Antioxidant and hypocholesterolaemic effects of Terminalia arjuna tree-bark powder: a randomised placebo-controlled trial.

OBJECTIVE: To evaluate the antioxidant and hypocholesterolaemic effects of Terminalia arjuna tree bark (a popular cardiotonic substance in Indian pharmacopoeia) and to compare it with a known antioxidant, vitamin E, we performed a randomized controlled trial. METHODS: One hundred and five successive patients with coronary heart disease (CHD) presenting to our centre were recruited and using a Latin-square design divided into 3 groups of 35 each. The groups were matched for age, lifestyle and dietary variables, clinical diagnosis and drug treatment status. None of the patients was on lipid-lowering drugs. Supplemental vitamins were stopped for one month before study began and American Heart Association Step II dietary advice was given to all. At baseline, total cholesterol, triglycerides, HDL and LDL cholesterol and lipid peroxide estimated as thiobarbituric acid reactive substances (TBARS) were determined. Group I received placebo capsules; Group II vitamin E capsules 400 units/day; and Group III received finely pulverized T. arjuna tree bark-powder (500 mg) in capsules daily. Lipids and lipid peroxide levels were determined at 30 days follow-up. RESULTS: Response rate in various groups varied from 86% to 91%. No significant changes in total, HDL, LDL cholesterol and triglycerides levels were seen in Groups I and II (paired t-test p > 0.05). In Group III there was a significant decrease in total cholesterol (-9.7 +/- 12.7%), and LDL cholesterol (-15.8 +/- 25.6%) (paired t-test p < 0.01). Lipid peroxide levels decreased significantly in both the treatment groups (p < 0.01). This decrease was more in vitamin E group (-36.4 +/- 17.7%) as compared to the T. arjuna group (-29.3 +/- 18.9%). CONCLUSIONS: Terminalia arjuna tree bark powder has significant antioxidant action that is comparable to vitamin E. In addition, it also has a significant hypocholesterolaemic effect.

Higher human kallikrein gene 4 (KLK4) expression indicates poor prognosis of ovarian cancer patients.

PURPOSE: Kallikrein gene 4 (KLK4, also known as prostase/KLK-L1), located on chromosome 19q13.4, is one of the newly discovered members of the human KLK-like gene family. This gene is up-regulated by androgens in the LNCaP prostatic carcinoma cell line and by androgens and progestins in the BT-474 breast cancer cell line. On the basis of its apparent association with hormonally regulated tissues, we have undertaken to examine the prognostic value of KLK4 expression in 147 malignant ovarian tissues. EXPERIMENTAL DESIGN: Tumors were pulverized, total RNA was extracted, and cDNA was prepared by reverse transcription. KLK4 was amplified by PCR using gene-specific primers, and its identity was verified by sequencing. Ovarian tissues were then classified as KLK4-positive or -negative, based on ethidium bromide visualization of the PCR product on agarose gels. RESULTS: KLK4 was found to be expressed in 69 (55%) of 147 of ovarian cancer samples. We found a strong positive association between KLK4 expression and tumor grade (P = 0.02) and clinical stage (P < 0.001). Univariate survival analysis revealed that patients with ovarian tumors positive for KLK4 expression had an increased risk for relapse and death (P = 0.003 and 0.001, respectively). Whereas knowledge of KLK4 status did not significantly increase the prognostic power of the multivariate models, additional analyses did determine that KLK4 was an independent unfavorable prognostic factor in patients with grade 1 and 2 tumors. CONCLUSIONS: Our findings indicate that KLK4 expression is associated with more aggressive forms of ovarian cancer.

[Isolation, purification and determination of the constituents of a toxic protein from plant amenone].

A toxic protein constituent named AME from the stems of plant amenone, have been isolated and purified. Through CM-SFF column chromatography and gel filtration on Sephacryl S-200 column with phosphate saline buffer as mobile phase. All of the operations were performed at 4 degrees C. The pulverized plant amenone material was soaked in phosphate saline buffer, homogenized, left standing overnight and then squeezed through coarse cloth by wringing. The supernatant was applied on the S-SFF column. Then, the column was eluted with the phosphate buffer containing 1 mol/L NaCl. The eluate was collected and dialyzed against water and phosphate buffer. The chromatography of the crude toxin dialyzed was carried out on the CM-SFF column with gradient elution of phosphate buffer containing NaCl. The fourth peak was collected and then applied on a gel filtration Sephacryl S-200 column using neutral phosphate buffer as mobile phase. The protein was further separated on two connected Protein 125 columns with mobile phase of 0.2 mol/L phosphate buffer (pH 6.5), the eluate was monitored at 280 nm on photodiode array detector. The protein presents typical the absorption spectrum of protein in ultraviolet region with the strong absorption at 280 nm and the weak absorption at 260 nm. The purity of peak of the protein was judged from the spectrum. The molecular weight of AME measured by two connected protein columns was approximately 35,000 D. The composition of amino acids was determined with OPA post-column derivatization/fluorescence detection.

Certified reference material for analytical quality assurance of minor and trace elements in food and related matrixes based on a typical Japanese diet: interlaboratory study.

A Certified Reference Material (CRM) was prepared at the National Institute for Environmental Studies (NIES), Japan, in collaboration with the National Institute of Radiological Sciences (NIRS), Japan, for the analytical quality assurance of minor and trace elements in food and related matrixes. The starting material for the CRM was all food served in 29 households in Japan over two 3-day periods in 1997-1998, and thus the CRM represented a typical Japanese diet. All foods (meals, snacks, and beverages) were homogenized, freeze-dried, pulverized, blended, dispensed into 1,100 bottles, and sterilized. The within- and between-bottle homogeneity of the prepared CRM was satisfactory for most of the elements. The concentrations of 14 elements (Na, Mg, K, Ca, Mn, Cu, Zn, As, Se, Sr, Cd, Sn, Ba, and U) were certified based on a collaborative analysis involving NIES, NIRS, and 20 other laboratories. Reference values were given for the concentrations of 12 additional elements (P, Cl, Fe, Co, Ni, Br, Rb, Mo, I, Cs, Pb, and Th). The elements certified and those given reference values include minerals, essential trace elements, contaminant elements, and long-lived radionuclides. Thus, this CRM is of practical value in the quality assurance of element analysis of foods and diets in nutritional, environmental, and radiological research.

[Effects of air staging with absorbents on trace metal during coal combustion].

Staged combustion was carried out on laboratory-scale pulverized coal combustion with different absorbents. The experiment indicated staged combustion increased emission of submicron particles, which went against the control of trace elements, especially for those of high volatile elements, such as Cu, Ni. The thermodynamics calculation also indicate the transformation of trace metal was different with different atmosphere, suboxidized and reduced species were more easily formed under reduced condition. In both conditions, absorbents show a certain absorptive ability to trace metal, and different absorbent had different ability. For unstaged combustion, kaolinite was the best for Co, Cr and Ni; dolomite for Be, and CaO for Cu. But for under staged condition, HZ- dolomite was the best for Be, Cr and Ni; Kaolinite for Co and Cu.

Microbial contaminants in the interiors of spacecraft components.

The hazard from microbiological contamination in the interiors of components of planetary-impacting spacecraft has been generally recognized, but techniques for experimentally evaluating the significance of this hazard have been unavailable. A goal in sterilizing early Mars-impacting payloads is the elimination of the possibility that a microbial stowaway will cause the extraterrestrial-life detection techniques falsely to indicate existence of Martian life, particularly if none exists. Another goal is the prevention of changes in Martian ecology. Several promising life-detection techniques were evaluated in an experimental study of methods to detect microbial populations inside spacecraft-type materials after pulverization or being dissolved. The type of material inspected has a greater effect on the sensitivity of the detection method than any other parameter studied. Through culturing abraded particles, a bacillus inoculum of 100 spores per milliliter can be detected in solid propellants. Fifty percent of an inoculum can be recovered from some solids soluble in nontoxic solvents. For many of the solids studied, however, the culturing techniques used were unable to detect inoculums of 10(6) spores per milliliter of solid. The techniques other than culturing did not discriminate between living and dead cells. As with products of the food and pharmaceutical industries, assurance of sterility in the final product never rests in a negative sterility test on a copy of the product. A negative sterility test is certainly most meaningful for a product for which a very sensitive sterility test has been developed. When a sensitive final-product sterility test is not available, assurance of sterility depends more heavily on other information such as the manufacturing history of the product.

[Predicting low NOx combustion property of a coal-fired boiler].

More attention was paid to the low NOx combustion property of the high capacity tangential firing boiler, but the NOx emission and unburned carbon content in fly ash of coal burned boiler were complicated, they were affected by many factors, such as coal character, boiler's load, air distribution, boiler style, burner style, furnace temperature, excess air ratio, pulverized coal fineness and the uniformity of the air and coal distribution, etc. In this paper, the NOx emission property and unburned carbon content in fly ash of a 600 MW utility tangentially firing coal burned boiler was experimentally investigated, and taking advantage of the nonlinear dynamics characteristics and self-learning characteristics of artificial neural network, an artificial neural network model on low NOx combustion property of the high capacity boiler was developed and verified. The results illustrated that such a model can predicate the NOx emission concentration and unburned carbon content under various operating conditions, if combined with the optimization algorithm, the operator can find the best operation condition of the low NOx combustion.

Hair analysis of seven bodybuilders for anabolic steroids, ephedrine, and clenbuterol.

Several bodybuilders, all winners of international competitions, were arrested for trafficking of a number of doping agents including anabolic steroids, ephedrine, beta-adrenergics, human chorionic gonadotropin, antidepressants, and diuretics. In accordance with the recent French law against doping, the judge asked to test seven bodybuilders to identify doping practices. Hair and urine specimens were collected for analysis. After decontamination, a 100 mg hair strand was pulverized in a ball mill, hydrolyzed, extracted, and derivatized to be tested by GC/MS for anabolic steroids, beta-adrenergic compounds, ephedrine, and other doping agents. Urine was analyzed for anabolic steroids and metabolites, beta-adrenergic compounds, ephedrine, and human chorionic gonadotropin, in addition to a broad spectrum screening with GC/MS. The following compounds were detected in urine: ephedrine (29 and 36 ng/mL, n = 2), clenbuterol (0.2 to 0.3 ng/mL, n = 3), norandrosterone (4.7 to 100.7 ng/mL, n = 7), norethiocholanolone (0.9 to 161.8 ng/mL, n = 6), stanozolol (1 to 25.8 ng/mL, n = 4), methenolone (2.5 to 29.7 ng/mL, n = 4), testosterone (3 to 59.6 ng/mL, n = 7), epitestosterone (1 to 20.4 ng/mL, n = 7) and ratio testosterone/epitestosterone >6 for four subjects (18.5 to 59.6). The following drugs were detected in hair: ephedrine (0.67 and 10.70 ng/mg, n = 2), salbutamol (15 to 31 pg/mg, n = 3), clenbuterol (15 to 122 pg/mg, n = 6), nandrolone (1 to 7.5 pg/mg, n = 3), stanozolol (2 to 84 pg/mg, n = 4), methenolone (17 and 34 ng/ml, n = 2), testosterone enanthate (0.6 to 18.8 ng/mg, n = 5), and testosterone cypionate (3.3 to 4.8 ng/mg, n = 2). These results document the doping practice and demonstrate repetitive exposure to anabolic compounds and confirm the value of hair analysis as a complement to urinalysis in the control of doping practice.

Injuries and illnesses among New York City Fire Department rescue workers after responding to the World Trade Center attacks.

Within minutes of the terrorist attacks on September 11, 2001, the Fire Department of New York City (FDNY) operated a continuous rescue/recovery effort at the World Trade Center (WTC) site. Medical officers of FDNY Bureau of Health Services (FDNY-BHS) responded to provide emergency medical services (see box). The collapse of the WTC towers and several adjacent structures resulted in a vast, physically dangerous disaster zone. The height of the WTC towers produced extraordinary forces during their collapse, pulverizing considerable portions of the buildings' structural components and exposing first responders and civilians to substantial amounts of airborne particulate matter. Fires burned continuously under the debris until mid-December 2001. Because of ongoing fire activity and the large numbers of civilians and rescue workers who were killed during the attacks, approximately 11,000 FDNY firefighters and many emergency medical service (EMS) personnel worked on or directly adjacent to the rubble and incurred substantial exposures (Figure). This report describes morbidity and mortality in FDNY rescue workers during the 11-month period after the WTC attacks and documents a substantial increase in respiratory and stress-related illness compared with the time period before the WTC attacks. These findings demonstrate the need to provide acute and long-term medical monitoring, treatment, and counseling to FDNY rescue workers exposed to this disaster and to solve supply, compliance, and supervision problems so that respiratory protection can be rapidly provided at future disasters.

Use of respiratory protection among responders at the World Trade Center site--New York City, September 2001.

The terrorist attacks on the World Trade Center (WTC) on September 11,2001, created an occupational health and safety challenge for New York City (NYC) firefighters and rescue workers responding to the disaster. Immediate respiratory hazards included explosions, fire, falling debris, and dust clouds containing particulate matter comprised of pulverized building materials. Ongoing risks included lingering particulate matter in the air and intermittent combustion products from initial and persistent fires beneath the rubble pile. Because the nature and extent of exposures in disaster situations are complex and difficult to characterize, the use of adequate personal protective equipment (PPE), including respiratory protection, is essential in protecting the health of firefighters and other rescue workers. During the weeks after September 11, the NYC Fire Department's Bureau of Health Services (FDNY-BHS) and CDC's National Institute for Occupational Safety and Health (NIOSH) organized a collaborative study to evaluate occupational hazards and exposures for these workers, including their use of respiratory protection. This report summarizes the results of that study, which indicate that the majority of firefighters did not use adequate respiratory protection during the first week of the rescue/recovery operation.

[Development of prodioxin ointment].

There was a purpose to create a composite ointment of proteolytic and antimicrobial activity, the formulation of which would contain the proteolytic enzyme procelan and the antimicrobial preparation with wide spectrum of action dioxidin. During the development of prodioxin ointment by means of biological experiments the optimal concentration of dioxidin was evaluated; it was 1 percent. The optimal dispersity value of dioxidin particles was estimated by dialysis through a semipermeable membrane. The best release of dioxidine from prodioxin ointment occurred, when it was pulverized to particles with diameter 5-10 mm. The method of assay of dioxidin was modified for the ointment. The physicomechanical properties of the prodioxin ointment: flowability, colloidal stability, thermostability and rheological properties were examined. The investigated ointment is non-newtonian in character, e.g. an elastic and plastic system and may be intended for use in dermatological practice. Medicinal enzymic and antimicrobial effects of the prodioxin ointment were compared with the healing effect of Iruksol ointment. The medicinal effect of the prodioxin ointment exceeded that of Iruksol ointment by one day. Judging by the results of the physicochemical analysis a conclusion can be made that the prodioxin ointment is stable for two years at room temperature.

[A vibrating bed aerosol generator for fibrous and powder dust particles (author's transl)].

A fluidized bed with defined mechanical vibrations has been used for the pulverization of powders of fibrous and spherical particles. By means of this process, it was possible to prepare for practical purposes useful aerosols with constant concentrations. In certain size ranges the concentrations as well as the fiber and particle size distribution were adjustable and could be varied as a function of physical properties of the vibrating fluidized bed.

The weathering of natural and artificial stones in the baroque church of S. Pietro in Valle (Fano-Italy).

The church of San Pietro in Valle in Fano is with good reason considered one of the outstanding examples of baroque art in Central Italy. Currently the church is in a serious degradation status. For this reason, and in order to devise a correct conservative restoration project, a scientific study has been carried out, aimed at characterizing mortars used for the application of marble decorations and the different degradation products occurring in the church (i.e. pulverizations, encrustations, salt efflorescences, and condensation water). In the following, results obtained analyzing the different sample typologies by optical microscopy, X-ray diffraction, electron scanning microscopy with energy-dispersion micro-analytical system, ion chromatography, and head-space solid phase microextraction-gas chromatography-mass spectrometry will be reported.

Changes in embryonic 8-cell nuclei transferred by means of cell fusion to mouse eggs.

Metaphase II and activated mouse oocytes were fused with 8-cell blastomeres, and morphological changes in the transferred nuclei were followed using light and electron microscopy. In metaphase II oocytes, blastomere nuclei underwent premature chromosome condensation (PCC) typical for S-phase nuclei: chromatin pulverization. Then an abortive spindle was formed without evident microtubule organizing centers. Blastomere chromosomes condensed to a lesser degree than meiotic chromosomes and lacked mature functional, trilaminar kinetochores. After parthenogenetic activation of these oocytes, blastomere chromosomes followed, in synchrony with oocyte chromatin, a similar route of changes (anaphase, telophase) and then reformed interphase nuclei of the pronuclear type. Remodeling of 8-cell nucleus thus occurred, but the integrity of the chromatin set was frequently disturbed by formation of micronuclei. If blastomere fusion with oocytes was done close to activation (either before or after parthenogenetic stimulation), the chances of remodeling of the nuclei decreased, because PCC was not regularly induced in all oocytes. In hybrids produced 60 min or later after oocyte activation, blastomere nuclei were maintained in interphase without any structural modifications. Multiple experiments in the mouse have shown that the nuclei from 8-cell stage transferred to enucleated oocytes and egg cells are not capable of substituting for pronuclear functions. Possible reasons for impaired functional reprogramming of 8-cell nucleus in the mouse are discussed in light of our present findings on the morphology of nuclei transferred before and after oocyte activation.
