Immunosuppression in murine malaria. II. The primary response to bovine serum albumin.

The primary antibody response to alumadsorbed bovine serum albumin was depressed in CBA mice infected with Plasmodium yoelii yoelii. Responses initiated within approximately 3 weeks of this malaria infection were reduced in quantity, but not in avidity. Responses initated later were normal. A depressed primary response was also seen in infected compared to control splenectomized mice; this was accompanied by impaired priming for a secondary response.

Disarming the bluebottle: treatment of Physalia envenomation.

Clinical tests of the efficacy of various treatments of bluebottle (Physalia physalis) stings were conducted on 20 volunteers. Vinegar and aluminium sulphate solution (Stingose) gave a significant reduction in pain 15 minutes after application when compared with the control sites which were treated with sea water. The application of methylated spirits caused a significant increase in pain compared to salt water.

Adjuvant effect of bacterial LPS and/or alum precipitation in responses to polysaccharide and protein antigens.

A conjugate of a hapten (NIP) and a strongly antigenic protein chicken gamma globulin (CGG), when injected in soluble form into mice, induced weak primary responses, as weak as responses induced by conjugates of NIP (or other haptens) to polysaccharides Ficoll or alpha (-1-6) dextran. Mean concentrations of anti-hapten antibodies on day 14 varied within the range of 37-105 micrograms/ml (C57BL mice) or 14-38 micrograms/ml (CBA mice). The NIP-protein conjugate administered in alum-precipitated form induced 100 times higher primary antibody responses. Alum-precipitation of NIP-Ficoll made it a modestly stronger antigen than soluble NIP-Ficoll. When lipopolysaccharide (LPS) was injected together with any of the soluble antigens, the mice produced plenty of anti-hapten antibodies regardless of whether the antigen was hapten-polysaccharide or hapten-protein conjugate. Concentrations on day 14 varied from around 400 micrograms/ml to approximately 1600 micrograms/ml. LPS had a similar adjuvant effect on antidextran responses. LPS alone induced a polyclonal immunoglobulin production, and the immunoglobulin produced included 'anti-NIP' or 'anti-dextran' detectable in the solid phase antibody assay. These 'antibodies' induced by LPS alone were almost totally mercapto-ethanol-sensitive and poorly detectable by Farr assay or the bacteriophage assay. The response to the LPS+antigen combination was specific for the antigen and included both mercapto-ethanol-sensitive and resistant antibodies.

[Dynamics of homocytotropic antibody production in pigs].

Homocytotropic antibody (HTA) production in pigs was stimulated by sensitization with alum precipitated equine serum or with equine serum in Freund's complete adjuvant. Two doses of appropriate antigen were given in 7 days interval simultaneously with heat inactivated suspension of Bordetella pertussis organisms. HTA were detected in 6 of 10 pigs sensitized with alum precipitated serum and in 4 of 6 pigs sensitized with serum in Freund's complete adjuvant. In pigs sensitized with alum precipitated serum HTA appeared at day 6 after the first antigen administration. In pigs sensitized with serum in Freund's complete adjuvant HTA appeared at day 18 after the first antigen administration. Time course of appearance of HTA was about 10 days in both groups. Alum precipitated serum elicited considerably higher titres of HTA than equine serum in Freund's complete adjuvant. The passive cutaneous anaphylaxis activity of the HTA sera was located in the 7S fraction.

[Immune responsiveness of high and low antibody producing lines of mice against an antigen non-cross-reacting with the selection antigen (author's transl)].

Mice genetically selected for high (HL) or low (LL) antibody responsiveness to sheep erythrocytes (SE) also differed in the immune response to other antigens. We have studied in details the non-specific effect of the selection on responsiveness to bovine seralbumin (BSA) that is an antigen non cross-reacting with SE. BSA has been used either heat-aggregated (H-BSA) or alum-precipitated (Al-BSA). The characteristics of the response to H-BSA and to SE both injected intravenously were similar in several aspects: ranges of interline difference in agglutinin response were equivalent; minimal immunogenic doses were lower in HL than in LL mice. As for SE, the use of an adjuvant (Al-BSA) increased the response of LL more than that of HL mice, so the interline difference decreased. The interline difference between minimal immunogenic doses was weaker with Al-BSA than with H-BSA (100 times and 10,000 times respectively). The threshold dose for induction of immunologic memory was lower in the HL than in LL mice. For Al-BSA this threshold dose was higher than the minimal immunogenice dose in both lines; for H-BSA it was higher (in HL) or equal (in LL) to the minimal immunogenic dose. The results of this article confirm that the selective breeding for SE responsiveness has modified the antibody response to BSA in a similar way. The group of genes controlling the level of antibody response are also responsible for the induction of immunological memory.

The immunogenicity of K99 antigen in whole cell bacterins of Escherichia coli.

K99 antigen in Escherichia coli whole cell bacterins was immunogenic in rabbits and mice. Mice vaccinated subcutaneously and bled three weeks later had a serum antibody response which was dose dependent. The dose response on dilution of bacterins was shown to be mainly due to dilution of K99 antigen, rather than the reduction in adjuvant or bacterial cell concentration. The procedure appears to be a satisfactory one for immunogenicity testing of bacterins containing K99 antigen.
