Chloride transport in bullfrog gastric mucosa.

Omeprazole uncouples histamine-stimulated hydrogen and chloride secretion in the frog gastric mucosa. This uncoupling results in a large increase in PD and Isc due to the unmasking of an electrogenic Cl- flux. Using a selective anoxia technique, we have attempted to define the cellular origin of this electrogenic Cl- flux. Sixteen bullfrog gastric mucosae were mounted in Ussing chambers. Under short-circuit conditions, PD, Isc, R, and unidirectional 36Cl flux were determined every 15 min. Acid secretion (10(-5) M histamine) was measured by hand titration (0.1 N NaOH). With both sides of the mucosae oxygenated, the addition of omeprazole (10(-4) M) resulted in cessation of H+ secretion and the unveiling of an electrogenic Cl- flux. Subsequent serosal anoxia (5% CO2-95% N2 nutrient side/30% O2 secretory side) resulted in a drop in JClnet associated with an appropriate fall in Isc. These changes were reversible with reoxygenation: (Table: see text). Bullfrog gastric mucosae which are partially oxygenated on the secretory side only (serosal anoxia) do not secrete acid in response to histamine. Therefore, the JClnet which is inhibited in these experiments by serosal anoxia may originate in the gastric glands approximately equal to 3.0 mu eq/cm2 X hr). The JClnet which remains during serosal anoxia may originate in the surface epithelial cells (approximately equal to 1.0 mu eq/cm2 X hr).

Fura-2 handling in a polarized epithelial barrier: the toad urinary bladder.

Toad bladders sacs were placed inside quartz cuvettes. When fura-2 AM was added to the mucosal compartment, low temperature (4 degrees C) almost completely blocked the transepithelial transfer of fluorescence observed at 20 degrees C (20 degrees C = 371 +/- 56, 4 degrees C = 29 +/- 29 fluorescence intensity in arbitrary units (FIAU), excitation at 340 nm, emission at 510 nm). Simultaneously, fluorescence accumulation inside the tissue was significantly higher (20 degrees C = 25 +/- 5, 4 degrees C = 91 +/- 24% increase on basal levels (%IBL)). When fura-2 AM was added to the serosal side, low temperature also reduced the serosal to mucosal transfer (20 degrees C = 149 +/- 36, 4 degrees C = 61 +/- 35 FIAU). Nevertheless, in this situation tissue accumulation, that was significantly higher that the one observed when fura-2 AM was added to the mucosal side, was reduced at low temperature (20 degrees C = 300 +/- 30, 4 degrees C = 48 +/- 7 %IBL). Spectral analysis of the mucosal and serosal compartments indicated that free fura-2 was transferred from the intracellular to the serosal compartment, but not to the mucosal one. These results indicate that fura-2 appears as a useful tool to evaluate the cellular distribution and traffic of polycyclic charged and non-charged molecules.

Cadaverine and aminoguanidine potentiate the uptake of histamine in vitro in perfused intestinal segments of rats.

Examination of the serosal fluid following in vitro luminal perfusion of rat intestinal segments with 1 mg/ml [3H]histamine for 2 hr showed that histamine constituted only 22.1% of the total serosal radioactivity. The remainder of the radioactivity was comprised of histamine metabolites. When equimolar amounts of either aminoguanidine and cadaverine were added to the luminal perfusate, the percentage of the serosal radioactivity as histamine increased to 67.0 and 60.4%, respectively. However, when equal amounts of histamine and anserine were added to the luminal perfusate, only 30.6% of the 3H translocated within 2 hr was [3H]histamine. In all cases, the gross translocation rate based on the percentage of total serosal radioactivity for total radioisotope [( 3H]histamine plus [3H]histamine metabolites) was unchanged by the addition of these substances to the luminal perfusate. The results indicate that the potentiation of histamine toxicity by putrefactive amines, such as cadaverine, results from the inhibition of histamine metabolism which leads to increased uptake of unmetabolized histamine. The results do not support the hypothesis that potentiation occurs via an overall increase in the absorption of histamine and its metabolites due to some disruption in the barrier function of the intestine.

On the mechanism of stimulation of H+ transport in gastric mucosa by Ca++ ionophore A23187. I. Pathways for cytosolic calcium increase.

The pathways for cytosolic Ca++ increase under A23187 stimulation of H+ secretion were studied in the isolated gastric mucosa of the toad Bufo marinus. A23187 produced a more potent stimulation of secretion when added to the mucosal side which did not contain calcium. Measurements of ionophore incorporation by fluorometric methods indicated that A23187 incorporates into oxyntic cells intracellularly. The presence of divalent cations inhibited incorporation. This may be the reason for a more potent action when A23187 was added from the mucosal side. Withdrawal of calcium from serosal solution largely inhibited the secretory response to A23187 added to the mucosal side. Reintroduction of calcium into the serosal side in the presence of ionophore elicited H+ secretion. The results are consistent with an uptake of A23187 from the mucosal side into cellular organelles and basolateral membranes. Calcium entry through the serosal side may be responsible for triggering secretion. Although A23187 likely releases calcium from intracellular stores, its rate of release may not be sufficient to bring about a full stimulation of secretion in serosal-Ca++-free conditions.

The effect of TGF alpha on intestinal solute transport.

The effect of transforming growth factor alpha (TGF alpha) and epidermal growth factor (EGF) on 3-O-methylglucose transport was examined in vitro under short-circuited conditions in stripped rabbit jejunum. Mucosal EGF, 60 ng/ml, stimulated a significant increase in net 3-O-methylglucose transport (Jnet 0.67 +/- 0.15 vs. 0.90 +/- 0.15 microEq/cm2/h; P less than 0.03; n = 6) due to an increased mucosal to serosal flux (Jms 1.2 +/- 0.2 vs. 1.5 +/- 0.2 microEq/cm2/h; P less than 0.03). In contrast, TGF alpha, when applied to both mucosal and serosal surfaces at concentrations of either 60 (n = 6) or 150 (n = 9) ng/ml had no effect on either mucosal to serosal (Jms) or net transport (Jnet) of 3-O-methylglucose. TGF alpha did induce a significant increase in the serosal to mucosal flux (Jsm 60 ng/ml 0.44 +/- 0.02 vs. 0.51 +/- 0.03, 150 ng/ml 0.55 +/- 0.03 vs. 0.64 +/- 0.05 microEq/cm2/h; P less than 0.05). When brush border surface area was examined after exposure to either 60 ng/ml TGF alpha or saline vehicle for 2 h in in vivo isolated jejunal loops no significant difference was found (control 53 +/- 1.9; n = 35 vs. TGF alpha 52 +/- 1.9 microns 2; n = 29). Bioactivity of transforming growth factor alpha was assessed by an gastric acid secretion bioassay and found to be intact. These data provide further evidence for separate and distinct functional roles for these peptides in some biological systems.

Free peritoneal graft for repair of severe seromuscular defect of bowel: from experiment to clinical practice.

BACKGROUND: Severe seromuscular tear of the bowel with impending perforation following enterolysis or trauma is not uncommon in gastrointestinal surgery. It is sometimes complicated with enterocutaneous fistula, intra-abdominal abscess or free perforation, especially of the ileus. In addition, direct serosal repair is often impossible, or complicated by lumen compromise. Serosal patch repair also reportedly is also associated with complications such as dislodgement of the sutured bowel loop, fistula formation, adhesion ileus, or volvulus. Free peritoneal graft (FPG), as in split-thickness skin graft (STSG), can be taken by underlying healthy tissue within 24 to 48 h and used for repair of such defect. PATIENTS AND METHODS: In this study, FPG was used as an alternative to serosal patch repair or resection to cover severe defects of the colon, rectum, duodenum, or small bowel in 30 consecutive patients. All defects were over 8 x 4 cm in size and involved 30 to 80% of the circumference of the bowel wall. Each defect was deep enough to expose the thin mucosal layer, with the intestinal content visible through it. These defects could have been successfully treated by resection or repair using a serosal or mucosal patch instead of FPG. After making sure that the mucosa associated with the defect was alive and not perforated, a patch of intact peritoneum with pre-peritoneal tissue from the lateral abdominal wall (slightly smaller than the defect) was superimposed, with the edges simply sewn to the margin of the defect. RESULTS: All patients recovered uneventfully, with no untoward effects determined, even after long-term follow-up. CONCLUSION: We believe that FPG provides a feasible, simple, effective, economic, and safe alternative for repair of severe seromuscular defects; potentially making it widely applicable in clinical practice.

Amendments to the theory underlying Ussing chamber data of chloride ion secretion after bacterial enterotoxin exposure.

Bacterial enterotoxins may cause life-threatening diarrhoeal fluid loss in part because they stimulate enterocytes to secrete fluid into the small intestine as well as preventing normal fluid uptake. Abnormal chloride ion secretion is believed to provide the osmotic driving force for the inappropriate fluid movement. Evidence for enhanced chloride secretion consists of isotopic flux measurements in Ussing chambers, the standard apparatus for permeation studies. Flux from the lumen of the intestine is assumed to be determined solely by absorptive processes and flux towards the lumen solely by secretory processes. Bacterial enterotoxin increased flux towards the lumen is taken as an evidence of enhanced secretion. Examination of the flux equation solutions shows that the existing theoretical treatment of the Ussing chamber consists of the super-imposition of two contradictory unidirectional models. In contrast, the present analysis shows that a measured 'unidirectional' flux contains information both about absorptive and secretory processes, regardless of which flux is measured. Reciprocity is predicted for the fluxes, as decreases in the absorptive processes will cause increases in apparent secretory flux. Data from the literature show that mucosal-to-serosal chloride ion flux in rabbit ileum after exposure to secretagogues correlates inversely and highly significantly (r=0.74, n=17, p<0.001) with increases in serosal-to-mucosal chloride ion flux. As a category of evidence, flux data do not provide conclusive evidence of enhanced chloride secretion after exposure to enterotoxins, since an apparently enhanced serosal-to-mucosal flux would also be noted after inhibition of the mucosal-to-serosal flux. As interruption of absorptive processes can be misinterpreted as enhanced secretion in the Ussing chamber, this is a serious deficiency in the evidence for direct enterotoxin enhancement of the intestinal chloride ion channel as a basis for diarrhoeal disease.

Endogenous E-type prostaglandins in regulation of basal alkaline secretion by amphibian duodenum in vitro.

Segments of proximal duodenum from Rana catesbeiana, stripped of external muscle and mounted as a tube in a glass chamber, alkalinized the luminal-side bathing solution at a rate of 1.70 +/- 0.16 microEq/cm . h (n = 18, gross surface area approximately 1.5 cm2/cm). A single change of the serosal-side bathing solution for fresh solution reduced the rate of titratable alkaline secretion, which achieved a new steady state after 45 min amounting to 66% +/- 2% of the initial rate; transmucosal potential difference (lumen negative) fell from 10.6 +/- 1.2 to 8.8 +/- 1.1 mV. Concentrations of E-type prostaglandins in the serosal-side solution measured by radioimmunoassay were 8.5 nM (3 ng/ml) before, 0.17 nM 5 min after, and 1.7 nM 90 min after the solution change (n = 8). Reapplication of the original bathing solution 90 min after the initial change reestablished original secretory rate and potential difference. The increases in alkaline secretion and potential difference were comparable in magnitude and profile to those induced by serosal administration of 10 nM prostaglandin E2. Addition of the metabolic inhibitor 2,4-dinitrophenol (100 microM, serosal side) reduced basal alkaline secretion to 30% +/- 7% of the initial rate and abolished the potential difference (n = 8). These data demonstrate that endogenous prostaglandin E production by an isolated preparation of amphibian duodenum accounts for a proportion of alkaline secretion that is equivalent to 50% of metabolism-dependent basal secretion.

A serous lined antireflux valve: in vivo fluorourodynamic evaluation of antireflux continence mechanism.

PURPOSE: We evaluate the fluoroscopic and urodynamic performance of a new serosal lined antireflux continence valve (T pouch valve) using an in vivo animal model. MATERIALS AND METHODS: Intestinal reservoirs were constructed using an in vivo pig model. Multiple serosal lined valves of varying diameters and tunnel lengths were evaluated in the acute (immediately after construction) and chronic (4 weeks postoperatively) settings. Video fluorourodynamic evaluations were performed to identify valve leak point pressures and optimal tunnel length-to-luminal diameter ratios. RESULTS: Four serosal lined valves tapered to a diameter of 18 and 30F in 1 and 2 cm. tunnels were evaluated. Immediately following construction of the valves no leakage was observed during reservoir filling (maximal pressure 50 cm. water) or external compression maneuvers to simulate a Valsalva maneuver (maximal intra-reservoir pressure 100 cm. water). Identical fluorourodynamic studies performed after a 4-week recuperation demonstrated leakage through the 30F, 1 cm. tunnel valve during reservoir filling (intra-reservoir pressure 19 cm. water). Increasing the tunnel length of the 30F system to 2 cm. (tunnel length-to-luminal diameter ratio 2:1) eliminated all evidence of valve leakage up to a maximal intra-reservoir pressure of 100 cm. water. CONCLUSIONS: These studies demonstrate the urodynamic characteristics of a new serosal lined valve. This easily constructed mechanism serves as a reliable antireflux (afferent) or continence (efferent) system.

Cocaine permeability and metabolism in colonic T-84 epithelial cell line.

We studied the uptake, transport and metabolism of cocaine in human intestine using the colonic T-84 monolayers as a model. T-84 cells were grown in DMEM/Ham's F-12 medium containing 6% newborn calf serum at 37 degrees C on 1.0 microm collagen inserts. The cells develop into a polarized monolayer with the apical surface facing the upper chamber and the basolateral surface facing the lower. The monolayers develop a transepithelial resistance of > or = 600 ohms cm2 in 7 days. Varying concentrations of cocaine HCI was added in a serum free medium to the luminal side only, and after 30 min and 60 min samples from the luminal and serosal aspect were removed for analysis. Cocaine and its metabolites were measured by GC/MS. Cocaine transport across T-84 monolayers increased linearly with increasing concentration of cocaine, with no significant difference between 30 min and 60 min of exposure. Transepithelial resistance did not change even at 800 ng of cocaine suggesting no effect on monolayer viability. The metabolites, benzoylecgonine (BE) and ecgonine methyl ester (EME) but not norcocaine were detected in both luminal and serosal side. The concentrations of BE and EME in the luminal side were significantly higher than in the serosal. Combined recovery of cocaine, BE and EME from luminal and serosal sides were 52 - 80% of total added cocaine. While fresh medium did not metabolize cocaine, media previously exposed to the monolayer (cell-free medium) caused a significant breakdown into BE and EME, suggesting that esterases may be released into the medium. These results indicate transfer of cocaine across this model of intestinal epithelial cell line is by simple diffusion and is concentration dependent. These studies imply that cocaine in swallowed amniotic fluid can be absorbed by the fetal gastrointestinal tract.

Mucosal type mast cells express complement receptor type 2 (CD21).

Fragments of complement component C3 generated upon activation of the cascade play an important role in the induction and regulation of immune responses. Receptors interacting with various fragments of this versatile complement protein are expressed on a wide variety of cell types, including lymphocytes, macrophages, dendritic cells, follicular dendritic cells, granulocytes, erythrocytes and consequently, C3-products may influence several biological functions at different sites of the body, where complement activation occurs. Regarding the expression of various C3-receptors on mast cells, mainly rodent serosal type mastocytes have been investigated so far. It has been known for a long time that C3a triggers the release of mediators of immediate type hypersensitivity via binding to serosal-type cells. Complement receptor type 1 (CR1/CD35) and type 2 (CR2/CD21) interacting with the larger activation products, such as C3b and C3d, have so far been shown on serosal type mast cells only. In this study, the expression of CR1/2 on mucosal type mast cells is demonstrated. Using mouse CR1/2 specific single chain antibodies and the natural ligand C3d in cytofluorimetric measurements, we show that the rat mucosal mast cell line RBL-2H3 and mouse bone marrow-derived mast cells (BMMC) express CD21. RT-PCR experiments carried out with mouse CR1 and CR2 specific primers show CD21, but not CD35 specific products in BMMC. It is also demonstrated that, in contrast to serosal type mast cells, mucosal mastocytes do not express CD19. In an attempt to reveal the possible function of CR2 on mucosal type mast cells, the effect of receptor-clustering was tested regarding degranulation, Ca-response and IL-6 production, but no CR2-mediated change was detected in any of these processes.

Intestinal Na+/glucose cotransporter-mediated transport of glucose conjugate formed from disaccharide conjugate.

Intestinal absorption of beta-disaccharide (cellobiose, maltose and lactose) conjugates of p-nitrophenol (p-nitrophenyl beta-disaccharide) were examined in terms of the hydrolysis of disaccharide conjugate to monosaccharide conjugate and the transport of monosaccharide conjugate by Na+/glucose transport carrier (SGLT1). beta-Cellobioside, beta-maltoside and beta-lactoside of p-nitrophenol (p-NP) were hydrolyzed to p-nitrophenyl beta-glucoside (p-NPbeta glc) on the mucosal side, and p-NPbeta glc appeared on the serosal side. Although p-NP beta-disaccharide, p-NP and p-NP glucuronide also appeared on the serosal side, their amounts were much lower than that of p-NPbeta glc. The amount of p-NPbeta glc transported to the serosal side was decreased in the presence of phloridzin (transport inhibitor of SGLT1) and in the absence of Na+ (a cosubstrate of SGLT1), indicating that p-NPbeta glc was formed from p-NP beta-disaccharide on the mucosal side and transported to the serosal side by SGLT1. Furthermore, the absorption clearance of p-NPbeta glc, which was formed from p-NP beta-cellobioside and p-NP beta-lactoside by lactase-phloridzin hydrolase (LPH), was much higher than that of p-NPbeta glc itself, although the absorption clearance of p-NPbeta glc, which was formed from p-NP beta-maltoside by maltase was similar to that of p-NPbeta glc itself. These results indicated that p-NPbeta glc was transported by the vectorial cooperation of SGLT1 with LPH from mucosal p-NP beta-cellobioside or p-NP beta-lactoside.

Inhibitory effect of phloridzin and phloretin on glucuronidation of p-nitrophenol, acetaminophen and 1-naphthol: kinetic demonstration of the influence of glucuronidation metabolism on intestinal absorption in rats.

Intestinal glucuronidation and absorption of p-nitrophenol (p-NP), acetaminophen (APAP) and 1-naphthol (alpha-NA) in the presence of phloridzin (inhibitor of Na+/glucose cotransporter) and phloretin (aglycone of phloridzin) were studied. Glucuronides of p-NP, APAP and alpha-NA appeared on both the serosal and mucosal sides. The amounts of glucuronides on the serosal side were decreased in the presence of phloridzin and phloretin. p-NP, APAP and alpha-NA appeared on the serosal side as well, and the amounts of p-NP, APAP and alpha-NA on the serosal side were increased by the presence of phloridzin and phloretin. Furthermore, the intestinal glucuronidation and absorption of alpha-NA at various concentrations were studied in the presence and absence of phloretin. Metabolic clearance was decreased in the presence of phloretin, and the absorption clearance was increased. The higher concentrations of alpha-NA caused higher absorption clearance. The lower the metabolic clearance, the higher the absorption clearance. The relationship between glucuronidation metabolism and absorption in intestine was kinetically analyzed by the metabolic inhibition model. Complete inhibition of glucuronidation improved the intestinal absorption of alpha-NA, and the absorption clearance increased to 7.17 microliter/min/cm. The formation of phloretin and an unknown metabolite from phloridzin were observed. An unknown metabolite from phloretin was observed, and was suppressed by the presence of alpha-NA. This suggests that phloridzin was hydrolyzed to phloretin, which was metabolized to glucuronide, and thereby inhibited glucuronidation of p-NP, APAP and alpha-NA.

Site-specific absorption of M100240 and MDL 100,173 in rats evaluated using Sweetana-Grass diffusion chamber technology.

INTRODUCTION: M100240--an acetate thioester of MDL 100,173--a dual angiotensin-converting enzyme (ACE)/neutral endopeptidase (NEP) inhibitor is in Phase II development for cardiovascular conditions. The absorption characteristics of M100240 and MDL 100,173 were evaluated in specific regions of the rat gastrointestinal (GI) tract. METHODS: Solutions of M100240 and MDL 100,173 were prepared using mucosal Kreb's solution. Four male Sprague-Dawley rats, fasted before tissue procurement, were sacrificed using 95% CO2 and 5% O2. The entire small and large intestine were removed, rinsed in serosal Kreb's solution, and segments mounted onto the Sweetana-Grass diffusion chamber. Test compounds were added to the donor compartment and drug-free oxygenated serosal Kreb's solution to the receiving compartment. The temperature of the chambers was maintained at 37 degrees C and supplied with 95% O2/5% CO2. Samples were removed (0.5 ml) at 0, 30, 60, 90, 120, 180, and 240 min from the serosal side. The volume was maintained with drug-free warm serosal Kreb's solution. Samples were diluted with rabbit plasma and analyzed by liquid chromatography/mass spectrometry. Apparent permeation values [Papp (cm/min)] of M100240 and MDL 100,173 through the duodenum, jejunum, ileum, and colon were determined. RESULTS: The mean Papp of M100240 and MDL 100,173 was highest in the duodenum: 2.29 x 10(-4) +/- 2.40 x 10(-4) and 1.66 x 10(-4) cm/min +/- 8.33 x 10(-5), respectively. The mean Papp was lowest in the colon: 3.61 x 10(-6) +/- 3.44 x 10(-6) and 1.62 x 10(-5) +/- 3.21 x 10(-6) cm/min for M100240 and MDL 100,173, respectively. Absorption of MDL 100,173, however, was evident throughout the rat GI tract. DISCUSSION: M100240 and MDL 100,173 are predominantly absorbed from the duodenum in the rat GI tract. MDL 100,173 is also absorbed from the jejunum, ileum, and colon. These results, consistent with data obtained in humans, demonstrate the potential predictive value of the Sweetana-Grass model for site of absorption assessments.

Vitamin absorption in the in vivo intestine of normal and infected (Hymenolepis diminuta: Cestoda) rats.

Uptake and serosal transfer of the vitamins thiamine, riboflavin and folic acid have been studied in vivo in normal and parasitized rats infected with Hymenolepis diminuta (Cestoda). Regional differences in intestinal uptake of all three vitamins in both uninfected and parasitized animals were not satistically significant. In the parasitized intestine mucosal uptake and serosal transfer of thiamine were significantly inhibited, with increased mucosal accumulation of the vitamin as luminal thiamine concentration increased. Apparent increased riboflavin mucosal uptake in parasitized animals, was not matched by the reduced serosal transfer, suggesting adsorption of the vitamin in the unstirred aqueous layers. Mucosal uptake of folic acid increased in the parasitized gut; serosal transfer and mucosal accumulation were not affected. These results, indicating vitamin malabsorption associated with infection by H. diminuta, are consistent with the parasite inhibiting mucosal passive transport mechanisms. This conclusion is supported by the changes in net water fluxes associated with vitamin uptake in the parasitized intestine.

Translocation of human calcitonin in respiratory nasal epithelium is associated with self-assembly in lipid membrane.

We studied the mechanisms involved in the translocation of human calcitonin (hCT) through excised bovine nasal mucosa (net mucosal-to-serosal permeability approximately 10(-)5 cm s-1). To determine structural requirements for the suggested vesicular internalization two carboxyfluorescein-labeled (fl) hCT fragments, the C-terminal fragment [Nalpha-fl]hCT(9-32) and the N-terminal fragment [Lys(fl)18]hCT(1-24) were synthesized. In presence of the endocytosis inhibitor cytochalasin D mucosal-to-serosal and serosal-to-mucosal hCT permeabilities were equal. Pathway visualization by confocal laser scanning microscopy showed punctated fluorescence indicating vesicular internalization of both hCT and [Nalpha-fl]hCT(9-32). In contrast, the N-terminal fragment lacking the beta-sheet forming C-terminus (25-32) was not internalized. Circular dichroism showed that, when interacting with neutral and negatively charged liposomes, hCT adopts beta-sheet conformation. In a concentrated aqueous solution, beta-sheet formation induces hCT self-assembly and fibrillation. High partitioning of hCT into lipid bilayer membranes was reflected by an apparent partition coefficient log D(pH 7.4) = 2.5 (liposome-buffer equilibrium dialysis). We propose that the high lipid partitioning and beta-sheet formation result in C-terminus-restricted supramolecular self-assembly of hCT and [Nalpha-fl]hCT(9-32) in lipid membranes. Vesicular internalization is suggested to be associated with self-assembly induced perturbation of the lipid bilayer. Condensed hCT self-assemblies may explain the high capacity of net mucosal-to-serosal hCT permeation, which compares favorably with the low transport capacity of receptor-mediated endocytosis.

Effect of cyclic AMP on acidification in the isolated turtle bladder.

Cyclic AMP (10 mM) has been demonstrated to inhibit hydrogen ion secretion in the isolated turtle bladder. These experiments were designed to study the effect of cyclic AMP on hydrogen ion secretion in the isolated turtle bladder using both the pH stat and the reverse short circuit current techniques. Sodium transport was measured as the short circuit current. Studies were carried out at 0% CO2 and 1% CO2 at pH 7.4. Cyclic AMP (10 mM) was added to either the serosal or mucosal solutions, and hydrogen ion secretion was measured from 0 to 120 min. In the presence or absence of carbon dioxide, cyclic AMP and dibutyryl cyclic AMP had no effect on hydrogen ion secretion in fasted turtles. The addition of theophylline (10 mM) to the serosal solution, with or without cyclic AMP had no effect on proton secretion. Sodium transport was unchanged from control following serosal or mucosal addition of 10 mM cyclic AMP in the presence or absence of carbon dioxide. In chronically bicarbonate-loaded turtles proton secretion was the same as control fasted turtles. In these animals, however, serosal administration of 10 mM cyclic AMP significantly stimulated bicarbonate secretion. Stimulation of bicarbonate secretion occurred in the presence of a 20 mM bicarbonate gradient. When there was no bicarbonate gradient, cyclic AMP was without effect; cyclic AMP had no effect on bicarbonate permeability when measured in the presence of acetazolamide. These results indicate that cyclic AMP has no effect on hydrogen ion secretion or sodium transport in the isolated turtle bladder when studied at two different rates of acidification (0 and 1% CO2). Cyclic AMP appears to stimulate active bicarbonate secretion.

Absorption of 5-fluorouracil and related pyrimidines in rat small intestine.

The transport of 5-fluorouracil, uracil and thymine has been studied with isolated jejunal loops of rat small intestine. High performance liquid chromatography was used to identify the pyrimidines and measure their concentrations. When the lumen of the intestine was perfused with 5-fluorouracil or uracil at 0.1 mmol/l or 0.2 mmol/l, the concentration in the serosal secretions was significantly higher than that in the lumen. For thymine the serosal concentration exceeded that in the lumen only at 0.1 mmol/l. Analysis of the mucosal tissue water at the end of the perfusion demonstrated that when the intestinal lumen was perfused with any one of the three pyrimidines at 0.1 mmol/l or 0.2 mmol/l the concentration within the tissue was significantly above that in the lumen. After an initial lag period linear rates of transport from the lumen to the serosal secretions were obtained for all three pyrimidines over a 10-fold concentration range from 0.1 mmol/l to 1 mmol/l. Uracil and thymine inhibited the transmural transport of 5-fluorouracil. The transport of 5-fluorouracil was also studied with a vascularly perfused preparation of rat small intestine. At 0.1 mmol/l the rate of transmural transport of the drug in this preparation was substantially higher than in the jejunal loops. This difference was eliminated by adding 5-fluorouracil to the vascular perfusate, suggesting that the higher transport rate in the vascularly perfused preparation was due to the lower serosal drug concentrations in the mesenteric circulation of the perfused intestine. At a concentration of 5 mmol/l 5-fluorouracil inhibited water transport in the isolated loops and transmural D-galactose transport in the vascular perfusions.
