites were separated from the liver matrix using an automated extractor/concentrator. The resulting extract was then purified by high-performance liquid chromatography, and the metabolites were characterized and quantitated by gas chromatography-mass spectrometry.

Chemical changes of organic compounds in chlorinated water. XVI. Gas chromatographic-mass spectrometric studies of reactions of tricyclic aromatic hydrocarbons with hypochlorite in dilute aqueous solution.

The products of aqueous chlorination reactions of tricyclic aromatic hydrocarbons (fluorene, carbazole, dibenzofuran, anthracene, phenanthrene and some methyl derivatives) with hypochlorite have been determined by gas chromatography-mass spectrometry. They included chloro-substituted, oxygenated (quinones) and hydroxylated (phenols) compounds, and products of addition which were readily formed at ambient temperature. The extent of the reactions was shown to depend on the chlorine dose, the solution pH, the initial concentrations of both compounds and the structures. Monochlorinated compounds and quinones were shown to be present in chlorinated water under the conditions utilized for water treatment.

Nordic laboratory intercomparison of supercritical fluid extraction for the determination of total petroleum hydrocarbon, polychlorinated biphenyls and polycyclic aromatic hydrocarbons in soil.

Two developed supercritical fluid extraction (SFE) methods [one for the determination of total petroleum hydrocarbon (TPH) and polychlorinated biphenyls (PCBs), and one for polycyclic aromatic hydrocarbons (PAHs) and creosote components in soil] were evaluated in a Nordic laboratory intercomparison study with 11 participating laboratories. The interlaboratory comparison showed that excellent recoveries can be obtained with SFE for PAHs and PCBs compared to the solvent extraction. For the TPH, the recoveries were significantly higher than those achieved with solvent extraction. The accuracy, expressed as the relative standard deviation, was higher than expected (generally 8-25% for PAHs, 6-20% for PCBs and less than 18% for TPH with a few very high values, especially for PCBs), but not different from the other intercomparison studies. Difference between liquid- and solid-phase collection in SFE was found to be significant only for more volatile PAH components such as naphthalene and fluorene. For PCBs and TPH, there were some variation in the results obtained with the two trapping methods.

Analysis and confirmation of synthetic anorexics in adulterated traditional Chinese medicines by high-performance capillary electrophoresis.

Six synthetic anorexics, clobenzorex, diethylpropion, fenfluramine, methamphetamine, phenylpropanolamine and phentermine, which can be found as adulterants in traditional Chinese medicines were assayed simultaneously by high-performance capillary electrophoresis. The electrolyte was a buffer solution containing 120 mM phosphate buffer (NaH2PO4/H3PO4, pH 2.0) and 15% acetonitrile. Applied voltage was 16 kV and temperature was 30 degrees C. Fluoren-2,7-diammonium chloride was used as an internal standard and detector set at 200 nm. The recoveries of the synthetic anorexic adulterants in traditional Chinese medicinal formula using C8-SCX mixed solid-phase extraction were studied. Several traditional Chinese medicinal powders obtained from clinics were also studied by the above HPCE method and confirmed by GC-MS. Clobenzorex, diethylpropion and fenfluramine were found and determine in these samples.

Sample cleanup and reversed-phase high-performance liquid chromatographic analysis of polar aromatic compounds in groundwater samples from a former gas plant.

A method for the analysis of the polar aromatic compounds 1H-quinolin-4-one (Q), 10H-acridin-9-one (A), 5H-phenanthridin-6-one (P) and 9H-fluoren-9-one (F) in aqueous solutions has been developed. The method comprises steps for sample preparation (solid-phase extraction, cleanup) and analytical determination by means of reversed-phase high-performance liquid chromatography (RP-HPLC). For the cleanup step the suitability of two different sorbents (alternative A: silica gel, alternative B: LiChrolut EN) was investigated. Alternative B depicted several advantages, in particular higher sorbent capacity, faster and less complicated handling, higher recovery and better reproducibility. For Q, A and P, reproducibility of all method steps is better than 13%, with recovery rates ranging from 76% to 105% (n=3). Alternative B was applied to groundwater samples from a former gas plant. The analytes A and P could be detected at concentrations in the micro/l range.
