Effect of dextran-S (alpha, 1-3 dextran) on the growth of plasmacytomas MOPC-104E and J558.

The murine plasmacytomas MOPC-104E and J558 secrete IgM-lambda and IgA-lambda, respectively, which are antibodies to alpha, 1-3 dextran, a constituent of dextran-S (DEX-S) extracted from Leuconostoc mesenteroides. A single i.p. injection of 10 microgram DEX-S into BALB/c mice from 7 days before and up to 3 days after the implantation of 5 x 10(3) plasmacytoma cells protected the BALB/c mice from developing the tumors. Immunization with other antigens, such as Escherichia coli lipopolysaccharide, inulin (alpha, 1-6 linkage), and dextran T-10 (no known alpha, 1-3 linkages) did not protect the mice from developing the tumors. Growth of LPC-1 was not affected by DEX-S. The mechanism of this growth inhibition is unknown. It does not appear to depend solely on binding of antigen to tumor cells, and the limits of the effective time intervals between antigen and tumor injection suggest dependence on the presence of antibody to alpha-1,3 dextran.

Effect of subcutaneously administered 2,6,10,14-tetramethylpentadecane on plasmacytoma growth.

2,6,10,14-Tetramethylpentadecane (pristane), s.c. injected simultaneously with plasmacytoma inocula, enhances the transplantability of tumor cells. The effect is dose and time dependent. The enhancement is shown only by plasmacytoma (MOPC-315 and MPC-11) and not by the murine lymphosarcoma and chondrosarcoma tested. Various mechanisms, such as stress and depression of humoral and cellular immunity, have been considered.

Plasmacytoma of the breast.

Extramedullary plasmacytomas are unusual tumors that generally represent the initial manifestation of systemic multiple myeloma. I studied a plasmacytoma of the breast in a patient who has remained free of symptoms nine years after initial diagnosis.

Intracranial plasmacytoma associated with multiple myeloma.

A 58-year-old man with multiple myeloma presented with a 3-month history of a sensation of occipital pressure, transient blurring of vision and increased bone pain. Funduscopic examination revealed disc swelling, tortuous veins and superficial retinal hemorrhages simulating early hyperviscosity syndrome. Clinical investigation and computerized tomography, however, indicated that an intracranial plasmacytoma was the cause of his papilledema. Cobalt 60 beam therapy and vincristine were added to his treatment regimen. Seven weeks later the intracranial tumour had almost totally disappeared. Symptomatic cerebral compression from an intracranial plasmacytoma associated with systemic disease is uncommon. The diagnosis and management are reviewed.

Type C virus particles in X63-Ag8.653 mouse plasmacytoma cells and in hybridomas derived from it.

In addition to type A virus particles which are common in Balb/c derived mineral oil induced plasmacytomas, type C virus particles have been found in the plasmacytoma cell lines X63-Ag8 and X63-Ag8.653 used in the hybridoma technique. In hybridomas obtained after fusion of X63-Ag8.653 cells with Balb/c spleen cells, C particles were expressed in 5 out of 13 different clones examined, whereas A particles were always present. Virus expression and antibody production do not exclude each other.

Plasmacytomas in rhesus monkeys.

Plasmacytomas were found in two adult male rhesus monkeys. Involvement of vertebral bone marrow, skin, kidney, and soft tissues was present in one case and infiltration of the stomach, lymph nodes, and retroperitoneum in the other. Both tumors were seen to be typical plasmacytomas by light and electron microscopy. Strong IgA immunoreactivity was demonstrated in the tumor cells of one case and weak reactivity in the other using rabbit anti-human immunoglobulin sera and the immunoperoxidase technique. Also, IgA was demonstrated in tumor extracts and in the serum of one case.

Solitary plasmacytoma in the cerebellopontine angle.

A case of solitary plasmacytoma of the skull base growing into the cerebellopontine angle is reported. Myeloma protein of the lambda type light chain was identified in the patient's cerebrospinal fluid. The difficulty in differentiating solitary plasmacytomas of the skull base from other tumors in this location, such as chordoma and meningioma, is discussed.

Intrasellar solitary plasmacytoma terminating in multiple myeloma: a report of a case including electron microscopical study.

A case of multiple myeloma is reported that presented as a solitary plasmacytoma of the bone of the sella turcica. The diagnosis was established by electron microscopy in conjunction with the immunoperoxidase technique. The patient succumbed 37 months after the diagnosis was made. The relation between solitary plasmacytoma of bone and multiple myeloma is discussed. The differential diagnosis of intrasellar tumors may pose serious problems to the surgical pathologist. In difficult cases the value of the immunoperoxidase technique and electron microscopy is emphasized.

Quantitative immunofluorescent analysis of surface phenotypes of murine B cell lymphomas and plasmacytomas with monoclonal antibodies.

In this study, a large series of murine B lymphomas and plasmacytomas were examined by quantitative flow cytometry analysis using a panel of monoclonal antibodies against murine differentiation antigens. These cell lines appear to represent subpopulations of lymphoid cells arrested at distinct stages of differentiation. In general, the pattern of reactions of these monoclonal antibodies with the B cell neoplasms was comparable to the reactions seen with normal cells in the same lineage. The Thy-1.2, Lyt-2, and T-30 differentiation antigens were not detected on any B lymphoma or plasmacytoma. However, certain surface Ig-positive B lymphomas do express the Lyt-1 antigen. With respect to other differentiation markers examined, including E2, F1, ThB, Lgp-100 (Ly 9.1), G2, and T-200 (Ly 5), the reaction of the B cell tumors reflected the expression of these markers on comparable normal cells. This investigation also emphasized the marked intratumor and intertumor heterogeneity that is observed when cloned cell lines are analyzed quantitatively for a large number of surface markers. Thus, this approach may be particularly useful in defining heterogeneity in maturation states within cloned tumor cell lines.

Thyroid-gland plasma cell neoplasm (plasmacytoma).

A 61-year-old man with a primary thyroid plasmacytoma was studied. Intracytoplasmic monoclonal immunoglobulin (IgG-kappa) was demonstrated in tissue sections, using the immunoperoxidase technique. After histologic diagnosis of plasmacytoma, serum immunoelectrophoresis revealed a monoclonal component (IgG-kappa). Roentgenographic skeletal survey and bone marrow examination gave normal results. The patient received postoperative therapy with radiation and drugs. As of this writing, he is alive 20 months after diagnosis, without evidence of tumor.

Plasmacytomas of the NZB mouse.

Plasmacytomas were readily induced in NZB mice by three i.p. inoculations with pristane (2, 6, 10, 14-tetramethylpentadecane). In comparison with comparable induction regimens in BALB/c mice a) the latent period for plasmacytoma development was significantly longer in NZB's; b) the frequency of IgA paraproteins produced by NZB tumors was much reduced and that of IgG (IgG1, IgG2a, IgG2b, IgG3Y was significantly increased; and c) the frequency of carbohydrate-binding paraproteins was 10-fold lower. There was a high frequency of primary ascites containing more than one paraprotein, but none of 25 such tumors produced more than one immunoglobulin class on serial transplantation.

Genetics of susceptibility to pristane-induced plasmacytomas in BALB/cAn: reduced susceptibility in BALB/cJ with a brief description of pristane-induced arthritis.

BALB/cAn and BALB/cJ inbred strains of mice were separated 43 yr ago, but are still very closely related because they carry the same allelomorphs at many loci. When injected 3 times i.p. with 0.5 ml of pristane (2, 6, 10, 14 tetramethylpentadecane) 11% of strain BALB/cJ develop plasmacytomas in contrast to 61% in BALB/cAn sublines. BALB/cJ first injected with pristane as neonates also developed a reduced number of plasmacytomas as compared with BALB/cAn. About 20% of pristane-treated BALB/cAn and 70% of pristane-treated BALB/cJ develop arthritis, which first appeared in the ankle joints after a latent period of 4 mo or more.

Experimental plasmacytoma in mice. II. Qualitative studies of myeloma proteins.

Qualitative studies on paraproteins present in ascites and/or in serum of mice with induced plasmacytoma were performed by the method of immunoelectrophoresis. Myeloma proteins were found in 75 out of 116 plasmacytomas. Among them in 53 cases production of IgA, in 6-IgG3, and in 1 case IgG was detected. In 10 out of 75 samples, two paraproteins present in one ascites were detected.

Experimental plasmacytoma in mice. I. Induction, morphological and biological characteristics.

The induction of plasmacytomas in BALB/c mice by Bayol F or Pristan is described. In some experiments mice treated with Bayol F were antigenically stimulated by repeated injections of allogenic C57BL/6 spleen cells. The incidence of plasmacytoma was 57% in Bayol F treated mice and 59% in the mice subjected to sustained antigenic stimulation. However, the tumors appeared about 6 months earlier in mice stimulated with the allogeneic spleen cells.

A murine plasmacytoma model for screening active drugs for human myeloma.

The basic problem of current animal models is the inability to select and rate the relative effectiveness of new drugs of myeloma of man. As a first step in broaching this problem, results of clinical drug trial studies were used for predicting activity of the drugs in the MOPC 104E plasmacytoma model. Comparison of the model with human myeloma and six additional murine plasmacytomas for drug response indicates close correlation between the reported results in man and the expected results in the animal models. The MOPC 104E model was versatile because drug effects could be rapidly measured in vivo in individual animals. The effect of the drug on the rate of growth, the rate of regression, the doubling time, the number of tumor cells killed, and the tumor and drug toxicities on the host could be continuously measured. These measurements were used to select active drugs and to rate the relative activity of the different drugs for myeloma.

A solitary plasmacytoma in a dog with progression to a disseminated myeloma.

Solitary plasmacytomas are rare occurrences in dogs, consequently their potential for malignancy is undetermined. A solitary plasmacytoma was removed from the perianal region of a dog. The dog was clinically normal at that time, but was killed one year later as a result of hind limb stiffness and uremia. At the postmortem examination a disseminated myeloma was found, involving the vertebral column, liver, spleen, bone marrow and visceral lymph nodes.

Extramedullary plasmacytomas presenting as mediastinal masses: clinicopathologic study of two cases preceding the onset of multiple myeloma.

We present two cases of extramedullary plasmacytoma presenting as a mediastinal mass and preceding the onset of full-blown multiple myeloma. The patients are a 62-year-old woman who presented with progressive dyspnea and left-sided chest pain and a 59-year-old asymptomatic man. In both patients, radiographic studies revealed a posterior and anterior mediastinal mass, respectively. Surgical resection of the tumor was performed in the two cases. The tumors were characterized by a well-circumscribed proliferation of plasma cells surrounded by residual lymph nodal tissue. Immunohistochemical studies on paraffin sections demonstrated lambda light chain restriction. Follow-up in our patients revealed that both of them developed multiple myeloma after 6 months and 2 years, respectively. One patient received treatment with melphalan and prednisone and is currently alive and well without evidence of disease, 2 years after diagnosis. The second patient died 4 years after resection of his tumor with evidence of disease in lumbar spine, skull, and lungs. Extramedullary plasmacytoma presenting as a mediastinal mass may precede the onset of full-blown multiple myeloma; therefore, institution of early systemic therapy in these patients may be of value in preventing further progression of the disease.
