A fast and stable photochromic switch based on the opening and closing of an oxazine ring.

[reaction: see text] We have designed a molecular switch based on the photoinduced opening and thermal closing of an oxazine ring. Ultraviolet excitation of this molecule induces the cleavage of a [C-O] bond to form a p-nitrophenolate chromophore in less than 10 ns with a quantum yield of ca. 0.1. The photogenerated isomer reverts thermally to the original oxazine within 50 ns. Our photochromic switch survives more than 3000 excitation cycles without decomposing, even in air-saturated solutions.

Synthesis and quantitative structure-activity relationships of N-(3-oxo-3,4-dihydro-2H-benzo[1,4]oxazine-6-carbonyl)guanidines as Na/H exchange inhibitors.

N-(3-Oxo-3,4-dihydro-2H-benzo[1,4]oxazine-6-carbonyl)guanidines 4 were prepared and tested for Na/H exchange inhibitory activities in order to clarify the structure-activity relationship (SAR). Quantitative SAR (QSAR) analysis of 6-carbonylguanidines 4 indicated that the length of the 4-substituent was parabolically related to activity and that the calculated optimum 4-substituents were propyl, ethyl and isopropyl groups. This SAR was similar to the SAR of the 2- and 4-substituents of 7-carbonylguanidine derivatives 3, although the position relative to the essential guanidinocarbonyl group was different. Larger 2-substituents, such as a phenyl group were unfavorable. The most potent derivative in this series was N-(4-isopropyl-2,2-dimethyl-3-oxo-3,4-dihydro- 2H-benzo[1,4]oxazine-6-carbonyl)guanidine 4 g, with an IC50 value of 0.12 microM. The methanesulfonate salt (KB-R9032) of 4g had excellent water-solubility and showed anti-arrhythmia activity against a rat acute myocardial infarction model. KB-R9032 was selected for further investigation as a therapy for ischemia-reperfusion induced injury.

Effect of some biologically interesting substituted tetrahydro-1,4-oxazines on drug metabolising enzymes and on inflammation.

The effect on hepatic drug metabolising enzymes was evaluated for three representative structures and that were selected from a series of substituted oxazine derivatives designed to possess particular pharmacological properties such as analgesic, antioxidant and hypolipidemic activity. In addition, since xenobiotic metabolism, reactive oxygen and nitrogen species, atherosclerosis and inflammation are interrelated and mutually affected, the effects of and on acute inflammation in vivo and lipoxygenase activity in vitro were also investigated. It was found that treatment of rats with caused induction of cytochrome P450, enhancement of the metabolism of aminopyrine in vitro and of zoxazolamine and hexobarbital in vivo. Compound appeared to induce particularly erythromycin N-demethylation, while, a nitric ester, reduced the catalytically active cytochrome P450, although it increased the metabolism of specific cytochrome P450 substrates, i.e. 4-nitrophenol and erythromycin. Compounds and with strong hypolipidemic and antioxidant properties, reduced acute inflammatory response in two inflammation models and inhibited lipoxygenase activity in vitro. These results are helpful in optimising the biological profile as well as the potential applications of substituted oxazines.

A sensitive gas chromatographic mass spectrometric assay for a novel dopamine agonist (+)-trans-3,4,4A,5,6,10B-hexahydro-4-propyl-2H-naphth(1,2-B)(1,4) oxazin-9-ol in human plasma.

(+)-trans-3,4,4A,5,6,10B-Hexahydro-4-propyl-2H-naphth(1,2-B)(1,4) oxazine-9-ol is a novel potent dopamine agonist. A sensitive and specific gas chromatographic/mass spectrometric assay procedure has been developed for the determination of the dopamine agonist at low picogram per millilitre levels in human plasma. The method comprises an extraction of the agonist from human plasma and subsequent derivatization of the phenolic functionality with pentafluoropropionic anhydride. The derivative is quantified by gas chromatographic and mass spectrometric detection using selected ion monitoring. The assay is linear over the concentration range 10-1000 pg ml-1.

Flow cytometry of DNA content using oxazine 750 or related laser dyes with 633 nm excitation.

The laser dyes oxazine 750 (OX750), LD700, and rhodamine 800 (R800) can be used in an instrument employing a low-power helium-neon laser source for flow cytometry of DNA content in ethanol-fixed or detergent-permeabilized cells. Cells in near-isotonic medium are stained with 10-30 microM dye, and fluorescence excited at 633 nm is measured at wavelengths above 665 nm. The dyes do not appear to stain RNA, and the intensity of DNA staining is not changed when 2 microM Hoechst 33342 is added to cells simultaneously with a red-excited dye. The effects on fluorescence of addition of DNA to LD700 or R800 in aqueous solution are strongly influenced by the base composition of the DNA; binding mechanisms remain to be determined.

A simple method for typing clinical isolates of herpesvirus simplex: replication in the presence of 2H-1,3-oxazine-2,6 (3H)-dione (Oxauracil).

A rapid simple method for the typing of wild HSV isolates based on selective inhibition of HSV-2 replication by the pyrimidine analogue oxauracil (2H-1,3-oxazine-2,6(3H)-dione) has been developed. Chi-square analysis of the results of typing of 30 wild strains by serum neutralization tests, immunofluorescence, and oxauracil-inhibition indicates no significant differences in assignment of types to the isolates. The oxauracil-inhibition test obviates confusion of typing resulting from use of immune sera containing cross-reactive antibodies. We conclude oxauracil typing is a simple, rapid, and precise test which can be performed in any laboratory capable of isolating HSV.

Evaluation of the effectiveness of 2H-1,3-oxazine-2,6(3H)-dione (oxauracil) in the treatment of herpes simplex virus (HSV)-induced acute encephalitis in mice.

Treatment of suckling mice with the pyrimidine analogue 2H-1,3-oxazine-2,6(3H)-dione (oxauracil) proved successful in reducing mortality associated with HSV-2-induced encephalitis. Oxauracil did not decrease mortality associated with HSV-1 infection; however, there was an increased survival time and a decrease in the amount of infectious virus recovered from the brains of HSV-1-infected animals.

Synthesis and antimicrobial evaluation of quaternary salts of 4-phenyl-1,2,3,6-tetrahydropyridine and 3,6-dimethyl-6-phenyl-tetrahydro-2H-1,3-oxazine.

Fifteen predominantly alkyl bromide quaternary salts of 1-substituted 4-phenyl-1,2,3,6-tetrahydropyridine and 10 from 3,6-dimethyl-l-phenyltetrahydro-2H-1,3-oxazine were synthesized. None was effective against the parasitic protozoan Eimeria tenella and the helminth trichostrongyle nematode. Nearly all inhibited Gram-positive and Gram-negative bacteria; maximum efficiency was obtained with nonyl through dodecyl bromide salt derivatives. Antifungal effectiveness paralleled these results. The oxazinium salt analogs were inhibitory in an in vitro peridontal microorganism screen. The decyl bromide derivative at 0.05% in drinking water prevented dental plaque and reduced calculus deposition in rats but not in hamsters fed cariogenic diets. A 0.01% concentration of the tetrahydropyridinium analog caused increased plaque in rats compared to nonmedicated control animals.

An oxazine reagent for derivatization of carboxylic acid analytes suitable for liquid chromatographic detection using visible diode laser-induced fluorescence.

This study reports the use of Nile Blue, an oxazine dye, as a derivatization reagent that fluoresces in the far-red spectral region and is suitable for derivatization with carboxylic-acid-containing analytes. Model carboxylic acid analytes such as benzoic acid, acetic acid, phenylacetic acid and hexanoic acid have been reacted as acid chlorides to form Nile Blue derivatives. The synthesis product of the Nile Blue benzoic acid derivative was confirmed using electrospray-mass spectrometry, infrared spectrometry, 1H and 13C nuclear magnetic resonance, reversed phase liquid chromatography (RP-HPLC), normal phase-thin layer chromatography, and spectral characterization. The synthesized Nile Blue derivatives, separated from reaction by-products with RP-HPLC, all demonstrated an approximately 10-fold drop in molar absorptivity and relative quantum yield. In addition, a 40 nm increase in Stokes shift was observed. A portion of the fluorescence was regained through post-column ionization of the Nile Blue benzoic acid derivative at pH 12. A RP-HPLC limit of detection of 88.25 fmol on column has been reported with conventional fluorescence detection-post-column ionization of the Nile Blue benzoic acid derivative. A limit of detection of 1.99 fmol on column (3.98 x 10(-11) M) has been demonstrated for the Nile Blue benzoic acid derivative with the use of a laboratory-constructed visible diode laser fluorescence detector.

Design and synthesis of phenethyl benzo[1,4]oxazine-3-ones as potent inhibitors of PI3Kinasegamma.

The Type 1 PI3Kinases comprise a family of enzymes, which primarily phosphorylate PIP2 to give the second messenger PIP3, a key player in many intracellular signaling processes [Science, 2002, 296, 1655; Trends Pharmacol. Sci.2003, 24, 366]. Of the four type 1 PI3Ks, the gamma-isoform, which is expressed almost exclusively in leukocytes [Curr. Biol., 1997, 7, R470], is of particular interest with respect to its role in inflammatory diseases such as rheumatoid arthritis (RA) and chronic obstructive pulmonary disease (COPD) [Mol. Med. Today, 2000, 6, 347]. Investigation of a series of 4,6-disubstituted-4H-benzo[1,4]oxazin-3-ones has led to the identification of single-digit nanomolar inhibitors of PI3Kgamma, several of which had good cell based activity and were shown to be active in vivo in an aspectic peritonitis model of inflammatory cell migration.

N-Acyl-1,2,3,4a,5,10b-hexahydro-[1]benzopyrano-[3,4-b][1,4]oxazine-9-carbonitriles as bladder-selective potassium channel openers.

Optically active N-acyl-5,5-dimethyl-1,2,3,4a,5,10b-hexahydro-[1]benzopyrano[3,4-b][1,4]oxazine-9-carbonitriles 2-22 were synthesized as rigid analogues of cromakalim. The (4aR, 10bR)-N-benzoyl derivative (-)-11 was identified as a bladder-selective KCO (IC50, bladder = 8.2 microM, C50, portal vein = 34.5 microM). Among the analogues of 11 with substitution on the benzoyl moiety, the 3-methyl analogue (-)-14 showed highly potent and selective activity at portal vein (IC50, bladder = 279 microM, IC50, portal vein = 0.54 microM). The 4-bromo analogue (-)-19 (IC50, bladder = 2.0 microM, IC50, portal vein = 8.1 microM) and the 4-hydroxy analogue (-)-21 (IC50, bladder = 3.8 microM, IC50, portal vein = 75 microM) showed enhanced activity at the bladder, while maintaining unprecedented bladder selectivity in vitro. The N-benzenesulfonyl analogue (-)-22, a bioisoster of (-)-11, showed similar activity at the bladder with enhanced selectivity (IC50, bladder = 11.6 microM, IC50, portal vein = 120 microM).

Mechanism of inactivation of chymotrypsin by 5-butyl-3H-1,3-oxazine-2,6-dione.

5-Butyl-3H-1,3-oxazine-2,6-dione (1) inactivates chymotrypsin. The extent of inactivation is dependent upon the concentration of 1. Upon dilution of the inactivated enzyme, catalytic activity is partially restored. Reactivation is a biphasic process. An initial relatively rapid phase (k = 1.8 X 10(-2) min), whose amplitude is dependent upon the extent of dilution, is observed. Maximally, 60-65% of the catalytic activity can be recovered. The rapid phase is followed by a slow phase (k approximately 1 X 10(-3) min-1). With 1 labeled with 14C at C-2, it was shown that two forms of inactive enzyme are formed, E.1 and E.1'. 14C label is retained in E.1 but is no longer present in E.1'. Presumably, C-2 is lost as CO2. The following reaction sequence is proposed for the inactivation of chymotrypsin: E + 1 in equilibrium E.1 CO2----E.1'----E + 1''. The probable structures of E.1, E . 1', and 1'' are shown in Scheme I in the text.

Synthesis and dopamine agonist properties of (+-)-trans-3,4,4a,10b-tetrahydro-4-propyl-2H,5H-[1]benzopyrano [4,3-b]-1,4-oxazin-9-ol and its enantiomers.

The dopamine agonist profile of (+-)-trans-3,4,4a,10b-tetrahydro-4-propyl-2H,5H-[1]benzopyrano [4,3-b]-1,4-oxazin-9-ol (16a) and its enantiomers (16b-c) was examined. Racemic 16a exhibited moderate affinity for the dopamine (DA) D2 receptor labeled with the DA antagonist ligand [3H]haloperidol and moderate in vivo activity; it attenuated gamma-butyrolactone-stimulated DA synthesis, decreased DA neuronal firing of substantia nigra DA neurons, and inhibited exploratory locomotor activity in rats, a profile consistent with a DA autoreceptor agonist mechanism of action. The (+)-enantiomer 16b possessed greater DA receptor affinity with the agonist ligand [3H]-N-propylnorapomorphine than with the antagonist ligand. In rats it potently inhibited DA synthesis and neuronal firing and also inhibited exploratory locomotion. The (-)-enantiomer, on the other hand, did not have significant activity in any of these tests. This profile indicates that like many other rigid DA agonists, the dopaminergic activity resides in one enantiomer, in this case the (+)-enantiomer 16b. On the basis of single-crystal X-ray analysis of a key intermediate, the absolute configuration of 16b was found to be 4aR, 10bR.

3-Aryl- and 3-hydroxy-3-aryloctahydropyrido[2,1-c][1,4]oxazines. Synthesis, stereochemistry, and central nervous system pharmacological actions.

A series of substituted 3-aryl- and hydroxy-3-aryloctahydropyrido[2,1-c][1,4]oxazines has been synthesized for purposes of investigating potentially useful CNS pharmacological actions of this novel heterocyclic system. The preferred conformation of the bicyclic system of the parent compounds, 1 and 2, has been shown to be trans with equatorial orientation of the 3-phenyl substituent in each compound. The various substituted aryl analogues of this series are depressants of motor activity in mice, and certain analogues exhibit significant anticonvulsant and appetite suppressant activity in test animals.
