[Immunochemical characteristics and serologic properties of lipopolysaccharides isolated from various Brucella species using various extraction methods].

The results of the comparative analysis of LPS isolated by different methods of extraction from the cultures of several Brucella species differing in their virulence are presented. Purified LPS preparations have been obtained from Brucella virulent and vaccine strains by using such methods as water-phenol extraction, Boivin's method, mild alkaline hydrolysis of the antigen according to Boivin's procedure. The presence of certain relationship between the method used for the extraction of Brucella LPS to be compared and their chemical composition, immunological characteristics and serological activity has been established. As shown in this investigation, in the process of the preparation of a highly sensitive diagnosticum for the passive hemagglutination test the use of LPS obtained from Brucella virulent strains, but not from the vaccine strain, by the method of mild alkaline hydrolysis according to Boivin's procedure is expedient. The data presented in this work indicate that the soluble complex of lipid A obtained from Brucella LPS has been found to possess serological activity. The results of the study of the serological properties of lipid A indicate that the lipid component may also play a certain role in the manifestation of the serological activity of Brucella LPS.

A comparison of serological, cellular and DNA-RFLP methods for HLA matching in the selection of related bone marrow donors.

Serological, cellular and DNA-RFLP (restriction fragment length polymorphism) methods of determining HLA compatibility between 10 leukaemic patients and potential related bone marrow donors were systematically compared. DR beta/DQ alpha/DQ beta/DNA-RFLP typing of these families gave results in agreement with those obtained by serological methods (matching for HLA-A, -B and -DR), supported by mixed lymphocyte culture (MLC) data, indicating the validity and accuracy of DNA-RFLP matching in transplantation. However, a significant minority of four leukaemic patients plus two healthy individuals were not clearly HLA-DR typable by serology, but all such individuals were easily typable by DNA-RFLP. These results were supported by MLC data, where available. In addition, all data were in agreement with previously reported correlations between DNA-RFLPs and HLA-DR serology, allowing unambiguous assignment of HLA-DR types where these were previously in doubt. These results demonstrate the value of DNA-RFLP HLA class II DR and DQ typing in leukaemic patients requiring marrow transplantation who are not clearly typable by traditional methods and suggest that this approach should constitute an important element of future HLA matching programmes for bone marrow transplantation.

Influenza B, Q fever, and the consequences of febrile illness occurring during jungle warfare training: a clinical and serological study.

Seventy-one Royal Marines undergoing jungle warfare training were studied clinically and serologically to determine the incidence and consequences of febrile illness. During acclimatization and the early part of the deployment the incidence of illness having an impact on training was low; during the latter part an outbreak of influenza B occurred, with 25 cases diagnosed clinically. Only 12 of these were confirmed serologically, probably because the outbreak was still in progress when the second samples were taken. A further four subclinical cases were uncovered by the serological study. Five cases of heat exhaustion occurred, one Marine suffering two episodes. Four patients required case-vac from the jungle. Three Marines were found to have serological evidence of previous acute Q fever, including the patient who had two episodes of heat exhaustion. This incidence is higher than expected, and warrants further investigation.

[Detection of streptococci from various serological groups in animals].

During the period from 1985 to 1988 we determined 228 strains of streptococci isolated from samples of various sorts of biomaterials, mainly animals. From this set of streptococci we classified 207 strains into serological groups according to the Lancefield classification and about 30% strains into species by means of serological and biochemical methods. Most of the strains were allocated to group C (37.28%) and group Q (17.39%). 89 streptococci strains originated from pigs, 40 strains from horses, 13 from cattle, 12 from dogs, 9 from poultry and 8 from coypu. The other streptococci strains originated from other animal species or from different material. Most serological groups of streptococci were determined in pigs, less in cattle (7), in poultry (6), in dogs (6), in horses (5) and in coypu (3). Streptococci of the serological group Q were determined in as many as 9 animal species, group C in 8 species and group G in 6 species.

[Serology of HIV infection].

Serological testing aimed at the detection of antibodies against the human immunodeficiency virus (HIV) is now performed throughout the world to screen stored blood for transfusion, for the diagnosis of HIV infection and of AIDS, and to determine the prognosis of AIDS patients. The present report, based on 258,090 serum samples screened for HIV antibodies, gives an overview of the theoretical and practical aspects of both modern screening methods and confirmation tests for HIV1 and HIV2, the demonstration of serological antigen and IgM, and HIV serology of the cerebrospinal fluid. Knowledge of HIV serology appears to be important for any physician, and in particular for the dermatologist, since HIV infection (like syphilis) can cause a variety of dermatological manifestations mimicking other dermatoses.

Comparative study of various serological criteria in malaria survey.

The determination of the most appropriate serological criterion related to the blood slide examination in malaria surveys was carried out in western Thailand, an area of low persistent malaria transmission. Six different serological criteria were assessed against the blood slide results among 5001 paired blood samples of one-month interval. The indices used in the analysis were sensitivity, specificity and kappa coefficient and the blood slide examination was used as the reference test. It was apparent that a titre of greater than or equal to 1:1024 was the most appropriate serological criterion related to the blood slide results. This serological criterion can be used to indicate the high or low malaria exposure group, particularly in an area of low transmission.

[Statistical relations between blood and cerebrospinal fluid serology in patients with syphilis].

In any case of latent syphilis without conclusive history, an examination of the cerebrospinal fluid (CSF) is generally recommended. The present paper deals with the question whether particular findings obtained by serologic tests on the blood can supply sufficient proof to rule out any specific involvement of the central nervous system in an individual patient. Serologic investigation on 148 blood-CSF sample pairs revealed striking correlations between several blood and CSF parameters on a high level of statistical significance. Based on blood serology data alone, a multi-dimensional, non-parametric statistical test (the k-nearest-neighbor method) was able to rule out specific involvement of the CSF in more than one third of the patients. We consider our approach useful in certain cases where CSF examination should rather be avoided, especially in patients with impaired general condition due to other diseases. Yet, in any case of neurological irregularity, CSF examination should generally be carried out regardless of the results of blood serology.

Toxoplasma infection after human allogeneic bone marrow transplantation: clinical and serological study of 80 patients.

Systematic clinical and serological studies to evaluate the frequency of toxoplasmosis in bone marrow transplant recipients were performed in 80 consecutive patients. Antitoxoplasma antibody titres were measured in donors and recipients before transplant and subsequently post-transplant. Before bone marrow transplant, 54 recipients were seropositive and 26 were seronegative, whereas 35 donors were seropositive and 45 were seronegative. After bone marrow transplant, the frequency of clinical and serological manifestations of toxoplasmosis appeared closely related to the recipient's serological status before transplant. In the seronegative group of patients before transplant the incidence of toxoplasmosis was low: only two patients experienced seroconversion 3 months after bone marrow transplant and one developed clinical symptoms consistent with toxoplasmosis but without cerebral involvement. Clinical toxoplasmosis or secondary elevation of antibody titres was mostly observed in pre-bone marrow transplant seropositive patients; in this group, cerebral toxoplasmosis occurred in four patients and a significant secondary rise of antibody titres was observed in 16 patients. It thus appears that toxoplasmosis is most often related to a reactivation of latent cysts. Prophylactic treatment may be useful in patients presenting serological evidence of past or latent infection before bone marrow transplant.

[Dengue 2 in eastern Senegal: serologic survey in simian and human populations. 1974-85].

After the previously reported isolations of dengue 2 virus in eastern Senegal in 1974 and 1981-1982, a retrospective serological study on simian and human populations was carried out in the same area. We investigated 1,095 simian sera collected at regular intervals between 1974 and 1984 from wild caught monkeys and 1,783 human sera from young children less than 11 years old collected during punctual surveys after the rainy season from 1976 to 1985. Sera were tested using HAI test, CF test and for someone's ELISA for specific IgM antibodies. Serological data from monkeys corroborated the virus isolations and demonstrated the existence of two epizootics in 1974-1975 and 1981-1982. No CF antibodies were detected in children sera up to 1981 epizootic when about 11% of tested sera showed a probable infection by dengue 2 virus, no clinical dengue infections were notified by the medical staff. After 1982, serological results showed that the virus maintained in the same area until 1985. The mechanism of the circulation of dengue 2 virus in eastern Senegal is discussed on the basis of these serological results.

IFA serological surveys of malaria in north, north-west and south-west parts of Iran.

In the IFA serological surveys of malaria carried out in north, north-west and south-west parts of Iran during 1975-1982 altogether 9,132 subjects were studied for malaria antibodies and parasitaemia. Serological data indicated probable malaria transmission in a consolidation area where the autochthonous cases of malaria were reported a year after this serological study. Asymptomatic parasite carriers of P. malariae were found by IFA and parasite concentration techniques among the professional blood donors and the residents of a village without any recent malaria history. IFA results with P. falciparum antigen reflected the malaria histories in the studied areas of west Azerbaijan better than P. vivax antigen. The serological and parasitological findings in the nomads of Bakhtiary tribes showed that the nomads are more exposed to malaria infection in the winter quarters of Izeh area and they are also more under malaria control programme when they are living in this area. In comparison of IFA results of 438 paired plasma and dried blood samples tested with P. vivax and P. falciparum antigens, there was no significant difference between SPR in plasma and dried blood samples, however in the dried blood samples collected by malaria surveillance agents on filter paper SPR and GMRT were considerably low.

The serologic response to Meth A sarcoma vaccines after cyclophosphamide treatment is additionally increased by various adjuvants.

We have shown previously that the serologic response of BALB/c mice to immunization with BALB/c sarcoma Meth A cells can be more effectively augmented by pretreatment with cyclophosphamide (Cy) than by the use of adjuvants. The serologic response was directed against a highly restricted cell surface antigen, closely related to or identical with the unique transplantation antigen characteristic for this tumor. In this paper, we report the results of our attempts to obtain additional augmentation by using Cy and adjuvants together. For these studies, the optimal Cy dose, interval between Cy and vaccine administration, and vaccine cell number were determined. Mice were injected with Cy 25 mg/kg i.p., and 3 days later, with viable irradiated (10,000 rad) Meth A cells subcutaneously, under conditions in which only few mice produced antibody. Sera were tested for antibody with reactivity against Meth A by complement dependent cytotoxicity (CDCX), which predominantly detects IgM, and by the protein A (PA) and anti-IgG assays, which detect IgG. Of the various adjuvants tested, only monophosphoryl lipid A (MPLA) and CP20,961 resulted in significantly increased titers of reactivity in both the CDCX and PA assays over that obtained when using Cy alone. Although the mean titers observed with CDCX ranged between 1/160 and 1/320, no titer above 1/40 was observed with the PA assay. The specificity of the CDCX reactivity detected by the assay for the Meth A antigen was ascertained by absorption analysis of selected sera by using a panel of BALB/c spleen and tumor cell lines grown in vitro or in vivo. PA titers were too low to permit absorption analysis, and the titers obtained in the anti-IgG assay were lower still. Attempts to augment the anti-Meth A IgG response or to convert the IgM response to IgG were unsuccessful. The combined approach described here (i.e., vaccination with irradiated syngeneic tumor cells plus MPLA in Cy-pretreated mice) was also shown to be effective in augmenting the serologic response against two additional murine leukemia virus-negative sarcomas that are known to be less immunogenic, CMS4 and CMS5. It appears, therefore, that this combined approach may be applicable to stimulating serologic responses against a variety of tumor cell surface antigens.

Serologic abnormalities in spontaneous and drug-induced systemic lupus erythematosus.

Extensive serologic changes occur in systemic lupus erythematosus (SLE) which are probably secondary to unknow primary cause(s). The New Zealand hybrid mouse model most likely has a viral-induced disease and does not show many of the clinical features of the human disease. The best example of human SLE which provides a clue to etiology is the drug-induced type, particularly that due to procainamide. In these patients it is possible to study the development of serologic changes prior to the onset of clinical manifestations, and then observe regression of the clinical and serological changes on withdrawal of the medication. Although there is a rough correlation between the many serologic abnormalities and the clinical picture, enough exceptions exist, so that single tests such as serum complement, anti-DNA antibodies, per cent DNA binding, and others, cannot be used as a sine qua non for management. Care of the patient still remains a clinical problem guided by various laboratory procedures but not dependent on any one. Alkyating agents have a limited role in the treatment of lupus nephropathy and cutaneous vasculitis but azathioprine is probably of no value in SLE.

Serological types of Diplococcus pneumoniae isolated from the respiratory tract of children with cystic fibrosis and children with other diseases.

The distribution of serological types of D. pneumoniae was investigated in 40 strains isolated from 26 children with cystic fibrosis and 57 strains isolated from 39 children with other diseases. All strains were isolated from sputum or tracheal secretion. The strains from cystic fibrosis patients belonged to 14 different serological types, the most prevalent were 19F, 19A and 3. The strains from the other group of children belonged to 20 different serological types, the most prevalent were 23F, 19F and 11A. The differences between the two groups of patients as to the prevalences of types were small, and it is concluded that no special serological types of D. pneumoniae are associated with cystic fibrosis.

Serological analysis of H-2 mutations using monoclonal antibodies.

Nine H-2Kb and H-2Db mutants were examined for serological differences from the wild type C57BL/6 using a panel of monoclonal antibodies (mAbs). Several differences (between the Kb mutants) were established with these mAbs. The bm3 and bm11 mutants could be distinguished serologically even though they have very similar amino acid changes. Conversely, the bm 1 and bm 10 mice, which differ in their sites of Kb mutation, express a common serological mutation, suggesting that tertiary conformation is involved in antibody binding. Both bm13 and bm14 (H-2Db mutants) exhibited profound serological alterations in H-2Db molecules when tested with mAbs that bind determinants mapping to both the NC1 and C2M domains of the Db molecule. In addition, bm13 showed altered binding of two anti-H-2Kb monoclonals.

[Aujeszky's disease: serological responsiveness after vaccination of 6-10-week-old piglets with maternal antibody (author's transl)].

Trials were made to study the extent to which 6-10-week-old piglets with and without virus-neutralizing (VN) antibodies are capable of a serological response to vaccination. The piglets were inoculated with three different vaccines: two live vaccines (Aescovac Aujeszky and MK25 vaccine) and an inactivated vaccine (Geskyvac). The titre of virus-neutralizing antibody in the blood was determined for each vaccine in approximately 200 piglets at the time of inoculation as well as within four and twelve weeks after inoculation. The maternal antibodies present at the time of inoculation were found to have an inhibitory action on the serological response following inoculation. Of those piglets in which a maternal VN titre was observed using the above technique, only 9 per cent showed an increase in titre after inoculation. This proportion was 74 per cent in the serologically negative piglets. Marked variations in serological response to the different vaccines were observed. the most positive response was that induced by Aescovac. There was an obvious relationship between the VN titre following inoculation and immunity against intracerebral challenge with 2,000 TCID50 of Aujeszky disease virus. Animals showing titres of 1:4 or over were immune to this challenge.

Biochemical and serological studies on oral ureaplasma.

This study was conducted to study chemically and serologically the characteristics of the Ureaplasmas isolated from the human oral cavity. Two hundred and fifty-one healthy and 12 periodontitis subjects were examined for the incidence of the isolation of Ureaplasmas from their oral cavity. A total of twenty-six strains was isolated from the healthy human saliva. But no strains could be isolated from a variety of clinical specimens obtained from the patients. The serological properties of the isolates were tested by the method of metabolism inhibition test (MI test). Seven out of 26 isolates were serologically identical with either one of the ATCC standard strains. However, the serological types of the other strains could not be demonstrated by the MI test. The biological characteristics of 4 isolates and ATCC strains were tested by the usual method. The isolates did not metabolize glucose and arginine, while all strains hydrolyzed urea. On the other hand, none of the isolates lysed skimmed milk and gelatin. The proteolytic activity of the isolates could be demonstrated by using casein and horse serum proteins as substrates. Zymogram patterns from one of the isolates and Streptococcus salivarius were obtained by polyacrylamide gel electrophoresis of the cells lysed with digitonin or cell protein extracts. On the basis of the gel electrophoresis patterns, it is clear that the urease of the Ureaplasma is different from that of the Streptococcus salivarius.

[Effect of routes and doses of Salmonella abortus ovis inoculation on the abortive effect and serologic response of ewes].

Three successive experiments using a restricted number of Préalpes X Lacaune ewes were designed to form a procedure allowing the reproduction of an abortive infection with Salmonella abortus ovis. Effects of inoculations at mid-gestation were estimated by the duration of gestation, bacteriological excretion at lambing and evolutions of rectal temperatures and of serological antibodies. Serological responses were revealed with four tests: O or H agglutination tests in tubes, H agglutination or complement fixation tests in microtechnics. Intravenous inoculations of 10(9) salmonella induced lethal septicaemia. Intragastric inoculations of 10(7), 10(8), 10(9) or 10(10) salmonella did not produce effects related to inoculated doses; in particular the only abortion obtained in this way occurred after inoculation of 10(7) salmonella. Subcutaneous inoculation of 10(6) to 10(10) salmonella induced serological, bacteriological and clinical responses related to the inoculated doses: ewes with at least two febrile phases during the 20 days post-inoculation aborted, including two ewes not excreting salmonella at lambing. After intragastric or subcutaneous inoculation, and with the four serological tests, the initial raising of titers was followed by an early and rapid decrease, without a new rise round about the time of abortion; no salmonella was isolated from ewes autopsied three to six months after inoculation. It appeared that the subcutaneous inoculation of 10(10) S. abortus ovis viable to ewes at mid-gestation was able to frequently induce a sublethal but abortive infection.

Chlamydiosis in cattle and in man: an epidemiologic and serologic study.

In the Pardubice area, the systematic examinations for Chlamydia-specific antibodies in cattle were started in 1967. In the years 1967-1978 the detectable levels of antibody to Chlamydia were demonstrated in 16.1% of 38,394 sera from cattle and in 18.6% of 5,244 sera from slinking cows. In the year 1979 the percentage of seropositivity in cattle increased significantly to 23.5% of the 4,197 sera examined. Examinations in 1980 revealed that 23.1% of 4,042 sera from cattle, 34.4% of 319 sera from slinking cows and 17.9% of 209 sera from calves were found antibody positive. Over the period from 1974 to 1978 isolates of Chlamydia organisms were obtained from 11.7% of 452 bovine slinks and from 10% of 271 dead or prematurely slaughtered calves. In the years 1979-1981 a total of 775 individuals were subjected to medical and serological examinations for Chlamydia infection using the BIOVETA group antigen. The first group of examinees included 307 farm personnel, attendants to healthy breeds of cattle. Detectable levels of antibody to Chlamydia were recorded in 1.3% of clinically healthy individuals and in 12% of the 75 healthy, randomly selected cows taken care of by this group of personnel. The second group included 280 individuals who were in charge of slinking cows or calves with chlamydial pneumonia, or who were employed in a rendering plant. A total of 7.5% of antibody-positive individuals was found in this group of examinees. Of the 439 calves attended 17.9% had serologic evidence of infection and the 30 dead calves examined were isolate-positive: sera from 176 slinking cows showed positivity in 23.8% of cases. In the third group of 188 persons serving as controls 5.3% had serologic evidence of chlamydial infection. The statistical analysis of deviations from the Gaussian variance normal curve showed neither sex-related nor inter-group differences. At the time of serologic examinations of humans the epidemiologic situation in the given area was extremely favorable. The importance of a close cooperation between the public health and veterinary service personnel (notification of abortions in cattle) and the need of a due attention of gynecologists, dermatologists and urologists are emphasized.
