[A normal thyroid gland upon autopsy: a relatively uncommon finding].

In a series of 420 autopsies the thyroid glands have been weighed, serial sections made and examined histologically with at least one specimen per lobe. There is an age-dependent increase in mean thyroid weight. In the 7th decade the mean weight exceeds 29 g and weights above this are regarded as goitre. Between the 4th and 8th decade the incidence of goitre is 20-30%, after which the incidence rises steadily to 64% in the 10th decade. The commonest thyroid pathology is hyperplastic nodules, which are found in 39% of patients (49.4% of males and 33.4% of females). 2.4% of all patients have adenomas, 1.9% primary carcinomas and 2.8% thyroid metastases. Inflammatory infiltrates are observed in 6.6% of cases (9.4% of females and 4.4 of males). To establish the incidence of normal thyroid glands we have studied 840 serial autopsies. Only 25% of thyroids are normal (19% of females and 30% of males). The incidence of normal thyroid glands tends to decrease with age and in no decade does it reach 50%.

[Radiation exposure of the ocular lens and thyroid gland in digital subtraction angiography of brain-supplying arteries].

PURPOSE: To find out the individual level of radiation exposure of the ocular lens and thyroid gland during DSA of arteries supplying the brain, considering the indication. MATERIAL AND METHOD: The study was conducted on 72 patients during aortic arch and cerebral artery angiographies using thermoluminescent dosimetry. RESULTS: During 20 aortic arch angiographies the measured values were within non-critical limits, whereas during carotid, vertebral or cerebral four-vessel angiography of the left ocular lens exposure values of more than 500 mGy (max. 782 mGy) were stated in two of 52 cases. The highest measured exposure relating to the right ocular lens was 126 mGy, to the thyroid gland 88 mGy. However, in 51 of 52 cases thyroid gland doses of below 51 mGy were measured. In 71 of 72 cases there was a lateral difference between right and left lens exposure up to twice the measured dose for the right ocular lens during aortic arch angiographies and up to sixteen-fold in respect of the left lens during cerebral angiographies. CONCLUSION: In addition to the fluoroscopic time, number of images, fade-in and zoom some other dose-influencing factors, radiation geometry is especially decisive for organ exposure. The risk of cataracts or thyroid gland carcinomas/hypothyrosis can be neglected with median values of 40 and 70 mGy relating to the ocular lens and 17 mGy for the thyroid gland during cerebral angiographies.

[Peptide bioregulator efficacy in the correction of reduced thyroid gland function in the residents of Magadan Region].

Thyroid gland pathology is one of the most widespread diseases. Age is the main risk factor in the development of hypothyroid states, alongside with biogeochemical and climatic adverse factors of the environment. Disturbed hormonal status of the organism affects the quality of life in patients with subclinical and manifested hypothyroidism forms, especially in old age, and requires adequate correction methods. In this connection Thyramin bioregulator is noteworthy, exerting tissue specific effect on thyroid gland cells and enabling the restoration of the disturbed thyroid tissue function.

Overexpression of c-Ras in hyperplasia and adenomas of the feline thyroid gland: an immunohistochemical analysis of 34 cases.

Formalin-fixed, paraffin-embedded thyroid glands from 18 cats diagnosed with hyperthyroidism were evaluated immunohistochemically for overexpression of the products of oncogenes c-ras and bcl2 and the tumor suppressor gene p53. Fourteen thyroid glands from euthyroid cats without histologically detectable thyroid lesions were examined similarly as controls. Results from these investigations showed that all cases of nodular follicular hyperplasia/adenomas stained positively for overexpression of c-Ras protein using a mouse monoclonal anti-human pan-Ras antibody. The most intensely positively staining regions were in luminal cells surrounding abortive follicles. Subjacent thyroid and parathyroid glands from euthyroid cats did not stain immunohistochemically for pan-Ras. There was no detectable staining for either Bc12 or p53 in any of the cats. These results indicated that overexpression of c-ras was highly associated with areas of nodular follicular hyperplasia/adenomas of feline thyroid glands, and mutations in this oncogene may play a role in the etiopathogenesis of hyperthyroidism in cats.

[Interaction between the hypothalamo-hypophyseal system and the thyroid gland during different age periods].

Age peculiarities of interaction of the hypothalamo-hypophysial system and the thyroid gland with consideration to direct and feed-back control were studied. Significant alterations appear in old age in the hypothalamo-hypophysial system and the thyroid gland at the level of direct and feed-back control; there is an increase of sensitivity of the thyroid gland tissue to TSH, and of the hypothalamus-hypophysis--to the T4 effect. This rise of sensitivity is largely determined by the activation of the T4 deiodination processes in the adenohypophysis. The reactive capacity of the thyroid gland and of the hypothalamo-hypophysial complex to the corresponding influence is decreased.

[Age-related characteristics of the thyroid gland rheogram in healthy persons].

The right and left lobes of the thyroid gland were studied rheographically in 35 healthy persons (70 rheograms) of three age groups for the assessment of the blood flow intensity and functional condition of the thyroid gland vessels. It appeared that the circulation intensity through the thyroid gland decreased with the advance of age; there was also a reduction of elasticity of the vascular wall and an increase in the peripheral resistance of the blood flow in the gland.

Traumatic rupture of the thyroid gland--a case report.

Traumatic rupture of the thyroid gland is a rare event. This report describes such an injury, in this case caused by a kick from a horse. Blunt injury to the anterior structures of the neck involving the major blood vessels and larynx has been described, but major blunt injury to the thyroid gland is rare.

[Adenosquamous carcinoma originating from the thyroid gland--report of a case and review of the literature].

Adenosquamous cell carcinoma originating from the thyroid gland is extremely rare. Only a few cases have been reported by light-microscopic observations. Like pure squamous cell carcinoma of the thyroid gland, it is generally thought to originate from the thyroid follicular epithelium and to be concerned with squamous metaplasia of the follicular epithelium. In our case, papillary adenocarcinoma was found in the thyroid gland, with the transition to squamous cell carcinoma. Immunohistochemical study for thyroglobulin and keratin further implied he transition of adenosquamous cell carcinoma in the thyroid gland. The origins of squamous cells in thyroid carcinoma are briefly discussed.

Sonographic measurements of the neonatal thyroid gland.

The purpose of this study was to establish the normal dimensions of the thyroid gland in euthyroid neonates. Ultrasonographic evaluation of the thyroid gland was performed in the first week of life in 68 term neonates whose gestational ages at birth were 37 to 41 weeks and whose birth weights ranged from 2570 to 4790 g. Measurements of the thyroid lobes were done in transverse, anteroposterior, and longitudinal planes. All neonates had age-appropriate thyroid stimulating hormone levels at birth. Mean +/- 2 standard deviations for thyroid volume were 472 mm3 and 1430 mm3. The mean measurements for the sum of both lobes for transverse, anteroposterior, and longitudinal dimensions were 13.9 mm, 15.1 mm, and 34.6 mm, respectively. No significant correlation of these measurements was found for gestational age, birth weight, or thyroid-stimulating hormone levels. The correlation coefficient between these measurements and total volume was 0.64 or less. Term euthyroid neonates were found to have a thyroid gland volume ranging between 472 mm3 and 1430 mm3. In term neonates sonographic measurements of thyroid lobes should be obtained in all three planes for reliable assessment of thyroid volume.

Dimeric form of calcitonin gene-related peptide in the porcine thyroid gland.

In this study we investigated peptides that increase rat platelet cAMP in porcine thyroid gland. Gel filtration of extracts from porcine thyroid gland showed high and low molecular weight activity. Low molecular weight activity contained peptides, including calcitonin gene-related peptide (CGRP), vasoactive intestinal polypeptide (VIP) and peptide histidine isoleucine (PHI). We isolated a high molecular weight peptide (M. W. 11,000) showing potent activity able to increase rat platelet cAMP in porcine thyroid gland. The peptide's N-terminal sequence was determined to be Ser-X-Asn-Thr-Ala-Thr- by gas phase sequencer analysis, a sequence identical to that of porcine CGRP. The peptide had CGRP immunoreactivity as well as platelet cAMP elevating activity. By gel filtration HPLC, synthetic human CGRP (M. W. 3790) was eluted in a position corresponding to M. W. 5,500. These results suggest that judging from its high molecular weight the above peptide is a dimeric form of CGRP.

[Morphofunctional characteristics of the thyroid gland after correction of iodine and bromine deficiency].

Experiments were conducted on rats; morphofunctional changes of the thyroid gland in correction of iodine and bromine deficiency, depending on the regimen of the microelements administration in the course of a one-week cycle were studied. Daily administration of a one-week dose of iodine and bromine proved to be most effective in respect to the normalization of the morphofunctional condition of the thyroid gland in animals given iodine-deficient diet. The lowest thyroid gland weight, intensification of the synthetic processes in the thyrocytes, complete utilization of the thyroid gland iodine, and enrichment of the organism with trace elements were noted. Recommendations are given concerning the expediency of the change of regimen of iodine and bromine administration in group antigoiter prophylaxis.

Morphometric investigations on endocrine glands. VI. Point counting in th thyroid gland.

The component distributions in a dog thyroid section are found to be inhomogeneous in a different degree. Therefore special conditions are needed in morphometrical research of such tissues to get accurate results, e.g. a sufficiently large area. The net point distance may reach 400 microns for test series, and 1,000 or more hits are enough for an acceptable representative value. The often observed zonal heterogeneity for the colloid and epithelium is diminished and blurred if the relative colloid percentage is high. The interstitium values show an anisotropic distribution in the gland. The epithelium: colloid ratio seems to be higher around larger areas of interstitium in relation to the data of the total section, but no high correlation between these both parameters was observed.

Effect of radiophosphorus on the developing endocrine glands of Swiss albino mice. I. A study on the thyroid gland.

A study has been made on the effects of P-32 on the developing thyroid gland of Swiss albino mice. The series of experiments reveal that: (I) P-32 at the dose rate of 1.0 muCi/g body-weight to the seven-day pregnant mouse has almost no effect on the 18-day foetus or as late as 14-day postnatal thyroid. (II) With the same dose, when given to one-day-old mouse, signs of dead mitotic figures are noticed. The effects are more pronounced in the beginning, but after the fourth week no morphological changes are seen. (III) When a seven-day-old animal is injected with the same dose, dead cells are found in the earlier stages followed by the damage to the vascular structure of the epithelium. (IV) The 14-day injected group shows damage to the epithelium in the form of condensed nuclei and loss of colloid from the follicles. Replacement of follicles by fatty degeneration is evident. (V) A maximum susceptibility to P-32 irradiation is shown by 21-day-old injected animals, where epithelial desquamation and replacement of follicles by fatty degeneration are easily marked one week after the injection. (VI) In the last group, where P-32 is given at the 28th day after parturition, there may be a tendency of the thickening of the blood vessels. (VII) The females show a greater damage to P-32 injection than the males in the postnatal period studied.

Decreased basal and acute insulin-stimulated effect on the uptake of glucose and amino acid in vitro by thyroid glands from streptozotocin-diabetic rats.

Since experimental diabetes in rats and mice is associated with impairment of several aspects of thyroid function, we determined glucose and amino acid uptake in vitro by isolated thyroid glands from normal and streptozotocin-diabetic rats. Adult male Wistar rats weighing 150-200 g were used. Diabetes was induced by intraperitoneal injection of streptozotocin (STZ, 65 mg/kg body weight) and after five days only rats with blood glucose levels higher than 250 mg/dl were used. The thyroid glands were preincubated in Krebs-Ringer bicarbonate buffer in the presence or absence of insulin (0.7 nM to 7 muM) for 90 min and then incubated with the same concentration of the hormone or its vehicle plus 0.2 microCi of [1-14C]-2-deoxy-D-glucose ([14C]DG) or [1-14C] methylaminoisobutyric acid ([14C]MeAIB) for 15 to 180 min. The uptake of [14C]DG or [14C]MeAIB by the thyroid glands of normal rats increased as a function of incubation time, and the presence of insulin (7 microM) induced a significant increase of labelled DG from 3.30 +/- 0.11 to 4.16 +/- 0.12 and of labelled meAIB from 1.79 +/- 0.06 to 3.10 +/- 0.17 tissue/medium ratio (T/M) and after 45 min of incubation. The lowest concentration of insulin that increased both [14C]DG and [14C]MeAIB transport was 7 nM. Thyroid glands from STZ rats exhibited lower basal values of [14C]DG (4.03 +/- 0.11 T/M) or [14C]MeAIB uptake (1.05 +/- 0.05 T/M) than glands from normal rats (4.62 +/- 0.13 and 1.70 +/- 0.08 T/M, respectively). Insulin produced a stimulatory effect on the transport of both substrates in STZ rats. However, the maximal stimulating concentration of the hormone did not restore [14C]DG and [14C]MeAIB uptake to control values (4.89 +/- 0.17 in STZ rats versus 5.44 +/- 0.17 T/M in controls for [14C]DG, and 1.51 +/- 0.11 in STZ rats versus 2.19 +/- 0.10 T/M in controls for [14C]MeAIB). These results indicate that insulin exerts a direct action on the thyroid gland, and its absence or reduction affects thyroid metabolism, contributing, at least in part, to the abnormality in thyroid function associated with diabetes mellitus.

Ultrastructure of the thyroid glands of rats fed photomirex: an 18-month recovery study.

The thyroid glands, from rats which had received photomirex (0.05, 0.5, 5 or 50 ppm) or mirex (5 or 50 ppm) in their diets for 28 days and maintained for 18 months on clean diet, were examined by electron microscopy. A dose of 0.05-5 ppm photomirex resulted in an augmentation of principal follicular cell heights. Their cytoplasm exhibited a marked numerical increase of secondary lysosomes. In addition, at 5 ppm, follicular cells in some segments of the gland contained only a few secondary lysosomes. After dietary exposure to 50 ppm photomirex the number of lysosomal elements remained elevated in epithelial cell cytoplasm. Morphological aberrations in the follicles could not be detected after the 5 ppm mirex treatment. The most remarkable alterations in this experiment occurred in the thyroid glands of animals ingesting 50 ppm mirex where columnar follicular cells were engorged with deformed lysosomal bodies. Remaining components of the follicular cell architecture in all the treated groups were unaffected and were similar to those in the control group. These results demonstrated that following a 28-day dietary exposure of rats to photomirex (0.05-50 ppm) or mirex (50 ppm), alterations in the thyroid glands persisted for at least 18 months.

[Effects of excessive iodine on apoptosis and expression of bcl-2, bax genes in the cells of brain and thyroid gland in guinea pigs].

OBJECTIVE: To investigate the effects of excessive iodine intake on apoptosis and the expression of bcl-2, bax genes in the cells of brain and thyroid gland. METHODS: Guinea pigs, divided into two groups at random, were fed with distilled water containing iodine 50 microg/L (a control group with appropriate iodine) and 10,000 microg/L (a trial group with excessive iodine), respectively for six months. Apoptosis rate and expression of bcl-2, bax gene in the cells of cerebral cortex, hippocampus and thyroid gland in guinea pigs were quantitatively determined by flow cytometry and immunofluorescent techniques. RESULTS: The proportion of apoptosis in the cells of cerebral cortex, hippocampus and thyroid gland was significantly higher in the group with excessive iodine intake than that in the control one (P < 0.01). The level of expression of bcl-2 gene was notably decreased (P < 0.05), but that of bax was notably increased (P < 0.01), in the cerebral cortex, hippocampus and thyroid gland in the group with excessive iodine than in the control one. The ratio of bcl-2 to bax in the cerebral cortex, hippocampus and thyroid gland of the group with excessive iodine was decreased significantly (P < 0.01) than that of the control one. CONCLUSION: Excessive iodine intake may induce apoptosis in the cells of cerebral cortex, hippocampus and thyroid gland. The over-expression of bax gene and the down-regulation of bcl-2, which can then cause the decrease in a ratio of bcl-2 to bax, appear to play an important role in the course of apoptosis induced by excessive iodine intake.

[Participation of prostaglandins in the mechanism of hypothalamo-hypophyseal regulation of thyroid gland function].

Prostaglandins E (PGE) content was studied in the hypothalamus, hypophysis, thyroid gland and blood of rats after thyroliberin injections, thyrotropin administration, and thyroidectomy in experimental hyperthyroidism. Changes in the PGE level, cyclic adenosine monophosphate (cAMP), and in thyroxin secretion in the thyroid gland of man under the effect of thyrotropin and acetylsalicyclic acid were studied. The effect of acetylsalicylic acid on thyroxin and triiodothyronine secretion in healthy persons and those suffering from thyrotoxicosis was investigated. Analysis of the results obtained indicated that PGE played a significant role in the mechanism of the thyroid gland function regulation.
