Action of plasma and liver extract from adult mice on the mitotic activity of young mouse liver.

Inbred C3HS male mice, standardized for periodicity analysis were used. A hundred and seventy 25 +/- 2 days old mice were injected at 16:00 hs with saline, plasma or liver extract from 27 mice 90 days old. Controls were made at 08/16, 12/20, 16/24, 08/40, 12/44, 16/48, 08/64, 12/68 and 16/72 (time of day/time post-injection). The mitotic activity of the hepatocytes and litoral cells were determined. The injection of small doses of extract and plasma inhibits the mitotic activity of hepatocytes during the first and second following days. A compensatory wave appears in the third day. The extract inhibits the mitotic activity of litoral cells in the first day of control only, whereas the plasma inhibits this variable in the second and third day.

[Formulation, fabrication, and evaluation of a cow's milk formula to Complementary Feeding National Program in Chile].

The complementary feeding national programme (PNAC) delivers 2 kg-month of dried full-fat cow's milk (LP) to 0-1 year old infants. The purpose of this work was to design, produce and evaluate nutrient stability and shelf life of a modified LP milk formula (LPM), which seems nutritionally more adequate for infants and is of similar cost than LP, to be included into the PNAC. LPM has the following attributes: P% 12.0 (LP 21.8%): casein/whey proteins ratio: 40 to 50/60 to 50 (LP 80/20); more digestible fats; essential fatty acids: > or = 4% of total calories; vitamins and minerals which cover 100% daily requirements for 0 to 6 months old infants; protein and energy can be modulated between 1.5 and 2.25 g/100 ml and 50-75 kcal/100 ml respectively. Forty tons of LPM were produced in accordance with our specifications for a field study. Quality controls on the product showed the production feasibility of such a formula at a lower price than LP. To determine shelf life, periodical analysis of peroxides, available lysine, ascorbic acid and linoleic acid were carried out in LPM stored for 1 year at 20 degrees C and 37 degrees C. It was found that LPM has an optimum shelf life of 7 months when packed in the same material that is now being in use for LP.(ABSTRACT TRUNCATED AT 250 WORDS)

Direct measurement of pulmonary and systemic flow during assisted circulation.

During biventricular assistance as a bridge to cardiac transplantation, the flow data provided by the pumping systems were compared with flow data obtained with the Fick method. There was a difference between the data sets only in the first 20 hr of observation, with the Fick method giving higher values. During the same period, analysis of the arterial and pulmonary pressure traces showed pulsatile activity to be related with the electrocardiogram's T waves. In the long run, the flow data provided by both methods were no different and the T wave-related pulsatile activity disappeared. The authors concluded that the Fick method represents a useful tool when measuring total flow during biventricular support.

Late EEG after-effects in humans following hyperventilation. I. A comparison between intero- and exteroceptive influences.

Using a modification of the computer period analysis of the EEG (Aladjov and Daskalova 1987) we studied the late after-effects (until the 30th min) in human EEG (monopolar leads at F1, C3, P3 and 01) following two kinds of influences: a) short interoceptive one: 3 min hyperventilation (HV) and b) a series of 5 trains of visual light flashes (VS), presented in 5-7 min intervals. The changes in the ongoing EEG in the 2nd min after each VS and in intervals of 5 min after HV were compared with the values before the stimulation. VS did not elicit long-lasting EEG after-effects, which characterized the single interoceptive influence (HV) and consisted mainly of beta-2 (24-36 Hz) EEG activity increase in all studied cortical regions (Nikolov and Kisselkova 1988). Moreover, the exteroceptive stimulation (VS) had no essential influence on the late EEG after-effects after HV, when both kinds of stimulation were applied in the same experiment.

[Threshold values for chemicals to prevent disease].

Proper interpretation of threshold limit values should always take into account that such limits are not absolute, but rather subject to change depending on advances in scientific knowledge. Threshold limit values derived from toxicologic study are best suited to evaluation of health risks of chemicals in the environment. Although more toxicologic information than is currently available would be desirable for the establishment of limit values, this should not prevent agreement on limits for more substances. "Better" threshold limit values would be forthcoming from epidemiologic studies, which are particularly rare in the FRG. Prospective studies measure current exposure; but appearance of detrimental health effects generally requires a lengthy latency period (e.g., decades in the case of cancer or cardiovascular disease). Threshold limit values permit monitoring and, if necessary, restriction of anthropogenic activity. Such restrictions are necessary, as shown by severe health damage which has occurred in the past (e.g., angiosarcoma due to vinyl chloride or neurogenic damage due to mercury in the workplace, tumors due to arsenic in drinking water, and methemoglobinemia in infants due to nitrite or renal damage due to cadmium in food). Evaluation of potential detrimental health effects for threshold limit values in environmental media is difficult because the effective dose cannot be determined. Monitoring of such limit values, which have already been incorporated into West German law, is relatively easy to implement, however (e.g. continuous outdoor air quality sampling and measurement, and periodic analysis of drinking water and foodstuffs). Since such monitoring may be performed close to the source, preventive measures should be easy to implement. Biological threshold limit values (biological monitoring) are essential to effective evaluation of the health effects of chemicals. Such limits should be established for more substances. When biological limit values are exceeded, however, it is generally difficult to determine the source(s) of exposure. Since individuals themselves cause the most damage to their own health e.g., through consumption of alcoholic beverages, cigarette smoking, or poor eating habits; the establishment of threshold limit values designed to protect individuals from themselves would be the most effective method of preventing environmental disease.

[Adaptation of the circulatory system of newborn infants with intrauterine hypotrophy].

A study was made of adaptation of the circulatory system in 40 full-term newborns with grade II and III intrauterine++ hypotrophy during the early neonatal period. Analysis of the hemodynamics demonstrated the study group children to have arterial hypertension, BP lability, and high peripheral vascular resistance. The conclusion has been made about the necessity of classifying intrauterine hypotrophy newborns with the group at risk for adaptation failure in the circulatory system.

Metabolism of 7 beta-alkyl chenodeoxycholic acid analogs and their effect on cholesterol metabolism in hamsters.

The metabolism of 7-ethyl- and 7-propyl-chenodeoxycholic acids was studied in hamsters. Both bile acid analogs were absorbed efficiently by the intestine and secreted into the bile at rates similar to those of chenodeoxycholic acid. After intraduodenal administration into bile fistula hamsters, the 7-alkyl analogs were present in bile as the glycine and taurine conjugates. The glycine/taurine ratios were: chenodeoxycholic acid, 1.9; 7-ethyl analog, 0.3; and 7-propyl analog, 0.2. After oral administration, during a 21-day feeding experiment, the 14C-labeled analogs were recovered quantitatively in the feces. Chenodeoxycholic acid was largely 7-dehydroxylated to lithocholic acid in the intestinal tract. In contrast, the 7 alpha-hydroxy group of the 7-alkyl bile acids was completely resistant to bacterial action. 7-Ethyl-chenodeoxycholic acid was transformed in part to a compound tentatively identified as 7 alpha-hydroxy-3-oxo- 7 beta-ethyl-5 beta-cholanoic acid while 7-propyl-chenodeoxycholic acid was excreted unchanged. In the hamsters used, the 7-alkyl bile acid analogs did not inhibit the bacterial dehydroxylation of chenodeoxycholic acid. At the end of the 21-day feeding period, analysis of the gallbladder bile showed that 7-methyl-, 7-ethyl-, and 7-propyl-chenodeoxycholic acids accounted for 38, 31, and 12% of total bile acids, respectively. The 7-alkyl bile acids decreased cholesterol absorption; the 7-propyl analog caused significant decrease in serum and liver cholesterol concentration. These experiments demonstrate that the 7-ethyl- and 7-propyl chenodeoxycholic acids, just like the 7-methyl-analog, are absorbed by the intestine and participate in the enterohepatic circulation.(ABSTRACT TRUNCATED AT 250 WORDS)

[Methemoglobin level and morphology of the blood in children from the State Children's Home in Gda'nsk-Oliwa].

Blood methemoglobin levels and blood cell counts were carried out in 64 children of the State Children's Home is Gdańsk-Oliwa. An excessive consumption of nitrites and nitrites leading to the increase in methemoglobinemia and anaemia observe in the previous studies stimulated the present investigations. Methemoglobinemia was detected in 14.1% of children together with marked anaemia especially in the youngest children. The authors suggest that partial elimination of nitrites and nitrates from the diet and periodical analysis of the drinking water and food seem necessary.

Infant mortality in the Western Galilee, 1964-86.

The infant mortality rate (IMR) in the Jewish and Arab populations in the Western Galilee was studied during the 2-year period 1985-86, and compared with those of previous surveys conducted since 1964-65. The IMR declined steadily during the two decades, from 33.6 to 8.5/1,000 in the Jewish population and from 49.2 to 18.2/1,000 in the Arab population. The decline was noted in both neonatal and postneonatal periods. Analysis of the causes of death showed that enteric and respiratory infections ranked high in the first and second surveys (1964-65 and 1970) and decreased to a low level in both population groups in recent years, but were still relatively high among Arab infants living in villages. The decrease in the IMR seems to be the result of an improvement in the general living conditions of the population and the efficiency of the health services.

[Lipids in the plasma of voluntary blood donors. A 2-year retrospective study (1985-1987)].

In a regional blood transfusion centre, we observed 45 episodes of hyperlipidemic plasma among the 45,000 units produced from 12,000 unpaid volunteer blood donors during a two year period. Analysis of fasting serum cholesterol and serum triglycerides showed that 26 of the 45 were raised in either one or both of the values. Dietary instructions were given, in most cases through their general practitioners, who were informed of the results. Control of serum lipids taken in average 9.5 weeks later by the general practitioner showed a remarkable decrease in triglyceride and a slight decrease in serum cholesterol. Among the 26 we found one case of non insulin dependent diabetes mellitus, three possible and three chronic alcohol abusers. Based on these findings, fasting serum cholesterol and serum triglyceride are now analysed in donors, who repeatedly show hyperlipemic plasma.

The lpr gene is associated with resistance to engraftment by lymphoid but not erythroid stem cells from normal mice.

This study demonstrates cell lineage-specific resistance to engraftment involving lymphocytes but not erythrocytes by the spontaneously autoimmune MRL/lpr mouse strain. In these experiments, MRL/lpr mice were lethally irradiated (1000 R) and reconstituted with normal A-Thy bone marrow stem cells. Periodic analysis from 6 wk to 6 mo posttransplantation demonstrated that the T and B cells of these chimeras were derived from the MRL/lpr host. However, in the same A-Thy----MRL/lpr chimeras, erythrocyte repopulation was completely of A-Thy donor origin. In contrast, control MRL/+ (congenic mice that differ from MRL/lpr at the lpr locus and do not develop accelerated autoimmune disease) recipients were successfully repopulated in both the lymphoid and erythroid compartments by the A-Thy donor cells.

Depression of delayed-type hypersensitivity in mice with hypothalamic lesion induced by monosodium glutamate: involvement of neuroendocrine system in immunomodulation.

Delayed-type hypersensitivity (DTH) was depressed in mice that had been treated with monosodium glutamate (MSG) in their suckling period. Analysis of the DTH depression by use of the macrophage migration inhibition assay showed dysfunction of DTH effector T cells. The neuronal loss of nuclei in the hypothalamus, which elaborates the corticotropin-releasing factor and the hypersecretion of adrenocorticotrophic hormone, was observed in the MSG-treated mice. Therefore, DTH response may be modulated by the neuroendocrine system.

[The EEG preferential frequency as an indicator of the degree of wakefulness].

Ever since electroencephalographic research began, interest has been focused on problems of the physiology, pathology, and symptomatology of the relationship between sleep and wakefulness. Following preliminaries, the present paper describes a combined procedure for the determination of the EEG preferential frequency, and by random sampling statistically examines its suitability for indicating the fluctuations of activation level as a control measure. A trained volunteer gradually increased his degree of wakefulness, reliably producing parallel changes in the preferential frequency in three frequency bands. A standardised manual period analysis as control produced largely the same results. The preferential frequency has considerable value for indicating the degree of wakefulness.

The effects of vitamin B6 supplementation on premenstrual symptoms.

A double-blind controlled study of the effects of vitamin B6 supplementation on premenstrual symptoms was conducted. Fifty-five women who reported moderate to severe premenstrual mood changes participated in the study. Symptoms were monitored prospectively through daily home record-keeping over a one-month baseline period followed by two months of treatment. Subjects were randomly assigned to receive daily supplements of 150 mg of vitamin B6 or placebo over the entire two-month treatment period. Analysis of covariance suggested that even though vitamin B6 may improve premenstrual symptoms related to autonomic reactions (eg, dizziness and vomiting) and behavioral changes (eg, poor performance and decreased social activities), a significant amount of physical and affective symptomatology remained during the premenstrual phase. In light of recently reported, potentially toxic effects of low doses of vitamin B6, our results call for caution in using this therapy for premenstrual symptoms.

A controlled trial of pentoxifylline (Trental 400) in intermittent claudication: clinical, haemostatic and rheological effects.

A formulation of pentoxifylline (Trental 400) has been claimed to increase claudication distance by improving red cell deformability and decreasing blood viscosity and platelet aggregation. In order to test this claim 30 stable claudicants recruited from one population took part in a double-blind randomised trial of placebo versus pentoxifylline. Both placebo and pentoxifylline treated patients improved subjectively and increased their claudication distance over the study period. Analysis of the blood results showed that the only apparent change was a tendency towards increased red cell filtration in the group treated with pentoxifylline; but no significant difference was found between claudicants and normal controls in the degree of red cell filterability nor did red cell filterability correlate with claudication distance. Pentoxifylline did not affect claudication distance or have any useful effect on blood flow properties, calling into question both its efficacy and suggested mode of action.

Tandamine--a new norepinephrine reuptake inhibitor. Clinical, psychometric and quantitative EEG studies in depressed patients.

Tandamine (AY-23,946)- a new norepinephrine reuptake inhibitor with practically no serotonin potentiation, MAO inhibition and anticholinergic activity-was administered in doses from 75 to 200 mg to a group of 20 hospitalized depressed patients. Weekly clinical evaluation including the ECDEU global score. Hamilton score and Zung self-rating score demonstrated a slight improvement which became statistically significant between the 1st and 2nd weeks of therapy. While the 'thymoleptic' properties of the drug were weak, a pronouned 'activating/stimulatory' effect of the drug was noted. Psychometric tests showed an increase in attention, concentration and psychomotor activity as well as an improvement in FPI personality dimensions 'depression' and 'inhibition', while the Taylor anxiety score did not reveal any changes. Digital computer period analysis of the EEG demonstrated, 6 h after oral administration of 50 mg tandamine, a decrease of slow waves, a significant increase of fast activity as well as a significant attenuation of the amplitude variability. Such changes are reminiscent of the pharmaco-EEG profile of psychostimulatory drugs and may represent the neurophysiological correlate for the activating/stimulatory properties of tandamine. Our finding suggest that the well-tolerated drug may be of some benefit for retarded depressions.

Nandrolone decanoate (deca-durabolin) in primary Sjögren's syndrome: a double blind pilot study.

The efficacy and side-effects of Deca-Durabolin (DD) were tested, in a double blind fashion, in twenty female primary Sjögren's syndrome (1 degree SS) patients. Ten randomly assigned patients received DD (100 mg IM bi-weekly) for six months, and ten others placebo, for the same period. Analysis of the results revealed that the DD-treated patients showed a moderate improvement of subjective xerostomia, a significant decrease of the erythrocyte sedimentation rate (ESR), and an overall improvement of their feeling of well-being, -judged by themselves and the investigator subjectively-, when compared with the placebo group. All the sicca objective parameters (results of Schirmer's I test, slit lamp eye examination after rose bengal staining, stimulated parotid flow rate measurements and labial minor salivary gland histopathology) were not significantly altered in either group. The clinical side-effects were the expected ones, i.e. hirsutism, hoarseness and an increase in libido, more pronounced in the DD-treated group. At the end of the study, one DD-treated patient, developed a diffuse well-differentiated B-lymphocytic lymphoma, which regressed spontaneously three months later.

Sodium and potassium intakes amongst free-living adult Nigerians.

Sixteen adult male and female Nigerians, aged between 18 and 48 years, participated in a 7-d study designed to estimate mean daily intakes of sodium and potassium and to determine their food sources. Food logs were kept during the 7-d study period. Analysis of data showed a mean daily intake of 8.1 g table salt with a range of 3.8 to 18.1 g which provided a mean daily intake of 3.2 g sodium. Mean sodium intake from foods alone was 0.85 g +/- 0.08 while sodium nutrient density was 2.3 g/1000 kcal. Potassium intake was 2.86 g +/- 0.2 or 1.46 g/1000 kcal. Mean sodium/potassium ratio of the diets was 1.5. Discretionary use of salt was 82 per cent of total intake with table salt supplying 78 per cent. Cereals and products provided the highest proportion of natural sodium while starchy staples supplied over 40 per cent of the potassium in the diet.
