Bromide intoxication of horses, goats, and cattle.

During the summer and fall of 1973, a few horses, goats, and cattle in the Napa Valley of California became intoxicated by bromide via the ingestion of volunteer oat hay that had been cut from a field treated with methyl bromide, a soil fumigant. The bromide content of the hay ranged from 6,00 to 8,400 ppm. Signs of intoxication were lethargy, weakness, and ataxia. Animals experimentally fed the contaminated hay developed signs of intoxication between the 7th and 9th days.

For COPD a combination of ipratropium bromide and albuterol sulfate is more effective than albuterol base.

BACKGROUND: A combination metered-dose inhaler aerosol containing ipratropium bromide and albuterol sulfate has been reported to be more effective than either of its components in patients with chronic obstructive pulmonary disease. The dose of albuterol sulfate is equal in moles per liter to the dose of albuterol base used in the commercially available metered-dose inhalers. OBJECTIVE: To compare the safety and efficacy of the combination of ipratropium bromide and albuterol sulfate with a commonly prescribed albuterol metered-dose inhaler containing albuterol base alone. METHODS: Investigators at different sites performed a double-blind, 29-day trial involving 357 patients with chronic obstructive pulmonary disease. Efficacy measurements were taken at 15, 30, and 60 minutes after treatment with study medication and then hourly up to 6 hours on days 1 and 29 of the trial. The primary end point was improvement in forced expiratory volume in 1 second. Clinical status was followed up and safety monitoring was also performed. RESULTS: The combination produced a significantly greater peak and mean improvement in forced expiratory volume in 1 second over albuterol base alone on both test days. Similar changes were seen with forced vital capacity. Evaluations of clinical status were better for patients receiving combination therapy, and some improvements were statistically significant. The overall incidence of adverse effects was similar in the 2 treatment groups. CONCLUSION: We conclude that a combination of ipratropium bromide and albuterol sulfate is more effective at improving pulmonary function than albuterol base alone, with no potentiation of adverse effects.

Randomized trial of the addition of ipratropium bromide to albuterol and corticosteroid therapy in children hospitalized because of an acute asthma exacerbation.

OBJECTIVE: To determine whether the addition of inhaled ipratropium bromide to inhaled albuterol and systemic corticosteroid therapy was more efficacious than inhaled albuterol and systemic corticosteroids alone in the inpatient treatment of acute asthma exacerbations in children. DESIGN: Double-blind, randomized, placebo-controlled trial. SETTING: Pediatric inpatient unit of a tertiary urban hospital. PARTICIPANTS: Eighty children (aged 1-18 years) hospitalized because of an acute asthma exacerbation. INTERVENTION: Children were randomized to receive either nebulized ipratropium bromide, 250 microg, or nebulized isotonic sodium chloride solution, 1 mL. All children received albuterol and systemic corticosteroids. MAIN OUTCOME MEASURES: The primary outcome variable was a validated clinical asthma score, measured at baseline and every 6 hours for 36 hours. Secondary outcome measures included the forced expiratory volume in 1 second, the oxygen saturation, the number of doses of inhaled study drug, the time to an inhaled drug-dosing interval of 4 hours, and the length of the hospital stay. RESULTS: There were no differences between groups on baseline characteristics. The intention-to-treat analysis, using repeated-measures analysis of variance, showed no significant (P =.07) difference between the groups in the clinical asthma score over time. There were also no significant differences between groups on secondary outcomes. CONCLUSION: The addition of nebulized ipratropium bromide to nebulized beta(2)-agonist and corticosteroid therapy in the treatment of children hospitalized because of asthma (following intensive emergency department treatment) confers no extra benefit.

Demyelination and remyelination of the caudal cerebellar peduncle of adult rats following stereotaxic injections of lysolecithin, ethidium bromide, and complement/anti-galactocerebroside: a comparative study.

Experimentally induced demyelination due to the direct injection of gliotoxic agents has provided powerful models for studying the biology of remyelination. For the most part, these models have involved injection into white matter tracts of the spinal cord. However, the spinal cord has a number of limitations, such as the size of lesions that it is possible to make and its unsuitability for long-term direct cannulation for the delivery of putative remyelination-enhancing agents. In this study, we describe the natural history of three new models of demyelination/remyelination based on the stereotaxic injection of three gliotoxins: lysolecithin, ethidium bromide, and a combination of anti-galactocerebroside antibody and complement (GalC-ab/comp) into the caudal cerebellar peduncle of adult rats. All three agents produced large areas of demyelination with minimal axonal damage, which undergo extensive remyelination. Ethidium bromide- and GalC-ab/comp-induced lesions remyelinated more slowly than those induced by lysolecithin. The contribution to the remyelination of the lesion by Schwann cells reflects the degree of astrocyte damage incurred within the demyelinated area and is greatest for ethidium bromide-induced demyelination. These new models not only provide further insights into the mechanisms of CNS remyelination but also provide a valuable new resource for addressing a series of key issues relevant to current efforts to promote CNS remyelination either by the enhancement of intrinsic processes or by the transplantation of myelinogenic cells.

The measurements of propantheline ion in biological fluids after administering propantheline bromide to man.

The measurement of propantheline ion has been accomplished in urine and plasma following administration of propantheline bromide to man. Trideuteropropantheline bromide is added to the biological fluid to act as a carrier and internal standard to quantify the propantheline ion using multiple ion monitoring. Methane was used as reactant gas but following the discovery of exchange of the trideuteromethyl group when using methane, ammonia was used for later analyses. The determination of propantheline ion in urine and plasma after administration of propantheline bromide to man is described.

Organic chemistry on cold molecular films: kinetic stabilization of SN1 and SN2 intermediates in the reactions of ethanol and 2-methylpropan-2-ol with hydrogen bromide.

We prepared thin molecular films of ethanol and 2-methylpropan-2-ol on Ru(001) substrates at temperature of 100-150 K and examined their reactivity toward HBr. The reaction intermediates and products formed at the surfaces were unambiguously identified by the techniques of Cs(+) reactive ion scattering (RIS) and low-energy sputtering. The reaction on the ethanol surface produced protonated ethanol, which is stabilized on the surface and does not proceed to further reactions. On the 2-methylpropan-2-ol surface, protonated alcohol [(CH(3))(3)COH(2) (+)] and carbocation [(CH(3))(3)C(+)] were formed with the respective yield of 20 and 78 %. Alkyl bromides, which are the final products of the corresponding reactions in liquid solvents, have extremely small yields on these surfaces (< 0.3 % for ethyl bromide and 2 % for tert-butyl bromide). The results indicate that the reactions on frozen films are characterized by kinetic control, stabilization of ionic intermediates (protonated alcohols and tert-butyl cation), and effective blocking of the charge recombination steps in S(N)1 and S(N)2 paths. The implication of these findings for the molecular evolution process in interstellar medium is also discussed.

Cell cycle analysis by combining the 5-bromodeoxyuridine/33258 Hoechst technique with DNA-specific ethidium bromide staining.

Cells were grown in 5-bromodeoxyuridine(BrdUrd)-containing medium, harvested and stained with a combination of 33258 Hoechst and ethidium bromide for analysis in a two-parametrical flow cytometer. Both fluorescences were excited by UV laser light, and ethidium bromide was additionally excited by energy transfer from the Hoechst dye. The separation of the two fluorescences proved to be excellent, the projected histograms showing a quality comparable to the quality obtained by single dye staining. The ethidium bromide fluorescence gives information as to where the cell is located within the cycle of DNA replication, while the BrdUrd-quenched Hoechst fluorescence gives information as to where in the cycle the cell was located at the beginning of BrdUrd incorporation. In this way information is obtained concerning the distance a cell traveled through the cycle during the BrdUrd incubation time. This method may become a powerful tool in many investigations dealing with cell cycle perturbations in culture.

Single UV excitation of Hoechst 33342 and ethidium bromide for simultaneous cell cycle analysis and viability determinations on in vitro cultures of murine B lymphocytes.

Assessment of DNA content by flow cytometry has largely depended on staining techniques which do not permit exclusion of dead cells from the data set. During studies of B cell activation in vitro, the large number of nonviable cells greatly affects the cell cycle distribution and thus the accurate evaluation of proliferation flow cytometry. This report describes the development of two dual staining techniques which use Hoechst 33342 and ethidium bromide excited by a single UV source to eliminate dead cells from the DNA histogram of the viable cells in murine B cell cultures. Hoechst 33342 and 0.62 micrograms/ml of ethidium bromide permit the evaluation of cell cycle distributions on the viable cells with a ratio gate. The combination of Hoechst 33342 and 6.2 micrograms/ml ethidium bromide results in the resolution of the two populations due to fluorescence energy transfer with a single PMT. Using this technique we demonstrated the simultaneous determination of DNA and RNA content on viable cells using only two PMTs. Both these techniques can be performed on either a laser or an arc lamp flow cytometer where CVs of less than 7% and as low as 3.2% are normally achieved. Determination of the S phase using these techniques produces a high correlation with DNA synthesis determined by radiolabeled precursor determination. These techniques permit the use of flow cytometry to determine proliferation during B cell activation.

Asthma: comparative bronchodilator effects of ipratropium bromide and isoproterenol.

The effect of ipratropium bromide administered at two dosage levels, 40 and 80 mug, isoproterenol, 150 mug, and placebo using a metered dose inhaler was evaluated in ten adult patients with asthma in a double-blind, crossover study. The new atropine-like drug proved to be as effective a bronchodilator as isoproterenol in this study, although it had a later peak effect. Ipratropium bromide had a longer course of action than isoproterenol (4 hours compared to 1-2 hours) and was free of significant side effects. The larger dose of the new drug produced a slightly greater and longer-acting effect than the smaller dose. Ipratropium bromide seems to have had bronchodilator effects on both large and small airways.

Stimulation of 2-deoxy-d-glucose transport in control and virus-transformed cells by ethidium bromide.

Recently we demonstrated that ethidium bromide altered the plasma and subcellular membrane glycoproteins in control and virus transformed cells. It is reported here that ethidium bromide also stimulated the membrane associated process of sugar transport. The KM of the virus transformed cells and the ethidium bromide treated cells is the same as that of the control cells while the maximum velocity as compared to the control cells is significantly increased. The transport of 2-deoxyl-D-glucose was inhibited by glucose, cytochalasin B and neuraminidase but was unaffected by variations in cell density or pH of the incubation medium.

Commercial synthesis of alpha-D-cellobiosyl bromide heptaacetate.

A detailed commercial process for the synthesis and purification of the activated disaccharide, alpha-D-cellobiosyl bromide heptaacetate (1) was developed. Reaction of alpha-D-cellobiose octaacetate (CBO) with HBr in glacial acetic acid or in glacial acetic acid/methylene chloride combination affords alpha-D-cellobiosyl bromide heptaacetate in high yield and excellent quality. Process variables such as reaction solvent, reaction time, reaction temperature, HBr stoichiometry, isolation methods and product purification options were optimized for large-scale synthesis. alpha-D-Cellobiosyl bromide heptaacetate was successfully prepared in a commercial manufacturing plant.

Influence of steroids and stress on toxicity and disposition of tetraethylammonium bromide.

In female rats, pretreatment with dexamethasone acetate or triamcinolone reduced the toxicity and plasma concentrations of tetraethylammonium bromide while increasing its level in urine. Pretreatment with corticosterone acetate or pregnenolone-16alpha-carbonitrile shared none of these effects. Although starvation or restraint neither diminished the tetraethylammonium bromide concentrations in plasma nor accelerated its urinary excretion, its toxicity was diminished by the stress induced with spinal cord lesions, heat, cold, hydrocortisone, or reserpine as well as starvation or restraint. The protection offered against the toxicant by stress and by the potent glucocorticoids seemed to be mediated, at least partly, via different mechanisms. Stress-induced resistance to tetraethylammonium bromide could not be attributed to elevated plasma corticosterone levels, whereas glucocorticoid-induced resistance could be partially ascribed to increased urinary excretion of the toxicant.

Efficacy of salbutamol and ipratropium bromide in decreasing bronchial hyperreactivity in children with cystic fibrosis.

A proportion of patients with cystic fibrosis (CF) suffer from increased airway hyperreactivity but their response to bronchodilators is variable. Adrenergic agents may produce an increase, no change or a decrease in forced expiratory volume in 1 second (FEV1). We hypothesized that the variable response might be related to poor aerosol distribution caused by the presence of secretions. Therefore, in 11 children with CF and airway hyperreactivity the influence of pretreatment with either 0.9% saline, salbutamol, or ipratropium bromide on the methacholine challenge test was evaluated in a double-blind, randomized, cross-over study. FEV1 (mean +/- S.E.) did not change following pretreatment with saline, salbutamol, or ipratropium (1.64 +/- 0.22, 1.63 +/- 0.16 and 1.67 +/- 0.19, respectively). All patients demonstrated airway hyperreactivity with a PC20 below 8 mg/mL (geometric mean, 0.41 mg/mL) after saline pretreatment. Salbutamol inhalation significantly increased the PC20 to 1.24 mg/mL (P less than 0.01), but ipratropium bromide was found to be even more effective than salbutamol (PC20 = 7.37 mg/mL) (P less than 0.0001). We conclude that the variable response to bronchodilator is not secondary to impaired aerosol distribution since ipratropium bromide effectively blocked the response to methacholine. The improvement in PC20 without a change in baseline FEV1 following salbutamol suggests that the adrenergic agent altered the contractile mechanism of smooth muscle.

Physical, chemical and environmental properties of selected chemical alternatives for the pre-plant use of methyl bromide as soil fumigant.

Production and use of methyl bromide, a soil fumigant, are being restricted because of this chemical's deleterious effects on stratospheric ozone concentrations. Several products, some of which are currently used as soil fumigants, are being considered as possible replacements for methyl bromide, alone and in various combinations. Among these, 1,3-dichloropropene, methyl isothiocyanate generators such as metam-sodium, and chloropicrin are currently registered, while others such as methyl iodide and sodium azide are at different stages of the registration process. This review examines physicochemical properties, environmental fate, and metabolism of the various potential methyl bromide replacement products.

Evaluation of chemical and integrated strategies as alternatives to methyl bromide for the control of root-knot nematodes in Greece.

Current environmental awareness has led to a greater demand for alternative nematode control strategies. Three field experiments were established to compare management tactics on cucumber in commercial greenhouses naturally infested with root-knot nematodes (Meloidogyne spp). Cucumber rootstocks which have shown resistance to soil-borne diseases were tested to reveal any resistance/tolerance to root-knot nematodes, and integration of these rootstocks with nematicides was investigated. Metham-sodium and 1,3-dichloropropene (1,3-D) provided good control of nematode populations when their application was followed by the application of a non-fumigant nematicide such as cadusafos or oxamyl. Neither fumigant could provide season-long control of nematode populations, and a further application of cadusafos was required for satisfactory control. The efficacy of metham-sodium was significantly increased when injected into soil in comparison with its application through the drip irrigation system. The use of rootstocks resistant to soil-borne fungal pathogens used together with chemical means of nematode control provided promising results for their further use in integrated strategies as alternatives to methyl bromide. However, the latter was the superior treatment for the control of root-knot nematodes in soil infested with residues of galled roots. Dazomet, metham-sodium nor the non-fumigant nematicides oxamyl and fenamiphos could reduce nematode population as efficiently as methyl bromide. None of the chemicals tested except methyl bromide could enter galled roots and kill surviving nematodes.

Replacing methyl bromide in annual strawberry production with glucosinolate-containing green manure crops.

The use of biocidal green manure crops is an agronomic technique for amending soil with fresh organic matter containing volatile compounds active in controlling some soil-borne pests and diseases. Two new selections of the Brassicaceae family were cultivated, incorporated before planting strawberries and tested as an alternative to fumigation with methyl bromide. Two biocidal green manure crops (Brassica juncea L sel ISCI20, Eruca sativa Mill cv Nemat) containing glucosinolate-myrosinase systems, a conventional green manure (barley), untreated soil and a fumigated control were evaluated during two seasons. The effect of these soil management systems on subsequent strawberry performance was evaluated by monitoring yield and plant growth parameters. In both years, biocidal plant green manure treatments led to a fruit yield lower than with methyl bromide, but higher than with conventional green manure or untreated soil. These results confirm the good prospects for biocidal green manures, not only as an environmentally friendly alternative to methyl bromide in conventional agriculture, but also in organic agriculture as an alternative to conventional green manure crops.

Investigation of alternatives to methyl bromide for management of Meloidogyne javanica on greenhouse grown tomato.

The root galling index and the densities of eggs in roots and juveniles in soil of the root-knot nematode Meloidogyne javanica (Treub) Chitwood on tomato, and the effect of these on crop yield were assessed in greenhouse experiments applying various treatments at two different sites in Crete, Greece. Tomato crops were grown for four cycles by rotating nematode-resistant (first and third spring crops) with susceptible (second and fourth autumn crops) cultivars and receiving the following treatments: (a) untreated control; (b) methyl bromide application before the first and third crops; (c) application of the fungus Pochonia chlamydosporia (Goddard) Zare, Gams & Evans before planting the first, third and fourth crops with a supplementary application three weeks after the beginning of the fourth crop; (d) application of oxamyl in both sites and fenamiphos in site 1 only at the second and fourth crops; (e) combination of treatments (c) and (d). The fungus density in soil was monitored three weeks after application and at the end of each crop, when roots were lifted. Pochonia chlamydosporia had a variable establishment and did not control the nematode. Its pathogenicity on eggs was not demonstrated, as in all cases galls were big, with all egg masses inside and protected from infection. The methyl bromide treatment significantly reduced root galling and egg production compared to other treatments in all crop cycles and the yield of the fourth crop was significantly greater. Nematicides reduced nematode densities compared with untreated controls and the fungus treatment, but they were less effective than methyl bromide and resulted in increased yield in one site only.

Activity measurement and multiplicity detection of human secretory-type ribonuclease based on polycytidylic acid/ethidium bromide fluorescence.

Two analytical methods for human secretory-type ribonuclease, which are based on polycytidylic acid/ethidium bromide fluorescence, have been developed. The first is a method for measurement of secretory-type ribonuclease activity utilizing the radial diffusion of ribonuclease in a thin agarose gel plate containing polycytidylic acid and ethidium bromide. Ribonuclease activity was visualized as a dark circle on a fluorescent background under ultraviolet light after immersing the gel in a cooled acidic solution. The radius of the dark circle was proportional to the amount of the enzyme. This method allows quantitation of human secretory-type ribonuclease down to at least 5 x 10(-5) unit, which corresponds to 60 pg. Secretory-type ribonuclease activity in 18 different human tissues and body fluids was measured. The second method is a zymogram technique for detection of secretory-type ribonuclease after isoelectric focusing, which includes placing a dried agarose film containing polycytidylic acid and ethidium bromide on the focused gel. Human secretory-type ribonuclease (less than 3 x 10(-4) unit) was detected with a high band resolution on the same principle as that of the activity assay described above.
