Liposome delivery of aminoglycosides in burn wounds.

The current study evaluated the pharmacodynamics of topically applied antimicrobials incorporated into radiolabeled liposomes. Radiolabeled 125I-phenyldecanoic acid was used in the formulation of small unilamellar liposomes. Sephadex (cross-linked dextran beads) G-50 columns were run to determine the per cent of radioactivity incorporated into liposomes and persistence of radioactive tag on the liposome after two weeks (greater than 95 per cent) remained incorporated. On the day of the experiment, 31 adult Sprague Dawley rats were subjected to a 10 per cent total body surface area full thickness burn (Walker burn model). All rats were treated with topical application of 0.3 milliliters of tobramycin entrapped in 125I-liposomes, and burn wounds were covered with Opsite (Winfield Laboratories). This formulation resulted in each rat receiving 14 milligrams per kilogram of tobramycin with a specific activity of 10.41 microcuries. Rats were sacrificed at several time periods after burn injury (nine after 24 hours, 12 after 48 hours, 11 after 72 hours). At these time intervals, serum and tissue tobramycin levels were measured, burn dressing, burn tissue and splanchnic organs were harvested and radioactivity was assessed with a gamma scintillation counter to determine tissue concentration of 125I-liposome tobramycin. Concentration of tobramycin in the serum was negligible at 24, 48 and 72 hours postburn, but was significant in the burn tissues at these times. The radioactive recovery data demonstrated that the majority (greater than 90 per cent) of the recovered liposomes remained at the site of application (the burn wound). No splanchnic organs had greater than 2 per cent of the recovered 125I-liposomes at any time period. These data suggest that, in burn wounds, tobramycin incorporated into liposomes remain at the site of initial application, resulting in high local concentrations with little systemic absorption and confirm that liposomes provide an effective vehicle for delivery of antimicrobials at the site of the burn injury.

[3H-thymidine-paper strip-counting of in vitro lymphocyte transformation. II. Specific antigens in the whole blood assay (author's transl)].

Erythrocytes did not influence the harvesting procedure or the paper strip counting method in a liquid scintillation counter using the whole blood assay of lymphocyte transformation test. The leucocyte-erythrocyte-ratio influences the blastogenesis in vitro. Only 1--2 ml of peripheral blood are necessary to proof lymphocyte reactivity to mitogens or antigens. The storage of the blood is possible over a period of 24 hours (4 degrees C) if the absolute lymphocytes count remains constant.

Regional permeability of coenzyme Q10 in isolated rat gastrointestinal tracts.

The objective of the study was to identify the region with the maximum permeability for low bioavailable coenzyme Q10 (CoQ) in the gastrointestinal tract. To evaluate the regional differences in permeability, male Sprague-Dawley rats, 250-300 g, were anesthetized and the gastrointestinal segments were isolated. Stomach, duodenum, jejunum, ileum and colon tissues were mounted on a Navicyte side-by-side diffusion apparatus. Radiolabeled CoQ (1 microM in DMEM, pH 7.4, 37 degrees C was added to the donor side and the samples withdrawn from the receiver compartment at predetermined time intervals were analyzed using a scintillation counter. Membrane integrity was monitored by 14C-mannitol permeability. The apical to basal permeability coefficients (Papp x 10(-6), cm/s) were 0.32 +/- 0.13, 3.14 +/- 0.89, 1.36 +/- 1.4, 0.83 +/- 0.40, and 1.59 +/- 0.13, for CoQ through rat stomach, duodenum, jejunum, ileum, and colon tissues respectively. The basolateral to apical permeability coefficients (Papp x 10(-6), cm/s) were 1.6 +/- 0.2, 2.2 +/- 1.2, 0.88 +/- 0.12, 1.6 +/- 0.42, and 1.9 +/- 0.41 respectively. Therefore the region of maximum CoQ permeability is duodenum followed by colon and ileum. Jejunum and stomach regions also have fairly high permeability. Therefore CoQ formulations should be made with an aim to target the duodenum to get maximum dosage effect.

[Ocular penetration of UF-021, a new prostaglandin related compound, in the rabbit eye].

The ocular penetration and pigment affinity of topically applied UF-021, a newly developed prostaglandin-related compound, were investigated in rabbit eyes using 3H-labeled UF-021. Ten minutes to 24 hours after instillation of 0.12% of 3H-UF-021 solution into New Zealand white or pigmented rabbit eyes, the rabbits were sacrificed and the eyes were immediately enucleated. The extraocular muscles, conjunctiva, aqueous humor, cornea, iris, anterior and posterior sclera, ciliary body, lens, vitreous, retino-choroid complex, optic nerve were separated and blood samples were taken. The concentration of 3H-UF-021 in each sample was determined with a liquid scintillation counter. The peak concentration of UF-021 was obtained at 40 minutes and 1 hour after instillation in the iris-ciliary body and aqueous humor of albino rabbit. According to the pharmacokinetic analysis of the data, UF-021 showed good ocular penetration into the rabbit eye, and the permeability of the epithelial barrier for UF-021 was calculated to be 2.9 x 10(-3) cm/hr. The apparent elimination rate constant and the apparent absorption rate constant were also calculated to be 0.21 hr-1 and 1.28hr-1, respectively. The time course for 3H-UF-021 concentration in the eye of pigmented rabbit was almost the same as that in the eye of non-pigmented rabbit, indicating that UF-021 did not bind to pigmented tissues.

[Immune regulatory effect of human bone marrow mesenchymal stem cells on T lymphocyte].

To investigate the immune regulatory effects of human bone marrow mesenchymal stem cells on alloantigen T lymphocyte in vitro, human MSCs were isolated and expanded from bone marrow cells, and identified with cell morphology, and the phenotypes were assessed by immunohistochemistry and flow cytometry. As the stimulation factor of T lymphocytes proliferation, either PHA or dendritic cells isolated from cord blood were cocultured with CD2(+) T lymphocytes from peripheral blood mononuclear cells by magnetic beads with or without MSC in 96-well plats for seven days. T cell proliferation was assessed by [(3)H]-thymidine incorporation using a liquid scintillation counter. T cell subsets, Th1, Th2, Tc1 and Tc2 were analyzed by flow cytometry after co-culture of CD2(+) T cells with MSCs for 10 days. The results showed that a significant decrease of CD2(+) T cell proliferation was evident when MSC were added back to T cells stimulated by DC or PHA, and an increase of Th2 and Tc2 subsets were observed after co-culture of MSC with T lymphocytes. It is suggested that allogeneic MSC can suppress T cell proliferation in vitro and the cause of that was partly depend on interaction of cells and the alteration of T cell subsets.

[Improved radioligand assay of insulin autoantibody].

OBJECTIVE: To establish an improved radioligand assay of insulin autoantibody (IAA) and to investigate the diagnostic role of IAA in type 1 diabetes (T1DM). METHODS: 125I-insulin antigens were mixed with sera samples in the Eppendorf tubes. The immunocomplexes were precipitated with protein A-agarose, then washed with TBST buffer and counted the cpm value using liquid scintillation counter. The assay results were expressed by IAA index and compared with those of an international standardized laboratory and a domestic anti-insulin antibody kit. Sera of 32 recent-onset type 1 diabetic patients and 120 healthy controls were screened for IAA and the consistency, sensitivity, specificity and the diagnostic value of IAA were evaluated. RESULTS: (1) The intra-assay coefficient of variation (CV) was 5.8% - 8.3%, and the inter-assay CV 7.0% - 11.0%. The IAA indices of 41 samples tested in our laboratory and in an international standardized laboratory were strongly positively related (r = 0. 783, P < 0. 01) (100.0% concordance). Arrayed according to their antibody titers, the IAA indices of 82 samples measured in our laboratory and in a domestic anti-insulin antibody kit were also strongly positively related (r = 0.982, P < 0.0001) (89.0% concordance, Kappa value 0.813), but 9 samples were positive with low titer (index 0.05 - 1.00) by the radioligand assay or negative by the kit. 2) The IAA index cut-off point was determined according to the upper limit of 99.5% percentile of 120 healthy controls. The index of 0.051 or higher was defined as positive. The IAA positivity prevalence was 18.8% (6/32) in type I diabetes and 0.8% (1/120) in the healthy controls (P < 0.001). The positivity frequency of IAA was 28.6% (4/14) in patients younger than 15 years of age and the duration less than half a year, while 11.1% (2/ 18) in those older than 15. CONCLUSION: The improved radioligand assay for IAA has a high sensitivity and specificity and can be used in clinical practice, especially for the low-titer positive subjects.

[Enhancement of bacillus Calmette-Guerin attachment to the bladder wall: experimental study].

OBJECTIVE: To provide experimentally a scientific basis for enhancement of bacillus Calmette-Guerin (BCG) antineoplastic effect. METHODS: Thirty rabbits were equally divided into five groups (groups A, B, C, D, and E) and the bladder mucosa of every rabbit was injured by electrocautery, cryocautery and incision on the left lateral wall, right lateral wall and posterior wall respectively. Then different drugs were instilled into bladders. Group A: pure phosphate-buffered sodium instillation (PBS); group B: PBS and radiolabeled BCG (3H-BCG) instillation; group C and group D: epsilon-aminocaproic acid or para-aminomethyl benzoic acid and 3H-BCG instillation; group E: heparin and 3H-BCG instillation. After instillation, every injured bladder wall and non-injured wall (anterior wall of bladder) were surgically removed and digested. The quantity of BCG adherence was determined by liquid scintillation counter. RESULTS: The quantity of BCG attached to injured bladder wall was significantly higher than that of non-injured wall (P < 0.001). The mean level of group C or group D was significantly higher than that of group B (P < 0.001) while the mean level of group E was significantly lower than that of group B (P < 0.001). CONCLUSION: For experimental rabbits, intravesical instillation of fibrin clot stabilizer EACA or PAMBA can enhance BCG attachment to the bladder wall while fibrin clot inhibitor heparin inhibits it.

[Effect of human placenta derived mesenchymal stem cells on cord blood lymphocyte transformation].

OBJECTIVE: To study the effect of human placenta derived mesenchymal stem cells (MSCs) on the immune function of lymphocytes derived from human umbilical cord blood. METHODS: Mononucleated cells (MNC) were isolated from human placenta tissue perfusate by density gradient fractionation. Individual colonies were selected and cultured. The culture-expanded cells were characterized by immune phenotyping so as to identify the MSCs. The MSCs were cultured under conditions promoting differentiation to osteoblasts or adipocytes. MNCs were isolated from adult peripheral blood and human umbilical cord blood and cultured, then the adherent cells were excluded and the suspended cells, lymphocytes, were inoculated in the culture fluids of MSCs of different concentrations and phytohemagglutinin (PHA), a nonspecific mitogenic stimulant, was added for 84 hours (MSC + PHA groups), then (3)H-thymidine deoxyribose ((3)H-TdR) was added for 12 hours. The cells were collected and scintillation counter was used to calculate the counts per minute (cpm). Pure lymphocytes without MSC and stimulated by PHA were used as control group (non-MSC Group) and pure lymphocytes and pure MSCs without PHA were used as blank control groups (non-PHA Group). RESULTS: From human placenta MSCs were successfully isolated and exhibited fibroblast-like morphology. Flow cytometric analysis showed that the placental MSCs were a homogeneous cell population devoid of hematopoietic cells positive for CD29, CD44, CD73, CD105, CD166, and HLA-ABC positive and negative for CD34, CD45, and HLA-DR. They could be induced into adipocytes or osteocytes. The cpm value of the non-MSC Group was 171 855 +/- 31 454, significantly higher than that of non-PHA Group (26 453 +/- 5268). The cpm values of the different concentrations MSC + PHA groups were all significantly lower than that of non-MSC Group in a dose-dependent manner; when the dose of MSCs was 2 x 10(5) the suppression rate was 79.97% in PB and 64.06% in UCB. CONCLUSION: MSCs derived from postpartum human placenta, an important and novel source of multipotent stem cells, suppress blood lymphocyte proliferation, thus may be used to reduce graft -versus-host disease (GVHD) in recipients.

Permeability of arachidonic acid metabolites through the round window membrane in chinchillas.

Our previous studies showed that samples of middle ear effusion contain high concentrations of arachidonic acid metabolites (AAMs); both prostaglandins (PGs) and leukotrienes (LTs). Since abnormal levels of AAMs in the perilymph seem to be associated with salicylate ototoxicity and the round window membrane (RWM) is the most likely route of entry from the middle ear to the inner ear, it would be important to know the permeability of AAMs through the RWM. Permeability of AAMs was determined by applying labeled and standard representatives AAMs on the RWM and counting radioactivity or measuring concentrations of AAMs in perilymph at different time intervals. Using chinchillas as experimental animals, samples of perilymph were obtained through a fenestration made at the helicotrema of the cochlea after 0.5, 1, 2, and 4, hours from placing tritiated or standard PGE2, 6-keto-PGF1 alpha, LTB4, LTC4 and 15-hydroxyeicosatetraenoic acid (HETE) over RWMs. Radioactivity in the perilymph was determined by a scintillation counter and concentrations of AAMs were measured by reverse phase high performance liquid chromatography (RP-HPLC) and radioimmunoassay. AAMs, which are twenty carbon fatty acids, were found to cross the RWM readily. Radioactivity and concentrations of AAMs in perilymph were detected in 30 minutes and peaked by one hour from the application of AAMs on the RWM. The results of this study suggest that AAMs such as PGs and LTs present in the middle ear fluid can cross the RWM readily and enter into the perilymphatic space of the cochlea. Sensorineural hearing loss observed in chronic otitis media could be mediated by PGs and LTs in the middle ear effusion crossing over to the inner ear through the RWM.

Acute effects of exogenous nitric oxide on glucose uptake in skeletal muscle of normoglycaemic and diabetic rats.

BACKGROUND: The role of nitric oxide (NO) in the modulation of basal and insulin-stimulated glucose uptake remains controversial. The aim of this study was to investigate the role of NO released from its donors, S-nitrosoglutathione (GSNO) and S-nitroso-N-acetylpenicillamine (SNAP), on glucose uptake in skeletal muscle of normoglyceamic and type 2 diabetic rats. MATERIAL/METHODS: Skeletal muscle strips of type 2 diabetic and normoglycaemic Sprague-Dawley rats were incubated with or without various concentrations (200 microM, 500 microM, 1000 microM, 10 mM & 20 mM) of SNAP or GSNO in the presence or absence of insulin (100 nM or 10 microM). The associated radioactivity was determined by liquid scintillation counting in a Beckman LS6000 scintillation counter programmed for dual-channel counting. RESULTS: SNAP and GSNO at 1000 microM significantly elevated basal and insulin-stimulated 2-deoxyglucose uptake (P<0.05) in normoglycaemic and diabetic rats. Millimolar concentrations (10 & 20 mM) of GSNO and SNAP significantly decreased basal and insulin-stimulated glucose uptake in skeletal muscle strips of normoglycaemic and type 2 diabetic rats in a concentration-dependent manner (P<0.05). The inhibition of insulin-stimulated glucose uptake was greater in the diabetic rats using both NO donors compared with normoglycaemic rats (P<0.05). CONCLUSIONS: The stimulatory effect of micromolar concentrations of GSNO and SNAP enhances basal and insulin-stimulated glucose uptake in normoglycaemic and type 2 diabetic rats, however higher concentrations elicited an inhibitory effect in normoglycaemic and diabetic rats. This highlights the NO-glucose uptake mechanism as a possible potential therapeutic target in the treatment of type 2 diabetes.

[Characteristics of adenine nucleotide translocator in mitochondria of rat cerebral cortex during hypobaric hypoxia exposure].

The purpose of the present study was to explore the effects of hypoxic exposure on mitochondrial adenine nucleotide translocator (ANT) activity and its characteristics. Male Wistar rats were exposed to hypoxia in a hypobaric chamber simulating high altitude at 5 000 m for 1, 5, 15 and 30 d. Control rats were fed outside the hypobaric chamber. Rats were sacrificed by decapitation and mitochondria from the cerebral cortex were isolated by differential centrifugation at each time point. The ANT activity was detected by the atractyloside (ATR)-inhibitor stop technique. Mitochondria was initiated by addition of (3)H-ADP and terminated after 12 s by quick addition of ATR. The radioactivity was measured in a liquid scintillation counter. Nonspecific binding of (3)H-ADP to mitochondria was estimated by incubation of mitochondrial samples with ATR prior to the addition of (3)H-ADP. This blank was substracted from the measured radioactivities. The activity of ANT was expressed as nanomoles (3)H-ADP per minute per milligram protein. The ANT density was determined by titrating the rate of state 3 respiration with increasing concentrations of carboxyatractyloside (CAT). Mitochondria were pre-incubated with CAT in a respiratory medium before ADP addition to initiate state 3 respiration. Plots of O2 consumption versus CAT appeared biphasic with an increasing inhibitory segment followed by a steady respiration, indicating that state 3 respiration was completely inhibited. The density of ANT was determined by the amount of CAT required to completely inhibit state 3 respiration, assuming a 1:1 binding stoichiometry, which was expressed as ANT density per milligram mitochondria protein. (ATP+ADP) in mitochondria was measured by high performance liquid chromatography (HPLC). The results showed that there was an obvious decrease in the ANT activity during hypoxic exposure. The lowest ANT activity was seen in 5 d group. Partial recovery of ANT activity was observed in 15 and 30 d groups, but ANT activity was still lower than that in the control group (P<0.01). Compared with that in normoxic control group, no change of ANT density in mitochondria was observed in hypoxia group (P>0.05). The turnover number of ANT in control, 1, 5, 15, 30 d groups were 16.67, 1.90, 0.40, 1.81, 4.73 pmol ADP/(min.pmol ANT), respectively. However, (ATP+ADP) in mitochondria in 1, 5, 15, 30 d groups were 63.37%, 48.44%, 52.73%, 60.16% of control group respectively. Therefore, the turnover number of energy production and expenditure were reduced. These observations suggest that the change of ANT activity may be one of the mechanisms of cellular oxidative phosphorylation dysfunction during hypoxic exposure.

[Effects of Bushen Tiaochong Recipe contained serum on proliferation and secretion of ovarian granulocyte cultured with excessive androgen].

OBJECTIVE: To observe the effect of Bushen Tiaochong Recipe (BTR) on proliferation and secretion of ovarian granulocyte (OGC) cultured with excessive androgen by serum pharmacologic technique. METHODS: Follicular stimulating hormone (FSH) contained and BTR contained (high, moderate or low dose) serum prepared on 6-week-old Sprague Dawley rats by serum pharmacologic method were respectively added into cultured OGC cell-line after treatment with testosterone propionat (TP). Then the cells were incubated for 48 hrs. The quantity of 3 H-thymidine incorporated DNA was determined with a scintillation counter; the cell cycle distribution and proliferation index (PI) were estimated with flow cytometer (FCM); and the contents of estrodial and progestogen in culture fluid and intracellular cAMP were detected by radioimmuoassay. RESULTS: Excessive androgen obviously inhibited the proliferation and secretion of OGC, and reduced the intracellular cAMP. BTR contained serum could improve above-mentioned inhibition of androgen to promote the cell proliferation and secretion of E2 and P in dose-dependant manner. CONCLUSION: BTR could alleviate the inhibitory action of excessive androgen on OGC, the ovarian function may be adjusted by BTR through promoting follicular development, increasing the levels of estrogen and progestogen, improving the ratio of estrogen/androgen.

[Effect of three flavones on enzyme activity of recombinant human phosphoinositide 3-kinase p110beta catalytic subunit].

OBJECTIVE: To study the effect of three flavones-luteolin, apigenin and genistein on activity of recombinant human phosphoinositide 3-kinase (PI3-K) p110beta catalytic subunit. METHODS: Recombinant human P13-K p110beta catalytic subunit was expressed by gene engineering. PI3-K activity was assayed by incubation recombinant PI3-K p110beta with phosphatidylinostiol-4,5-bisphosphate and [gamma-32P] ATP; the 32P-radiolabeled lipids were extracted with cholroform and methanol, and assessed by scintillation counter. RESULTS: Luteolin and apigenin showed inhibition on the recombinant p110beta catalytic subunit with IC50 8. 65 micromol/L and 11.56 micromol/L, but genistein had no inhibition. CONCLUSION: Luteolin and apigenin are inhibitors of P13-K. The recombinant P13-K p1100 catalytic subunit may be used as a molecular target for simpler screening and development of more effective inhibitors of P13-K.

Tritium metabolism in man.

Tritium metabolism in human beings was studied in volunteers who had exposure to tritiated water accidentally, by measuring the organically bound tritium with liquid scintillation counter, in sperms and plasma proteins. 2% of the initial urinary tritium specific activity was incorporated as bound tritium in sperms. In plasma proteins, on the 20th day of exposure, tritium bound in globulin was 3 times higher than that of albumin, tritium bound in globulin was 3 times higher than that of albumin.

[The evaluation of the adhesive properties of dental materials by using radioactive-labelled bacteria].

Dental materials with different surfaces were incubated with 111In-oxine-labelled bacteria Streptococcus mutans 20 min respectively 2 h, 6 h and 24 h. To determine the amount of the bacteria accumulated on the specimens we measured the radioactivity with a scintillation counter. The same measuring was done after decontamination by the help of ultrasonication. The comparison of activities shows that different dental material surfaces adhere different amounts of bacteria. The adhesive properties are material and time dependent, but the surface processing has a lower influence on the bacterial adherence. The lowest degree of bacterial adherence has the cobalt base alloy Gisadent KCM 83. On the other hand the highest remaining activity was found after decontamination of cobalt base alloy and PMMA resin Kallocryl A.

Hemodynamic changes after hepatectomy in rats studied with radioactive microspheres.

It has been difficult to measure hepatic arterial blood flow and portal venous flow simultaneously, especially in small animals. Radioactive microspheres were used in this experiment to quantitate splanchnic hemodynamics after hepatectomy in rats. With a reference sample technique, a certain amount of radioactive microspheres was injected into the left ventricle. The reference sample was withdrawn from the femoral artery at a constant rate. The animal was killed with a bolus of saturated KCl. The kidneys and splanchnic organs were removed and weighed. The radioactivity of each organ was determined using a gamma scintillation counter. Organ blood flow was calculated by the following formula: [formula: see text] Immediately after partial hepatectomy, a decreased cardiac index from 32.31 +/- 10.12 to 23.44 +/- 3.21 ml/(min x 100g body weight) (p less than 0.05), decreased hepatic arterial blood flow from 0.40 +/- 0.12 to 0.33 +/- 0.03 ml/(min x g liver) (p less than 0.05), increased portal venous inflow from 0.90 +/- 0.30 to 2.20 +/- 0.26 ml/(min x g liver) (p less than 0.05) and increased total hepatic blood flow from 1.30 +/- 0.39 to 2.53 +/- 0.26 ml/(min x g liver) (p less than 0.005) were observed. With an intrasplenic injection of an additional amount of radioactive microspheres, the ratio of lung/(lung + liver) radioactivities indicated the degree of portal systemic shunt (PSS). Though the portal pressure was elevated after hepatectomy (8.80 +/- 0.7 vs 11.9 +/- 1.7 cm H2O, p greater than 0.05), the extent of PSS was negligible (0.02 +/- 0.01% vs 0.03 +/- 0.01%, p greater than 0.05). The radioactive microspheres with the reference sample technique is a simple, rapid, reliable and reproducible method for investigating the hemodynamic changes following partial hepatectomy.

Uptake of metallic mercury and mercuric ion by human erythrocytes.

Uptake of metallic mercury (Hg degrees) and mercuric ion (Hg2+) by erythrocytes was studied by incubating erythrocytes with various concentrations of radioactive metallic mercury and mercuric ion in phosphate-buffered saline (pH 6.8) or plasma at 25 degrees C for 30 min. Radioactivity taken up in the cytosol (endsome) and stroma were determined with a gamma scintillation counter. The radioactivity ratio of the mercury recovered in the cytosol fraction to metallic mercury incubated in the saline was significantly higher than the ratio of that to mercuric ion. Similar findings were observed in erythrocytes incubated with metallic mercury and mercuric ion in plasma, although the recovered radioactivity of mercury in the cytosol of erythrocytes incubated with metallic mercury or mercuric ion in plasma was less than that incubated in phosphate-buffered saline. Thus, erythrocytes incubated with metallic mercury took up a larger amount of mercury than those incubated with mercuric ion. Discussion is made on these findings.

[Effect of monoclonal antibody against methylcholanthrene-induced cytochrome P-450 forms on benzo(a)pyrene metabolism in hepatic microsomes of C57BL/10 mice].

In this study, hepatic microsomes from 5,6-benzoflavone induced C57BL/10 mice were used. To inhibit monooxygenase activities, the monoclonal antibody MAb 1-7-1 recognizing two isoenzymes of methylcholanthrene-induced cytochrome P-450 was applied. Microsomes were incubated with tritium labeled benzo(a)pyrene [G-3H]BP for 10 min at 37 degrees C. The incubation mixture contained: 50 mM potassium phosphate buffer, pH 7.25; 30 mM KCl; 3 mM MgCl2; 2 mM NADPH; 80 microM [G-3H]BP (specific activity 50 mCi/mmol); and monoclonal antibody MAb 1-7-1 or ascites fluid (NBS) containing nonspecific IgG as a control. The ratio of antibody protein/microsomal protein was 2:5. BP metabolites were extracted from incubation mixtures by ethyl acetate. The organic layer was dried over sodium sulfate, and evaporated under a stream of nitrogen. To separate BP metabolites HPLC technology was used. The column was eluted with methanol gradient (60-100%) for 45 minutes. The radio-activity of collected samples was determined using liquid scintillation counter. Differential inhibitory effects of MAb 1-7-1 on BP-metabolites formation were found, e.g. 7,8-diol was inhibited by 86.1% and quinones by 62.5%. The predominant metabolite, 3-OH-BP, was inhibited by 80.4%. Moreover, it was found that MAb 1-7-1 inhibition of benzo(a)pyrene hydroxylase activity (by 75.8%, as measured by fluorescent technique) was very similar to the inhibition of 3-OH-BP along with 9-OH-BP formation (as measured by HPLC).
