[Observation and analysis on mutation of routine STR locus].

OBJECTIVE: To observe and analyze the characteristic of mutation at STR locus. METHODS: 27 mutant genes observed in 1211 paternity testing cases were checked by PAGE-silver stained and PowerPlex 16 System Kit and validated by sequencing. RESULTS: Mutant genes locate on 15 loci. The pattern of mutation was accord with stepwise mutation model. The mutation ratio of male-to-female was 8:1 and correlated to the age of father. CONCLUSION: Mutation rate is correlated to the geometric mean of the number of homogeneous repeats of locus. The higher the mean, the higher the mutation rate. These loci are not so appropriate for use in paternity testing.

[Polymorphism of X-STR loci DXS7108 and DXS1196 in the Northern Polish population].

This paper describes results of a population study of two X-linked STR microsatellite markers: DXS7108 and DXS1196. 298 samples of DNA of unrelated persons (male and female) from the Northern part of Poland were analyzed. DNA was isolated using a non-enzymatic method. After amplification PCR products were separated by means of capillary electrophoresis using the ABI PRISM 310 Genetic Analyzer. The most common alleles of each locus were sequenced and used as a control ladder to type unknown samples. Testing for Hardy-Weinberg equilibrium (HWE) showed no significant deviation for these two loci. Statistical parameters (PD, HET, MEC) showed that examined systems are useful in forensic medicine.

STR-typing of human DNA from human fecal matter using the QIAGEN QIAamp stool mini kit.

The purpose of this study was to compare the effectiveness of the QIAGEN QIAamp Stool Mini Kit against a standard phenolchloroform procedure for the extraction, quantitation, and STR-typing of human nuclear DNA from human feces. Stools from six subjects were sampled by swabbing and excision. Samples extracted with the QIAamp kit gave a wide range of DNA yields, whereas those extracted by the organic method yielded no DNA. DNA was not recovered from one subject's stools by either procedure. The QIAamp extracts were amplified with the Profiler Plus and COfiler kits, and PCR inhibition was observed with DNA extracts that were further concentrated. Substitution of water or TE-4 for the QIAamp elution buffer eliminated most, if not all, of the inhibition. A modified QIAamp procedure was used to extract thirty samples, which were subjected to one of five environmental conditions. DNA was recovered from all of these samples, and typing results were obtained on 93% of the samples.

Identification and characterization of variant alleles at CODIS STR loci.

Short tandem repeat (STR) profiles from 32,671 individuals generated by the ABI Profiler Plus and Cofiler systems were screened for variant alleles not represented within manufacturer-provided allelic ladders. A total of 85 distinct variants were identified at 12 of the 13 CODIS loci, most of which involve a truncated tetranucleotide repeat unit. Twelve novel alleles, identified at D3S1358, FGA, D18S51, D5S818, D7S820 and TPOX, were confirmed by nucleotide sequence analysis and include both insertions and deletions involving the repeat units themselves as well as DNA flanking the repeat regions. Population genetic data were collected for all variants and frequencies range from 0.0003 (many single observations) to 0.0042 (D7S820 '10.3' in North American Hispanics). In total, the variant alleles identified in this study are carried by 1.6% of the estimated 1 million individuals tested annually in the U.S. for the purposes of parentage resolution. A paternity case involving a recombination event of paternal origin is presented and demonstrates how variant alleles can significantly strengthen the genetic evidence in troublesome cases. In such instances, increased costs and turnaround time associated with additional testing may be eliminated.

[STR polymorphism of the non-coding human growth hormone gene (HUMGHCSA) and its use in personal identification--a preliminary report].

The HUMGHCSA locus, located within the non-coding region of the human growth hormone gene on chromosome 17, exhibiting polymorphism of the number of repeats of two and four nucleotide motifs (CT and CTTT), but also polymorphism caused by nucleotide substitution, is one of the most polymorphic STR markers. The purpose of this study was to report the development of a system of fluorescent analysis for an automatic sequencer, to test its reliability and to develop biostatistical parameters of its usefulness in forensic examinations. The studies, carried out on a group of 200 non-related individuals, showed the presence of 24 different alleles, of 221 to 279 bp in length. Biostatistical calculations demonstrated that the expected heterozygosity was 0,898634 +/- 0,015091, PD - 0,980586, PIC - 0,887709, PE - 0,792767, PE for motherless cases - 0,6556701, and the average paternity index - 4,932625. Such results indicate a High usefulness of the studied HUMGHCSA locus for identification examinations.

[Preparation of proteolytic enzymes from the thermophilous actinomycete Actinomyces thermovulgaris str. T-54].

A preparation with a high proteolytic activity has been obtained by acetone precipitation from the culture liquid filtrate of the thermophilous actinomycete Actinomyces thermovulgaris str. T-54. The proteolytic (caseinolytic), fibrinolytic and thrombolytic activity of the preparation is comparable with that of trypsin and far superior to that of fibrinolysine. The preparation is stable at pH 5.0=9.5 and inactivated in the acid zone. The study of pH dependent proteolytic activity has shown acid, neutral and alkaline proteases in the preparation. It is relatively thermostable, and is completely inactivated for 10 min at 90 degrees. It is suggested that the preparation contains four enzymes or four enzymic groups that are different in their temperature sensitivity.

[Verification of the preparation of Streptococcus pyogenes str. A1 96/50 based on the results of growth properties of media].

For tests of the growth properties of media Czechoslovak standards prescribe a culture of the Streptococcus pyogenes strain Str. A1 96/50 which must be shaken with sea sand and diluted with a gelatin solution. The author found that shaking the culture with sea sand usually reduces significantly (P less than 0.05) the number of colonies in agar medium, as compared with a culture which did not have this treatment. The author also found that there is no significant difference (P greater than 0.05) in the number of colonies when the culture is diluted with a gelatin solution or isotonic sodium chloride solution. He proved that there is no significant difference (P greater than 0.05) in the scatter of the number of colonies in agar when the culture is shaken with sea sand or if it is diluted with a gelatin solution, as compared with a culture not shaken with sand or diluted with sodium chloride solution.

[Use of enzymatic hydrolysates of microorganism biomass in media for culturing Str. lactis, the producer of nisin].

Fermentative hydrolysates (FH) of the protein-vitamin complex (PVC) were used for the first time in media for cultivation of Str. lactis. It was found that the FH may be used in the media for cultivation of streptococci as a source of nitrogen instead of the yeast autolysate from baker's yeast and the level of nisin biosynthesis did not change. The replacement of the yeast autolysate by the FH is possible in both flasks with low volumes of the medium and 401 fermenters with 25 liters of the medium. The BVC FH is not a food raw material and its cost is low, which makes it advantageous.

[New anthracycline antibiotics taurimycins A and B produced by a Str. tauricus No. 7036-1 culture].

Taurimycins A and B are new anthracycline antibiotics belonging to the group of glycosides. They were isolated from the mycelium of culture 7036-1 classified with Str. tauricus. The empiral formula of taurimycin A is C14H51O16N,M+813. The empirical formula of taurimycin B is C41H49O16N,M+811. The absorption maxima in the UV spectra of the antibiotics (in alcohol solutions) were observed at 235, 257, 292, 466, 482, 494, 515 and 527 nm. The IR spectra showed absorption bands at 3450, 2945, 2870, 1740 and 1610 cm-1. Hydrolysis of the antibiotics with hydrochloric acid resulted in formation of an aglycone identified as epsilon-pyrromycinone.

Identification of Streptococcus mutans from human dental plaque using the RapID STR System.

The identification of Streptococcus mutans from dental plaque samples with rapid systems, such as the RapID STR System, has not been recommended by the manufacturers. In this study, we evaluated the use of the above method for the identification of 34 human dental plaque Streptococcus mutans strains which had been identified previously by conventional procedures. The results obtained demonstrate that 88.2% of the Streptococcus mutans strains tested were identifiable by the rapid method.

[Forensic application of two short tandem repeat (STR) polymorphic markers, FES and HUMTH01].

This paper reports polymorphisms of two short tandem repeats (STR) in the human c-fes/proto-oncogene (FES) and the human tyrosine hydroxylase gene (HUMTH01), and their application to paternity cases. Genomic DNA samples from 213 unrelated Japanese individuals and from 30 paternity cases were subjected to the polymerase chain reaction (PCR) and then the amplified products were electrophoresed in denaturing polyacrylamide sequencing gels. Allelic bands were detected with autoradiography. Seven alleles differing in the numbers of the repeat units were observed at both loci. Heterozygosity (%) and polymorphism information context were calculated, being 69% and 0.641 FES, and 70% and 0.647 for HUMTH01. The allele frequencies of the two STRs in this study were found to be similar to those previously reported on Asians but different from those in Caucasians and Blacks. When the STRs were applied to 30 paternity cases that had been already tested by using conventional markers, there were no discrepancies between the results of the STRs and of conventional markers. Furthermore, the two STRs could be successfully coamplified in multiplex PCR. These results suggest that the two STRs are useful in forensic work in Japan.
