Assessment of hydration status by urinary analysis of elite junior taekwon-do athletes in preparing for competition.

The aim of this study was to assess over time the hydration status of taekwon-do athletes during a preparatory camp. Measures of urine osmolality, conductivity, specific gravity and colour were made on the first urine sample passed by the participants (n = 32) in the morning before breakfast. The urinary measurements were made on samples collected at the beginning of the camp, 5 days later and one day before competition. Body mass was also measured at the same instants. Body mass (mean +/- s) was essentially the same on each of the measurement days (62.6 +/- 12.2, 62.7 +/- 12.3 and 62.2 +/- 12.6 kg, respectively). Mean urine osmolality at the beginning of the camp was relatively high (998 +/- 171 mOsmol . kg-1), suggesting that a significant number of the athletes were already hypohydrated. However, no significant differences were detected in urine osmolality at the three time points during the study. There were no significant differences in any of the four methods of urine analysis during the study (P > 0.05). The average values for all samples were 989 +/- 205 mOsmol . kg-1 for osmolality, 25.5 +/- 6.7 mS . cm-1 for conductivity, 1.017 +/- 0.010 g . cm-3 for specific gravity and 4 +/- 1 arbitrary units for colour. Correlation analysis between the different methods suggested moderately good agreement (correlation coefficient = 0.5-0.7) between all four measurement techniques (P < 0.01). The coefficients of variation for these techniques were relatively low (CV = 11.8 - 35.0%). The results of this study suggest that some of the taekwon-do athletes were slightly hypohydrated in the morning on each of the test days, but there was no evidence to suggest that most of the athletes further restricted their fluid intake to make weight. In addition, it appears that each of the four methods used gave essentially the same estimate of hydration status of these athletes.

The role of rehydration in the prevention of fat embolism syndrome.

We encountered three cases of young sportsmen developing fat embolism syndrome (FES) after sustaining isolated tibial shaft fractures whilst playing football. All fractures were treated with intra-medullary nails and all three patients were kept nil-by-mouth pre-operatively without intravenous fluids. Correction of shock is often quoted as an important factor in the prevention of FES. However, animal studies have shown that dehydration, as opposed to hypovolaemia, may also be of great importance. We therefore examined the specific gravity of the urine of 20 patients with musculoskeletal injuries sustained during sport. The mean urinary specific gravity was significantly higher than that of a control group of 10 members of staff. We emphasise the importance of adequate pre-operative rehydration, especially if injuries were sustained during heavy exercise, as this may reduce the risk of developing FES.

[A biological study of inbred Weiser-Maples guinea pigs--urinalysis, hematological and blood-chemical values and organ weights].

Urinalysis, hematological and blood chemical examinations and measurement of organ weights were carried out for the purpose of collecting background data on Weiser-Maples (WM) which was established as an inbred strain of guinea pigs. The results were compared with those for the commercial guinea pigs, Std: Hartley (H). In addition, the measurement methods for urinary specific gravity and whether centrifugation of urine influences the results of urinalysis or not were examined. The specific gravity values calculated from the volume and weight of urine were highly correlated with those from the refraction rate. The results of urinalysis of the supernatant of centrifuged urine were similar to those of urine not centrifuged, but the results suggested that the use of the supernatant provided a more accurate measurement of urine in guinea pigs. The positive rate for urinary protein was higher in the WM strain (38.9%) than in the H strain (0%). Although the WM strain showed higher or lower values than the H strain in some items of hematological and blood chemical examinations and organ weights, we could not regard them as characteristic of the WM strain.

Performance evaluation of three on-site adulterant detection devices for urine specimens.

The performance of three on-site adulterant detection devices that assess the integrity of urine specimens collected for drug-of-abuse testing was evaluated: the Intect 7, MASK Ultra Screen, and Adultacheck 4. Intect 7 simultaneously tests creatinine, nitrite, glutaraldehyde, pH, specific gravity, and the presence of bleach and pyridinium chlorochromate (PCC). Mask Ultra Screen tests creatinine, nitrite, pH, specific gravity, and oxidants, and Adultacheck 4 tests creatinine, nitrite, glutaraldehyde, and pH. Urine specimens were prepared with the Substance Abuse and Mental Health Administration regulated analytes at 50% above the cut-off concentrations. Stealth, Urine Luck, Instant Clean ADD-IT-ive, and KLEAR were added individually to the drug-added urine specimens so that their concentrations reflected the "optimum" usage reported in their package inserts and 25% above and below that optimum. Stealth is reported to be peroxidase; Urine Luck is believed to be PCC; Instant Clean ADD-it-ive reportedly contains glutaraldehyde, and Klear is a nitrite. The following diluents/adulterants were added at 25%, 33%, and 50% of the volume of drug-added urine: distilled water, bleach, ammonia, and vinegar. Of the devices tested, Intect 7 proved to be the most sensitive, and it correctly indicated the presence of adulterant or diluent in all samples tested. In order to do so, all indication pads had to be assessed in concert. Adultacheck 4 specifically assesses four characteristics of urine integrity and is therefore very limited in detecting the use of several popular adulterants that are commercially available. Although it correctly assessed the four characteristics, it did not detect the use of Stealth, Urine Luck, or Instant Clean ADD-it-ive. Mask Ultra Screen can potentially detect a broader range of adulterants than Adultacheck 4. However, in practice, it only detected them at levels well above their optimum usage, making it less efficacious than Intect 7. Clearly, the specific identification of an adulterant is a trade-off for sensitive detection of several adulterants.

Lessons from the space experiment SL-J/FMPT/L7: the effect of microgravity on chicken embryogenesis and bone formation.

Thirty fertilized chicken eggs preincubated for 0, 7, and 10 days on earth (10 eggs each) were flown in the space shuttle "Endeavour" and further incubated for 7 days under microgravity. Twenty out of 30 eggs (nine out of ten 10-day-old eggs; 10 out of ten 7-day-old eggs; 1 out of ten 0-day-old eggs) were recovered alive after landing. The only living embryo of the 0-day-old egg died 24 days after launch, and was comparable to a 16-day-old embryo when it died. The high mortality of the 0-day-old eggs appeared to be related to the specific inner structure of the egg. The yolk (specific gravity, 1.029) would not have separated from the albumen (1.040) during space flight. The subtle difference in specific gravity between the yolk and the albumen appeared to play a critical role in early chick embryogenesis. All the tissues, including cartilage and bone, were formed normally in 7- and 10-day-old chicken embryos during space flight. When the 7- and 10-day-old embryos recovered at Kennedy Space Center (KSC) were further incubated on earth, they hatched normally. No appreciable changes were recognized in these chicks. The reason for these unexpected results is not known. Further space experiments are needed to explain the failure of the effects of microgravity on chicken embryogenesis.

Physical properties of antibiotic aerosols produced by jet and ultrasonic nebulizers.

The drug stability and the physical properties of aerosol of five different antibiotics were compared for ultrasonic and jet-type nebulization. These antibiotics were used alone, as well as combined with betamethasone sodium phosphate and/or tyloxapol glycerol sodium bicarbonate. The parameters investigated were changes in appearance, pH, temperature, viscosity, specific gravity and the residual potency of the antibiotics. No significant change was shown in appearance, viscosity and specific gravity. Temperatures of the solutions in the ultrasonic nebulizer cup increased with time, though almost no change after 10 minutes in the jet nebulizer. For pH, with antibiotics alone in the ultrasonic nebulizer, some drugs showed an increase, and other drugs showed a decrease, while generally a little change in the jet nebulizer. For residual potency, one drug showed decreased potency in the aerosol, though it was increased in the remnant solution, in contrast, another drug showed a tendency to increased potency in the aerosol and decreased potency in the remnant solution in the ultrasonic nebulizer. In the jet nebulizer, drugs showed generally slight increased potency and no difference among drugs. It was suggested that, with ultrasonic nebulization, in some drugs the solvents were nebulized and the principal ingredient tended to remain behind, while in other drugs the opposite occurred so that the effects were not uniform.

Pentoxifylline does not reduce cerebral ischemic edema in hypertensive rats.

Continuous infusion of pentoxifylline 0.30 mg per kg per min starting 30 min after occlusion of the middle cerebral artery did not reduce the development of cerebral edema, as measured by specific gravity 6 h after occlusion in spontaneously hypertensive rats. On the contrary, in the parietal region the specific gravity was significantly lower in treated rats, indicating an increased water content. Thus, this study failed to show any beneficial effect of pentoxifylline in brain edema during early permanent ischemia.

[Actual evaluation of nutrition in different children age groups].

Children actual nutrition research in Novomoskovsk in different age groups 4-5, 9-10, 12-13 and 15-16 revealed the existence of the main food component shortage, nutrient and aminoacid disbalance, and also the reduction of the power value of the daily nutrition. Within the years the specific gravity of children with protein, fat and carbohydrate, shortage has the marked tendency towards the reduction, the specific gravity with the shortage of C, A, B2, B6, B12 vitamins and also with iron shortage among the girls has the tendency towards the increase.

Exploiting gravity in abdominal angiography.

The difference between the specific gravity of blood and contrast medium and the consequent tendency of contrast media to fill the lower tissues is exploited in abdominal angiography by special positioning of patients: prone for the coeliac, hepatic and mesenteric arteries, prone with the legs raised for abdominal aortography; lateral decubitus on the side of interest for renal angiography. The advantages are better parenchymal opacification, increased contrast and improved details. Gravity positioning saves time and material, and is easy to learn.

Studies on the capacity of human tonsillar lymphocyte subpopulations to produce interferon.

Tonsillar lymphocytes produced classical interferon (Type I) in response to NDV infection and produced immune interferon (Type II) in response to PHA, PPD or histo-incompatible antigens. Lymphocytes having higher specific gravity and having a greater proliferative response to mitogens or antigens produced more interferon than lymphocytes having lower specific gravity. There was no difference between heavy and light small tonsillar lymphocytes in regard to their sensitivity to the protective effects of interferon. As for the interferon produced in association with a cellular immune response, the peak of interferon production tended to appear earlier than the peak of 3H-TdR incorporation. It is suggested that tonsillar lymphocytes play an important role in human host defense against virus infections.

Correlation between human sperm hypo-osmotic swelling test and other semen parameters.

Ninety semen samples from male patients were studied. They were classified into 4 different groups; normospermic fertile, oligospermia, asthenospermia, and oligoasthenospermia. In addition to routine semen analysis, the hypo-osmotic swelling test was done for all these samples, measuring total protein and specific gravity. A significant positive correlation was observed between percent sperm swelling and its motility, its total protein, and specific gravity. Also a significant negative correlation was noticed between percent sperm swelling and its count. Although there were overlapping ranges, different groups showed significant relations of percent sperm swelling among themselves. It is postulated that the sperm quality and not its quantity is the cornerstone of the hypo-osmotic test and that the physico-chemical properties of the sperm influences its swelling.

Response of CEDIA amphetamines assay after a single dose of bitter orange.

Bitter orange has recently been substituted as an ingredient in many "ephedra-free" dietary supplements used for weight loss. The primary active ingredient in bitter orange is synephrine. Previous reports have documented false-positive results from ephedrine with urine amphetamine assays. Because of the similarity in chemical structure of ephedrine and synephrine, it is hypothesized that ingestion of a bitter orange supplement may have the potential to cause false-positive results with urine amphetamine assays. The purpose of this study was to determine the response of the CEDIA Amphetamines Assay after ingestion of bitter orange. Six healthy adult male volunteers were administered a single oral dose of Nature's Way Bitter Orange, a 900-mg dietary supplement extract standardized to 6% synephrine. Urine specimens were collected at baseline and 3 and 6 hours post-administration. Additional urine specimens were collected from 1 subject at 9, 12, and 15 hours after administration. All specimens were analyzed by the CEDIA Amphetamines Assay. Urine specific gravity and pH also were measured. All urine specimens demonstrated a negative response to the CEDIA Amphetamines Assay. Urine specific gravity ranged from 1.007 to 1.028, and pH ranged from 5.0 to 7.0; thus, reducing the possibility that the negative results were caused by diluted specimens or reduced excretion of synephrine into alkaline urine. This information will be of value when health care providers or those who interpret drug screens are asked to provide consultation regarding the interference of bitter orange supplements with the CEDIA Amphetamines Assay. A single-dose of Nature's Way Bitter Orange was not found to cause a false-positive response to the CEDIA Amphetamines Assay in 6 healthy adult male volunteers.

Microgravimetric analysis of nerve edema.

A technique for microgravimetric analysis of nerve edema was used to demonstrate increased water content in hexachlorophene neuropathy. The method was modified from one used previously to quantify brain edema by using density gradient columns prepared with two nonaqueous fluids of high and low specific gravity. This method revealed a wide separation between hexachlorophene-treated and control nerves. Gradient position was then related to specific gravity, which averaged 1.03426 in hexachlorophene-treated nerves versus 1.04921 for control nerves. This, in turn, was correlated with the percentage of water in the tissue. Water content of hexachlorophene-intoxicated nerves was approximately 10% greater than control nerves. A major advantage of this technique is its sensitivity in detecting edema in small tissue samples.

Density heterogeneity of human lung mast cells.

Suspensions of enzymatically dispersed human lung parenchymal mast cells were fractionated according to density by flotation through discontinuous Percoll gradients and examined for their responsiveness to release stimulants and pharmacologic agonists. Mast cells localized to all six density fractions (I-VI) examined: densities varied from specific gravities of 1.053 gm/ml to 1.123 gm/ml. Most (67%) lung mast cells localized to fractions III and IV, corresponding to specific gravities of 1.077 to 1.088 gm/ml, respectively. Histamine content increased with density from 2.7 +/- 0.3 pg per cell in fraction 1 to 4.8 +/- 0.7 pg per cell in fraction VI (mean +/- SEM; n = 19). Fraction III was least responsive to high concentrations of anti-IgE than to any other fractions and, along with fraction IV, the most responsive to ionophore A23187. All fractions released the arachidonate mediators prostaglandin D2 and leukotriene C4 in response to anti-IgE. In four of eight lungs tested, formyl methionine peptide (10(-6) to 10(-4) mol/L) weakly elicited histamine release (3% to 6%) in fractions I and II cells. Compound 48/80 (0.1 to 10 micrograms/ml; n = 3) failed to induce histamine release in any fractions. The cyclic adenosine monophosphate-active drugs, isoproterenol (10(-4) mol/L), dibutyryl cyclic adenosine monophosphate (3 mmol/L), and isobutylmethylxanthine (3 X 10(-4) mol/L) inhibited anti-IgE-induced histamine release from all fractions equivalently. Dimaprit (3 X 10(-5) mol/L) and cromolyn sodium (10(-5) -3 x 10(-3) mol/L) failed to significantly inhibit any fraction.(ABSTRACT TRUNCATED AT 250 WORDS)

Polyuria and polydipsia. Diagnostic approach and problems associated with patient evaluation.

Primary disorders of water balance (central diabetes insipidus [DI], nephrogenic DI, and psychogenic polydipsia) should always be considered in the differential diagnosis of polyuria and polydipsia. In general, animals with these disorders have only one laboratory abnormality: a low urine specific gravity. In most instances, the more common causes of polyuria and polydipsia (e.g., hyperadrenocorticism, chronic renal failure, pyelonephritis, pyometra) have specific and obvious abnormalities associated with the complete blood cell count, the serum chemistry profile, and urinalysis. In some cases, however, a low urine specific gravity may be the only abnormality associated with these more common findings. The workup for polyuria and polydipsia can be tedious, time-consuming, expensive, confusing, and not without significant patient morbidity, especially in those cases with normal or near-normal blood work. This article focuses on the diagnostic approach and problems associated with diagnostic testing in patients with disorders of water balance.

Influence of dietary calcium and phosphorus on metabolism and production in laying hens.

1. The effects of different dietary concentrations of calcium (24 to 56-9 g/kg) and phosphorus (4-5 to 14-2 g/kg) on production and some aspects of metabolism were studied in laying hens. 2. Treatments did not affect egg numbers, food consumption, conversion efficiency of food to egg, bodyweight gain or mortality. 3. Increasing dietary calcium (Ca) significantly increased plasma Ca and inorganic phosphorus (P), breaking strength at the radius and egg specific gravity and significantly decreased plasma alkaline phosphatase and egg weight. 4. Increasing dietary phosphorus increased plasma P and decreased egg specific gravity significantly. 5. Plasma Ca, P and alkaline phosphatase and radius breaking strength were suitable indices of the Ca status of the hens.

Haemolymph characteristics of a copper-tolerant decapod Orconectes virilis (Hagen) (Astacidae, Crustacea).

The mean haemolymph volume in a copper-tolerant, intermoult (C2-C4) adult (more than a year old) Orconectes virilis, as determined by [14C-carboxylic] inulin dilution method, ranged from 61.17 microliters in females (mean wet body weight = 4.34 g; mean carapace length = 4.43 cm) to 61.79 microliters in males (mean wet weight = 5.73 g; mean carapace length = 4.49 cm). The mean haemolymph volume per cent varied from 14.5 (in females) to 31.1 (in males). Haemolymph from males had an average pH of 7.35, osmotic pressure of 414.1 mmol/kg H2O, and a specific gravity of 1.0603, while that from females averaged a pH of 7.48, osmotic pressure of 433.6 mmol/kg H2O and a specific gravity of 1.0469. Mean total haemocyte counts ranged from 3,434/mm3 (1,000/mm3 halinocytes, 1,125/mm3 hemi-granulocytes and 1,309/mm3 granulocytes) in males to 4,905/mm3 (1,525/mm3 halinocytes. 1,490/mm3 hemi-granulocytes and 1,890/mm3 granulocytes) in females.
