Tentative fingerprint-efficacy study of Houttuynia cordata injection in quality control of traditional Chinese medicine.

To establish potent fingerprint for quality control of traditional Chinese medicine, Houttuynia cordata (Saururaceae) injection (HCI), the attempt on fingerprint-efficacy was developed in this study. HCI from ten different factories were determined by gas chromatography-mass spectrum (GC-MS) and classified by hierarchical clustering. The anti-inflammatory effect of HCI was characterized with the rat pleurisy model induced by carrageenin and the mice ear edema model by xylene. The results showed that anti-inflammatory effect of the injections from most of factories on the two models was significant. There was corresponding relationship between the fingerprint of HCI and efficacy to certain extent. The main common constitutes in injection from the factories that possess anti-inflammatory activity were analysed with GC-MS and identified using the NIST Mass Spectral Database. This common pattern of HCI based on the efficacy was helpful for the purpose of quality control.

Bimodal fingerprint capturing system based on compound-eye imaging module.

A compact imaging system called TOMBO (Thin Observation Module by Bound Optics) is proposed in which a microlens array is used for thin hardware configuration. This paper describes a fingerprint-capturing module as an application of the TOMBO. Experimental results by the TOMBO prototype system are shown to clarify the applicability of the TOMBO to the fingerprint capturing. Different types of biometrics, i.e., fingerprint and face images, are captured by the same hardware, which shows the extendability of the system for multimodal identification.

The frequency of fingerprint type in parents of children with Trisomy 21 in Japan.

Analysis of the frequency data of each fingerprint type (arch, ulnar loop, radial loop, and whorl) of the parents of children with Trisomy 21 (Fathers: 71; Mothers: 128) born between 1965 and 1970 obtained from the Tokyo Medical and Dental University Hospital was carried out. Japanese controls were taken from dermatoglyphics data in Japan. We conducted the Friedman test on each type of fingerprint between Japanese controls and parents of Trisomy 21 children. Results from a statistical analysis based on the above data showed significant differences, more arches (p < 0.0001) and fewer whorls (p < 0.05) in mothers of children with Trisomy 21. Among fathers of Trisomy 21 children, a significant difference was found in there being fewer whorls (p < 0.05) and ulnar loops (p = 0.06). Considering the mothers' fingerprints, we suspected that females with a higher frequency of arches and a lower frequency of whorls had a stronger possibility of bearing Trisomy 21 babies. On the other hand, in fathers of Trisomy 21 children, we considered that there would be a possibility of significant differences if cases in the sample were increased.

The quality assessment of compound liquorice tablets by capillary electrophoresis fingerprints.

Capillary electrophoresis (CE) is a powerful separation technique that is peculiarly able to determine the fingerprints of traditional Chinese medicine. The Capillary Electrophoresis Fingerprints (CEFP) of compound liquorice tablets (CLTs) was established to evaluate the quality of CLTs. The background electrolyte was a 50 mM sodium borate solution. The detection wavelength was 228 nm and a 14 kV voltage was applied. Hydrochlorothiazide served as an internal standard and temperature was 24-25 degrees C. The CLTs were extracted by a 50 mM borate solution containing 10% (v/v) methanol. The results showed that CE was a powerful tool for detecting of the fingerprints of traditional Chinese medicine and their constituents. The quality assessments were performed to compare the correlation coefficients (R) and cos theta (C) between each batch vector of CLTs and the total standard profile vector (TSPV), which was the mean vector of all the batch vectors of CLTs with 1.0. The nearer to 1.0 was the R or C, the higher was the similarity. The values of R and C between the TSPV and each batch vector of CLTs produced at different factories were all above 0.9150. The values of R and C between every two standard profile vectors (SPV) from different factories were all above 0.9420. A good stability and reproducibility of the CEFP of CLTs were obtained in the study. Further, we first put forward a new parameter Q to evaluate both the quantification accuracy and the qualitative accuracy for a CEFP, which was verified by determinations of GHIA, GHEA, MP and SB in CLTs. The CEFP was successfully employed to assess the quality of CLTs produced at Kunming Pharmaceutical Factory to be good.

Silent lupus nephritis with fingerprint deposits.

A 26-year old woman with diffuse active lupus nephritis showed no proteinuria. Electron microscopy revealed a typical fingerprint pattern in glomerular subendothelial electron dense deposits. Out of 21 renal biopsy cases of lupus nephritis, we found fingerprint deposits in 4 cases (19%) including the present case. All of the cases showed diffuse proliferative glomerulonephritis with overt proteinuria, except the present case. To our knowledge, the present case is the first reported case of active lupus nephritis, where fingerprint deposits were found, with normal urinalysis.

A new visibly-excited fluorescent component in latent fingerprint residue induced by gaseous electrical discharge.

A technique that exposes fingerprint residue to a gaseous electrical discharge in nitrogen followed by treatment with ammonium hydrogen carbonate vapors to produce fluorescence is investigated. Particular attention is made to fluorescence observed via laser illumination at 514 nm. Insight into the nature of the fluorescent components is achieved through the use of thin-layer chromatography (TLC) of fingerprint residue. Results reported indicate the fluorescence observed is from previously non-fluorescent fractions of the fingerprint residue, and TLC results point towards lipid derivatives as a possible source of the fluorescence.

Processes involved in the development of latent fingerprints using the cyanoacrylate fuming method.

Chemical processes involved in the development of latent fingerprints using the cyanoacrylate fuming method have been studied. Two major types of latent prints have been investigated-clean and oily prints. Scanning electron microscopy (SEM) has been used as a tool for determining the morphology of the polymer developed separately on clean and oily prints after cyanoacrylate fuming. A correlation between the chemical composition of an aged latent fingerprint, prior to development, and the quality of a developed fingerprint has been observed in the morphology. The moisture in the print prior to fuming has been found to be more important than the moisture in the air during fuming for the development of a useful latent print. In addition, the amount of time required to develop a high quality latent print has been found to be within 2 min. The cyanoacrylate polymerization process is extremely rapid. When heat is used to accelerate the fuming process, typically a period of 2 min is required to develop the print. The optimum development time depends upon the concentration of cyanoacrylate vapors within the enclosure.

Latent fingerprint detection using a scanning Kelvin microprobe.

Electrochemical interactions between latent human fingerprints and metal surfaces in ambient air are investigated using a scanning Kelvin microprobe. Inorganic salts present in sweat deposited at fingerprint ridges locally depassivate the metal surface producing a Volta potential decrease of greater than 200 mV. Volta potential patterns may persist for months and prints may be visualized by potential mapping, even when overcoated with a polymer layer. Because the Volta potential differences are produced by involatile salts they persist when the organic components of the fingerprint residue have been volatilized by heating to 600 degrees C.

Advanced solvent-free application of ninhydrin for detection of latent fingerprints on thermal paper and other surfaces.

This work presents the first known experiments of ninhydrin sublimation in vacuum to detect latent fingerprints on thermal paper. In this method, latent fingerprints become visible in rich detail without the background black staining known from the application of ninhydrin solutions to thermal paper. The method involves hanging the thermal paper samples 15 cm above a heating source with dispersed ninhydrin crystals in a vacuum chamber. The optimized conditions for ninhydrin sublimation are 50 mg ninhydrin, 2 to 5 mbar vacuum, and 150 degrees C heating source temperature for 30 min. The application of this method is also successful on the new euro notes. Latent fingerprints can be developed across the transitions from paper to optical variable device (OVD).

[Application of fingerprint chromatogram in quality control of Shen-Mai injection].

The theory and practice of traditional Chinese medicine require some comprehensive methods to assess quality of the Chinese herbal medication. Fingerprint chromatogram is one of the feasible approaches to evaluate the quality of Chinese herbal medication. So the fingerprint chromatogram of Shen-Mai injection was established by using reversed-phase high performance liquid chromatography. The chromatographic conditions were as follows: a Hypersil C18 column was used; the mobile phase was composed of water (A) and acetontrile (B) with linear gradient elution (0-50 min, 5%-95% B, volume fraction); the flow rate was 1.0 mL/min and the UV absorbance detection was set at 202 nm. The peak-area ratios of twenty-three fingerprint peaks and internal standard (diphenyl) were taken as the criteria for quality control. The quality differences in various batches and various manufacturers of Shen-Mai injections were investigated by projection discriminance based on principal component analysis. The results show the method developed is convenient, reliable and applicable for the quality control analysis of Shen-Mai injection.

[A novel fingerprint method for quality evaluation of Chinese medicinal plants based on analytical data visualization].

OBJECTIVE: To investigate new method for evaluating the quality of Chinese Medicinal Plants (CMP). METHOD: A visualization technique for representing instrumental analytical data was developed by applying the fundamental of Data Visualization, with Principal Component Analysis (PCA) and spatial projection transformation, original IR spectral data were projected into a low-dimensional subspace so that the dimensionality of original data space was decreased and tiny fingerprint features were extracted. The data set in the subspace was visualized by means of two-dimensional grayscale images. Consequently, the characteristic fingerprint for appraising the quality of CMP was obtained. RESULT: 42 mulberry root-bark samples from three different quality classes were identified with the proposed method, which showed that the fingerprint images had satisfactory resolution and classification accuracy as high as 90.5%. CONCLUSION: The proposed method is a useful technique for appraising the quality of CMP.

[IR-fingerprinting ordination comparison on different organs of three species of isodon].

The plants of the genus Isodon are of important medicinal values, being widely used in the production of the Chinese traditional and herbal drugs. Compared with the other identification methods, the identification of the Chinese traditional and herbal drugs using the fourier-transform infrared spectrometer with OMNI collector is simple and convenient, fast and accurate. Moreover, the extraction or break of the samples are not necessary in the identification using OMNI collector. In the present paper, fourier-transform infrared spectrometer with OMNI collector is applied to gain the IR-fingerprintings of eleven samples of Isodon inflexus, I. Lophanthoides and I. Macrocalyx. Based on the indices of wavenumber-absorbance, the differences of eleven IR-fingerprintings are compared by PCA (Principal Component Analysis). The results show that it is practical to apply PCA on the basis of IR-fingerprinting to compare the chemical differences of plant samples.

Transcriptome fingerprints distinguish hallucinogenic and nonhallucinogenic 5-hydroxytryptamine 2A receptor agonist effects in mouse somatosensory cortex.

Most neuropharmacological agents and many drugs of abuse modulate the activity of heptahelical G-protein-coupled receptors. Although the effects of these ligands result from changes in cellular signaling, their neurobehavioral activity may not correlate with results of in vitro signal transduction assays. 5-Hydroxytryptamine 2A receptor (5-HT2AR) partial agonists that have similar pharmacological profiles differ in the behavioral responses they elicit. In vitro studies suggest that different agonists acting at the same receptor may establish distinct patterns of signal transduction. Testing this hypothesis in the brain requires a global signal transduction assay that is applicable in vivo. To distinguish the cellular effects of the different 5-HT2AR agonists, we developed an assay for global signal transduction on the basis of high throughput quantification of rapidly modulated transcripts. Study of the responses to agonists in human embryonic kidney 293 cells stably expressing 5-HT2ARs demonstrated that each agonist elicits a distinct transcriptome fingerprint. We therefore studied behavioral and cortical signal transduction responses in wild-type and 5-HT2AR null-mutant mice. The hallucinogenic chemicals (+/-)-2,5-dimethoxy-4-iodoamphetamine (DOI) and lysergic acid diethylamide (LSD) stimulated a head-twitch behavioral response that was not observed with the nonhallucinogenic lisuride hydrogen maleate (LHM) and was absent in receptor null-mutant mice. We also found that DOI, LSD, and LHM each induced distinct transcriptome fingerprints in somatosensory cortex that were absent in 5-HT2AR null-mutants. Moreover, DOI and LSD showed similarities in the transcriptome fingerprints obtained that were not observed with the behaviorally inactive drug LHM. Our results demonstrate that chemicals acting at the 5-HT2AR induce specific cellular response patterns in vivo that are reflected in unique changes in the somatosensory cortex transcriptome.
