High concordance of karyotype analysis and RT-PCR for CBF beta/MYH11 in unselected patients with acute myeloid leukemia. A single center study.

Identification of the inversion 16 in patients with acute myeloid leukemia (AML) is of great practical value since these patients have a relatively favorable prognosis, especially when treated with high-dose cytarabine. We compared the results of cytogenetic analysis and reverse transcriptase-polymerase chain reaction (RT-PCR) for core binding factor (CBF) beta/myosin heavy chain (MYH11) in 241 unselected cases of AML. In contrast with other studies, we found a high concordance between these 2 methods. Eighteen of 241 patients showed a cytogenetic anomaly of the chromosome 16. We detected the fusion transcript by RT-PCR in all 18 cases and in 2 additional patients with AML without any cytogenetic anomaly of chromosome 16. One patient had a normal diploid karyotype, and the second patient showed a trisomy 22 in karyotype analysis, which often is associated with inv(16). Only 8 of 20 CBF beta/MYH11-positive patients had M4Eo morphologic features. The much higher discrepancy between cytogenetic analysis and RT-PCR in other studies, especially in AMLs other than M4Eo, possibly indicates the necessity for PCR screening regardless of the French-American-British classification.

Karyotypic analysis of adult pluripotent stem cells.

Three categories of precursor cells have been identified in postnatal mammals: tissue-committed progenitor cells, germ layer lineage-committed stem cells and lineage-uncommitted pluripotent stem cells. Progenitor cells are the immediate precursors of differentiated tissues. Germ layer lineage stem cells can be induced to form multiple cell types belonging to their respective ectodermal, mesodermal, and endodermal embryological lineages. Pluripotent stem cells will form somatic cell types from all three primary germ layer lineages. Progenitor cells demonstrate a finite life span before replicative senescence and cell death occur. Both germ layer lineage stem cells and pluripotent stem cells are telomerase positive and display extensive capabilities for self-renewal. Stem cells which undergo such extensive replication have the potential for undergoing mutations that may subsequently alter cellular functions. Gross mutations in the genome may be visualized as chromosomal aneuploidy and/or chromosomes that appear aberrant. This study was designed to determine whether any gross genomic mutations occurred within the adult pluripotent stem cells. Karyotypic analysis was performed using pluripotent stem cells purified from adult male rats using established procedures. Giemsa Banding was used in conjunction with light microscopy to visualize metaphase chromosome spreads. To date over 800 metaphase spreads have been analyzed. We found that the metaphase spreads averaged 42 chromosomes and concluded that these pluripotent stem cells isolated from adult rats have a normal karyotype.

Generation and multiplication of plantlets from callus derived from Haplopappus gracilus (Nutt.) Gray and their karyotype analysis.

Unopened flower heads of Haplopappus gracilis (2n = 4) provided primary explants for callus production and subsequent induction of organized growth. Callus was initiated from small (3-5 mm in length) floral buds with benzylaminopurine (BAP) (44.4 micromoles; 10 mg/l) and naphthalene acetic acid (NAA) (0.54 micromole; 0.1 mg/l). Lowering the BAP level to 4.44 micromoles (1 mg/l) but maintaining the NAA level, gave rise to organized but highly compressed shoot growing points from an otherwise undifferentiated callus mass. Shoots selected from such cultures were maintainable and could be proliferated by growing 1-1.5-cm stem tip cuttings on Murashige and Skoog basal medium (solidified with agar) containing 0.444 micromole (0.1 mg/l) BAP and 0.054 micromole (0.01 mg/l) NAA. The stem tip multiplication rates obtainable by these means permit reliable strategies for shoot multiplication or production of rooted plantlets. Prolonged subculture and maintenance of shoots on growth regulator-free medium leads to in vitro flowering and greatly reduces rooting capacity. Karyotype analysis of chromosomes from root tip cells at metaphase and chromosome measurements show that karyologically uniform plantlets (based on chromosome number and morphology) can be obtained.

Transmission modes of Pneumocystis carinii among rats observed by karyotype analysis.

To observe the transmission patterns of karyotype of Pneumocystis carinii (Pc) by rat colonies, three strains of rats, Sprague-Dawley(SD), Wistar(W) and Fisher(F) from various animal vendors, were suppressed of their immunity by injection of methyl prednisolone. They were kept for 5 to 13 weeks in 3 different animal rooms, A, B, and C. The purified organisms were prepared in low melting point agarose gel by embedded lysis method for pulsed field gel electrophoresis. Field inversion gel electrophoresis showed 2 patterns of the karyotype of Pc. The rooms A and C contained SD rats from the source P, and also the room A was used for F and W rats. However, Pc from all of the SD and F rats in the room A showed same karyotypes, the pattern I. The SD rats from different vendors, M and S, were reared in the room B, and shared the same Pc karyotypes, the pattern II. The rats of W strain were from the vendor M, and immune-suppressed in the animal room A. Five weeks after the experiment, the Pc showed the karyotype pattern II but the pattern became mixed with the type I after 7 to 8 weeks. The findings revealed that the animals born and reared in the same animal quarter harbored Pc with same karyotypes. If the animals were kept under immune-suppression in the same room with heavily infected hosts, they could be infected by Pc from their neighbors. The present experimental findings suggest that Pc is transmitted among rats through the air.

Multicolor spectral karyotype analysis of a transplantable human ileal carcinoid.

In this report we present the results of a combined cytogenetic and multicolor spectral karyotype (SKY) analysis of a transplantable human ileal carcinoid (GOT1). By using SKY it was possible to identify the origin and organization of all clonal marker chromosomes and to identify cryptic translocations not detectable by conventional chromosome banding. The stemline karyotype of low passage GOT1 cells was interpreted as 43,XX, der(1)del(1)(?), inv(2)(p25q13), del(3)(p21), del(5)(q13q31), del(6)(q13), -9, -13, -15, del(16) (q22). Analysis of the GOT1 cells after about 2.5 years of propagation in nude mice allowed us to follow the in vivo progression of this tumor. Relatively few additional rearrangements had occurred during this period, indicating that the GOT1 cells are genetically stable. Most of the abnormalities detected result in loss of whole or parts of chromosomes, suggesting that loss of multiple chromosomal regions, presumably containing tumor suppressor genes, might be important genetic events in ileal carcinoids.

Karyotype analysis and marker chromosomes of renal cell carcinoma.

Karyotyping was performed in 46 human renal cell carcinomas of various histological grades. Controls included chromosome analysis of normal renal parenchymal cells from the same patients. Various numerical chromosome aberrations were found as well as marker chromosomes. They are specific of the individual tumor but no single marker was identified occurring in all tumors tested. Only trisomy 3 was found in different tumors (31%), suggesting a more general character of this aberration. Histological dedifferentiation and large tumor size correlate with focusing of the karyotype towards distinct chromosomal modes indicative of distinct tumor cell subclones within these tumors. Their rapid growth, reflecting biological aggressiveness, is most probably responsible for the patients' poor prognosis.

Prenatally detected fetal myelomeningocele: is karyotype analysis warranted?

PURPOSE: To determine the prevalence of karyotype abnormalities in fetuses with prenatally detected spina bifida and evaluate the ability of prenatal sonography to enable prediction of chromosomal abnormalities. MATERIALS AND METHODS: Sonograms from 63 fetuses with prenatally detected spina bifida were reviewed, and associated sonographic abnormalities were recorded. Sonographic findings were correlated with autopsy or clinical findings when possible. RESULTS: Associated sonographic abnormalities were present in 15 (24%) of fetuses with spina bifida. Among 52 fetuses with known karyotypes, the prevalence of chromosome abnormalities was 17%. Karyotype abnormalities included trisomy 18, trisomy 13, triploidy, and translocation. Twenty-two percent of chromosomally abnormal fetuses had sonographically isolated spina bifida. CONCLUSION: Prenatal sonography can help predict most karyotypically abnormal fetuses with spina bifida, but approximately 20% will be missed with this technique alone in the second trimester. The authors believe cytogenetic analysis is justified in the setting of prenatally detected spina bifida.

Chromosome polymorphism in Astyanax fasciatus (Teleostei, Characidae). 1. Karyotype analysis, Ag-NORs and mapping of the 18S and 5S ribosomal genes in sympatric karyotypes and their possible hybrid forms.

Astyanax fasciatus may be characterized as a chromosomally diversified 'species' presenting distinct cytotypes, each with its specific variants. The sympatric and syntopic occurrence of different cytotypes reinforces the hypothesis in which A. fasciatus may represent a group of species currently placed under a single common designation. Specimens from three collection points spread along the Mogi-Guaçu River in southeast Brazil were examined in the present work: (1) near its headwaters (Ouro Fino--MG), (2) in the middle region of the river (Cachoeira de Emas, Pirassunun ga--SP) and (3) close to its confluence with the Pardo River (Barrinha--SP). The 2n = 48 chromosomes cytotype was found in all sampling points, while cytotype 2n = 46 was only encountered in Barrinha and Cachoeira de Emas. In the latter locality, cytotype 2n = 46 predominated; nevertheless, other karyotype forms with 2n = 45 and 47 chromosomes also occurred, besides a structural variant of cytotype 2n = 46. One specimen with 2n = 47 chromosomes was also found in Ouro Fino. The Ag-NOR analysis, as well as the location of the 18S and 5S ribosomal genes, were conserved in all cytotypes. The data indicate that the variant karyotypes are a consequence of interbreeding between the standard cytotypes (2n = 46 and 48) and/or its descendants. This suggests a karyotype plasticity for this species, where at least a few variant karyotypes would not have deleterious effects on their bearers.

Karyotypic analysis of the human monoblastic cell line U937.

Karyotypes of three sublines of the human cell line U937 showed considerable variation, but all contained four consistent marker chromosomes (i.e., 3q-, 11q-, 16p+, and 17p- chromosomes). The 11q- chromosome appeared to be derived from either an interstitial deletion in bands 11q21-23 or from a translocation with an unidentified chromosome. The presence of this chromosome was of particular interest because rearrangements of chromosome #11 at band 11q23 are often associated with malignancies of the monocytic lineage. The possible significance of these findings is discussed.

Profiling cytogenetic diversity with entropy-based karyotypic analysis.

Regardless the high degree of variation observed in solid tumor karyotypes, the use of diversity as a measurable phenomenon remains unexplored. Here we present a systematic cytogenetic analysis using Shannon's entropy as a measure for karyotypic diversity. Karyotypes from 14 epithelial tumor types (n = 1232) have the aneuploidy status scored, resulting in highly asymmetrical sample distribution, for which we determine the index of diversity (Shannon H') of structural and numerical chromosomal aberrations. Since karyotypic diversity is tissue-specific, this approach may give rise to new insights into the processes that may account for aneuploidy progression and solid tumor outcomes.

Cytogenetic studies of Hynobiidae (Urodela) XVI. Comparative C-banded karyotype analysis of Pseudohynobius flavomaculatus (Fei et Ye), Ranodon shihi (Liu) and Batrachuperus pinchonii (David).

Initial analysis of Pseudohynobius flavomaculatus chromosomes determined the chromosome number of this species to be 2n = 52. A re-examination of Ranodon shihi chromosomes detected 2n = 66 chromosomes, in contrast with a previous finding of 2n = 64. The C-banding patterns of these two species and that of Batrachuperus pinchonii were compared with each other. Regions of homoeology in the C-banding pattern among these three species represented 33.51-48.30% of the total length of their chromosomes. We also detected two types of chromosome rearrangement in hynobiid species based on the results of the present and previous cytogenetic studies.

Immature teratoma of the ovary grade 3, with karyotype analysis.

The relationship between dermoid cysts and immature teratomas has been investigated by study of gross and microscopic data. Karyotypes with banding analysis, already numerous for dermoid cysts, have recently been obtained for immature teratomas. This grade 3, Stage III immature ovarian teratoma was of probable premeiotic origin, as evidenced by chromosome heteromorphisms identical to those of the host cells. The relationship of this pattern to that of other immature teratomas and dermoid cysts is discussed.

Karyotype analysis of a human mammary sarcoma explant in vitro.

Karyotype of a fibroblastic SH-1 cell line derived from a human mammary sarcoma had 86 (80-92) chromosomes including 6 (4-7) markers, and a various number of double minutes. There were generally even numbers (e.g., 2, 4, etc.) of chromosomes in 17 intact and 3 common marker chromosomes. The t(3;14) and del(14), which had 2 each per s cell genome, were probably formed by the balanced translocation between nos. 3 and 14. Three other markers were single and contained a partial or entire no. 1. The presence of these latter markers was often accompanied by the decrease in the number of the intact no. 1, indicating that no. 1 was the only chromosome actively engaged in rearrangements during this stage of karyotypic diversification. Of the no. 2 and 7q that were over-represented on the per cell level, the 7q had a similar observation from many other solid tumors found in this and other laboratories. In contrast, none of the under-represented chromosomes had the concordant changes consistent to other tumors, although the frequent involvement of no. 9 in the chromosome rearrangements was seen in some human heteroploidies.

Karyotype analysis in Brachiaria (Poaceae) species.

This is the first karyotype characterization of Brachiaria species. Twelve accessions belonging to five species were analysed. The basic chromosome number was x = 9 and 7, the same reported for the tribe Paniceae. Variations in the chromosome number were observed in B. decumbens (2n = 18; 36) and B. humidicola (2n = 36; 42; 54). Chromosome numbers of 2n = 18 in B. ruziziensis and 2n = 36 in B. brizantha and B. jubata were recorded. Inter- and intraspecific karyotype differentiation of the accessions analysed was facilitated by variations in karyotypic symmetry. The karyotypes were generally considered symmetrical, with a tendency to asymmetry in the direction of the polyploids. It is suggested that addition, deletions and mainly polyploidy have been the most direct causes involved in the chromosome evolution of this genus.

DNA index and karyotype analysis in myelodysplasia.

DNA index (DI) determined by flow cytometry and karyotype determined by conventional methods were obtained on bone marrow samples from 43 haematologically normal subjects and 54 patients with myelodysplastic syndrome (MDS). Twenty one patients had a clonal karyotype abnormality but an additional five had a DI outside the normal range, showing evidence of aneuploidy that was not available from chromsome preparations. When patients were grouped into those with excess chromosomal material, those with diploid karyotypes, and those with a loss of chromosomal material, there was a significant difference among the mean DIs of each group, normal subjects being different from all patient groups. In these patients DI measurements were of value when carried out together with conventional chromsomal analysis in gaining the maximum amount of genetic information when a satisfactory karyotype might not be available or where failure of an abnormal cell population to proliferate might give an incomplete cytogenetic picture. The contribution of non-clonal chromsome loss to the DI is probably significant but has not been quantitated.

Chromosomal banding and karyotype analysis in an Ehrlich mouse ascites tumor cell line.

The karyotype and the banding pattern of chromosomes through different techniques following Giemsa staining have been worked out in an Ehrlich mouse ascites tumour cell line. The stemline cells show a mode at 74-76. A metacentric chromosome is present in almost all the cells. Some cells show a long telocentric chromosome with a possible secondary constriction. C-bands are restricted at the telomeric regions of the chromosomes excepting in the metacentric one. From the banding pattern it seems that the nucleolar organizers are located at the telomeric region. No band is associated with the second constriction of the long telocentric chromosome. It is suggested that the long metacentric chromosome has arisen out of a Robertsonian translocation involving end to end centromeric fusion of two non-homologue chromosomes.

[Karyotype analysis of Carpesium cernuum].

The paper reported the chromosome number, karyotype and chromosome volume of Carpesium cernuum. The normal diploid was 2n = 40, karyotype formula based on Levan's publication was k(2n) = 40 = 24 m + 12 sm + 2 st + 2T. According to the method of S. R. Kuo, the chromosome relative length was 2n = 40 = 8L + 6M2 + 16M1 + 10S, which belong to "2B" type according to the Stebbins' karyotype classification. The AS.K% was 62.35% from Arano's method. The total volume of chromosome groups was 13.76 microns 3.
