Purification and characterization of a protein tyrosine acid phosphatase from boar seminal vesicle glands.

A protein tyrosine phosphatase (PTPase) with acid phosphatase activity was purified (500-fold) from the fluid of boar seminal vesicles. Preparative purification was performed with a 3-step procedure, employing FPLC S-Sepharose Fast Flow, Mono Q and Superdex 75 column. Protein tyrosine acid phosphatase (PTAPase) was homogeneous by polyacrylamide gel electrophoresis (PAGE, SDS-PAGE). PTAPase is a glycoprotein which has a molecular weight of about 41-42 kDa. This enzyme was maximally active at pH 5.5, and its thermostability was less than 80 degrees C. The K(m) value for p-nitrophenylphosphate, a specific synthetic substrate, was 0.87 x 10(-3)M, however, higher substrate specificity was shown when phosphotyrosine (K(m)=0.37 x 10(-3)M) and protein fragments, such as gastrin (K(m)=0.0032 x 10(-3)M) and hirudin (K(m)=0.0075 x 10(-3)M), were used as substrates. Activity of PTAPase was inhibited by dephostatin, molybdate and orthovanadate by 100, 95 and 70%, respectively, when phosphotyrosine was used as the substrate. Immunofluorescence study has shown that the seminal vesicles are the only source of PTAPase in boar seminal plasma.

Effect of mesenchymal glandular inductors on the growth and cytodifferentiation of neonatal mouse seminal vesicle epithelium.

To investigate the developmental properties of glandular mesenchymal inductors along the cranial-caudal extent of the developing male urogenital tract, neonatal mouse seminal vesicle epithelium (SVE) was combined with mesenchyme of the seminal vesicle (SVM), urogenital sinus (UGM), bulbourethral gland (BUG-M), or bladder (BLM) and grafted under the renal capsule of adult syngeneic or athymic male mice. Both SVM + SVE and UGM + SVE tissue recombinants expressed SV histogenesis and SV secretory proteins. BUG-M + SVE recombinants exhibited extensive growth as evidenced by a 36-fold increase in wet weight and a 27-fold increase in DNA content; however, the glandular structures that were induced in the SVE lacked the convoluted mucosa typical of SV. Furthermore, neither SV nor prostatic secretory proteins were detected in these recombinants. SVE grown in association with BLM failed to develop altogether. Thus, the ability to promote SV histogenesis and function is distinctly different in mesenchyme of cranial (SVM and UGM) versus caudal (BUG-M) regions. This implies the existence of a glandular inductive field in the developing male urogenital tract within which inductive activity varies regionally.

Differential effects of 2-difluoromethylornithine and methylglyoxal bis(guanylhydrazone) on the testosterone-induced growth of ventral prostate and seminal vesicles of castrated rats.

2-Difluoromethylornithine totally prevented any increases in putrescine and spermidine concentrations in the ventral prostate of castrated rats during a 6-day testosterone treatment. Prostatic ornithine decarboxylase activity was inhibited by 80%, whereas S-adenosylmethionine decarboxylase was stimulated by more than 9-fold. In seminal vesicle, the inhibition of putrescine and spermidine accumulation, as well as of ornithine decarboxylase activity, was only minimal, and no stimulation of S-adenosylmethionine decarboxylase was observed. Administration of methylglyoxal bis(guanylhydrazone) to castrated androgen-treated rats resulted in a marked increase in concentrations of all prostatic polyamines. Prostatic ornithine decarboxylase activity was nearly 2 times and adenosylmethionine decarboxylase activity 9 times higher than that of the testosterone-treated animals. In contrast with ventral prostate, methylglyoxal bis(guanylhydrazone) treatment inhibited moderately the accumulation of spermidine and spermine in seminal vesicle, although both ornithine decarboxylase and S-adenosylmethionine decarboxylase activities were stimulated. Difluoromethylornithine inhibited significantly the weight gain of ventral prostate, but methylglyoxal bis(guanylhydrazone) produced a substantial increase in prostatic weight. These changes were largely due to the fact that the volume of prostatic secretion was greatly decreased by difluoromethylornithine, whereas methylglyoxal bis(guanylhydrazone) increased the amount of secretion. Treatment with difluoromethylornithine strikingly increased the methylglyoxal bis(guanylhydrazone) content of both ventral prostate and seminal vesicle, but even under these conditions the drug concentration remained low in comparison with other tissues. The results indicate that a combined use of these two polyamine anti-metabolites does not necessarily result in a synergistic growth inhibition of the androgen-induced growth of male accessory sexual glands.

[Early results following radical prostatectomy in patients with capsule invasion, seminal vesicle infiltration and micrometastases].

One hundred thirty-three patients with prostatic carcinoma underwent bilateral pelvic lymphadenectomy and radical retropubic prostatectomy between 1975 and 1988. Patients who had a localized prostatic carcinoma (less than or equal to T2b N0 M0 or less than or equal to B 2) and a small number (n = 10) with limited T3 carcinoma were considered for surgical therapy on the basis of a digital prostatic examination. Histological examination revealed locally advanced prostatic carcinoma in 89 patients with capsular infiltration or perforation and seminal vesicle involvement. Microscopic lymph node metastases were noted in 14 cases. Some patients with capsular perforation and seminal vesicle involvement received adjuvant therapy (orchiectomy or radiation). All patients with lymph node metastases were treated by orchiectomy. One local failure occurred among 24 patients with capsular infiltration within 42 months of follow-up. No failure occurred in stage pT3 disease (capsular perforation) with adjuvant therapy (n = 12) and in stage pT3 disease (seminal vesicle involvement) with (n = 9) and without (n = 12) adjuvant therapy after mean follow-up periods of 35, 42 and 52 months, respectively. Distant metastases occurred in 2 patients with stage pT3 disease (capsular perforation) without adjuvant therapy (n = 18) within a mean follow-up of 51 months, and 1 of these patients died of prostatic cancer. Distant metastases occurred in 3 patients with pT2-pT3N1 disease within a mean follow-up of 54 months: 1 of these patients died of prostatic cancer. Local failure was noted in 1 patient in this group.(ABSTRACT TRUNCATED AT 250 WORDS)

The removal of the seminal vesicles from the boar and the effects on the semen characteristics.

A technique is described for the removal of the seminal vesicles from the boar. The operation was carried out on twelve animals and six of the animals were subsequently trained for semen collection. The seminal plasma from the boars after surgery compared with normal litter mates had a more watery consistency and did not form the characteristic gelduring ejaculation. The sperm concentration was 49% lower while the total reduction of sperm number ejaculate was 78% in the experimental animals, but the ratio of living to dead spermatozoa remained unchanged. The concentrations of citrate and protein were significantly depressed in the seminal plasma of the animals after surgery and the pH increased; the osmolarity remained unchanged. In semination of gilts with the semen from experimental boars revealed no significant loss of fertility compared with the normal controls. Animals slaughtered up to 17 months after surgery showed no regeneration of the seminal vesicles.

Ki-ras mutations with frequent normal allele loss versus absence of p53 mutations in rat prostate and seminal vesicle carcinomas induced with 3,2'-dimethyl-4-aminobiphenyl.

We have developed a prostate carcinogenesis model in Fischer 344 rats using 3,2'-dimethyl-4-aminobiphenyl (DMAB) as a carcinogen to examine various potential modifying factors. In this study, mutational changes in the ras and p53 genes were assessed in DMAB-induced rat prostate and seminal vesicle carcinomas by single-strand conformation polymorphism analysis and subsequent direct DNA sequencing. Eight of 22 prostate adenocarcinomas (three of nine (33.3%) from the ventral lobe and five of 13 (38.5%) from the dorsolateral lobe, including three transplantable tumors) and one of 11 seminal vesicle adenocarcinomas (9.1%) demonstrated point mutations in the Ki-ras gene. One prostate malignant fibrohistiocytoma examined was negative. Among the positive cases, five (three ventral prostate carcinomas and two transplantable tumors) also showed loss of the normal allele. In contrast, other than one mutation in the p53 gene in the malignant fibrohistiocytoma, there were no mutations in the Ha-ras or p53 genes. These results indicate that mutational activation of the Ki-ras gene, but not of the Ha-ras or p53 genes may play a mechanistic role in prostate and seminal vesicle carcinogenesis by DMAB and that a loss of the normal allele of the Ki-ras gene may also be involved in the process.

Physical analysis of ejaculate to evaluate the secretory activity of the seminal vesicles and prostate.

The purpose of this study was to ascertain whether analysis of the physical properties of ejaculate also has any diagnostic potential for evaluating the function of these accessory sex glands. Diverse normal and abnormal states of coagulation, liquefaction, volume, viscosity and pH were studied with regard to the levels of biochemical markers of the seminal vesicles (fructose and inorganic phosphorus) and prostate (calcium, zinc and acid phosphatase). Fructose and inorganic phosphorus were significantly decreased in samples with absent or poor coagulation (p<0.001), volume < 2.0 mL (p=0.009 and p<0.001, respectively), hypoviscosity (p=0.013 and p<0.001), hyperviscosity (p=0.006 and p<0.001) and pH < or = 7.1 (p=0.018 and p<0.001). Also, fructose and inorganic phosphorus were significantly decreased in samples with liquefaction > 120 min (p=0.003) and pH > 8.0 (p<0.001), respectively. Calcium, zinc and acid phosphatase activity were significantly increased in samples with absent or poor coagulation (p<0.001), and significantly decreased in samples with volume > 5.0 mL (p=0.007, p=0.034 and p=0.011) and pH > 8.0 (p<0.001). Also, calcium and zinc were significantly increased in hypoviscous samples (p=0.012 and p=0.003), whereas the zinc concentration was significantly lower in hyperviscous samples (p=0.026). Using receiver operating characteristic (ROC) curve analysis, pH showed the highest predictive power to identify prostate dysfunction (83.6%) and simultaneous prostate and seminal vesicle dysfunction (98.8%). Physical analysis of ejaculate was also found to be clinically useful for evaluating the secretory activity of the seminal vesicles and prostate. Abnormal coagulation, liquefaction, volume, viscosity and pH strongly suggest gland dysfunction.

Structure of the vaginal plugs generated by normal rats and by rats with partially removed seminal vesicles.

Normal male rats generate vaginal plugs that appear to be firmly apposed to the vagino-cervical junction and permit a large number of spermatozoa to reach the uterus. A few spermatozoa form entangled masses inside these plugs, as revealed by light microscopy. Males in which the seminal vesicles have been partially removed produce plugs that are smaller and softer than those generated by normal males, and the plugs display a cup-like structure at the proximal end. The cup-like structure is completely filled with spermatozoa that exhibit a characteristic arrangement in relation to the plug material. In this situation, the number of spermatozoa that reach the uterus is very much reduced. Experiments were also carried out to explore the restoration of sperm transport by addition of a vaginal plug. Such experiments involved successive matings of individual females with a seminal vesicle-deprived male and with a vasectomized male (which generated the plug) and also the intravaginal injection of seminal vesicle secretions after mating with a seminal vesicle-deprived male. In none of the experimental situations was transport of spermatozoa to the uterus restored, and the plug consisted of a large quantity of trapped spermatozoa inside a mass of coagulated proteins. The results suggest that the structure of the plug depends on the amount of seminal vesicle secretion present in the ejaculate and that the vaginal plug must be formed immediately after deposition of the sperm if spermatozoa are to reach the uterus.

Regression of autophagic vacuoles in pancreatic acinar, seminal vesicle epithelial, and liver parenchymal cells: a comparative morphometric study of the effect of vinblastine and leupeptin followed by cycloheximide treatment.

Treatment of mice with both leupeptin (0.06 mg/g body wt) and vinblastine (0.05 mg/g body wt) for 2 h caused a many-fold enlargement of the autophagic-lysosomal compartment of pancreatic acinar, seminal vesicle epithelial, and liver parenchymal cells. In all three types of cells a predominance of large, dense bodies was seen after leupeptin treatment and that of typical autophagic vacuoles were seen after vinblastine treatment. An exponential decrease of the volume fraction of autophagic vacuoles was observed in leupeptin-treated cells after the administration of cycloheximide (0.2 mg/g body wt). The half-life of autophagic vacuoles estimated from the decay curve was 5.3, 5.7, and 6.6 min for pancreatic, seminal vesicle, and liver cells, respectively. Our data suggest that sequestered cytoplasmic material rapidly enters the lysosomes in leupeptin-treated cells and accumulates in this compartment. In contrast, no regression of the autophagic vacuole compartment of pancreatic and seminal vesicle cells was observed after the administration of cycloheximide to animals pretreated with vinblastine, and only a slight decrease was seen in liver cells. These observations show that the lifetime of autophagic vacuoles is prolonged by vinblastine resulting in their accumulation in the cells. However, our measurements also lend support to the view that in addition to the accumulatory effect on undegraded cytoplasmic material, stimulation of sequestration may play a role in the enlargement of the autophagic lysosomal compartment after treatment with leupeptin as well as with vinblastine in all three types of cells investigated.

Ultrastructure of the epithelium of the seminal vesicles after the application of the preparation stimulating the secretory function of the prostate gland.

In the electron microscopic picture of the seminal vesicles in rats some changes within Golgi apparatus and within vesicles of secreta were found after the application of preparation stimulating the secretory function of prostate an not being a hormone from the group of androgens. Hyperplasia of Golgi structures and changes of its localization in the cell as well as the increase in number and capacity of vesicles of secreta give evidence of the intensified secretory processes taking place in the cells of seminal vesicles.

Complex carbohydrate histochemistry of the lateral prostate and seminal vesicle of the guinea pig.

A systematic histochemical study of the complex carbohydrates of the lateral prostate and seminal vesicle of the guinea pig has been made. The complex carbohydrates of the guinea pig male accessory sex glands were partially characterized by various conventional carbohydrate histochemical methods including periodic acid-Schiff, selective periodate oxidation-Schiff reaction, Alcian blue staining at pH 2.5 and 1.0, and high iron diamine. The results indicated that neutral glycoconjugates with 1,2-glycol groups and sialic acids were present in the luminal border and apical cytoplasm of the glandular cells, basement membrane and connective tissue in the lamina propria of the lateral prostate. Similar patterns were demonstrated in the seminal vesicle except that there were relatively fewer or no neutral carbohydrates in the apical cytoplasm of the vesicular epithelial cells. The epithelial basement membrane and connective tissue at the epithelial-stromal interface of both glands were rich in acidic and sulphated glycosaminoglycans. Partial characterization by bovine testicular hyaluronidase indicated the presence of chondroitin sulphates in the lamina propria of the glands.

Evidence for a role of prolactin in prostate and seminal vesicle growth in immature male rats.

To determine if prolactin secreted endogenously by anterior pituitary grafts could augment male accessory organ weights, single anterior pituitary grafts were placed under the kidney capsule of male rats, whereas control animals received a graft of muscle. Three weeks after transplantation, the animals were sacrificed by decapitation and a significant increase in plasma prolactin was observed, which was accompanied by a highly significant increase in the weights of seminal vesicles, ventral and dorsal prostates, and adrenals. To determine the importance of testicular steroids in the response, animals were castrated. The increases in prolactin, seminal vesicle and ventral and dorsal prostate weights still occurred whether or not a small dose of testosterone replacement therapy was employed. In the presence of the pituitary graft, adrenal weight usually increased in these animals as well. To rule out a requirement for adrenal steroids in the response, adrenalectomized-castrate animals were also studied, and the increases in prolactin and prostate weights still occurred although there was no significant increase in the size of the seminal vesicles. To rule out the participation of other pituitary hormones, pituitary grafts were also placed in hypophysectomized animals, and in these animals, there was also a significant increase in prolactin, accessory sex organ and testicular weights, but the adrenals did not increase in size. The grafts failed to alter gonadotropin titers in any experiment. It is concluded that single anterior pituitary grafts are capable of secreting sufficient prolactin to increase the size of the sex accessories and sometimes the testes and adrenals of the rat. Furthermore, the effects on the male sex accessory organs were demonstrable in the absence of testicular or both testicular and adrenal steroids. The data suggest that prolactin may play a physiological role in the growth of the testes, adrenals and sex accessories.

Is there "metabolic" DNA in the mouse seminal vesicle?

THIS STUDY WAS DESIGNED TO ANSWER THE QUESTION: Is H(3)-thymidine uptake by nuclei of the mouse seminal vesicle evidence for DNA synthesis and mitosis, or does it signify some "metabolic" function of DNA unrelated to chromosome duplication? Mice were given an intraperitoneal injection of H(3)-thymidine. Six hours later Feulgen squashes of the seminal vesicle epithelium were made and covered with autoradiographic stripping film. The silver grains above labeled nuclei were counted, and the Feulgen dye contents of these same nuclei were determined photometrically after removal of the grains from the emulsion. Unlabeled nuclei were also measured. The dye contents of non-radioactive nuclei form a unimodal distribution, indicating that polyploidy is absent from this tissue. The radioactive nuclei fall into two groups. In the first, the average dye content is the same as that of the cold nuclei (2C). In the second, the values range from 2C to 4C. In the 2C to 4C group the grain count is proportional to the dye content, showing that incorporation is correlated with synthesis. The radioactive 2C nuclei arose by mitosis during the course of the experiment. This is shown by the following facts: (1) They frequently occur in pairs. (2) They average smaller than unlabeled 2C nuclei. (3) Their average grain count is approximately half that of the 4C nuclei. (4) Labeled division figures are found. (5) A mitotic rate estimated from the number of labeled 2C nuclei accords reasonably well with one based on the number of observed mitoses. Since the incorporation of thymidine accompanies DNA synthesis and precedes mitosis, there is no reason to postulate a special "metabolic" DNA in this tissue.

[Seminal vesicle cyst with ipsilateral renal agenesis: a case report].

A 26-year-old man was referred to our hospital for further evaluation of a cystic lesion in the pelvis. He was completely asymptomatic, although he experienced gross hematuria 10 months before his visit. Drip infusion pyelography (DIP) and abdominal computerized tomography (CT) showed a cystic structure behind the bladder and absence of the left kidney. Transrectal ultrasonography (TRUS) suggested left ejaculatory duct obstruction and seminal vesicle deformity. On cystoscopy a large bulging mass under the left side of the bladder was seen. The left half of the trigone and left ureteral orifice were absent. Vasovesiculography demonstrated dilated and tortuous left seminal vesicle. Three days after this procedure, the patient suffered left epididymitis. It did not respond rapidly to antibiotic therapy, so percutaneous drainage of the cyst was performed. He has been completely free of symptoms and no seminal vesicle cysts have been detected on TRUS at five months follow-up.

[Seminal vesicles: echographic findings in 244 subjects. Evaluation of anatomic data and correlations with pathologic prostatic-vesicular parameters].

Over a 2-year period (1989-90), about 400 cases were examined (out of nearly 2,500 US examinations of the prostate), 244 of which had subsequent bioptic, surgical or autoptic confirmation. This paper reports the anatomic, clinical and pathologic data relative to the seminal vesicles of 244 subjects who underwent transrectal US of the prostate for different clinical indications. Our biometric values were slightly different from those reported in literature, the latter being based on autoptic findings. Agenesis and monolateral ectopia were confirmed to be extremely rare: 4 cases only (1.6%) in our series. A typical finding was the nearly perfect symmetry; in size and structure, of the two seminal vesicles in the same subject; as a matter of fact, a loco-regional pathologic condition was detected in all the patients (14) with asymmetry. A hundred per cent agreement was observed between the US patterns of the seminal vesicles and the prostatic patterns in case of acute/subacute prostatic inflammation involving also the 2 glands: this parameter can be used to follow the clinical course of the inflammatory process.

Carbohydrates in the epithelium lining the seminal vesicle of the rat as studied by histochemical methods of light and electron microscopy.

In the epithelium lining the seminal vesicle of the rat, carbohydrates have been studied by means of histochemical methods of light and electron microscopy. In light microscopy, the free surface and granules of different sizes in the distal cytoplasm of the epithelial cells and lumenal masses of secretions were shown to exhibit positive reactions for carbohydrates with 1,2-glycol and acidic groups and sialic acid residues. In electron microscopy, the surface coat of the plasma membrane, certain elements of the Golgi complexes, dense cores of secretory granules and glycogen particles in the epithelial cells were found to exhibit positive reactions for carbohydrates with 1,2-glycol groupings. The correlated structures and functions of the carbohydrate-containing components have been discussed in the light of the known physiology of the seminal vesicle in the rat.

Pharmacological characterization of the postjunctional alpha-adrenoceptors of the rat isolated seminal vesicle.

In an attempt to define the pharmacological characteristics of the postjunctional alpha-adrenoceptors of the rat seminal vesicle, responses to certain phenylethanolamine and imidazoline agonists were investigated, in vitro, under experimental conditions outlined by Furchgott (1972), using alpha 1-selective, non-selective and alpha 2-selective adrenoceptor antagonists. Adrenaline (ADR), noradrenaline (NA) and phenylephrine (PE) produced concentration-dependent contractions. In many experiments the concentration-response (C-R) curves had a distinct "shoulder" at the level of 60-80% of the maximum response (Emax), a situation reminiscent of the rat anococcygeus muscle and the rat basilar artery. The relative potencies of ADR:NA:PE, derived from their EC50 values, were 4.07:1:0.26. In contrast clonidine, oxymetazoline and naphazoline failed to contract the tissue even in concentrations up to 1 X 10(-3) M. In fact the imidazoline derivatives prevented responses to the phenylethanolamines. The antagonist action of clonidine, against phenylephrine, was studied in detail. Prazosin, phentolamine, yohimbine, corynanthine and clonidine all caused a rightward displacement of the C-R curves for NA without depressing Emax. The Arunlakshana and Schild plots of the data were linear and had slopes not significantly different from unity. The pA2 estimates obtained were 9.17 (9.13-9.21) for prazosin, 8.58 (8.07-9.09) for phentolamine, 6.70 (6.44-6.98) for yohimbine and 7.05 (6.81-7.30) for corynanthine. Clonidine had a pA2 value of 6.60 (6.55-6.67) against phenylephrine. On the basis of results obtained with antagonists, the postjunctional alpha-adrenoceptors of the rat seminal vesicle could be firmly placed in the gross category of "alpha 1".(ABSTRACT TRUNCATED AT 250 WORDS)
