Synthesis and characterization of camphorsulfonyl acetate of cellulose.

Novel cellulose derivatives were prepared from reacting (1R)-(+)-camphor-10-sulfonic chloride (CSC) with cellulose acetate (CA) in acetone and triethylamine. The reaction conditions, including reaction time and reactant molar ratios, were optimized. The structure of the products was confirmed by means of 1H NMR, 13C NMR, FT-IR and elementary analysis. The techniques were also used to determine the degree of the substitution of camphorsulfonyl groups (DSCS). The data calculated from 1H NMR, 13C NMR, percent grafting (G %) and elementary analysis coincided with those from chemical analysis. Compared to cellulose acetate, the cellulose derivatives exhibited decreased thermal stability, improved solubility in organic solvents and enhanced enantioselectivity towards tyrosine isomers. The solubility and enantioselectivity increased with increasing degrees of camphorsulfonyl substitution.

Concentrations and specific loads of UV filters in sewage sludge originating from a monitoring network in Switzerland.

Many substances related to human activities end up in wastewater and accumulate in sewage sludge. The present study focuses on the analysis of widely used UV filters 3-(4-methylbenzylidene) camphor (4-MBC), octyl-methoxycinnamate (OMC), octocrylene (OC) and octyl-triazone (OT) in sewage sludge originating from a monitoring network in Switzerland. Mean concentrations in stabilised sludge from 14 wastewater treatment plants were 1780, 110, 4840 and 5510 microg/kg dry matter for 4-MBC, OMC, OC and OT, respectively. Specific loads in sewage sludge show that UV filters originate mainly from private households, but surface runoff and industries may be considered as additional sources. This indicates that besides use for sunscreens and cosmetics UV filters might occur in plastics and other materials and be released to the environment by volatilization or leaching. Differences between the modeled per capita loads of UV filters in sewage sludge and the observed specific loads in sewage sludge are probably due to erroneous figures of production volumes, degradation and sorption during wastewater treatment as well as degradation processes during transport in the sewer or sludge treatment. Thus, further research is needed to elucidate the fate of UV filters after application and release into the environment. Other compounds used as UV filters should be included in future studies.

Determination of polydimethylsiloxane-air partition coefficients using headspace sorptive extraction.

Polydimethylsiloxane-air partition coefficients (K(PDMS-A)) were determined using direct headspace analysis and headspace sorptive extraction (HSSE) with polydimethylsiloxane-coated (PDMS) stir bars. The partition coefficients were investigated for three compounds, p-dichlorobenzene (PDCB), naphthalene and camphor, all of which sublimate at room temperature and find use as moth repellents. In order to determine the K(PDMS-A) values of these compounds, the air concentration and the concentration present on PDMS, both at equilibrium, were measured. The results indicate that PDMS-air partition coefficients are proportional to octanol-air partition coefficients. Thus, the latter could be used to estimate the extraction efficiency of PDMS for these compounds in air. Alternatively, octanol-air partition coefficients for organic compounds could be estimated from the PDMS-air partition coefficient values. As expected, the PDMS-air (or octanol-air) partition coefficient increased with decreasing temperature. Importantly, the partition coefficients determined at saturated vapor pressures were lower than the values determined at lower analyte concentrations, with the differences being greater for compounds with larger partition coefficients. Consequently, caution should be exercised when applying K(PDMS-A) values determined at high analyte concentrations to measurements at lower concentrations, especially when the partition coefficients are large.

Evaluation of the photostability of different UV filter combinations in a sunscreen.

Development of photostable sunscreens is extremely important to preserve the UV protective capacity and to prevent the reactive intermediates of photounstable filter substances behaving as photo-oxidants when coming into direct contact with the skin. Thus, the objective of this study was to evaluate the photostability of four different UV filter combinations in a sunscreen by using HPLC analysis and spectrophotometry. The formulations that were investigated included four different UV filter combinations often used in SPF 15 sunscreens. The UV filter combinations were: octyl methoxycinnamate (OMC), benzophenone-3 (BP-3) and octyl salicylate (OS) (formulation 1); OMC, avobenzone (AVB) and 4-methylbenzilidene camphor (MBC) (formulation 2); OMC, BP-3 and octocrylene (OC) (formulation 3); OMC, AVB and OC (formulation 4). In the photostability studies, 40 mg of each formulation were spread onto a glass plate and left to dry before exposure to different UVA/UVB irradiation. Exposed samples were then immersed in isopropanol and the dried film dissolved ultrasonically. The filter components in the resulting solution were quantified by HPLC analysis with detection at 325 nm and by spectrophotometry. In this study, the four UV filter combinations showed different photostability profiles and the best one was formulation 3 (OMC, BP-3 and OC), followed by formulations 4, 1 and 2. In addition, OC improved the photostability of OMC, AVB and BP-3.

The conditioned enhancement of neutrophil activity is catecholamine dependent.

Neutrophil activity was elevated in the conditioned mice for the first time through an established conditioned training process. Catecholamines were proved to be important in the regulation of this conditioned innate immunity. In the study, the camphor odor (as the conditioned stimulus, CS) and poly I: C (as the unconditioned stimulus, US) was used to conditionally elevate the activity of the splenic neutrophils. The mechanism(s) responsible for the conditioned enhancement of neutrophil activity was further investigated using the neurochemical blocking assay and immunohistochemical analysis. Results showed that the neutrophil activity was significantly enhanced through the conditioned training process; both reserpine and 6-hydroxydopamine (6-OHDA) significantly blocked this conditioned innate immunity at the conditioned recall stage. Dexamethasone (Dex), however, showed no effect on the conditioned neutrophil response. Tyrosine hydroxylase (TH)-positive cells significantly increased in the locus coeruleus (LC), hypothalamus, and cortex but not in the spleen of the conditioned animals. These results indicate that during the conditioned recall stage, the brain signals the splenic neutrophils via the sympathetic nervous system (SNS) by releasing the peripheral catecholamines in spleen. The activation of the SNS, on the other hand, is also under the influence of catecholamines released in the LC. The hypothalamic pituitary (HP) axis, on the other hand, plays no role in the regulation of the conditioned neutrophil response.

More than cool: promiscuous relationships of menthol and other sensory compounds.

Several temperature-activated transient receptor potential (thermoTRP) ion channels are the molecular receptors of natural compounds that evoke thermal and pain sensations. Menthol, popularly known for its cooling effect, activates TRPM8--a cold-activated thermoTRP ion channel. However, human physiological studies demonstrate a paradoxical role of menthol in modulation of warm sensation, and here, we show that menthol also activates heat-activated TRPV3. We further show that menthol inhibits TRPA1, potentially explaining the use of menthol as an analgesic. Similar to menthol, both camphor and cinnamaldehyde (initially reported to be specific activators of TRPV3 and TRPA1, respectively) also modulate other thermoTRPs. Therefore, we find that many "sensory compounds" presumed to be specific have a promiscuous relationship with thermoTRPs.

A randomized trial of Korodin Herz-Kreislauf-Tropfen as add-on treatment in older patients with orthostatic hypotension.

In a randomized, double-blind, placebo-controlled, parallel group, phase III clinical trial efficacy and safety of Korodin, a combination of natural D-camphor and an extract from fresh crataegus berries, was investigated in patients 50 years and older with orthostatic hypotension. At visit 1 eligibility of patients was checked and a placebo medication was given to all patients. At visit 2 orthostatic hypotension had to be reconfirmed, then the patient was randomized either to Korodin or placebo, study medication (25 drops) was applied once and then outcome was measured. After 7 days of home treatment with daily 3 x 25 drops outcome was measured at visit 3. Systolic blood pressure (SBP), diastolic blood pressure (DBP) and heart rate (HR) were documented 10, 5, 2 and 0 min before as well as 1, 3, 5, 8, and 10 min after getting in the upright position at visit 1, at visit 2 before and after application of study medication and at visit 3. Primary outcome was the change of mean arterial blood pressure (MAP) from just before standing up to the nadir within the first 3 min after standing up. Secondary outcome variables were SBP, DBP, HR, quality of life (SF-12) and seven typical signs and symptoms of orthostatic hypotension. The study was performed in a rehabilitation clinic and in two doctor's practices in Germany from November 2002 to May 2003. During this time, 57 patients were admitted to the study, 39 patients were eligible and randomized, 38 patients were treated according to protocol and evaluated, 21 patients with Korodin and 17 patients with placebo. After a single application the median decrease of MAP was 11.4 mmHg for Korodin and 14.0 mmHg for placebo. Compared to baseline, the median MAP improved 4.3 mmHg for Korodin and 0.3 mmHg for placebo. After 1 week of treatment the decrease of median MAP after standing up was 9.3 mmHg for Korodin and 13.3 mmHg for placebo. Compared to baseline, the improvement was 5.9 mmHg for Korodin and 1.6 mmHg for placebo. Efficacy of 1 week treatment was significant. For the single application a superiority of Korodin over placebo was seen; however, it was not significant. All secondary outcome variables confirmed these findings, except for the physical summary score in the quality of life evaluation (SF-12 questionnaire). Only one adverse event occurred, but this was not serious and without relationship to the study medication. The other safety variables (SBP, DBP, HR, ECG, physical examination) did not show any problems. This study demonstrates that Korodin is efficacious for orthostatic hypotension in patients over 50 years.

Biosynthesis of the irregular monoterpene artemisia ketone, the sesquiterpene germacrene D and other isoprenoids in Tanacetum vulgare L. (Asteraceae).

The incorporation of [1-13C]-labeled glucose into the irregular monoterpene artemisia ketone, the regular monoterpenes camphor and beta-thujone, the sesquiterpene germacrene D, the diterpene trans-phytol and beta-sitosterol and isofucosterol has been studied in axenic cultures of Tanacetum vulgare L. (Asteraceae). Quantitative 13C NMR spectroscopic analysis of the resulting labeling patterns showed that the isoprene units of the monoterpenes and the diterpene are formed via the methylerythritol phosphate (MEP) pathway, whereas the isoprene building blocks of the sesquiterpene and the sterols originate from the mevalonic acid (MVA) pathway.

Crystal structures of cyanide complexes of P450cam and the oxygenase domain of inducible nitric oxide synthase-structural models of the short-lived oxygen complexes.

The crystal structure of the ternary cyanide complex of P450cam and camphor was determined to 1.8A resolution and found to be identical with the structure of the active oxygen complex [I. Schlichting et al., 2000, Science 287, 1615]. Notably, cyanide binds in a bent mode and induces the active conformation that is characterized by the presence of two water molecules and a flip of the carbonyl of the conserved Asp251. The structure of the ternary complex of cyanide, L-arginine, and the oxygenase domain of inducible nitric oxide synthase was determined to 2.4A resolution. Cyanide binds essentially linearly, interacts with L-Arg, and induces the binding of a water molecule at the active site. This water is positioned by backbone interactions, located 2.8A from the nitrogen atom of cyanide, and could provide a proton required for O-O bond scission in the hydroxylation reaction of nitric oxide synthase.

The ability of various chemicals to elicit olfactory beta-waves in the pyriform cortex of meadow voles (Microtus pennsylvanicus) and laboratory rats (Rattus norvegicus).

Previous research has shown that, in freely moving laboratory rats, the odors of benzyl alcohol, camphor, carvacrol, isopentenyl methyl sulfide, 2-propylthietane, salicylaldehyde, trimethylthiazoline, and xylene (plus other compounds) elicit high amplitude bursts of roughly 20-Hz waves (beta-waves) in the olfactory bulb and pyriform cortex. Since all these compounds are effective antifeedants in a variety of small herbivores, a more extensive test of the effect of the odor of a variety of antifeedants on pyriform cortex activity in meadow voles (Microtus pennsylvanicus) was undertaken. A beta-wave response was reliably elicited in the vole pyriform cortex by the odors of isopentenyl methyl sulfide (a component of fox odor), 2-propyl thietane (a component of stoat odor), and xylene but not by the odors of 29 other compounds. The results indicate: (a) that there are clear interspecific differences in the olfactory reactions of the pyriform cortex and (b) that not all antifeedants elicit beta-waves in the pyriform cortex.

The oxidation of naphthalene and pyrene by cytochrome P450cam.

Mutants of the heme monooxygenase cytochrome P450cam in which Y96 had been replaced with hydrophobic residues, have been shown to oxidise naphthalene and pyrene with rates one to two orders of magnitude faster than the wild-type. Naphthalene was oxidised to 1- and 2-naphthol, probably via the 1,2-oxide intermediate. In the case of the Y96F mutant, naphthalene was oxidised at a rate comparable to camphor. Pyrene oxidation gave 1,6- and 1,8-pyrenequinone with no evidence for attack at the K-region, in contrast to mammalian enzymes. The results show that the Y96 residue plays a key role in controlling the substrate range of P450cam.

Surface-modified mutants of cytochrome P450cam: enzymatic properties and electrochemistry.

We report the electrochemistry of genetic variants of the haem monooxygenase cytochrome P450cam. A surface cysteine-free mutant (abbreviated as SCF) was prepared in which the five surface cysteine residues Cys-58, Cys-85, Cys-136, Cys-148 and Cys-334 were changed to alanines. Four single surface cysteine mutants with an additional mutation, R72C, R112C, K344C or R364C, were also prepared. The haem spin-state equilibria, NADH turnover rates and camphor-hydroxylation properties, as well as the electrochemistry of these mutants are reported. The coupling of a redox-active label, N-ferrocenylmaleimide, to the single surface cysteine mutant SCF-K344C, and the electrochemistry of this modified mutant are also described.

Trace analysis of organic volatiles in water by gas chromatography-mass spectrometry with glass capillary columns.

Traces of volatile organic materials in water have been concentrated by gas phase stripping and adsorption onto a porous polymer. A simple all-glass sampling device is proposed which allows efficient concentration at elevated temperatures. Sample transfer from absorbent into a gas chromatographic column is effected by a simple one-step procedure involving heat desorption. The capacity of the absorbent has been determined for a number of model substances which are found in water. Under the sampling conditions used, compounds being less volatile than benzene are usually quantitatively retained, with some exceptions. Separations were effected with highly efficient glass capillary columns. Water samples, collected from a small number of locations, included both tap water and untreated waters. A number of volatiles have been determined by gas chromatography-mass spectrometry in drinking water and in a river which flows through an industrialized area. The drinking water examined contains a large number of chlorinated and brominated compounds whereas the river water is largely free from this class of substances. Derivatives of camphor and terpenes have been identified in this particular river.

Structure of cytochrome P450eryF: substrate, inhibitors, and model compounds bound in the active site.

Much of our understanding of P450 reaction mechanisms derives from studies on P450cam, a bacterial camphor hydroxylase. P450cam has served as the model for understanding detailed structure/function relationships in mammalian P450 enzymes, which have not proved amenable to x-ray crystallographic techniques. To expand and improve the P450 model, we solved the structure of P450eryF, a cytochrome P450 involved in erythromycin biosynthesis. The overall structure of P450eryF is similar to that of P450cam, but differs in the exact positioning of several alpha-helices, which results in the enlargement of the substrate-binding pocket. P450eryF also differs from P450cam in having alanine in place of the highly conserved threonine residue in the active site. To assess the role of this alanine residue, two mutant forms of P450eryF and a substrate analog were examined. Our findings suggest that P450eryF has evolved an active site that utilizes the substrate to assist in catalysis. In addition, the enlarged substrate binding pocket of P450eryF enables P450eryF to bind certain steroid compounds and azole-based steroid hydroxylase inhibitors. Crystals have been obtained for P450eryF complexed with the antifungal drug ketoconazole, and the high-resolution structure has been determined.

Changes in pH at the dentin surface in roots obturated with calcium hydroxide pastes.

The purpose of this study was to determine the pH, after defined periods of time, in cavities prepared in the facial surface of the cervical, middle, and apical regions of roots obturated with calcium hydroxide pastes. Root canal instrumentation was performed on 40 recently extracted, single-rooted human teeth. Cavities 1.5 mm in diameter and 0.75 mm in depth were prepared in the cervical, middle, and apical regions of the facial surface of each root. Teeth were randomly divided into four groups. One group was left unobturated and served as a control. The three remaining groups were obturated with either aqueous calcium hydroxide, calcium hydroxide mixed with camphorated monochlorophenol. or Pulpdent pastes. Access cavities and apical foramina were closed with Cavit. Each tooth was stored individually in a vial containing unbuffered isotonic saline. pH at the surface was measured in the cervical, middle, and apical cavities at 0 and 3, 7, 14, 21, 28, 45, 60, 90, and 120 days. Results indicate that hydroxyl ions derived from calcium hydroxide pastes diffused through root dentin at all regions over the experimental period of 120 days. The pattern of pH change at the tooth surface was similar in all regions of the root, regardless of the type of calcium hydroxide paste used. This was a rapid rise in pH from a control value of pH 7.6, to greater than pH 9.5 by 3 days, followed by a small decline to pH 9.0 over the next 18 days, before finally rising and remaining at, or above pH 10.0 for the remainder of the experimental period. Pulpdent paste in the apical region was the only exception in this pattern, producing a pH rise nearly one full unit below the other pastes, pH 9.3. These results indicate that, for all pastes tested, a high pH is maintained at the root surface for at least 120 days.

Intracanal medicaments: evaluation of the antibacterial effects of chlorhexidine, metronidazole, and calcium hydroxide associated with three vehicles.

The use of an intracanal medicament may be helpful in eliminating remaining bacteria that survived inside root canals after complete chemomechanical preparation. This study evaluated the antibacterial activities of medicaments that act by means of contact, and not by vapor release, against obligate and facultative anaerobic bacteria commonly found in endodontic infections. The medicaments used were 0.12% chlorhexidine gel; 10% metronidazole gel; calcium hydroxide plus distilled water, calcium hydroxide plus camphorated paramonochlorophenol (CPMC); and calcium hydroxide plus glycerin. An agar diffusion test was used, and the zones of bacterial inhibition around each medicament were recorded and compared. The results revealed that calcium hydroxide/CPMC paste was effective against all bacterial strains tested. Chlorhexidine was also inhibitory to all strains. It was about as effective as calcium hydroxide/CPMC paste against most of the strains. Metronidazole also caused inhibition of growth of all obligate anaerobes tested and was more effective than calcium hydroxide/CPMC against two strains. Calcium hydroxide mixed with distilled water or glycerin failed to show zones of bacterial inhibition, probably because of limitations of the agar diffusion test.

Intermolecular electron transfer in cytochrome P450cam covalently bound with Tris(2,2'-bipyridyl)ruthenium(II): structural changes detected by FTIR spectroscopy.

Using Fourier transform infrared spectroscopy (FTIR) we have monitored the changes in the protein structure following photoinduced electron transfer from Ru(bpy)(3)(2+) covalently attached to cysteine 334 on the surface of cytochrome P450cam (CYP101). The FTIR difference spectra between the oxidized and reduced form indicate changes in a salt link and the secondary structure (alpha-helix and turn regions). Photoreduction was carried out in the presence of carbon monoxide in order to prove the reduction of the heme iron by means of the appearance of the characteristic CO stretch vibration infrared band at 1940 cm(-1) for the camphor-bound protein. This infrared band has also been used to estimate electron transfer rates. The observed rates depend on the protein concentration, indicating that intermolecular electron transfer occurs between the labeled molecules.

Effect of UV screens and preservatives on vitellogenin and choriogenin production in male medaka (Oryzias latipes).

Ultra violet (UV) screens and preservatives are widely and increasingly used in cosmetics and pharmaceuticals. In the present study, we examined the estrogenicity of 4-methyl-benzylidene camphor (4-MBC), octyl-methoxycinnamate (OMC), and propyl paraben (n-propyl-p-hydroxy-benzoate; PP), among UV screens and preservatives, using male medaka (Oryzias latipes), in regard to production of vitellogenin (VTG) and choriogenin (CHG) which are known to be estrogen-responsive gene products. First, using a VTG enzyme-linked immunosorbent assay (ELISA) system, we determined the increase in VTG plasma concentration in medaka due to exposure to 4-MBC, OMC, and PP, and compared this concentration to the non-treated control. Next, we found increases in mRNA expression levels of VTG subtypes VTG-1 and VTG-2, and CHG subtypes CHG-L and CHG-H, in liver due to exposure to 4-MBC, OMC, and PP compared to the non-treated control. In addition, we also found increased mRNA expression levels of estrogen receptor (ER) alpha, among sex hormone receptors in the liver, due to exposure to 4-MBC, OMC, and PP compared to the non-treated control. In this study, we showed that 4-MBC, OMC, and PP have estrogenic activity in fish.
