Sol-gel encapsulated enzyme arrays for high-throughput screening of biocatalytic activity.

We developed versatile low-cost arrays of sol-gel-encapsulated enzymes (referred to as solzymes) suitable for repeated assays of bioactivity or enzyme inhibition. Sol-gel microstructures containing active enzymes were stabilized on glass at moderate pH and room temperature without harsh calcination. A multi-well bilayer of polydimethylsiloxane was used to support the solzyme array and contain the reaction medium. Each of the 147 microwells has a working volume of 5 muL and contains 50 mug of immobilized enzyme. The solzyme arrays maintained high activity through repeated applications and exhibited superior thermostability compared to soluble enzymes. Among the enzymes used were lipases, glucose oxidase, and horseradish peroxidase. Twenty different lipases and proteases were also used to prepare a hydrolase array, for which bromthymol blue served as a generic indicator of activity. The relative activities of the encapsulated hydrolases correlated closely with those of the soluble hydrolases, illustrating that sol-gel encapsulation preserved the hierarchy of enzyme activity. The development of solzyme arrays paves the way to higher throughput screening of diverse proteins and enzymes, including those that are available only in trace amounts.

Short prereduced anaerobically sterilized (PRAS) biochemical scheme for identification of clinical isolates of bile-resistant Bacteroides species.

The rapid identification of isolates of bile-resistant Bacteroides species has clinical and therapeutic relevance because of differences in their patterns of susceptibility and virulence. Five hundred twenty-one strains of bile-resistant Bacteroides species that were previously identified by conventional biochemical methods were reexamined to determine the minimum essential parameters necessary for correct identification. Rapid tests for bile resistance, indole production, and catalase were combined with a novel scheme for biochemical determination of saccharolytic activity on arabinose, trehalose, rhamnose, and/or xylan that included the postincubation addition of bromthymol blue for visual pH determination. Organisms were inoculated into prereduced anaerobically sterilized (PRAS) carbohydrates directly from plates, and identification was complete within 24 h of obtaining a pure culture. Ninety-three percent of bile-resistant Bacteroides species from routine clinical specimens were identified correctly by this scheme; a small number of other indole-positive strains, B. splanchnicus, B. eggerthii, and B. stercoris, were misidentified as B. uniformis.

Solid-phase optoelectronic sensors for biochemical analysis.

Simple solid-phase optoelectronic sensors for penicillin, urea, and glucose are described. Triphenylmethane dyes such as bromcresol green and bromthymol blue were derivatized with glutathione and co-immobilized with appropriate enzymes to a transparent membrane sandwiched between a red-light-emitting diode and a silicon photodiode with integral amplifier. In the presence of the corresponding substrates, catalytic action in the enzyme-dye membrane perturbs the local pH and causes characteristic color changes in the membrane which are monitored as a rise or fall in the output voltage of the detector system. With enzymes such as penicillinase, urease, and glucose oxidase, the response of the optoelectronic sensors is extremely reproducible over the concentration range 0-10 mM penicillin G, urea, or D-glucose, respectively. This report describes the construction and operation of these simple, inexpensive, and reagentless optoelectronic sensors.

Intracellular pH measurements in gastric mucosa.

Intracellular pH was measured in bullfrog gastric mucosa using a pH-indicator dye, bromthymol blue (BTB), with a spectrophotometric technique. Studies showed that BTB is taken up by the gastric mucosa and bound to intracellular components. The binding of BTB was shown to cause a shift in the pKa of the dye from the solution value of 6.95 to a value of 8.0. During the nonsecreting state, intracellular pH was estimated to be 7.4 (metiamide inhibition) or 7.1 (SCN inhibition). During active secretion of acid, intracellular pH increased with increasing secretion rates, reaching values in excess of pH 8. Using preparations from which the surface epithelial cells had been removed, it was shown that at least a portion of the alkaline response to stimulation occurs in the oxyntic or tubular cells. The results are interpreted in view of existing models for the chemical reaction involved in gastric acid secretion.

Cryptococcus neoformans serotype groups encountered in Oklahoma.

Eighty clinical and 28 soil isolates of C. neoformans obtained in Oklahoma were separated into A-D and B-C serotype groups utilizing creatinine-dextrose agar with bromthymol blue. Previously, serotype B-C clinical isolates have been frequent only in patients from Southern California where as many as 50% of the isolates are of this type. In contrast, in patients from the rest of the United States the B-C frequency has been only 6%. Of the 80 C. neoformans isolates from Oklahoma patients, 12 (15%) were serotype B-C. One-half of these 12 Oklahoma patients with serotype B-C isolates had no history of any travel to California, and were long-time residents of Oklahoma. All 28 soil isolates of C. neformans from Oklahoma in this study were serotype A-D. Since serotype B-C recovery from a soil sample has never been reported, attempts are in progress to isolate serotype B-C from the environments of these patients from Oklahoma.

The biochemical basis for the distinction between the two Cryptococcus neoformans varieties with CGB medium.

The biochemical basis for the reaction to canavanine-glycine-bromthymol blue (CGB) agar by Cryptococcus neoformans var. gattii and C. neoformans var. neoformans was investigated. All of the var. gattii isolates tested were found to utilize glycine as the sole source of carbon and nitrogen and were resistant to L-canavanine. Only 11% of the serotype D isolates of var. neoformans utilized glycine as the sole source of carbon and nitrogen, but these were all sensitive to canavanine. Nineteen percent of the serotype A isolates of var. neoformans were able to assimilate glycine, and 81% of the glycine users were resistant to canavanine. However, these canavanine-resistant, glycine-assimilating, var. neoformans isolates failed to grow when they were cultured on a medium containing glycine and canavanine. Unlike the var. neoformans isolates, all of the var. gattii isolates tested grew on a medium that contained both of these compounds. Glycine-utilizing isolates exhibited good uptake of the amino acid, and a glycine-cleaving enzyme was discernable in the isolate. The isolates that fail to utilize glycine accumulated the amino acid at a rate which was barely 15% of that seen in the glycine users, and no glycine-cleaving enzyme was apparent within the 48-hr incubation period. When a cell-free extract (which had been derived from a glycine-utilizing isolate), was incubated with 14C-labeled glycine, ammonia, radiolabeled CO2, and serine were produced. The glycine decarboxylase activity of the cell-free extract was found to be enhanced by the addition of dithiothreitol, tetrahydrofolate, pyridoxal phosphate, and nicotinamide adenine dinucleotide (NAD). The ammonia released during glycine cleavage seems to be responsible for the positive reaction on CGB medium.

Immunological control of drug absorption from the gastrointestinal tract: effect of local anaphylaxis on the intestinal absorption of low molecular weight drugs in the rat.

Intestinal absorption of various drugs was examined by means of in situ recirculation technique during local anaphylaxis. The antibody was determined by passive cutaneous anaphylaxis technique in rats immunized once or three times. The optimal condition of local anaphylaxis was determined by the leakage of Evans Blue. The most significant increase in leaks of the dye was observed by the intraluminal challenge with 400 mg of ovalbumin for 10 min in ovalbumin-immunized rats, and this condition was chosen as the optimal condition of local anaphylaxis. Under this condition, intestinal absorption of caffeine, phenylbutazone, and bromthymol blue (BTB) significantly decreased in ovalbumin-immunized rats compared with the control, whereas no significant effect was noted in the intestinal absorption of salicylic acid, quinine, pralidoxime iodide (2-PAM), tetracycline, and phenol red. In normal rats, no significant decrease was obtained in the intestinal absorption of caffeine, phenylbutazone, and BTB. On the other hand, the decreased absorption of BTB was not found in ovalbumin-immunized rats by the intraluminal challenge with bovine gamma-globulin. Furthermore, there was no significant change in the decreased absorption of BTB between rats immunized once and three times. The most effective condition for decreased BTB absorption was observed by the intraluminal challenge with 200 mg of ovalbumin for 10 min in ovalbumin-immunized rats, which almost correlated with the data of Evans Blue leakage. From these observations, it appears that the mucosal immune responses affect the intestinal absorption of low molecular weight drugs.

Colorimetric determination of peptide antibiotics: in-process assay of cyclic octapeptidic antibiotics in fermentation broths.

A spectrophotometric method is presented for monitoring the biosynthesis of a new complex of cyclic octapeptidic antibiotics in fermentation broths. The method is based on the extraction of antibiotic from alkaline broth with butanol. An ion-pair, formed between the octapeptides and bromthymol blue, is extracted into chloroform from a solution buffered to pH 7.5. The absorbance of the colored solution is measured at 420 nm. Results are in good agreement with those obtained by microbiological assay. The method is also applicable to other peptidic antibiotics such as polymyxin B and gramicidin.

Isolation of saprophytic Cryptococcus neoformans from Puerto Rico: distribution and variety.

Until the present decade, no studies had been conducted in Puerto Rico on the saprophytic distribution and variety of Cryptococcus neoformans. Samples (522) of pigeon droppings from 14 western towns were tested for the presence of C. neoformans. The yeast was recovered from 24.7% (129 isolates) of the samples, representing 10 of the 14 towns studied. All environmental isolates were identified as C. neoformans var. neoformans using canavanine-glycine-bromthymol blue (CGB) agar. The yeast was isolated from 79.4% of the samples in one town, Isabela. The average number of yeast cells isolated from sites within this municipality was 5.1 x 10(5) per gram of pigeon droppings. This was 2.6 times the average number of yeast cells of C. neoformans isolated from sites in other towns. In addition, the yeast was isolated from four patients with the acquired immune deficiency syndrome (AIDS), each of whom died of cryptococcal meningitis. Each of these poorly encapsulated isolates was identified as C. neoformans var. neoformans using CGB agar. The results of this investigation demonstrate that C. neoformans var. neoformans is prevalent in Puerto Rico.

Rapid diagnosis of premature rupture of membranes using a new kit employing anti-AFP monoclonal antibody.

A kit using mouse monoclonal antibody against human alpha-fetoprotein (anti-AFP kit) has been developed. Cervical secretion (n = 34) and vaginal fluid (n = 15) was obtained from 30 pregnant volunteers. All 31 specimens obtained from women with intact membranes tested negative while all 18 specimens from women with premature rupture of membranes (PROM) tested positive. The newly developed kit allowed rapid and accurate diagnosis. We compared the kit results with those obtained by pH determination (nitrazine, bromthymol blue test) and measurement of AFP concentrations by enzyme-linked immunoassay (EIA).

Isolation, characterization, and esterase and CO2 hydration activities of ubiquitin from bovine erythrocytes.

Ubiquitin was isolated from bovine erythrocytes by a relatively simple procedure involving extraction with chloroform and ethanol, chromatography on DEAE-cellulose, and gel filtration. Amino acid and partial sequence analyses showed it to be identical to previously isolated material. Ubiquitin released p-nitrophenolate from p-nitrophenyl acetate, but did not cleave other esterase substrates that were tested. It had a turnover number of 116 mmol for p-nitrophenyl acetate at pH 7.7 and 30 degrees C, and this activity was relatively stable to heat treatment. Electrophoretic studies indicated that the ubiquitin was sequentially acetylated by p-nitrophenyl acetate, as judged by the appearance of more anodically migrating components. The reactions of ubiquitin with p-nitrophenyl acetate at pH 7.0 were biphasic and consisted of (a) an initial phase, during which the release of p-nitrophenol resulted from monoacetylation of the ubiquitin and from ubiquitin-catalyzed hydrolysis of the ester; and (b) a second phase, during which the release of p-nitrophenol resulted only from the breakdown and reformation of the acetyl-enzyme complex. Ubiquitin also showed CO2 hydration activity and could be localized following gel electrophoresis by the CO2-bromthymol blue staining method. The strong inhibitor of carbonic anhydrase, acetazolamide, also inhibited the CO2 hydration activity and p-nitrophenyl acetate activity of ubiquitin. An antibody against this protein did not precipitate bovine carbonic anhydrase II. The esterase activity of ubiquitin was much higher than those previously reported for the carbonic anhydrases.

Extractive spectrophotometric methods for the determination of nifedipine in pharmaceutical formulations using bromocresol green, bromophenol blue, bromothymol blue and eriochrome black T.

Four simple, sensitive and accurate spectrophotometric methods have been developed for the determination of nifedipine in pharmaceutical formulations. These methods are based on the formation of ion-pair complexes of amino derivative of the nifedipine with bromocresol green (BCG), bromophenol blue (BPB), bromothymol blue (BTB) and eriochrome black T (EBT) in acidic medium. The coloured products are extracted with chloroform and measured spectrophotometrically at 415 nm (BCG, BPB and BTB) and 520 nm (EBT). Beer's law was obeyed in the concentration range of 5.0-32.5, 4.0-37.5, 6.5-33.0 and 4.5-22.5 microg ml(-1) with molar absorptivity of 6.41 x 10(3), 4.85 x 10(3), 5.26 x 10(3) and 7.69 x 10(3) l mol(-1) cm(-1) and relative standard deviation of 0.82%, 0.72%, 0.66% and 0.68% for BCG, BPB, BTB and EBT methods, respectively. These methods have been successfully applied for the assay of drug in pharmaceutical formulations. No interference was observed from common pharmaceutical adjuvants. Statistical comparison of the results with the reference method shows excellent agreement and indicates no significant difference in accuracy and precision.

A simple spectrophotometric method for the determination of beta-blockers in dosage forms.

A simple, extraction-free spectrophotometric method is proposed for the analysis of some beta-blockers, namely atenolol, timolol and nadolol. The method is based on the interaction of the drugs in chloroform with 0.1% chloroformic solutions of acidic sulphophthalein dyes to form stable, yellow-coloured, ion-pair complexes peaking at 415 nm. The dyes used were bromophenol blue (BPB), bromothymol blue (BTB) and bromocresol purple (BCP). Under the optimum conditions, the three drugs could be assayed in the concentration range 1-10 microg ml(-1) with correlation coefficient (n = 5) more than 0.999 in all cases. The stoichiometry of the reaction was found to be 1:1 in all cases and the conditional stability constant (K(F)) of the complexes have been calculated. The free energy changes (DeltaG) were determined for all complexes formed. The interference likely to be introduced from co-formulated drugs was studied and their tolerance limits were determined. The proposed method was then applied to dosage-forms the percentage recoveries ranges from 99.12-100.95, and the results obtained were compared favorably with those given with the official methods.

A new spectrophotometric method for the determination of finasteride in tablets.

A simple, rapid, accurate, precise and sensitive colorimetric method for the determination of finasteride in tablets is described. The proposed methods are based on the formation of ion-pair complexes between the examined drug with bromophenol blue (BPB), bromocresol green (BCG) and bromothymol blue (BTB), which can be measured at the optimum lambda(max). Beer's law is obeyed in the concentration ranges 3.0-15.0, 3.0-15.0 and 5.0-20 microg/mL with BPB, BCG and BTB, respectively. The detection limits of FIN was found to be 1.16 microg/mL for BPB, 1.17 for BCG, 1.76 microg/mL for BTB. All the methods gave similar results and were validated for selectivity, linearity, precision and sensitivity. The proposed methods were directly and easily applied to the pharmaceutical preparation with accuracy, resulting from recovery experiments between 100.11 and 100.33% for BPB, 100.17 and 100.67% for BCG and 100.33 and 100.60% for BTB methods. The low relative standard deviation values indicate good precision and high recovery values indicate accuracy of the proposed methods. The proposed methods have been applied to the determination of drug in commercial tablets. Results obtained from the analysis of commercial preparations with the proposed methods are in good agreement with those obtained with the official HPLC method.

Extractive spectrophotometric methods for determination of diltiazem HCl in pharmaceutical formulations using bromothymol blue, bromophenol blue and bromocresol green.

Three simple and sensitive extractive spectrophotometric methods have been described for the assay of diltiazem hydrochloride either in pure form or in pharmaceutical formulations. The developed methods involve formation of coloured chloroform extractable ion-pair complexes of the drug with bromothymol blue (BTB), bromophenol blue (BPB) and bromocresol green (BCG) in acidic medium. The extracted complexes showed absorbance maxima at 415 nm for all three methods. Beer's law is obeyed in the concentration ranges 2.5-20.0, 2.5-10.0 and 2.5-12.5 microg ml(-1) with BTB, BPB and BCG, respectively. The methods have been applied to the determination of drug in commercial tablets and capsules. Results of analysis were validated statistically and through recovery studies.

Effects of pH indicators on various activities of chromatophroes of Rhodospirillum rubrum.

1. The effects of pH indicators on activities for ATP hydrolysis in the dark and ATP-Pi exchange in the dark were examined with chromatophores from Rhodospirillum rubrum. Of thirty-one pH indicators tested, eleven (metanil yellow, 2, 4-dinitrophenol, ethyl orange, bromocresol green, resazurin, neutral red, bromthymol blue, alpha-naphtholphthalein, o-cresolphthalein, phenolphthalein, and alizarin yellow G) almost completely inhibited the activities for ATP formation and ATP-Pi exchange at concentrations of 1 mM, and were studied in detail. 2. Of the eleven pH indicators, those other than alpha-naptholphthalein, o-cresolphthalein and phenolphthalein, when assayed at appropriate concentrations, inhibited ATP-Pi exchange, but not ATP hydrolysis. In ATP-Pi exchange, these eight pH indicators at the concentrations described above were competitive against Pi, and non-competitive against ATP. The remaining three kinds of pH indicators were non-competitive against either Pi or ATP, when assayed at concentrations of the dyes that inhibited both activities. 3. The amounts of pH indicators bound with chromatophores were measured. No correlation was found between the amounts of the bound dyes and the extents of their inhibition of either ATP formation or ATP-Pi exchange. 4. Ethyl orange (pKa=4.1) and 2, 4-dinitrophenol (pKa=3.9) stimulated ATP hydrolysis to the greatest extent. The latter dye was hardly bound with chromatophores. 5. The stimulatory effects of pH indicators on ATP hydrolysis were hardly affected by extraction of quinones from chromatophores. 6. Most of the pH indicators stimulated both succinate-cytochrome c2 and NADH-cytochrome c2 reductions in the dark. 7. The mechanism of uncoupling of the electron transfer system and the phosphorylation system by pH indicators and the mechanism of the coupling are discussed.

Reversible conversion from Ca(2)+-ATPase activity to Mg(2)+- and Mn(2)+-ATPase activities of coupling factor purified from acetone powder of Rhodospirillum rubrum chromatophores.

It is known that the coupling factor purified from the acetone powder of chromatophores from Rhodospirillum rubrum shows ATPase activity in the presence of Ca(2)+, but not in the presence of Mg(2)+ or Mn(2)+. The present study deals with conditions, under which the Ca(2)+-ATPase activity is reversibly converted into Mg(2)+- and Mn(2)+-ATPase activites with the purified coupling factor. 1. Of the pH indicators tested, 6 kinds coverted the Ca(2)+-ATPase activity into Mg(2)+- and Mn(2)+-ATPase activities in the order, ethyl orange greater than tropaeolin 000 greater than or equal to metanil yellow greater than tropaeolin 00 greater than ethyl red greater than or equal to bromthymol blue. 2. Of the detergents tested, those other than Triton X-100 and Brij 58 caused the conversion described above; dodecylsulfonate was most effective, whereas dodecylpyridinium chloride was moderately effective. 3. 2,4-Dinitrophenol stimulated approximately two-fold the Ca(2)+-ATPase activity, but not the Mg(2)+- or Mn(2)+-ATPase activity at all. However, in the presence of dodecylpyridinium chloride, the pH indicator remarkably stimulated the Mg(2)+- and Mn(2)+-ATPase activities, accompanied with a partial inhibition of the Ca(2)+-ATPase activity. Methyl red and ethyl red showed similar effects. 4. All the nucleoside triphosphates tested can serve as the substrate. ATP was most effective for the Ca(2)+-ATPase activity, whereas dATP was most effective for the Mg(2)+- and Mn(2)+-ATPase activities induced by ethyl orange. 5. In the presence of ethyl orange, the ATPase activity was induced by various divalent cations in the following order of effectiveness, Mg(2)+ greater than Zn(2)+ greater than CO(2)+ greater than Mn(2)+ greater than Ni(2)+. 6. The mechanism of the reversible conversion from the Ca(2)+-ATPase activity to the Mg(2)+- and Mn(2)+-ATPase activities by pH indicators and detergents is discussed.

Evaluation of test sensitivity and test specificity of three field methods for quantification of chloroquine in urine using liquid chromatography as reference method.

We have evaluated three quantitative colorimetric methods [bromthymol blue (BTB), Haskins, and Saker-Salomons (S-S)] for measurement of concentrations of chloroquine (CQ) in urine. A graphical evaluation of test sensitivity and test specificity at different concentrations of CQ in authentic urine samples was used. Liquid chromatography (LC) was used as a reference method. The Haskins method showed the highest test specificity (89%) and test sensitivity (97%), at a discrimination value of 2 mumols/L. At the same sensitivity (97%) as the Haskins method, S-S and BTB methods had test specificities of 49 and 13%, respectively. The correlation coefficients between the LC method and the colorimetric methods were all higher than 0.94 at a urinary concentration range of 2-100 mumols/L. The intra- and interassay variations for the colorimetric methods were all uniformly less than 15% at 2 mumols/L, but the Haskins method showed less variation than the other two methods.
