Using a cell line derived from human lung cancer (AOI), we successfully established human xenografts in KSN nude mice, which showed high incidence of multiple spontaneous metastases. The highest incidence of metastasis in untreated hosts was observed in the spleen followed by the lungs and lymph nodes. The rate of metastasis reached 100% in the mice bearing large sized tumors, when metastasis to any organ or tissue was counted. Experimental radiotherapy caused remarkable redistribution of metastatic foci among different organs. Lung metastasis tended to decrease, while metastasis to the liver and the kidney was increased after radiotherapy. Radiation-induced production of cytokines was speculated to be responsible for such an alteration of metastasis pattern.
Focal lesions of the spleen are due to multiple causes dominated by malignant lymphoma. The authors study the contribution of sonography in the diagnosis and etiology of these lesions based on a retrospective study of 38 cases, over an eight-year period. Hematological malignancies splenic metastases and splenic hydatid disease are the commonest lesions in this series. Sonography provides an etiologic diagnosis in the presence of highly suggestive features (hydatic cyst, hematomas) without requiring other investigations. The needle biopsy is necessary for any hypoechoic lesion, suggestive of a malignant etiology. CT is essential when a hemangioma is suspected.

A case is presented which, 9 years after surgical removal of a scirrhous breast cancer, exhibited morphologically atypical metastasis in the liver with fibrosis and reduction of liver parenchyma, as well as typical features of cirrhosis including portal hypertension and esophageal varices. Problems posed by the differential diagnosis and pathogenesis of this rare type of metastasis (known in the literature as metastatic carcinomatous cirrhosis) are briefly discussed.

DNA topoisomerase I (topo I) was purified 124-fold from the spleen of a patient which was diffusely infiltrated by malignant lymphoma. Extracts prepared from the lymphoma tissue demonstrated elevated topo I activity. The purification uses a new two step chromatographic method involving hydroxylapatite and Fast Protein Liquid Chromatography mono S. The purified enzyme has a molecular weight of 67 kilodaltons as determined by sodium dodecyl sulfate denaturing gel electrophoresis. Western blotting confirms the identity of the protein as topo I. The purified lymphoma topo I has a similar specific activity as topo I isolated by the same procedure from normal placenta. By using a new quantitative radiolabeled DNA relaxation assay, it was estimated that each molecule of topo I is able to relax 9.6 molecules of supercoiled DNA per min at 30C. In addition it was found that both the placental topo I and the lymphoma topo I were each 50% inhibited by 8 microns camptothecin and that the drug stimulates the lymphoma topo I to cleave supercoiled plasmid DNA. These findings indicate that the elevation of topo I measured in crude extracts of this human malignant lymphoma can be entirely accounted for by the increase in topo I protein in the tumor. Furthermore, the sensitivity of the lymphoma topo I towards camptothecin suggests that therapy with topo I directed agents might be beneficial in this tumor.
Several epidemiological investigations have suggested an increased incidence of lymphoma, leukemia, and brain tumor in residents living near power transmission lines. However, some observers failed to confirm such a positive correlation. To evaluate the effects of extremely low frequency (ELF) magnetic fields on leukemogenesis, an experimental animal model was used, in which thymic lymphoma/leukemia was induced by dimethylbenz(alpha)anthracene (DMBA) injected subcutaneously into the interscapular region of newborn mice within 24 h after birth. Beginning at the second week of age, 165 mice were exposed to 50 Hz magnetic field at 1 mT, 3 h/day, 6 days/week for 16 weeks, and 155 animals exposed to sham conditions. All surviving animals were killed by cervical dislocation at the age of 32 weeks and were examined pathologically. The results showed that the incidences of advanced thymic lymphoma, complicated with lymphomatous leukemia, were 21.8 and 23.9% in the two groups, respectively, without statistically significant differences. But dense metastatic infiltration by lymphoma cells into liver in the field exposure group greater (50%) than that in the sham-exposure group (16.2%) was observed (chi 2 = 9.847, P < 0.01). To determine whether ELF acts as a tumor promoter, further experiments are required.

Mucinous cystic neoplasms (MCN) of the pancreas are mucin-producing cystic tumors with an ovarian-like stroma (OLS). In the present study MCN were obtained from 27 patients. These MCN were derived from 22 pancreas, three livers, spleen, and mesentery. MCN in various organs have common clinicopathological profiles, being unilocular or multilocular cystic tumors, with a fibrous capsule and lined by mucin-secreting epithelium associated with an underlying subepithelial OLS. The OLS showed strong positivity for alpha-smooth muscle actin (alpha-SMA) and vimentin and weak, focal positivity for desmin. Both estrogen receptors and progesterone receptors were expressed in the nuclei of OLS cells. In addition, 20 ovarian MCN and 13 normal ovaries were studied with particular attention to the stroma. The stroma of ovarian MCN was strongly immunopositive for alpha-SMA and vimentin and focally positive for desmin, whereas normal ovarian stroma was immunonegative for both alpha-SMA and desmin. The OLS of MCN mentioned here was similar to the septa of ovarian MCN but not to ovarian stroma. In conclusion, MCN in various organs should be lumped together as 'extra ovarian' MCN. The OLS was identified on the basis of myofibroblastic proliferation both in response to neoplastic development and dependent on hormones.
