Detection of serum cardiac myosin light chains in acute experimental myocardial infarction: radioimmunoassay of cardiac myosin light chains.

To develop a more specific plasma test for myocardial infarction, antibodies specific for cardiac myosin light chains (CM-LC) were elicited that showed less than 3% cross-reactivity with skeletal muscle light chains. These antibodies were used to develop a radioimmunoassay for CM-LC that had a sensitivity of 20 ng (+/- 4 SD; P less than 0.001). Normal dog plasma showed no measurable concentrations of CM-LC (n = 6). Plasma samples from 10 dogs with experimental myocardial infarction produced by persistent left anterior descending coronary artery (LAD) occlusion were obtained at 0, 2, 4, 6, 24, 48 and 72 hours. CM-LC were first detectable in all 10 animals 6 hours after occlusion (97.98 +/- 14 ng/ml [mean +/- SEM]; P less than 0.001). Maximal CM-LC levels were usually obtained between 24 and 48 hours. Sham-operated open chest dogs (0--48 hours, n = 3) showed no measurable CM-LC in the plasma samples. Another group of 10 dogs were subjected to 5 hours of LAD occlusion, followed by reperfusion. In four dogs, CM-LC were detectable as early as 1 hour after reperfusion (81.88 +/- 37.75 ng/ml serum). Sera from all 10 dogs showed elevated levels of CM-LC (199.75 +/- 24.0 ng/ml) by 24 hours. Peak CM-LC concentrations were obtained in five dogs at 24 hours (247.0 +/- 35.28 ng/ml) and in another dog at 120 hours (245 ng/ml). Histochemical infarct size was determined to be 0.5--10% of the left ventricular mass at seven days by triphenyltetrazolium chloride staining. The specificity and sensitivity of this radioimmunoassay for detection of CM-LC, unique proteins to the heart, may be valuable in the diagnosis of myocardial infarction.

Optical masks prepared by using a liquid-crystal light valve for light-induced photorefractive waveguides.

The optical mask prepared by using a LCLV (liquid-crystal light valve) are first proposed for light-induced photoreactive waveguides in photorefractive materials. Employing this technique, various waveguide structures can be fabricated, e.g., Y- or multiple-branches waveguides, fiber-like waveguides, and Mach-Zehnder-like switches, and even whole optical circuits may be formed. A Y-branches waveguide and a fiber-like waveguide were demonstrated in a LiNbO3:Fe crystal. Several technical problems, such as intensities, resolutions, writing speed, and so on, were also discussed in detail. Using a LCLV with a fast response and a writing beam with a high intensity, the waveguide structures may be changed in real time.

Coupling of light from an LED into a thin light guide by diffractive gratings.

Ring-shaped and radial diffractive gratings are designed with rigorous diffraction theory to couple light of a nearly monochromatic LED into a thin planar light guide on the bottom side. The theoretical coupling efficiencies for ring-shaped and radial gratings are 41% and 66%, respectively. Optimized diffractive elements are manufactured with direct electron-beam lithography and reactive-ion-etching into SiO2 substrates. Good agreement between experimental and theoretical results for selected radial gratings is reached. Furthermore, the mass production tests using injection molding are carried out with good replicability.

White LED light coupling into light guides with diffraction gratings.

Radial diffractive gratings are used to couple light of a white LED into a light guide. Theoretical coupling efficiencies are evaluated with rigorous diffraction theory in a pure conical mounting. It is shown that when the refractive index of the grating increases from 1.46 to 2.05 the incoupling efficiency increases from 42% to 63%. Also, with the increasing refractive index the incoupling efficiency is shown to become more nearly uniform over the visible spectrum. Experimental results for the incoupled efficiencies and the color coordinates of the incoupled spectra are introduced for refractive indices n=1.46 and n=1.56.

Light extraction efficiency of a top-emission organic light-emitting diode with an Yb/Au double-layer cathode and an opaque Si anode.

We have computed the transmittances of four types of cathode--Yb/Au, Al/Au, Yb/Ag, and Al/Ag double layers--and the light extraction efficiencies of the top-emission organic light-emitting diodes with these cathodes, respectively, based on the characteristic matrix method and the dissipation spectrum model. Computations show that the Yb/Au cathode has a markedly higher transmittance than the other three types of cathode when the Yb and Au thicknesses in the Yb/Au cathode are, respectively, equal to the Al (or Yb) and Au (or Ag) thicknesses in the other three types of cathode. The power lost to the Yb/Au cathode due to the surface plasmon polaritons is the lowest, and hence the device with the Yb/Au cathode has the highest extraction efficiency. The transmittances for the four cathodes are also measured experimentally.

Ultraviolet light hardening in polymorphous light eruption--a controlled study comparing different emission spectra.

Polymorphous light eruption (PLE) is a common disorder characterized by a delayed, abnormal response to ultraviolet (UV) radiation, with a varied morphology of itching efflorescences on sun-exposed areas of the skin. Thirty-one PLE subjects were treated with either UVA (340-400 nm) or UVA and UVB (300-400 nm) phototherapy during spring 1987 (10 exposures to UV light). They were randomly allocated to these 2 groups. For subjects of the UVA group, the applied dose corresponded to their individual minimal tanning dose; for subjects of the UVA and UVB group it corresponded to approximately 3/4 of their individual minimal erythema dose. The sun protection effect was studied by a high dose of UVA (80-160 J/cm2; 340-440 nm) after the treatment period, by analysing the histidine content of the stratum corneum and the urocanic acid photoisomerization, and by evaluating the subjects' diaries. The patients were asked to expose their skin to sunlight at least 3 times after UV hardening in the following 2-10 weeks. The results of both the UVA provocation and of the natural sun exposure confirmed the success of UV hardening without the occurrence of severe side effects. The content of histidine and of its metabolite urocanic acid in stratum corneum was significantly increased during the treatment. These data are interpreted to be biochemical markers for improved sun protection.

[The patient with polymorphous light dermatosis. Skin type, hardening and other light-associated markers].

From 1985 to 1989 we interviewed 312 patients suffering from polymorphous light eruption (PLE). The interviews were based on a questionnaire dealing with the various light-dependent factors that exacerbate the disease. Of 90 patients who were tested with artificial UV-A and UV-B irradiation sources, 60 reacted with typical PLE lesions: (a) 27 patients to UV-A alone, (b) 12 to UV-B alone, and (c) 21 to both UV-A and UV-B. Using UV-A provocation tests we were able to determine the anamnestic criteria indicating a possible UV-A induction of PLE, e.g. occurrence in the shade, no protection from window glass, no benefit from conventional sunscreens, and occurrence in solaria. The period from experimental irradiation to induction of skin lesions was shorter in skin types I and II than in skin type III and IV. Hardening phenomenon was reported by 37% of our patients. Of the UV-A-positive patients, 38% showed the first presentation of PLE lesions at the height of summer, as against 64% of the total number of patients questioned. Additional lesions at non-irradiated skin sites occurred in 25% of our patients, the frequency rising with increasing duration of the tendency to PLE.

Polymorphous light eruption: successful reproduction of skin lesions, including papulovesicular light eruption, with ultraviolet B.

Thirty patients suffering from polymorphous light eruption (PLE), selected by criteria pointing to UVB sensitivity, were phototested for reproduction of skin lesions. Twenty-seven patients (90.0%) had symptoms compatible with or very similar to papulovesicular light eruption (PVLE). Of the 30 patients, 56.7% developed typical lesion of PLE at the test site by provocative phototesting with UVB. Eruptions of the immediate onset type were apt to be reproduced by multiple repeated irradiation, but those of the delayed onset type were reproduced by a single high-dose exposure. In addition, pruritus at the test site seen in all patients was thought to be an important diagnostic symptom. It was obvious that UVB played an etiological role in PLE, including PVLE.

Independent expression of a regulatory idiotype on heavy and light chains. A further immunochemical analysis with anti-heavy and anti-light chain antibodies.

The heavy (H) and light (L) chains of murine monoclonal autoantibody 62 reacting with thyroglobulin independently express idiotypic (Id) determinants that are very similar if not identical with the Id62 expressed on the intact protein. In this report, we describe the production and characterization of rabbit antibodies to isolated H62 and L62 chains to further prove that individual chains express Id62 in an immunogenic form. The results demonstrate that both chains are capable of eliciting antibodies that, after appropriate adsorption, behave like conventional anti-Id62 antibodies prepared against the intact antibody molecule. By direct radioimmunoassay binding, competition of Id binding and Western blot anti-H62 and anti-L62 antibodies identify as Id-positive the same group of IgG1, bind in a reciprocal fashion to H- and L-chains of parental monoclonal antibody 62, and detect Id62-positive polyclonal serum autoantibodies to thyroglobulin. We conclude that monoclonal antibody 62 expresses independently a similar Id on both polypeptide chains and the intact antibody molecule, or its isolated chains, induce qualitatively similar anti-Id responses. These results are discussed in light of the possible structure/function implication such autoantibodies may have within the Id network.

Topographical relationships between the monovalent and divalent cation binding sites of des-1-41-light chain bovine plasma protein C and des-1-41-light chain-activated bovine plasma protein C.

The paramagnetic effect of Mn2+ on the longitudinal relaxation rate (T1)-1 of 205Tl+, when both cations are bound to des-1-41-light chain bovine plasma protein C (GDPC) and its activation product, des-1-41-light chain-activated bovine plasma protein C (GDAPC), has been assessed by 205Tl+ NMR spectroscopy. A substantial shortening of the T1 for Tl+ bound to either protein was observed in the presence of Mn2+, an effect not noted upon substitution of Mn2+ with the diamagnetic cation Ca2+, which is known to bind to these proteins in a similar fashion to Mn2+. This paramagnetic effect was employed to estimate distances between the monovalent and divalent cation sites in these proteins, approximately 6.7 +/- 0.2 A with GDPC and 8.3 +/- 0.2 A in GDAPC. These data suggest that a conformational alteration occurs upon activation of GDPC which leads to an increase in the distance between the monovalent and divalent cation sites.

Papulovesicular light eruption. A defined subset of polymorphous light eruption.

A distinctive photodermatitis is seen commonly in tourists visiting Hawaii. Analysis of 150 cases revealed that the eruption is acute in onset following exposure to sunlight, is confined to exposed areas, and mainly affects young to middle-aged white women. The clinical presentation is remarkably uniform, consisting of papules, papulovesicles, or vesicles. These findings are consistent with a diagnosis of polymorphous light eruption (PMLE) and the histologic picture supports this diagnosis. Other clinical variants of PMLE were not seen in our patients, however, which suggests that this condition is more monomorphous than polymorphous. We suggest papulovesicular light eruption as a suitable name for this common and distinctive subset of PMLE.

[Intensive transillumination and spot-lighting with a new fiber optic light source (author's transl)].

An equipment for intensive transillumination is described which is able to provide approximate 8 fould higher angular emittance and 2.5 fould higher flux compared with customary used tools. The diameter of the fiber optic is about 1 mm and the angle of emittance of 30 or 60 degrees can be chosen. Even the experimental use of fiber optic with smaller diameter for intravitreal illumination has shown higher light intensity compared with other available light sources.

Effects of a myosin light chain kinase inhibitor, wortmannin, on cytoplasmic Ca2+ levels, myosin light chain phosphorylation and force in vascular smooth muscle.

Biochemical studies have shown that wortmannin is an inhibitor of myosin light chain (MLC) kinase (Nakanishi et al. (1992) J. Biol. Chem. 267: 2157-2163). To investigate the role of MLC kinase in smooth muscle contractions, we examined the effects of wortmannin on isolated smooth muscles of the rat aorta. Wortmannin (1 microM) decreased MLC phosphorylation and the amplitude of contractions induced by high K+ (72.7 mM) to a level seen at rest. This occurred without a change in cytosolic Ca2+ levels ([Ca2+]i). In contrast, wortmannin only partially inhibited the sustained contractions induced by phenylephrine (1 microM) and prostaglandin F2 alpha (PGF2 alpha, 10 microM) without a change in the [Ca2+]i. On the other hand, wortmannin (1 or 10 microM) reduced the increase in MLC phosphorylation induced by phenylephrine and PGF2 alpha to a level seen at rest. In the absence of external Ca2+, caffeine (20 mM) induced a transient increase in [Ca2+]i and force with an increase in MLC phosphorylation. Wortmannin completely inhibited the increase in MLC phosphorylation and contraction induced by caffeine without affecting the increase in [Ca2+]i. In the absence of external Ca2+, phenylephrine induced a small transient increase in [Ca2+]i, MLC phosphorylation and generation of force. This was followed by a small sustained contraction without an increase in [Ca2+]i and MLC phosphorylation. Wortmannin (1 microM) inhibited the transient phase of the contraction and the increase in MLC phosphorylation without affecting the transient increase in [Ca2+]i nor the sustained contraction. Wortmannin inhibited the Ca2(+)-induced contraction in permeabilized rat mesenteric artery, although it did not inhibit the Ca(2+)-independent, ATP-induced contraction in the thiophosphorylated muscle. These results suggest that wortmannin inhibits MLC phosphorylation due to an increase in the entry of Ca2+ or through the release of Ca2+ from the sarcoplasmic reticulum. The results also suggest that the activation of receptors by norepinephrine and PGF2 alpha induces a contraction via a MLC phosphorylation-independent pathway or through a pathway which is dependent on the resting level of MLC phosphorylation. We conclude that wortmannin is a useful tool in studies of the physiological role of MLC kinase.

The variable coupling between force and myosin light chain phosphorylation in Triton-skinned chicken gizzard fibre bundles: role of myosin light chain phosphatase.

The mechanism responsible for the regulation of smooth muscle tone at low levels of myosin light chain (MLC) phosphorylation is still poorly understood. According to one model, so-called latchbridges, which contribute to force maintenance at low levels of MLC phosphorylation, are generated by dephosphorylation of attached and phosphorylated crossbridges. The model predicts that the force generated for a given level of MLC phosphorylation depends on the activity of the MLC phosphatase. We tested this hypothesis by reducing the activity of the phosphatase by at least 80% in two ways: inhibition with okadaic acid and extraction. Under both conditions, higher levels of MLC phosphorylation were required to support a given level of force, suggesting a decreased flux of attached phosphorylated to attached dephosphorylated crossbridges, as predicted by this model. Although, under both conditions, the relationship between force and MLC phosphorylation was shifted to the right, the curves did not superimpose as would have been expected if the phosphatase activity were the only determinant of the coupling between force and phosphorylation. In the extracted fibres, two more proteins, calponin and SM22, were significantly reduced in addition. Therefore, these proteins might be involved in modulating the relationship between force and MLC phosphorylation.

The role of myosin light chain kinase-dependent phosphorylation of myosin light chain in phorbol ester-induced contraction of rabbit aorta.

We investigated the role of 20 kDa myosin light chain (MLC20) phosphorylation in contractions following protein kinase C (PKC) activation by 12-deoxyphorbol-13-isobutyrate (DPB) in rabbit aortae. DPB induced a sustained contraction and phosphorylation of MLC20 independent of a change in cytosolic Ca2+ ([Ca2+]i). Phosphorylation on Ser19 of MLC20, which is a target site of MLC kinase (MLCK), was 9.2 +/- 5.1% and 22.3 +/- 4.9% of the phosphorylation caused by KCl, at 5 and 30 min of application of DPB, respectively. When KCl-precontracted muscles were rinsed with Ca2+-free, EGTA solution, [Ca2+]i rapidly declined, MLC20 was dephosphorylated and the tension decreased. If DPB was present in the Ca2+-free solution, the relaxation and the dephosphorylation of either total MLC20 or Ser19 were inhibited. The phospholipase A2 inhibitor ONO-RS-082 partially antagonized the effects of DPB on the tension and the MLC20 dephosphorylation. In Ca2+-free solution, DPB induced a contraction smaller than that in normal solution without an increase in MLC20 phosphorylation, and the contraction was also sensitive to ONO-RS-082. These results suggest that a part of MLC20 phosphorylation following PKC activation is due to inhibition of MLC20 phosphatase and the phosphorylation is responsible for the contraction. Furthermore, a mechanism independent of [Ca2+]i and phosphorylation may play a significant role in the PKC-dependent contraction. The involvement arachidonic acid is suggested, not only in the inhibition of dephosphorylation but also in the Ca2+-independent regulation of contractile proteins.
