Neurochemical identification of A13 dopaminergic neuronal projections from the medial zona incerta to the horizontal limb of the diagonal band of Broca and the central nucleus of the amygdala.

Studies utilizing fluorescent histochemical techniques first revealed that A13 dopaminergic (DA) perikarya located in medial zona incerta (MZI) project to various regions within the hypothalamus; accordingly, these DA neurons were designated the 'incertohypothalamic' DA neuronal system. More recently, it has been shown that the anterograde neuronal tract tracer Phaseolus vulgaris leucoagglutinin, after injection into MZI, is identified in nerve terminals outside of the hypothalamus; for example, in horizontal limb of the diagonal band of Broca (HDB) and central nucleus of the amygdala (cAMY). The purpose of the present study was to determine, using neurochemical techniques, if A13 DA neurons project to the HDB and cAMY. Concentrations of dopamine and one of its metabolites 3,4-dihydroxyphenylacetic acid (DOPAC) were determined in HDB and cAMY following: (1) electrical stimulation of MZI, (2) electrolytic lesion or knife ablation of MZI, and (3) administration of gamma-hydroxybutyric acid (GHBA) into MZI. For comparison, similar measurements were made in nucleus accumbens (N. Acc.), a terminal region of A10 DA neurons in the ventral tegmental area (VTA). Electrical stimulation of MZI increased DOPAC concentrations in HDB and cAMY, whereas electrolytic or ablative lesions of MZI decreased dopamine concentrations in both of these regions. By contrast, neither stimulation nor lesion of MZI had any effect on DOPAC or dopamine concentrations in N. Acc. Intracerebral injection of GHBA into MZI increased dopamine concentrations in MZI and HDB, but not in cAMY or N. Acc. Intracerebral administration of GHBA into VTA increased dopamine concentrations in HDB and N. Acc., but not in MZI or cAMY.(ABSTRACT TRUNCATED AT 250 WORDS)

The effects of AMPA-induced lesions of the medial septum and vertical limb nucleus of the diagonal band of Broca on spatial delayed non-matching to sample and spatial learning in the water maze.

These experiments investigated in the rat the impact on spatial delayed non-matching to sample and on acquisition of the Morris water maze of (i) AMPA-induced lesions of the medial septal nucleus, which produced a marked reduction of hippocampal choline acetyltransferase activity and acetylcholine levels (measured using in vivo dialysis) together with lesser reductions in cholinergic markers in the cingulate cortex and (ii) similar AMPA-induced lesions of the vertical limb nucleus of the diagonal band of Broca (vDB), which produced more marked reductions in cholinergic markers in the cingulate cortex than in the hippocampus. Medial septal lesions produced a delay-dependent deficit in spatial working memory, while lesions of the vDB resulted in a delay-independent performance deficit. In addition, rats with vDB lesions adopted biased response strategies during the imposition of long delays. Neither lesion significantly affected the acquisition of a spatial reference memory task, the Morris water maze. The results are discussed in terms of cholinergic- and GABAergic-dependent functions of the hippocampal formation and cingulate cortex in spatial short-term and reference memory.

Prolactin- and testosterone-induced inhibition of LH secretion after orchidectomy: role of catecholaminergic neurones terminating in the diagonal band of Broca, medial preoptic nucleus and median eminence.

Central catecholaminergic neurones projecting to specific hypothalamic structures are involved in stimulating and inhibiting the activity of the GnRH-containing neurosecretory neurones. Both testosterone and elevated circulating prolactin (PRL) levels inhibit postcastration LH release. Three groups of adult male rats were orchidectomized and adrenalectomized, received corticosterone replacement and were: (i) administered purified ovine PRL (oPRL; 2400 microgram/s.c. injection) or (ii) its diluent, polyvinylpyrrolidone (PVP), every 12 h, or (iii) received physiological testosterone replacement for 2 days. At 0, 2 and 6 days postcastration, norepinephrine (NE), epinephrine (E) and dopamine (DA) turnover were estimated by the alpha-methyl-p-tyrosine method in three micro-dissected hypothalamic structures: the diagonal band of Broca at the level of the organum vasculosum of the lamina terminalis (DBB(ovlt)), the medial preoptic nucleus (MPN) and the median eminence (ME). In control (PVP-treated) rats, serum LH concentrations increased eightfold at 2 and 6 days postcastration and this rise was prevented by testosterone. oPRL treatment transiently suppressed LH secretion at 2 but not 6 days postcastration. Castration significantly decreased basal rat PRL (rPRL) levels at 2 and 6 days and testosterone administration partially prevented this effect. NE turnover in the ME and E turnover in the MPN increased markedly at 2 and 6 days postcastration, and testosterone replacement for 2 days prevented these increases. Thus, noradrenergic neurones innervating the ME and adrenergic neurones innvervating the MPN may drive postcastration LH secretion by providing stimulatory afferent input to the GnRH neurones. It was striking to observe that oPRL blocked the increases in both ME NE and MPN E turnover at 2 but not 6 days postcastration. Hence, oPRL may transiently suppress LH release by an inhibitory action on these NE and E neurones. DA turnover in the DBB(ovlt) was significantly decreased by 6 days postcastration. Testosterone-treated (2 days postcastration) and oPRL-treated (2 and 6 days postcastration) rats exhibited turnover values indistinguishable from day 0 controls. Hence, the A14 dopaminergic neurones, which synapse on GnRH neurones in the rostral preoptic area and may exert an inhibitory effect on them, are positively regulated by PRL and perhaps by testosterone as well. Autoregulatory feedback suppression of endogenous rPRL secretion by oPRL was observed both 2 and 6 days postcastration. In contrast to the A14 dopaminergic neurones, turnover in the A12 tuberoinfundibular dopaminergic (TIDA) neurones innervating the ME increased significantly by 6 days postcastration in control rats while oPRL administration further increased ME DA turnover at both 2 and 6 days. Hence, autofeedback regulation of rPRL secretion persists through at least 6 days of oPRL exposure temporally associated with markedly increased turnover in the TIDA neurones. In summary, our results support the hypothesis that the inhibitory effect of PRL on postcastration LH release is mediated by suppression of the activity of NE neurones innervating the ME and E neurones terminating in the MPN which, with time, become refractory to continued PRL exposure.

Castration decreases single cell levels of mRNA encoding glutamic acid decarboxylase in the diagonal band of broca and the sexually dimorphic nucleus of the preoptic area.

Using quantitative in situ hybridization histochemistry (ISHH), we determined the effect of castration on single cell levels of glutamic acid decarboxylase (GAD) mRNA in discrete hypothalamic regions of the male rat brain associated with the control of gonadotropin secretion. A 48-base oligodeoxynucleotide probe was used to detect with equal affinity the two isoforms of GAD message, GAD65 and GAD67. GAD message also was quantitated in a number of selected areas of the brain to contrast GAD gene expression amongst several populations of GABAergic neurons. Comparison of 11 brain regions demonstrated a 9.3-fold range in the quantity of single cell GAD mRNA with levels being highest in the amygdala and the diagonal band of Broca, moderate in the piriform cortex, caudate nucleus, substantia innominata, globus pallidus, cingulate cortex and medial septal nucleus, and lowest in the lateral septal nucleus and the medial preoptic nucleus (MPN). Castration markedly reduced single cell GAD mRNA levels in the DBB and the MPN, two discrete hypothalamic structures known to contain dendritic fields, cell bodies, and axons of GnRH neurons projecting to the median eminence. A striking finding was a dense core of steroid-sensitive GABAergic neurons within the MPN comprising the sexually dimorphic nucleus of the preoptic area (SDN-POA). Similar to the MPN as a whole, the amount of GAD mRNA expressed by cells in the SDN-POA of sham operated control rats was greater than in castrated animals. GAD mRNA levels were inversely related to serum LH titers, suggesting a role for these neurons in the mechanism controlling gonadal steroid negative feedback on LH secretion. This report provides the basis for future work to determine if GAD65, GAD67 or whether both isoforms are affected by gonadal steroid input.

Diffusible factors from rat arcuate nucleus and Broca's diagonal band nucleus increase size and neurite outgrowth, respectively, of cultured melanin-concentrating hormone containing neurons.

Using a co-culture model, we showed that diffusible factors from arcuate nucleus (AN) specifically increased the number and the size of hypothalamic neurons producing melanin-concentrating hormone (MCH). In this model neurite outgrowth and contacts between MCH neurons and dopaminergic neurons were also prominently increased, as compared to control lateral areas of the posterior hypothalamus (LH) primary cultures. These effects were mediated in part by AN glia but also by neurons of both fetal and adult AN. AN glia produced diffusible factor(s) mainly responsible for an important MCH neurite outgrowth and expressed inhibiting factors, preventing the adhesion of LH cells on AN glial cells. Furthermore, we report here a nerve growth factor-like effect from Broca's diagonal band on MCH hypothalamic neurons.

GABAergic neuronal activity and mRNA levels for both forms of glutamic acid decarboxylase (GAD65 and GAD67) are reduced in the diagonal band of Broca during the afternoon of proestrus.

There is considerable evidence that GABAergic neurons play an important role in the regulation of gonadotropin-releasing hormone (GnRH) secretion, and that these neurons may mediate the feedback actions of gonadal steroids on GnRH neurons. The aim of the present study was to investigate whether endogenous changes in ovarian steroid secretion during the estrous cycle influenced GABAergic neuronal activity in the preoptic region of the hypothalamus, and in other steroid-sensitive brain regions. Intact, adult female rats were sacrificed at various times during the days of metestrus or proestrus. GABAergic neuronal activity was estimated by measuring the rate of accumulation of GABA in microdissected brain regions after pharmacological inhibition of GABA degradation. Concentrations of mRNA for both forms of glutamic acid decarboxylase (GAD65 and GAD67) were quantified in microdissected brain regions by a microlysate ribonuclease protection assay. In the diagonal band of Broca at the level of the organum vasculosum of the lamina terminalis (DBB(ovlt)), GABAergic neuronal activity was significantly reduced during the afternoon of proestrus compared with the morning of either proestrus or metestrus. In the lateral septal nucleus, GABAergic neuronal activity was significantly increased in the afternoon of proestrus compared with the morning. There were no significant effects of time of day or day of estrous cycle in the medial preoptic nucleus, median eminence, ventromedial nucleus, suprachiasmatic nucleus, medial septal nucleus, hippocampus (CA1 region), or cingulate cortex. In the DBB(ovlt), mRNA levels for both GAD65 and GAD67 were significantly reduced in the afternoon of proestrus compared with the afternoon of metestrus. By contrast, there was no change in GAD65 and GAD67 mRNA levels in the cingulate cortex at any of the times examined. These results demonstrate that GABAergic neuronal activity, and mRNA levels for both GAD65 and GAD67, are reduced in the DBB(ovlt) during the afternoon of proestrus. These results support the hypothesis that decreased GABAergic neuronal activity in this region plays a major permissive role in the generation and maintenance of the estrogen-induced LH surge.

Prostaglandin E receptor EP3 subtype is involved in thermal hyperalgesia through its actions in the preoptic hypothalamus and the diagonal band of Broca in rats.

The effects of microinjection of prostaglandin E2 (PGE2) (0.5 fg-500 pg/0.2 microl) into the medial part of the preoptic area (MPO) on nociception were studied using a hot-plate test in rats. The intraMPO microinjection of PGE2 only at non-pyrogenic doses (5-50 fg) reduced the paw-withdrawal latency, suggesting hyperalgesia. Maximal reduction was obtained 15 min after the injection of PGE2 at 50 fg. Subsequently, to determine which types of prostanoid receptors are involved in the hyperalgesic effect of PGE2 in the MPO, we administered PGE2 receptor agonists, i.e., 17-phenyl-omega-trinor PGE2 (an EP1 receptor agonist), butaprost (an EP2 receptor agonist) and M&B28767 (an EP3 receptor agonist) into the MPO and observed the nociceptive behavior. The intraMPO injection of M&B28767 (0.005 fg-50 pg), like that of PGE2, exhibited a U-shaped dose response characteristic, i.e., a significant decrease of the paw-withdrawal latency only at 0.05-5 fg with the maximal response at 0.5 fg. However, neither the administration of EP1 (0.5 fg-50 ng) nor EP2 (0.5 fg-500 pg) agonists had any effect on nociception. The microinjection of M&B28767 at 0.5 fg into the other parts of preoptic hypothalamus (the lateral part of the preoptic area and the median preoptic nucleus) and the diagonal band of Broca (DBB) produced hyperalgesia similar to the intraMPO-induced hyperalgesia in terms of magnitude and time course. Microinjection of M&B28767 (0.5 fg) into either the paraventricular nucleus, the ventromedial hypothalamus, the lateral hypothalamic area or the septal nucleus had no effect on nociception. These findings suggest that PGE2 at non-pyrogenic doses in the brain induces hyperalgesia, at least in part, through its actions on EP3 receptors in the preoptic hypothalamus and the DBB in rats.

Parvalbumin in the medial septum/diagonal band complex of the African green monkey Cercopithecus aethiops.

The distribution of the calcium-binding protein Parvalbumin (PV), a marker for a subpopulation of GABAergic neurons, was studied in the septal complex of the African green monkey, Cercopithecus aethiops. PV-immunoreactive neurons were found in relatively small numbers in the medial septum and in large numbers in the nucleus of the diagonal band (BROCA). They were extremely rare in the dorsal and intermediate parts of the lateral septum. PV-immunoreactive neurons in the medial septum and the nucleus of the diagonal band were mostly fusiform when located close to the midline and round to polygonal in more lateral locations. Some perikarya of immunopositive as well as immunonegative cells were covered with immunostained boutons whereas others were not. Our present descriptive data suggest that there are no principal differences in PV-containing neurons in the septal region between non-human primates and rodents which have been analyzed in great detail in previous studies.

Evaluation of the true precocious puberty rats induced by neonatal administration of Danazol: therapeutic effects of nourishing "Yin"-Removing "Fire" Chinese herb mixture.

BACKGROUND: Nourishing "Yin"-Removing "Fire" Chinese Herb Mixture, a traditional herb-based formulation, has been successfully used for the management of idiopathic true precocious puberty (IPP) for more than thirty years. Precocious puberty rat model by neonatal administration of Danazol was used to investigate the effects of the herb mixture on the advanced sexual development of the rats, and the expression of hypothalamic gonadotropin-releasing hormone (GnRH), which is the important regulator for the hypothalamus-pituitary-gonadal axis, particularly at puberty. METHODS: Female Sprague-Dawley rats were divided into five groups: intact normal (N), IPP model (M), vehicle with no IPP (V), IPP model exposed to herb mixture (HM) and IPP model exposed to saline (S). Rats at 5 days of age were given a single subcutaneous injection of 300 microgram of Danazol dissolved in 25 microliter vehicle of propylene glycol-ethanol (1:1, v/v), to establish the precocious puberty model. From the day 15, rats in HM and S groups were continuously fed with either Nourishing "Yin"-Removing "Fire" Chinese Herb Mixture 2 ml or saline 2 ml, until 3 consecutive regular estrous cycles were established. The day of vaginal opening and the day of setup regular estrous cycle of the rats were observed. Blood concentration of estrogen was determined by radioimmunoassay. Immunohistochemistry and RT-PCR analysis were used to explore the expression of GnRH. RESULTS: The day of vaginal opening and first estrous showed significant advancement in M compared with N and V (p < 0.05, respectively). The blood estrogen level increased significantly in M compared with those in other groups (about 28 days of age, at the time of vaginal opening in M rats) (p < 0.05, respectively). GnRH cells in rostral medial septum (MS), Broca diagonal band nucleus (DBB) and the medial preoptic area (MPOA), were calculated. The number in M was less than those in N and V (p < 0.05, respectively). The number was significantly higher in HM than that in M (p < 0.05). The GnRH mRNA expression increased significantly in M compared with that in N and V (p < 0.05). CONCLUSION: The true precocious puberty model by neonatal administration of Danazol in female rats showed augmented expression of hypothalamic GnRH; the Nourishing "Yin"-Removing "Fire" Chinese Herb Mixture down-regulated the increased GnRH expression, and significantly delayed the sexual development of the precocious puberty rat.

[Neural connections in the septum of lizards studied by the method of horseradish peroxidase axonal transport].

Septal connections in lizards (Ophisaurus apodus) were studied by means of the massive iontophoretic HRP injection into the septum. Reciprocal connections between the septum and both dorsal and mediodorsal cortex, projections of the nucleus of Broca diagonal band and anterior thalamic nuclei (mainly dorsolateral) to the septum were revealed. Reciprocal connections of the septum with the preoptic area, periventricular hypothalamic structures containing dopaminergic neurons, as well as with the nuclei of the mammillary complex were shown to be the most massive. HRP injection into the mammillary complex revealed mainly anterolateral location of neurons projecting to the mammillary nuclei and dorsolateral terminal field of mammillo-septal projections. Some peculiarities and common features in hodological organization of the septum in reptiles and mammals are considered.

[Telencephalic sources of the afferents of the main olfactory bulb in the frog Rana temporaria].

Following horseradish peroxidase iontophoretic application into the main olfactory bulb (MOB) retrograde neuronal labeling was examined in the telencephalon in the frog. Labeled neurons, the sources of the MOB afferents are found in the mitral cell layer of the contralateral MOB, pallial and some subpallial areas. Very heavy labeling is observed in the pars ventralis of the lateral pallium, and to a lesser extent in the medial pallium, pars dorsalis of the lateral pallium and in the dorsal pallium. In subpallium labeled neurons are found in the eminentia postolfactoria, the rostral part of the medial septal nucleus, and in the nucleus of the ventro-medial telencephalic wall, which is probably homologous to the nucleus of the diagonal band (Broca) of mammals. No labelled neurons were found in the caudal portion of the MOB granular layer, usually referred to as the anterior olfactory nucleus. The arrangement of the MOB centrifugal innervation in amphibians is discussed in comparison with that in mammals.

[Study of the connections of the hippocampal (mediodorsal) cortex of Ophisaurus apodus by means of horseradish peroxidase axonal transport].

Origin of afferent projections of the hippocampal (mediodorsal cortex in lizards (Ophisaurus apodus) cells was revealed by studying retrograde transport of horseradish peroxidase. Most labelled neurons were located in n. dorsolateralis anterior thalami, corpus mamillaris, n. raphe superior, ipsilateral dorsal cortex and contralateral hippocampal cortex. Less neurons projecting to the hippocampal cortex were observed in the ventromedial part of basal telencephalon (tuberculum olfactorium, n. accumbens, nucleus of diagonal Broca band), hypothalamic preoptic area, mesencephalic ventrotegmental area. Labelled terminals of hippocampal efferents were found in the septum, thalamus, hypothalamus (mainly ipsilaterally) and bilaterally in the hippocampal and dorsal cortex. Both afferent and efferent connections of the hippocampal cortex in lizards are similar to those in mammals.

Estimation of the total number of cholinergic neurons containing estrogen receptor-alpha in the rat basal forebrain.

This study was undertaken to estimate the total number of cholinergic cells and the percentage of cholinergic cells that contain estrogen receptor-alpha (ER alpha) in the rat basal forebrain. Double immunostaining for choline acetyltransferase (ChAT) and ER alpha was carried out on 50-microm-thick free-floating sections. Because routine mounting method causes considerable flattening of the sections, we embedded immunostained sections in Durcupan, an epoxy resin known to cause virtually no shrinkage. When this procedure was used the section thickness was well preserved, individual cells could be clearly identified, and subcellular localization of ER alpha immunoreactivity was easy to verify. Cell counting in these sections revealed that the rat basal forebrain contains 26,390 +/- 1097 (mean +/- SEM) cholinergic neurons. This comprises 9674 +/- 504 in the medial septum-vertical diagonal band of Broca, 9403 +/- 484 in the horizontal diagonal band of Broca, and 7312 +/- 281 in the nucleus basalis. In these nuclei, 60%, 46%, and 14% of the cholinergic neurons were co-localized with ER alpha, respectively. We believe that our results are an improvement on existing data because of the better distinction of individual neurons that the Durcupan embedding method brings.

Localization of amino acids, neuropeptides and cholinergic markers in neurons of the septum-diagonal band complex projecting to the retrosplenial granular cortex of the rat.

Retrograde labelling was combined with immunohistochemistry to localize neurons containing choline acetyltransferase, gamma-aminobutyric acid (GABA), glutamate, leu-enkephalin, neurotensin, and substance P-like immunoreactivity in the projection pathways from the septum-diagonal band complex to the retrosplenial granular cortex in the rat. Injections of horseradish peroxidase conjugated to subunit B of cholera toxin (CT-HRP) into the retrosplenial granular cortex resulted in retrogradely labelled neurons in the ipsilateral nuclei of the diagonal band of Broca, especially in the horizonatal nucleus of the diagonal band, and small numbers of CT-HRP-labelled neurons were also found in the medial septal nucleus. In the horizontal and vertical nuclei of the diagonal band of Broca, 90-95% of CT-HRP-labelled neurons (35-45 per section) were immunoreactive for choline acetyltransferase and small numbers of retrogradely labelled neurons (2 to 4-5 per section) were also immunoreactive for GABA, glutamate, neurotensin, leu-enkephalin, or substance P. In the medial septal nucleus approximately 75-80% of the retrogradely labelled neurons (8-10 per section) were immunoreactive for choline acetyltransferase and up to 25% of the CT-HRP labelled neurons (1-3 per section) in the medial septal nucleus also displayed GABA-, glutamate-, neurotensin-, leu-enkephalin-, or substance P-immunoreactivity. These results suggest that the complexity of the neurotransmitter(s)/neuromodulator(s) of septum-diagonal band complex projections to the retrosplenial granular cortex should be taken into account when considering the mechanisms of cortical activation.

AF64A affects septal choline acetyltransferase but not parvalbumin immunoreactive cells.

Rats received bilateral intracerebroventricular (ICV) infusions of either AF64A (1.5 nmol/ventricle; n = 9) or vehicle (3.0 microliters/ventricle; n = 7). Four weeks later, the animals were anesthetized and their brains processed to visualize and quantify choline acetyltransferase (ChAT) immunoreactive (IR) and parvalbumin-IR GABAergic neurons in the septal complex by immunocytochemistry (PAP method). AF64A significantly reduced the number of ChAT-IR perikarya in the medial septum (28%), ventral limb of the diagonal band of Broca (30%), and horizontal limb of the diagonal band of Broca (20%), but did not affect the number of parvalbumin-containing GABAergic neurons in any of the septal subdivisions. These results provide further evidence that AF64A is a selective cholinotoxin.
