[Effect of vitamin D3 on the formation of proteoglycans and tissue calcification].

Nonspecific calcinosis engendered by vitamin D3 is associated with extraosseous calcium deposits, in connection with which proteoglycans (PG) can be observed. After administration of vitamin D3 tissue samples from cardiac muscle and aorta of rats were taken and traced ultrahistochemically by means of the acriflavin-phosphotungstic acid contrast-method. The calcification began first with an increase in PG in the cell membranes of the sarcolemma, in the ground substance, and in the capillaries of the interstice. Subsequently circularly formed concentric layers of calcium encrustations appear in the above-mentioned locations. Large amounts of PG were also observed prior to the formation of crystalline fascicular calcium deposits in the ground substance and in the elastic fibers of the media of the aorta. This process began at the outer edges of the media and continued towards their interior, resulting in a loosening of the bond between the elastic fibers and the sarcoplasmic continuations of the smooth muscle cells. The extraosseous calcification resulting after the administration of vitamin D3 can therefore be considered not only as a result of hypercalcemia, but also as a result of stimulation of the production of ground substance through vitamin D3. This form of calcification represents a characteristic pathophysiological process which is differentiated from the dystrophic form in that it begins without previous cell damage.

Establishment and characterization of two human mixed mesodermal tumor cell lines from the same patient.

A cell line designated HIRS -BM was established from fluid aspirated from the sternal bone marrow of a 16-year-old female. Another cell line ( HIRS -PB) was derived from the peripheral blood of the same patient. Both lines grew well, multilayering rapidly without contact inhibition, and 62 serial passages were successively done within 28 months. Both cultures contained spindle- or fibrous-shaped cells that revealed neoplastic and pleomorphic features, and these cells were characterized as possessing cross-striations in the cytoplasm. The cross-striations were detected by phosphotungstic acid hematoxylin stain. Some elongated cells were stained positively with anti-myoglobin by use of periodic acid-Schiff methods. The primary tumor in the uterus was diagnosed as a mixed mesodermal tumor composed of adenocarcinoma and rhabdomyosarcoma cells. The karyotype exhibited hyperploidy and large submetacentric marker chromosomes, and the modal chromosome number was 84. No difference was found between the 2 cell lines except for growth behavior and heterotransplantability . HIRS -BM cells grew more rapidly and were highly transplantable. The HIRS -BM cells were transplanted into the subcutis of BALB/c nude mice and produced mixed mesodermal tumors resembling the uterine tumor, while the HIRS -PB cells could not be transplanted. Due to the histogenesis of the mixed mesodermal tumor being's obscure with histologic observations only, this study was performed to obtain data by tissue culture of the tumor and resulted in support of the combination theory reported in the literature in regard to tumor.

[Morphologic aspects of intravascular disseminated coagulation. Apropos of 100 cases].

The authors report a morphological study of one hundred cases of disseminated intravascular coagulation (D.I.C.) of various origins. In each case laboratory findings demonstrated an authentic consumption coagulopathy. In all cases organ biopsies were performed either in vitro (rarely), or mainly immediately after death or at autopsy. Optic microscopy was performed for all of them (3 stains were chosen: hemalum eosin saffron, Mallory's phosphotungstic acid hematoxylin stain, P.A.S.). For a smaller number of cases ultrastructural microscopy and immunofluorescence were performed. As far as microscopic findings are concerned, the authors insist particularly upon intravascular microthrombi (fibrinous type and hyalin type). Haemorrhages and particularly necrosis, because of multiple origin, are not detailed. The repartition of these microthrombi is particularly detailed (in connection with the different organs, etiologies, clinical and/or biological findings, treatments). A review of literature is reported.

Changes in the length and width of the postsynaptic density, the width of the intersynaptic density and the synaptic cleft in the cerebral cortex synapses of rats exposed to prolonged aerogenic hypoxia during early ontogenesis. An electron microscopic morphometric study.

Infant rats, together with the female, were exposed daily from birth to the age of 17 days (except for the 6th, 7th, 13th and 14th day) to a simulated attitude of 7,000 m. On the 18th day, 20 hours after the last exposure to hypoxia, the animals were perfused. In each of 118 synapses from the two control animals and 162 synapses from the two experimental animals (type I synapses, second cortical layer of the somatosensory cortex, impregnated with phosphotungstic acid), we measured A) the maximal thickness of the postsynaptic density and B) the minimal thickness of the postsynaptic density, C) estimated the thickness of the postsynaptic density and measured D) the distance between the pre- and the postsynaptic density, E) the thickness of the intersynaptic density and F) the length of the postsynaptic dense projection. In the experimental animals we found value A) to be 25% lower, value B) 28% lower, value C) 30% lower, value D) 5% lower, value E) 2.5% lower and value F) 17% higher than in the controls. The differences are statistically significant (evaluated by a t-test). It was found that, under our experimental hypobaric conditions, the maturation of synapses is retarded or modified.

Ultrastructure of bacteriophages used for typing of Staphylococcus aureus.

This paper reports the results of a study on the morphology, size and homogeneity of the bacteriophages used for typing Staphylococcus aureus. Fifty-five bacteriophages were studied. Preparations for the electron microscope were made directly from lysates or from purified and concentrated bacteriophages, and were contrasted with neutralized phosphotungstic acid (KPT) and examined under an electron microscope at x 40 000 microscopic magnification. The studied bacteriophages differed with regard to size and shape. The bacteriophages were classified into two morphological groups: BI and BIII. In the BI group, there morphological types were distinguished: B14, B15 and B16. Only one morphological type was distinguished in the BIII group, designated by the symbol BIII2. Only lysates of 42C, 70 and D bacteriophages contained virions of two morphological types: BI6 an BIII2. Until now, it is not clear whether the bacteriophages are a mixture of two different virions, or whether they represent variation in the size of their heads. Of the 55 bacteriophages studied, 25 were examined by other investigators. The results showed that of the 25 bacteriophages 5 belonged to different morphological types. It was concluded that the sets of standard bacteriophages should be checked by means of the electron microscope.

A coat of glycoconjugates on the inner surface of the lysosomal membrane in the rat kidney.

After perfusion fixation of the rat kidney with glutaraldehyde, and postfixation of the renal cortex with osmium-low ferrocyanide (40 mM OsO4 +6 mM K4Fe(CN)6 in 0.135 M phosphate buffer, pH 8.0), secondary lysosomes of proximal tubule cells carry a coat of electron dense material on the inner surface of the lysosomal membrane. This coat separates matrix and membrane of lysosomes, and corresponds in location and width to the electron translucent halo of conventionally processed lysosomes in TEM. The material which forms the coat, is stained by phosphotungstic acid at pH 0.3, and by periodic acid - thiocarbohydrazide - silver proteinate more intensively than the cell surface coat of the same cell; it contains a high concentration of hydroxyl, vicinal-glycol and alpha-aminoalcohol groups.

Acute influence of smoking on plasma lipoproteins.

This study was designed to investigate acute effects of smoking on plasma lipoproteins. Twelve fasting smokers inhaled the smoke of two cigarettes. Blood samples were taken before and 30 min after smoking. Triglycerides decreased in plasma from 136.8 +/- 39.8 to 108.0 +/- 34.9, in VLDL from 79.8 +/- 31.4 to 58.4 +/- 28.5 mg%. Cholesterol fell in HDL3 from 29.9 +/- 5.0 to 26.6 +/- 5.8 mg%. Determination of HDL-cholesterol with the phosphotungstic acid precipitation method revealed a reduction from 45.7 +/- 10.2 to 40.3 +/- 11.1 mg% after smoking. All C-apoproteins decreased in VLDL as an entity. Apoprotein A-I was diminished in plasma from 137.5 +/- 23.1 to 118.2 +/- 25.4, and in HDL3, from 97.1 +/- 12.1 to 85.0 +/- 13.1, apoprotein A-II in plasma from 28.6 +/- 8.9 to 24.7 +/- 6.9 and in HDL3 from 21.1 +/- 8.4 to 17.4 +/- 5.7 mg%. Zonal ultracentrifugation of D less than 1.063 g/ml lipoproteins confirmed the influence of smoking on VLDL. In LDL, an increase in cholesterol esters, mainly cholesterol linoleate, from 57.3 +/- 30.2 to 67.4 +/- 25.9 mg% was observed after smoking. The HDL2 subclass was shifted toward a slightly lighter hydrated density, but the major effect of smoking was seen in HDL3. Total cholesterol decreased from 30.5 +/- 7.3 to 22.3 +/- 6.6 mg%, due to a fall of all major cholesterol esters from 23.7 +/- 7.2 to 15.8 +/- 6.9 mg%. HDL3-phospholipids were reduced from 74.9 +/- 37.0 to 46.0 +/- 23.9 mg%.(ABSTRACT TRUNCATED AT 250 WORDS)

Dictyosome polarity and membrane differentiation in outer cap cells of the maize root tip.

Outer rootcap cells of maize produce large numbers of secretory vesicles that ultimately fuse with the plasma membrane to discharge their product from the cell. As a result of the fusion, these vesicles contribute large quantities of membrane to the cell surface. In the present study, this phenomenon has been investigated using sections stained with phosphotungstic acid at low pH (PACP), a procedure in plant cells that specifically stains the plasma membrane. In the maize root tip, the PACP also stains the membranes of the secretory vesicles derived from Golgi apparatus to about the same density that it stains the plasma membrane. Additionally, the membranes of the secretory vesicles acquire the staining characteristic while still attached to the Golgi apparatus. The staining progresses across the dictyosome from the forming to the maturing pole, thus confirming the marked polarity of these dictyosomes. Interestingly, the PACP staining of Golgi apparatus is confined to the membranes of the secretory vesicles. It is largely absent from the central plates or peripheral tubules and provides an unambiguous example of lateral differentiation of membranes orthogonal to the major polarity axis. In the cytoplasm we could find no vesicles other than secretory vesicles bearing polysaccharide that were PACP positive. Even the occasional coated vesicle seen in the vicinity of the Golgi apparatus did not stain. Thus, if exocytotic vesicles are present in the maize root cap cell, they are formed in a manner where the PACP-staining constituent is not retained by the internalized membrane. The findings confirm dictyosome polarity in the maize root cap, provide evidence for membrane differentiation both across and at right angles to the major polarity axis, and suggest that endocytotic vesicles, if present, exclude the PACP-staining component.

[Alcoholic hyaline-like bodies in nerve cells of the caudate nuclei in various diseases of children].

Intracytoplasmic hyaline bodies were noticed in nerve cells of the caudate nuclei of the infantile diseases: Leigh's encephalomyelopathy (12-year-old, male), Wilson's disease (14-year-old, male), Morquio's disease (12-year-old, male), pseudoulegyric type of hepatocerebral degeneration (19-year-old, male) and Schilder's disease (15-year-old, male). The hyeline bodies were noticed as linear, striate, dendritic or antler-like in form and stained pink or red with hematoxylin-eosin, orange or red with tmasson trichrome, and dark blue or purple with phosphotungstic acid hematoxylin. The characteristics of the neuronal inclusions have much resemblance to the "alcoholic hyaline-like bodies" which were described by K. Kojima in the caudate nuclei of alcoholics. The results may indicate that the presence of the hyaline bodies in the caudate nucleus is not pathognomonic for alcoholic intoxication, but may suggest the presence of a certain common metabolic disorder among the described diseases.

Histochemical demonstration of glucagon and serotonin by a fluorescent method in the rat submaxillary gland.

The submaxillary glands of young rats were examined in vivo and in vitro. Yellow fluorescence in some cells treated with o-phthalaldehyde was observed and they yielded a positive reaction on staining with phosphotungstic acid haematoxylin indicating the presence of glucagon. These cells were localized between secretory endpieces of the gland. In rats treated with L-5-hydroxytryptophan (L-5-HTP) cells similar in shape and size showed yellow fluorescence after the action of formaldehyde vapour. The results of histochemical studies and ultrastructural analysis seem to indicate the presence of cells similar to APUD endocrine cells in the rat submaxillary gland.

The paraxial structure of the flagellum of trypanosomatidae.

The flagella of several Trypanosomatidae (Trypanosoma cruzi, Herpetomonas samuelpessoai, Leptomonas samueli, Herpetomonas megaseliae, and Crithidia harmosa) were studied. Besides the axoneme, they have a filamentous, latticelike structure, the paraxial or paraflagellar rod. This structure was not observed in the flagellum of the intracellular spheromastigote form of Trypanosoma cruzi, but it appeared when the transformation of epimastigote and trypomastigote stages occured. Cross sections of the flagella show that the paraxial structure maintains a fixed position relative to the axonemal microtubules, being localized in a region comprised between doublets 4 and 7. Flagella of T. cruzi and H. samuelpessoai were isolated using the detergent lubrox PX. Most of them did not have a flagellar membrane. However, the paraxial structure remained associated with the axoneme. By negative staining, short projections were seen connecting the paraxial structure to the axonemal microtubules. The paraxial structure did not stain with phosphotungstic acid as occurs with the peripheral doublet microtubules, but it is formed by microfilaments longitudinally oriented in relation to the axoneme crossed in two directions by oblique filaments which make an angle of 45 degrees with the longitudinal ones.

The histomorphology of pancreatic islets in the bandicoot, Bandicota bengalensis (Gray).

Histomorphology of pancreatic islets are first time reported in the Bandicoot, Bandicota bengalensis. Histological observations are made with conventional staining techniques on the pancreatic islets of Bandicota bengalensis. The endocrine tissues present all over the pancreatic mass through large number of islets are confined to spleenic region and exhibited various shaped from round, oval, oblong to irregular. Three main types of cells viz., A-1, A-2 and B cells are observed. The commonly used silver techniques failed in differentiation of A-1 and A-2 cells. However, A-1 cells were stained with toluidine blue, while A-2 cells with phosphotungstic acid haematoxylin. Beta cells are more in number and situated in the centre of the islet surrounded by peripheral alpha cells. Presence of a large number of B cells in an islet indicates the vital role in insulin in the blood sugar homeostasis of the bandicoot.

[Human ovarian myosarcoma and its cell culture (author's transl)].

One case of a very rare myosarcoma of ovarium origin was observed and establishment of its cell lines from the fluid obtained by paracentitis was accomplished. This report is on the determination of the tumor and the biological characteristics of its cultured cells. (Determination of the tumor) (1) This tumor was diagnosed as a malignant of the right ovary, using bimanual examination, selective angiography of the uterine artery, ultrasonic tomography, and abdominal ascitic cell examination. (2) The morphological type of tumor was based findings of hematoxylin eosin. Silver, Mallory and phosphotungstic acid hematoxylin stainings. The tumor was diagnosed as an ovarian sarcoma and probably myogenic. However, a clear cross strain could not be demonstrated. (Cell biological characteristics) (1) Morphological characteristics of cultured cells: a) Phase-contrast microscopy and Papanicolaou staining revealed round or short and spindle shaped cells, with markedly enlarged nucleoli. These proliferated without exhibiting any tendency of contact inhibition. Also frequently recognized were giant cells, multinuclear cells, an some bizarre such as a starfish-shaped cell. b) Electron microscopy revealed that bundles of fibrils regarded as myofibrils existed in the cytoplasma, with dense patches within them, but a cross strain could not be demonstrated. (2) Morphological characteristics of the tumor cells grown in a nude mouse: a) The tumor cells demonstrated similarities to the original tissues by of H.E., Silver, and Mallary stainings. b) A number of fibrils recognized by the electron microscopic observation were 100 A in width and were arranged in a concentric circles. In addition to the light microscopic findings, the above findings indicated that this cell line originated from myogenic sarcoma. (3) Biological characteristics of the cultured cells: a) The number of chromosomes varied widely and spread aneuploidically, the highest chromosome number was 76. b) Doubling time, Saturation density, and plating efficiency was 31.2 hr, 1.2 x 10(5)/cm2 and 43.9% respectively. c) These cells had a high sensitivity for trypsin and started morphological change rapidly.

Simple, direct measurement of lipoprotein X in serum.

This relatively simple method for quantitative estimation of lipoprotein X is based on assay of phospholipids in precipitates of lipoprotein X. We first remove other lipoproteins by precipitation with phosphotungstic acid solution. Lipoprotein X is then precipitated from the supernatant fluid with an alkaline solution of magnesium ion, and phospholipid is determined in this precipitate. Results are linearly related to concentration of lipoprotein X. The CV is less than 3%. Results correlated well with those for agar gel electrophoresis.

Influence of triglyceride-rich lipoproteins on nephelometry of apolipoprotein A-I.

Measurement of apolipoprotein A-I in native human sera by endpoint or rate nephelometry may give falsely high results for sera that have a high triglyceride content. Such erroneous determinations of apolipoprotein A-I can be avoided if one instead assays supernates of sera from which apolipoprotein B-containing particles have been precipitated with phosphotungstic acid and MgCl2. A high triglyceride concentration in serum does not impair the completeness of the precipitation of apolipoprotein B-containing lipoproteins or the analytical recovery of apolipoprotein A-I. In a prospective epidemiologic study, by using rate nephelometry we found that there were different mean concentrations for apolipoprotein A-I in supernates of sera from men (1.36 g/L) and women (1.49 g/L). Furthermore, the known risk factors for atherosclerosis showed different correlations with apolipoprotein A-I than with high-density lipoprotein cholesterol.

[Reorganization patterns in the synaptic architectonics of the neocortex in the postresuscitation period].

Staining of brain tissue with phosphotungstic acid was used to assay interneuronal contacts in the molecular layer of the cerebral cortex of rats with a history of clinical death because of hemorrhage and asphyxia. Common regularities of reconstruction of neocortical architectonics during the resuscitation period were defined. Marked reduction in the number of synapses was discovered in the neocortex. The main form of destruction of the contacts was the light type of destruction. Hypertrophy of the remaining synapses occurs in the early periods. The processes of neosynaptogenesis get activated on day 7. Both the types of structural recovery of the synaptic pool (hypertrophy and neoformation of the contacts) were fairly marked in the long-term period.

Rhabdomyoma: ultrastructural features and distribution of desmin, muscle type of intermediate filament protein.

A case of adult type rhabdomyoma in the neck region of a 77-year-old man is presented. In electron microscopy, some tumour cells showed irregularly orientated sarcomere formation with distinct Z-bands. All cells contained rod-like inclusions with attached thin cytoplasmic filaments. When stained with antibodies against desmin many tumour cells showed a diffuse peroxidase reaction, but the tumour also contained cells with cross striations. After phosphotungstic acid hematoxylin staining, dense cytoplasmic inclusions were seen; these structures showed strong affinity to antibodies against desmin. Our results apparently confirm the suggestion that the rod-like inclusions in rhabdomyoma cells are modified Z-bands. The desmin staining patterns in the present tumour may represent different stages in disorderly proliferation and degeneration of rhabdomyoma cells.

[DNA packing inside viral particles of Agrotis segetum (Schiff) turnip moth granulosis virus].

The structure of high molecular (88 million daltons) DNA of turnip moth granulosis virus obtained in the linear and cyclic forms was studied. Electron microscopic examination of the fine structure of DNA isolated from virus particles treated with n-toluene sulphonate-N-cyclohexyl-N'-beta-4 (methyl morpholine)-ethylcarbodiimide were carried out. Modified DNA areas were visualized by two methods: with P32 protein and phosphotungstate acid. In both cases a period of recurrence close to the length of the virus particle was revealed. This study confirmed experimentally the previously suggested model of the existence of structural disorders in DNA caused by bends of superspiral molecule at the ends of the virus particle.
