Evolution of the differential regulation of duplicate genes after polyploidization.

In the 50 million years since the polyploidization event that gave rise to the catostomid family of fishes the duplicate genes encoding isozymes have undergone different fates. Ample opportunity has been available for regulatory evolution of these duplicate genes. Approximately half the duplicate genes have lost their expressions during this time. Of the duplicate genes remaining, the majority have diverged to different extents in their expression within and among adult tissues. The pattern of divergence of duplicate gene expression is consistent with the accumulation of mutations at regulatory genes. The absence of a correlation of extent of divergence of gene expression with the level of genetic variability for isozymes at these loci is consistent with the view that the rates of regulatory gene and structural gene evolution are uncoupled. The magnitude of divergence of duplicate gene expressions varies among tissues, enzymes, and species. Little correlation was found with the extent of divergence of duplicate gene expression within a species and its degree of morphological "conservatism", although species pairs which are increasingly taxonomically distant are less likely to share specific patterns of differential gene expression. Probable phylogenetic times of origin of several patterns of differential gene expression have been proposed. Some patterns of differential gene expression have evolved in recent evolutionary times and are specific to one or a few species, whereas at least one pattern of differential gene expression is present in nearly all species and probably arose soon after the polyploidization event. Multilocus isozymes, formed by polyploidization, provide a useful model system for studying the forces responsible for the maintenance of duplicate genes and the evolution of these once identical genes to new spatially and temporally specific patterns of regulation.

Sources of variation in the output of locust spiracular motoneurones receiving common synaptic driving.

1. The closer muscles of the left and the right spiracles of a thoracic segment are both innervated by two motoneurones, which spike in a variety of patterns during expiration. This paper seeks to explain the origin of these patterns. 2. No direct coupling between the two motoneurones is revealed. In an isolated thoracic ganglion both motoneurones spike at different frequencies with no tendency for their spikes to become synchronized. 3. The two closer motoneurones in one segment receive common, patterned depolarizing synaptic potentials during expiration caused by interneurones relaying information from the metathoracic ganglion. 4. The closer motoneurones of all the thoracic segments receive the same pattern of synaptic potentials from these interneurones. Despite this, the spiracles of one segment may remain shut while those on other segments continue to open and close rhythmically. 5. The interplay between common synaptic driving, the threshold of the motoneurones for spike initiation, and the tendency for a motoneurone to spike at a particular frequency even in the absence of interneuronal driving, explains the various patterns of spikes during expiration. Common synaptic driving imposes the same basic pattern of commands on all the motoneurones, but the individual motoneurones determine the final pattern of motor spikes. 6. To be effective in producing a patterned output, an input pattern must operate within narrow limits on either side of the threshold of the motoneurone. If the depolarization is too large, a high frequency of unpatterned spikes will result; if too small, then either there will be no output or a low frequency of spikes will result whose patterning will be affected by other inputs.

A putative mechanism for bog patterning.

The surface of bogs commonly shows various spatial vegetation patterning. Typical are "string patterns" consisting of regular densely vegetated bands oriented perpendicular to the slope. Here, we report on regular "maze patterns" on flat ground, consisting of bands densely vegetated by vascular plants in a more sparsely vegetated matrix of nonvascular plant communities. We present a model reproducing these maze and string patterns, describing how nutrient-limited vascular plants are controlled by, and in turn control, both hydrology and solute transport. We propose that the patterns are self-organized and originate from a nutrient accumulation mechanism. In the model, this is caused by the convective transport of nutrients in the groundwater toward areas with higher vascular plant biomass, driven by differences in transpiration rate. In a numerical bifurcation analysis we show how the maze patterns originate from the spatially homogeneous equilibrium and how this is affected by changes in rainfall, nutrient input, and plant properties. Our results confirm earlier model results, showing that redistribution of a limiting resource may lead to fine-scale facilitative and coarse-scale competitive plant interactions in different ecosystems. Self-organization in ecosystems may be a more general phenomenon than previously thought, which can be mechanistically linked to scale-dependent facilitation and competition.

Diversity of palmar pattern ridge counts in Iranian populations.

We analyzed bilateral palmar prints of 3,158 individuals from 13 Iranian populations of diverse origins for palmar pattern ridge counts. Right palms did not show greater counts than the left in the majority of the populations studied. Highly significant interpopulational variation was observed for most palmar areas of both males and females. Dendrograms of males and combined sexes showed a good fit with the ethnohistoric background of the populations studied. The results thus demonstrate variation in the palmar pattern ridge counts of populations of diverse origins and may be used as a good measure of population distance.

A transmissible developmental block in Tetrahymena thermophila.

When the amicronucleate mutant BI3840 of Tetrahymena thermophila is mated with normal micronucleate cells, it receives a pronucleus from its partner but there is no further nuclear development and the conjugants separate, retaining their original macronuclei. Both of these sexually mature exconjugants and any cells with which they are mated show an unconditional block in macronuclear development. Although prezygotic nuclear divisions, nuclear transfer and post-zygotic nuclear divisions appeared normal upon cytological analysis of Giemsa-stained conjugants, macronuclear development was invariably aborted. Since the original macronucleus was resorbed, the cells were rendered amacronucleate and they died. When no macronuclear development was initiated, as in crosses with the aneuploid strain A* (III), the exconjugants were viable and retained their original macronuclei. This pattern was invariant with three different strains serving as the original micronuclear source, and in the case of one non-BI3840 exconjugant, persisted for over 200 cell generations. Exconjugants from a cross of one of the micronuclear donors with strain A* (III) did not show arrested development when crossed. It thus appears likely that there is conjugal transfer of non-nuclear information originating in BI3840 which is self-replicating and which causes an arrest in macronuclear development.

Differential immunohistochemical reactions of carcinoid tumors.

Neuron-specific enolase, S-100 protein, and Leu 7 are present in cells showing neuroendocrine differentiation. Leu 7 recognizes myelin-associated glycoprotein, and recent evidence suggests that it further recognizes a subset of neurosecretory granules. Forty-six primary and metastatic carcinoid tumors from various sites were evaluated immunohistochemically with antisera to neuron-specific enolase, S-100, and Leu 7. Only one tumor, a rectal carcinoid, failed to stain with neuron-specific enolase. The remaining cases showed four patterns of staining: nuclear, diffuse cytoplasmic, globular cytoplasmic, or mixed. The pattern of reaction did not correlate with either the embryonic origin or histologic pattern of the tumor. Two patterns of staining were obtained with anti-Leu 7: diffuse cytoplasmic, or dot or ring-like deposition of reaction product. The latter pattern was entirely confined to appendiceal carcinoids. Reactivity with S-100 was in the form of dense positivity in individual cells having a stellate outline. Less intense cytoplasmic positivity and, uncommonly, a mixture of both types of staining were also seen. These histochemical results may lend support to recent evidence suggesting the existence of identifiable subsets of neurosecretory granules in neuroendocrine tumors, including carcinoids.

Cytosolic distribution of villin in M cells from mouse Peyer's patches correlates with the absence of a brush border.

BACKGROUND & AIMS: The follicle-associated epithelium (FAE) of Peyer's patches mainly consists of two cell types: absorptive enterocytes with a brush border and M cells without this apical specialization. To study the controversial ontogeny of M cells (mesenchymal vs. epithelial origin), the expression pattern of tissue-specific cytoskeletal proteins, markers of cell origin that play a crucial role in the specific shape of epithelial cells and brush border assembly, was investigated. METHODS: The localization of cytokeratins, vimentin, and villin was determined on mouse FAE using immunocytochemistry and electron microscopy. RESULTS: Epithelial-specific cytokeratins were expressed in both absorptive enterocytes and M cells, whereas vimentin was not detected in mouse FAE. Villin, a tissue-specific, actin-binding protein of the brush border, was expressed in the two cell types. This protein had an unusual cytoplasmic distribution in FAE cells lacking a brush border and in cells having an intraepithelial pocket filled with lymphocytes. CONCLUSIONS: The presence of villin and the absence of vimentin in M cells support the intestinal origin of M cells. The cytoplasmic distribution of villin provides a new identification criteria for M cells and reflects the reorganization of the F-actin network, which correlates with the inability of M cells to assemble a brush border.

[Alternative German clusters for the core conflictual relationship theme method (CCRT)].

The Core Conflictual Relationship Theme Method (CCRT) is without doubt one of the most widely used relationship structure instruments in the field of psychodynamic psychotherapy research. Despite the important and sustained criticism of the German translation of the original Anglo-Saxon description of clusters and of standard categories, research in German-speaking countries still relies on this translation. This study aimed at developing an alternative German cluster version while maintaining the existing set of standard categories. This alternative cluster structure for the three CCRT components showed higher inner consistency while at the same time making more clinical sense. It also is promising in that it can be used with CCRTs scored using the original translation of the instrument. This first investigation of relationship patterns using the original and alternative clusters confirmed the increase of discriminant and convergent validity of the CCRT-method.

Inferences on the role of insertion in a mutation accumulation experiment with Drosophila melanogaster using RAPDs.

The genetic variability for RAPDs band pattern was studied in a set of 157 mutation accumulation (MA) lines of Drosophila melanogaster. These MA lines were derived from the same isogenic base population and subsequently maintained by full-sib mating during 132 generations. The ancestral pattern of the original isogenic base can be unambiguously established as the consensus pattern of the MA lines and, because these lines are expected to be homozygous, dominance for band pattern is not a concern. Only repeatable changes in band pattern were considered. The number of ancestral bands detected implies that nine-nucleotide targets are enough for repeatable PCR amplification. Compared with the ancestral pattern, one MA line lost one band and two MA lines gained a new one. These results can be accounted for by the insertion of transposable elements occurring at a rate 0.07 < i < 0.21 per whole haploid genome and generation. This range is typical for Drosophila and consistent with the previously observed mobility for the roo family, supporting the generality of previous estimates of spontaneous mutation rates for morphological and fitness traits based on these MA lines. The sequence of one of the new bands suggests that the Idefix family is also active in the lines.

DNA replication by a DNA-membrane complex extracted from Bacillus subtilis: site of initiation in vitro and initiation potential of subcomplexes.

A DNA-membrane complex extracted from Bacillus subtilis was studied further as a model system for initiation of bacterial DNA replication in vitro. Of three subcomplexes purified from the crude complex by a combination of CsCl and sucrose gradient centrifugation, the synthetic capability of only one was inhibited significantly by streptovaricin, a known inhibitor of RNA primer formation. A selective enrichment in the level of this subcomplex was obtained by manipulating a thymine-requiring mutant. The synthetic capabilities of an enriched and nonenriched DNA-membrane complex were compared in the presence and absence of streptovaricin. Although the rate and extent of DNA synthesis per unit of protein were approximately the same in the absence of the antibiotic, there was a much greater inhibition of synthesis shown by the enriched complex in the presence of streptovaricin. Although the amount of DNA present in the putative initiation subcomplex was less than 0.3 to 0.4% of the total DNA present in the crude complex, such DNA, except for a few quantitative differences, was still representative of genomic DNA. Newly synthesized DNA hybridized to specific origin- and non-origin-derived restriction fragments of the B. subtilis genome. However, when an elongation inhibitor (ddCTP) was added, hybridization of such DNA to almost all of the nonorigin fragments disappeared or was reduced drastically, whereas origin region hybridization patterns remained strong. The highest level of hybridization in the origin region occurred with a BamHI (B7) restriction fragment of 5.6 kilobases that has been implicated by others as one site initiation in vivo (N. Ogasawara, M. Seiki, and H. Yoshikawa, Nature (London) 281:702-704, 1979; S. J. Seror-Laurent and G. Henckes, Proc. Natl. Acad. Sci. USA 82:3586-3590, 1985).

Malignant peripheral nerve sheath tumor of the vulva. A case report.

BACKGROUND: Malignant peripheral nerve sheath tumors (MPNSTs) are rare tumors in the head and neck, and their location in the genitalia, especially the vulva, is very rare. CASE: An 81-year-old woman with a solid, round vulvar tumor, 2 cm in diameter, underwent a radical hemivulvectomy after recurrence of MPNST with no evidence of neural involvement or stigmata of neurofibromatosis. The tumor cells were positive for S-100 protein and negative for cytokeratin, actin smooth muscle, HMB45 and alpha-1-antitrypsin. The biopsy diagnosis was MPNST with clear surgical margins. The patient was followed for nine months after surgery, without evidence of disease. CONCLUSION: MPNSTs constitute a small fraction of peripheral nerve tumors. Their site of origin, immunohistochemical staining pattern and ultrastructural morphology are thought to indicate a schwannian origin; however, the histogenesis of the tumor remains controversial. A definite relationship to a peripheral nerve may or may not be present. Diagnosis of MPNST is based on the pathologic appearance, immunohistochemistry and clinical features, such as predominant cell type (spindle cell), nerve involvement or differentiation, and association with neurofibromatosis disease. There have been few documented cases of MPNST arising in the vulva.

[Computer technology of gene geographic analysis of a gene pool: III. Derivation of trend surfaces].

A computer technology for obtaining a set of trend (background) maps on the basis of an original geographic map is reported. These trend maps reveal the patterns of distribution of the mapped trait. The technology allows one to shape the reliefs of the genetic surfaces that reflect patterns having different origin and appearing at different times, via changing the parameters of algorithms for averaging and approximation. The set of maps, which is obtained with the use of approximation methods, simulates the hypotheses of the through trend and of the mono-, di-, or polycentric distribution of the trait. New methods for deriving the trend surfaces on the basis of averaging throughout the area of either the "moving window" or the window of variable size ("changing window") are proposed. The informative significance of maps that are obtained with the use of Chebyshev polynomials of high (up to several tens of) degrees is demonstrated.

Variability in end-tidal PCO2 and blood gas values in humans.

The aims of this study were: (1) to determine the within-subject, between-day variability in end-tidal P(CO2) (P(ET,CO2)); (2) to determine whether the within-subject, between-day variability in P(ET,CO2) was caused by variations in metabolic acid-base status; and (3) to determine whether between-subject variations in blood gas variables arose predominantly through variations in respiratory or renal pH control mechanisms. Fourteen healthy males were studied, of whom 12 provided adequate data for further analysis. Each subject was studied on at least six different occasions, with each visit separated by at least 1 week. On each occasion, P(ET,CO2) was determined over a 4-10 min period using a fine nasal catheter taped just inside the nose, and an arterialised capillary blood sample was obtained from each ear and analysed for blood gases. The following results were obtained. (1) P(ET,CO2) showed a standard deviation (S.D.) for the within-subject, between-day variation of 1.06 mmHg. (2) Less than 5% (P = NS) of the variability in P(ET,CO2) could be explained by underlying variations in metabolic acid-base status. (3) The variation in blood gas values between individuals did not fit a pattern associated with either a predominantly respiratory or a predominantly renal origin. Furthermore, the pattern of variation in the blood gas data suggested that variations in the renal controller for pH and the respiratory controller for pH were not independent. The precise origins of variability in blood gas regulation remain obscure.

Pattern formation in plant embryogenesis: a reassessment.

Pattern formation in embryogenesis generates the basic body plan of flowering plants by establishing the diversity of position-dependent cell fates characteristic of the seedling. The concept of pattern formation, which was originally based on the analysis of Arabidopsis pattern mutants, is re-examined in the light of recent data that address the origin of the axis of polarity, the origin of the primary meristems as elements of the apical-basal pattern and the formation of the radial pattern of tissue layers. The available evidence from genetic, molecular and experimental approaches supports the notion that embryonic pattern formation occurs in steps involving communication between clonally unrelated cells.

The murine ependymoblastoma: growth pattern and survival in C57Bl/6J mice.

The murine ependymoblastoma is a transplantable tumor of cerebral origin. The growth pattern and survival times of the murine ependymoblastoma implanted peripherally and intracranially in non nude C57Bl/6J mice have been found to be predictable and consistent when examined by means of Tumor Cell Dose Assessment (end point solution), Tumor Growth and Survival Assessment. The results suggest that a greater tumor cell dose is required to generate peripheral tumor take than brain tumor take. This difference may result from a greater immunologic response to tumor implanted peripherally than into the immunologically privileged brain.

Relationship between steroid excretion patterns and breast cancer incidence in Israeli women of various origins.

A nationwide study of the steroid excretion patterns in postmenopausal Israeli migrant women demonstrated differences between high- and low-risk groups for breast cancer in the following variables: age at first parturition, number of pregnancies, number of live births, height, and weight. The direction of the differnces was in line with those observed for breast cancer patients. The groups also differed in the exretion of estriol, 17-ketosteroids, and allotetrahydrocortisol. Multiple regression analysis revealed that the exretion of estriol was significantly lower in population groups in whom breast cancer incidence was high. Possibly this trend--which has also been observed in adolescent and premenopausal women--reflected environmental influences on peripheral estrogen metabolism.

The origin of the oblique effect examined with pattern adaptation and masking.

The decreased visibility of obliquely oriented patterns as compared to horizontal or vertical ones is termed the oblique effect. The origin of the oblique effect in the chain of visual processing was examined by comparing the potency of oblique adapting gratings to the potency of horizontal ones. Oblique gratings (which were less visible but of equal physical contrast) were as powerful or more powerful than horizontal gratings as adapting stimuli. Obliquely oriented stimuli also produced a slightly stronger tilt aftereffect than stimuli near the cardinal axes. These results suggest that the diminished neural representation of oblique stimuli arises in the human cortex, rather than from impairments of sensitivity or resolution in the initial geniculo-cortical projection.

Fingerprint pattern frequencies in schizophrenics. Importance of ethnic origin and plexus visualization score ratings.

Our previous work with schizophrenics has suggested that the plexus visualization score (PVS), a characteristic derived from in vivo observation of skin capillaries, may be helpful in distinguishing between biologically different groups of patients. Preliminary fingerprint data demonstrated a significant difference in the frequency of whorls between high PVS and low PVS schizophrenics. A survey of fingerprints in control subjects revealed, however, that their ethnic background, even within the presumably 'mixed' Caucasian population of the USA, should also be taken into account. In the present study, the fingerprint pattern frequencies were, therefore, studied in 242 schizophrenics selected on the basis of both their ethnic origin and PVS ratings. Our results demonstrate a statistically significant difference in fingerprint pattern frequencies between high PVS and low PVS schizophrenics, even when they are matched for ethnic origin. The effect of the ethnic factor on fingerprint pattern frequencies in this patient sample is similar to the one previously reported for normal subjects.
