Rhizoma Typhonii Praeparatum has been compared with Rhizoma Typhonii in outward characteristics, microscopic examination of the processed products and by physical and chemical tests. Some specifications to control the quality of the Rhizoma Typhonii praeparatum have been established.
Aluminium phosphate was precipitated from AlCl3-H3PO4 and NaOH, NH3H2O and NaHCO3--Na2CO3 mixture up to different pH values. The precipitates were dried at room temperature, 40 degrees and 105 degrees and their neutralizing properties (rate of neutralization and acid-consuming capacity) against 0.l N HCl were investigated. It was found that these properties depend on the nature of the reagents used for the formation of AlPO4, the pH and also the temperature of drying. The best antacid properties shows the AlPO4 preperation obtained from AlCl3-H3PO4 and NaHCO3-Na2CO3. For comparison, the therapeutic preparate "Aluphos" was also investigated.
Cow milk was experimentally contaminated with a vaccine strain of poliovirus type I. A concentration procedure was utilized to assay for the virus in a milk sample using adsorption on aluminium sulphate at pH 4.5-5.5, aluminium sulphate concentration being 0.15 g. 1(-1), followed by elution with 0.1 M Na2HPO4 pH 9.5 and subsequent detection in cultured VERO cells (kidney cells from the monkey Cercopithecus aethiops). The yield of the virus ranged from 14 to 58% of the inoculated amount, the mean value being 27%.
There has been no report that IgE antibodies can be raised in guinea pigs by immunizations through respiratory tracts. We dropped a 0.1 ml mixture of 50 micrograms mite extract and aluminium hydroxide gel (Alum) into each of both nostrils of 5 guinea pigs. A mite extract alone was dropped into the nostrils of another 5 guinea pigs. The immunization was done every week. IgE antibodies were detected in one of guinea pigs sensitized 5 times with Alum + mite and also in another guinea pig in the same group sensitized 10 times. No IgE antibodies were detected in the animals immunized with mite alone.

A recombinant, truncated HSV type 1 glycoprotein D secreted by Chinese hamster ovary cells (rgD1) was used to compare the ability of several adjuvants to stimulate protective immunity in guinea pigs. Adjuvants tested included CFA, aluminum hydroxide (alum), a lipophilic derivative of muramyl tripeptide (MTP-PE), and a muramyl dipeptide (MDP) covalently conjugated to rgD1. Animals were immunized three times with rgD1 plus the various adjuvants and antibody titers were determined by ELISA. Four weeks after the last immunization, the animals were challenged intravaginally with HSV type 2 and were monitored daily for clinical signs of disease, including frequency and severity of herpetic lesions, incidence of urinary retention, and mortality during the 14-day post-challenge observation period. Animals immunized in the foot-pad with rgD1 formulated with CFA showed the highest antibody titers. Animals immunized in the footpad with rgD1 using MTP-PE in a 4% squalene formulation, alum, or rgD1 conjugated to MDP showed mean antibody titers that were 57, 16, and 13% of the CFA titers, respectively. Immunization with rgD1 plus MTP-PE, alum, or rgD1-MDP conjugate by the i.m. route elicited lower antibody titers than the footpad route of immunization. Results of the viral challenge indicated that clinical symptoms of the groups immunized with rgD1 with CFA or MTP-PE as adjuvant were similar in magnitude and were markedly reduced compared with unimmunized control groups. Animals immunized with rgD1 combined with alum or rgD1-MDP conjugate showed clinical symptoms significantly more severe than the CFA or MTP-PE groups. The protective immunity observed after i.m. immunization of animals with rgD1 and MTP-PE was only slightly lower than animals immunized with the same Ag-adjuvant combination in the footpad. The results indicate that MTP-PE is an effective adjuvant for the recombinant herpes gD vaccine.

The immunogenicity of a malaria peptide presented in various forms was tested in terms of the quality and quantity of the antibody response in rabbits. Peptide conjugated to a protein carrier, diphtheria toxoid (DT), required strong adjuvants (e.g. muramyl dipeptide, MDP and Freund's adjuvant, FCA) to elicit high levels of antibody with high avidity. Alum was a poor adjuvant, producing the lowest titre and avidity of antibody compared with all the other groups. Peptide expressed in vaccinia (and without carrier) produced intermediate levels of antibody but the avidity of the antibodies produced were comparable to that produced by peptide conjugates given with muramyl dipeptide.
Mice were protected against subsequent infection with Schistosoma mansoni by intradermal or s.c. vaccination with killed schistosomula or soluble parasite extracts and bacillus Calmette-Guérin (BCG). Treatment with i.p. immunization was somewhat less effective, whereas i.m. vaccination failed to elicit protective immunity. The level of resistance induced by intradermal immunization was influenced by the strain of BCG used, and isolated BCG cell walls did not reliably substitute for whole BCG organisms as adjuvant. Bordetella pertussis vaccine and saponin were also able to function as adjuvants for protective immunity in this model, whereas other immunopotentiators including Corynebacterium parvum and aluminum hydroxide were ineffective. No correlation between resistance to challenge infection and antibody levels was detected. Animals immunized intradermally using either protective or non-protective adjuvants all showed minimal humoral reactivity against schistosomulum surface Ag but strong IgG response to soluble parasite components including paramyosin, which is the major serologically recognized Ag in mice vaccinated intradermally with schistosome Ag plus BCG and is protective in this model. In contrast, a strong correlation was observed between resistance and Ag-specific cell-mediated reactivity, including IFN production by T lymphocytes in vitro and macrophage activation in vivo. These results further substantiate the hypothesis that protection in this model is based on cell-mediated immune effector mechanisms. Moreover, they may be of general relevance in the design of vaccination protocols using other Ag or against other infectious agents.

Thirty-five ragweed-sensitive patients received immunotherapy with either alum-precipitated aqueous extracts (AP) or modified ragweed tyrosine adsorbate vaccine (Pollinex-R). Symptoms decreased with AP therapy only. Immunologically, there was a blunted specific IgE response and little specific IgG increase associated with both groups. Alum-precipitated aqueous extracts appeared to be more efficacious than Pollinex-R.

In 18 ragweed hay fever patients who received injections of alum-precipitated pyridine (Allpyral) ragweed extracts in maintenance doses of between 6000 and 8000 P.N.U. at intervals of between two and four months over a two-year period, the prolonged interval did not seem to influence either the clinical response or the tendency to reaction, or result in significant change in immunological tolerance as determined by antibody studies, which included fluorescent antibody assays, as well as hemagglutination and RAST technics.
