Identification of structural components associated with cytostatic activity in MCF-7 but not in MDA-MB-231 cells.

The National Cancer Institute's Developmental Therapeutics Program maintains the screening results obtained in 60 standardized cancer cell lines and contained 37,836 compounds for this study. This dataset has shown to be an outstanding resource for the development of structure-activity relationship (SAR) models describing anticancer activity. We report here a novel SAR modeling approach based on a subtractive protocol to develop models that describe cell type-specific molecular descriptors of cytotoxicity. The goal of this approach is to separate features associated with antiproliferative activity to many cell lines from those that effect only a specific cell type. To assess this approach, we developed SAR models for cytostatic activity against the human breast cancer cell lines MCF-7 and MDA-MB-231 and one differential activity model for compounds that were potent cytostatic agents in MCF-7 cells but relatively inactive against MDA-MB-231 cells. The models were between 72 and 84% accurate when challenged with compounds not in the learning sets. Structural features associated with the differential activity model highlighted how the use of this approach can selectively identify chemical moieties associated with potent cytostatic action to MCF-7 but not to MDA-MB-231 cells. We surmise that outgrowth of this method can facilitate the development of SAR models with sufficient resolution and clarity to identify chemical moieties associated with antiproliferative activity to selective individual cancer types while being innocuous to other cell types.

Examining structured representation and designated fiscal support for women's health in the U.S. Department of Health and Human Resources.

The United States Department of Health and Human Services (DHHS) is committed to monitoring, protecting, and improving the health of the nation. We examine the structure established within DHHS to address the health of women; review initiatives generated by women's health offices, advisors, and coordinators within DHHS agencies; and contrast the budgets provided to women's health offices with those of the parent DHHS agencies. Data were obtained from DHHS and other public government documents, DHHS websites, contact with agency personnel, and literature review. Significant clinical, research, and educational efforts important to the health of women have resulted from representation for women's health within the DHHS. Yet, structured representation and designated fiscal support necessary to maintain and expand these efforts are variable and not guaranteed across agencies. Only one Office of Women's Health and one Senior Advisor position are supported by statute, one director's position for an Office of Women's Health has been downgraded in government rank, and two other women's health positions had their reporting structure changed, making them less centrally located in their respective agencies. During the last 4 years of unprecedented growth within DHHS, only one Office of Women's Health received consistent increases in budgeted dollars. There is a clear need to support and stabilize representation for women's health within DHHS in order to maintain current productive efforts, coordinate existing and developing initiatives, and integrate new topics of importance to women's health into each agency. This can be accomplished by establishing structured offices by statute and ensuring future funding commensurate with the mission of each office.

Effect of environmental enrichment upon resource holding power in fish in prior residence situations.

Resource holding power (RHP), as expressed by gaining dominance, can be affected by extrinsic and intrinsic factors. Extrinsic factors that increase the RHP include e.g. prior exposure to the contest area. The pay-off asymmetry hypothesis was tested according to which there is an asymmetry in the value the resident's territory has for the resident and intruders, i.e. the resident loses more than intruder opponents when losing the contest over a territory in which it has invested much energy during the exploration of food and shelter resources, during the settlement of conflicts with territorial neighbours, etc. This asymmetry will increase the resident's chance to win the fight over intruders. If the resident-intruder asymmetry is enhanced by presenting resource-rich territories to e.g. fish (structurally diverse or 'rich' aquariums), the hypothesis predicts that the resident's dominance chance (probability to win the fight) will be higher than in structurally 'poor' aquariums. It was found that residents were proportionately more often dominant (fight winners) than intruders in the rich compared to the poor aquariums in all seven tested fish species. This demonstrates that a high territory resource value (aquarium enrichment) significantly facilitates the expression of the resident's dominance advantage (prior residence effect). In contrast to dominance, aggressive behaviour before the dominance settlement did not generally differ between residents and intruders when compared in rich and poor aquariums. This suggests that dominance in itself and aggression prior to dominance settlement are at least partially guided by different motivational systems.

Consumers' knowledge about their health insurance coverage.

This paper describes how much families know about their health insurance coverage and investigates whether consumer education and simplified benefit structures would improve knowledge. Families' perceptions about their insurance benefits were measured in two household surveys administered in six sites. Knowledge was assessed by comparing families' responses with policy data collected from the carrier. The vast majority of families understand insurance policies that specify one or two parameters in their benefit provisions. However, more complex payment structures are not well understood. Increased exposure to information in the plans leads to increased knowledge which suggests that education programs could improve the general level of knowledge. We conclude that if market strategies for allocating medical resources are pursued, simplifying insurance benefit structures and educating consumers about their insurance benefits would aid consumers in making more informed economic choices about medical care.

Why are structured interviews so rarely used in personnel selection?

By adopting the theory of planned behavior, this study tried to predict human resources managers' (N = 79) intentions toward unstructured and structured interview techniques. Managers evaluated case descriptions of both techniques and were interviewed about their own practices. The data revealed stronger intentions toward unstructured interviewing than toward structured interviewing, which was consistent with their own practices in selecting staff, which appeared to be rather unstructured. l. Ajzen's (1991) theory appeared to be a useful framework for predicting managers' intentions. In particular, attitudes and subjective norms were predictive of intentions to engage in either method. Only intentions toward the unstructured case were related to managers' actual behavior.

A patient orientation program at a comprehensive cancer center.

PURPOSE/OBJECTIVES: To determine which method of delivery of a cancer orientation program contributed to higher levels of satisfaction and lower levels of anxiety for newly diagnosed patients with cancer and each patient's support person. DESIGN: A randomized study of patients with cancer and caregivers into one of three delivery methods for an orientation program or a control arm. SETTING: A National Cancer Institute-designated comprehensive cancer center in the midwestern United States. SAMPLE: Newly registered patients with cancer diagnoses and their identified support people. METHODS: The intervention consisted of an orientation video and booklet delivered by three separate methods: class, drop-in sessions, or information mailed to homes. Participants completed questionnaires before the intervention and three weeks after the intervention. MAIN RESEARCH VARIABLES: State and trait anxiety, satisfaction, understanding of the organization, awareness and use of resources, and stress and coping. FINDINGS: The most successful accrual arms were the mailed intervention and control groups. The mailed intervention group compared to the control group reported higher levels of satisfaction with the cancer center, satisfaction with resources, understanding of the cancer center's structure, and satisfaction with healthcare professionals' communication with them. Fewer intervention group participants reported a lack of awareness of specific resources, and a larger percentage of the intervention group used available resources. Fewer benefits were noted with caregivers. CONCLUSIONS: The mailed intervention was successful in improving several patient outcomes. It was shown to be especially helpful to those with high trait anxiety. IMPLICATIONS FOR NURSING: A mailed orientation program can be a useful approach for increasing satisfaction with services.

Dealing with an information overload of health science data: structured utilisation of libraries, distributed knowledge in databases and Web content.

The organizational structures of web contents and electronic information resources must adapt to the demands of a growing volume of information and user requirements. Otherwise the information society will be threatened by disinformation. The biomedical sciences are especially vulnerable in this regard, since they are strongly oriented toward text-based knowledge sources. Here sustainable improvement can only be achieved by using a comprehensive, integrated approach that not only includes data management but also specifically incorporates the editorial processes, including structuring information sources and publication. The technical resources needed to effectively master these tasks are already available in the form of the data standards and tools of the Semantic Web. They include Rich Site Summaries (RSS), which have become an established means of distributing and syndicating conventional news messages and blogs. They can also provide access to the contents of the previously mentioned information sources, which are conventionally classified as 'deep web' content.

Biogeography and community structure of North American seed-harvester ants.

Seed-harvester ants are a dominant and conspicuous insect group throughout arid portions of the southwestern United States and northern Mexico, and they include approximately 75 species. Intense study in the late 1970s and early 1980s led to the paradigm that interspecific competition for limited seed resources is the primary factor that structures seed-harvester ant communities. This review attempts to adjust this paradigm, suggesting that interspecific competition for food is probably less important than previously thought, whereas intraspecific competition is common and strong. Abiotic-habitat factors also have important effects on local species composition and attributes. This review also emphasizes several aspects of seed-harvester ants that have been poorly studied, including historical factors, adaptive radiation, and biogeography, to provide a more detailed evolutionary background for understanding existing species assemblages.

A theoretical and empirical analysis of context: neighbourhoods, smoking and youth.

Numerous studies are currently addressing the issue of contextual effects on health and disease outcomes. The majority of these studies fall short of providing a theoretical basis with which to explain what context is and how it affects individual disease outcomes. We propose a theoretical model, entitled collective lifestyles, which brings together three concepts from practice theory: social structure, social practices and agency. We do so in an attempt to move away from both behavioural and structural-functionalist explanations of the differential distribution of disease outcomes among areas by including a contextualisation of health behaviours that considers their meaning. We test the framework using the empirical example of smoking and pre-adolescents in 32 communities across Québec, Canada. Social structure is operationalised as characteristics and resources; characteristics are the socio-economic aggregate characteristics of individuals culled from the 1996 Canadian Census, and resources are what regulates and transforms smoking practices. Information about social practices was collected in focus groups with pre-adolescents from four of the participating communities. Using zero-order and partial correlations we find that a portrait of communities emerges. Where there is a high proportion of more socio-economically advantaged people, resources tend to be more smoking discouraging, with the opposite being true for disadvantaged communities. Upon analysis of the focus group material, however, we find that the social practices in communities do not necessarily reflect the "objectified" measures of social structure. We suggest that a different conceptualisation of accessibility and lifestyle in contextual studies may enable us to improve our grasp on how differential rates of disease come about in local areas.

Eutrophication, Fisheries, and Consumer-Resource Dynamics in Marine Pelagic Ecosystems.

Anthropogenic nutrient enrichment and fishing influence marine ecosystems worldwide by altering resource availability and food-web structure. Meta-analyses of 47 marine mesocosm experiments manipulating nutrients and consumers, and of time series data of nutrients, plankton, and fishes from 20 natural marine systems, revealed that nutrients generally enhance phytoplankton biomass and carnivores depress herbivore biomass. However, resource and consumer effects attenuate through marine pelagic food webs, resulting in a weak coupling between phytoplankton and herbivores. Despite substantial physical and biological variability in marine pelagic ecosystems, alterations of resource availability and consumers result in general patterns of community change.

The AEDMI project, an example of prototype development for a clinical decision helping system.

The AEDMI project, carried out in the Bellvitge University Hospital, near Barcelona, is proposed as an example of clinical decision system prototype development. Several difficulties have been encountered to develop this project. One of them was to find the adequate understanding for the project and proper funding for personnel resources. The support by FIS grants and from the hospital structures was essential to the survival of the project; however the resources always have been below the real needs for development. The difficulties with clinical coding systems, the excessive expectancies upon prototypes, and the lack of culture in information methods were other relevant hinders. The complexity of clinical realm, the uncertainty of medical decisions, and the current trends to substitute the clinical information by data obtained from electronic devices, or analytical monitoring of patients, are additional problems for clinical information managing.

Obligatory medical insurance in Russia: the participants' perspective.

The Russian Federation adopted a nation-wide system of obligatory medical insurance in 1993 in an effort to earmark a targeted source of funding for health care and to reverse a steep decline in health outcomes. The author conducted a survey in 1995-1996 of managers of two of the new institutional participants in Russia's health insurance scheme: Territorial Health Insurance Funds and private medical insurance companies. The survey results reveal deep dissatisfaction with the level of financing provided by the new system; continuing confusion and substantial regional variation in the implementation of the insurance legislation; fierce bureaucratic and institutional infighting between the major players, stemming primarily from controversy over delineation of responsibilities and ongoing battles for control over resources; promising hints of competition and other market-based incentives emerging from the current chaos; and broad agreement that further structural reform must accompany increased infusions of resources in order for significant systemic improvements to be realized.

[Prognostic significance of arterial blood pressure levels and the effectiveness of the treatment of arterial hypertension in a non-organized male population 40-59 years of age in Moscow].

Isolated working rat hearts were subjected to a 3-hour ischemia followed by reperfusion. The defense effects of the heart cooling to 6-7 and 12-15 degrees C were assessed according to the degree of postischemic recovery of hemodynamic (arterial pressure, heart rate, coronary flow, cardiac output) and biochemical (ATP, phosphocreatine, Pi, and lactate content) characteristics and electron microscopic evidence. The energy resources of the myocardium and its cellular structures were effectively preserved during one-hour ischemia at 12-15 degrees C. At 6-7 degrees C, the energy resources of the hearts were better preserved in long-term ischemia, where the functional characteristics were recovered to the same extent as in the 12-15 degrees C hypothermia, but some cell damage was observed. The obtained findings suggested that the temperature of 12-15 degrees C should be the temperature of choice for heart cooling in cardiosurgery.

Constructing ontology-driven protein family databases.

MOTIVATION: Protein family databases provide a central focus for scientific communities as well as providing useful resources to aide research. However, such resources require constant curation and often become outdated and discontinued. We have developed an ontology-driven system for capturing and managing protein family data that addresses the problems of maintenance and sustainability. RESULTS: Using protein phosphatases and ABC transporters as model protein families, we constructed two protein family database resources around a central DAML+OIL ontology. Each resource contains specialist information about each protein family, providing specialized domain-specific resources based on the same template structure. The formal structure, combined with the extraction of biological data using GO terms, allows for automated update strategies. Despite the functional differences between the two protein families, the ontology model was equally applicable to both, demonstrating the generic nature of the system. AVAILABILITY: The protein phosphatase resource, PhosphaBase, is freely available on the internet (http://www.bioinf.man.ac.uk/phosphabase). The DAML+OIL ontology for the protein phosphatases and the ABC transporters is available on request from the authors. CONTACT: kwolstencroft@cs.man.ac.uk.

Measuring hospital medical staff organizational structure.

Based on organization theory and the work of Roemer and Friedman, seven dimensions of hospital medical staff organization structure are proposed and examined. The data are based on a 1973 nationwide survey of hospital medical staffs conducted by the American Hospital Association. Factor analysis yielded six relatively independent dimensions supporting a multidimensional view of medical staff organization structure. The six dimensions include 1) Resource Capability, 2) Generalist Physician Contractual Orientation, 3) Communication/Control, 4) Local Staff Orientation, 5) Participation in Decision Making, and 6) Hospital-Based Physician Contractual Orientation. It is suggested that these dimensions can be used to develop an empirical typology of hospital medical staff organization structure and to investigate the relationship between medical staff organization and public policy issues related to cost containment and quality assurance.

Light-induced mass turnover in a mono-species community of mixotrophs.

We formulate a simple model for growth of a facultative photoautotroph with chemoheterotrophic capabilities. The organism is described by zero, one or three reserve components, and one structural component, all taken to be generalized compounds. The rules of synthesizing units are used for interactions among the uptake processes of the various nutrients and light (parallel processing), and for the merging of autotrophic and heterotrophic activities (sequential processing). For simplicity, we focus on the assimilation of inorganic carbon, inorganic nitrogen and light, and of two organic compounds (dead reserves and dead structure) that originate from aging. The process of resource recycling in a closed environment, as driven by light, and its links with community's structure (amount of biomass) is analysed in this simplest of all communities. Explicit analytical expressions for the steady states show how structure and function depend on the system parameters light, total carbon and total nitrogen. The behaviour resembles the Monod model for the Canonical Community, a three-species ecosystem consisting of producers, consumers and decomposers. If trophic preferences of a mixotroph are allowed to follow a random walk across generations, a trophic structure evolves where mixotrophs coexist with auto- and heterotrophs. Depth profiles are presented for the implied steady-state concentrations of dissolved inorganic carbon and nitrogen.

IMGT gene identification and Colliers de Perles of human immunoglobulins with known 3D structures.

A new database, IMGT/3Dstructure-DB, was developed and implemented in the IMGT (international ImMunoGeneTics database) information system (http://imgt.cines.fr) to provide a unique expertised resource on immunoglobulin and T-cell receptor structural data. Corresponding protein sequences were annotated with IMGT tools, which allow the precise identification of the genes expressed in these proteins, and the description of framework and complementarity determining regions according to the IMGT standardized nomenclature and IMGT unique numbering. Two-dimensional graphical representations of the V-DOMAINs, designated as Colliers de Perles, are automatically produced. A query Web interface allows interactive search of the IMGT/3D structure-DB data. In this article, IMGT gene identification and Colliers de Perles of human immunoglobulins with known 3D structures in the Protein Data Bank are presented.
