Inhibitory effects of mesoionic 3-aryl substituted oxatriazole-5-imine derivatives on vascular smooth muscle cell mitogenesis and proliferation in vitro.

1. The effects of oxatriazole-type (GEA 3162 and GEA 5624) nitric oxide (NO) donors on mitogenesis and proliferation were studied in vascular smooth muscle cell (VSMC) culture. The effects of the GEA-compounds were compared with well-known NO-donors 3-morpholinosydnonimine (SIN-1) and S-nitroso-N-acetylpenicillamine (SNAP). 2. All NO-donors released NO and increased the production of cyclic GMP concentration-dependently. The production of cyclic GMP was inhibited by the guanylate cyclase inhibitor, ODQ (1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one). 3. The NO-donors inhibited basal and serum-induced DNA synthesis concentration-dependently. The GEA-compounds were needed in concentrations 10 times lower than SIN-1 and SNAP. GEA 3162, SIN-1 and SNAP were also able to inhibit serum-induced cell proliferation. GEA 5624 was ineffective. The antimitogenic effect of NO-donors was not reduced by inhibiting the guanylate cyclase. 4. These results suggest that NO inhibits serum-induced DNA synthesis and proliferation of VSMC by a cyclic GMP-independent mechanism. The oxatriazole-type NO-donor GEA 3162 was found to be a more potent inhibitor of mitogenesis and cell proliferation than SIN-1 and SNAP.

Lewis base effects in the Baylis-Hillman reaction of imines with cyclohex-2-en-1-one and cyclopent-2-en-1-one.

In the Baylis-Hillman reaction of N-benzylidene-4-methyl-benzenesulfonamide with cyclohex-2-en-1-one or cyclopent-2-en-1-one, we found that, in the presence of a catalytic amount of DMAP, the Baylis-Hillman reaction can be greatly accelerated to give the normal Baylis-Hillman adduct 1 or 3 in good or very high yields: moreover, using PBu3 as a Lewis base in the reaction of N-benzylidene-4-methylbenzenesulfonamide with cyclopent-2-en-1-one, the normal Baylis-Hillman adducts 3 could be obtained in very high yields within 5 h, however, using PBu3 or DBU as a Lewis base in the reaction of N-benzylidene-4-methyl-benzenesulfonamide with cyclohex-2-en-1-one, beside the normal Baylis-Hillman adduct 1 abnormal Baylis-Hillman adduct 3-aryl-2-[(4-methylphenyl)sulfonyl]-2-azabicyclo[2.2.2]octan-5-one 2 was formed at the same time; the substituent's effects were also examined.

Aza-pinacol rearrangement: acid-catalyzed rearrangement of aziridines to imines.

A series of di-, tri-, and tetra-substituted N-tosylaziridines [N-(toluene-p-sulfonyl)aziridines] 1, prepared by aziridination of the corresponding alkenes with N-[(tolyl-p-sulfonyl)imino]phenyliodinane (TsN = IPh), was found to undergo a BF3-catalyzed rearrangement (aza-pinacol rearrangement) under mild conditions to give the corresponding N-tosylimines 2 generally in satisfactory yields.

Ring forming reactions of imines of 2-aminobenzaldehyde and related compounds.

By addition of organometallic reagents to 2-aminobenzonitriles followed by quenching with suitable electrophiles (acyl halides, aldehydes and ketones), several types of 6-membered benzofused aromatic and non-aromatic nitrogen heterocycles could be obtained. Rearrangements leading to 1,2-dihydro-3H-1,4-benzodiazepin-3-ones and preparation of various quinazolines are described.

Origin of 1,3-induction in the addition of alkyl lithium to imines bearing an N-stereogenic center.

The origin of diastereoselectivity in the addition of alkyl lithium to chiral Schiff bases has been investigated experimentally and theoretically and the formation of the major diastereomer can be explained from the energy minimized structure of the Schiff base in which the phenyl group has been found to orient in such a manner that it posed lesser steric hindrance to the incoming nucleophile as compared to the alkyl group.

The first examples of metal-mediated addition of a phosphorus imine to nitriles; the preparation and X-ray crystal structures of [PtCl4{NH=C(Et)N=PPh3}2] and [PtCl2(EtCN){NH=C(Et)N[double bond, length as m-dash]PPh3}].

Imino(triphenyl)phosphorane, Ph3P=NH (1), reacts with nitrile complexes of Pt(IV) to generate hydrolytically sensitive [PtCl4{NH=C(R)N=PPh3}2](R=Me 2a, Et 2b, Ph 2c), and with the Pt(II) complex [PtCl2(EtCN)2] to give [PtCl2(EtCN){NH=C(Et)N=PPh3}](3) and [PtCl2{NH=C(Et)N=PPh3}2](4); X-ray crystallography performed upon (2b) and (3) confirms the presence of an imine/nitrile addition ligand bound by the terminal nitrogen.

Direct synthesis of (imine)platinum(II) complexes by iminoacylation of ketoximes with activated organonitrile ligands.

The metal-mediated iminoacylation of ketoximes R1R2C=NOH (1a R1 = R2 = Me; 1b R1 = Me, R2 = Et; 1c R1R2 = C4H8; 1d R1R2 = C5H10) upon treatment with the platinum(II) complex trans-[PtCl2(NCCH2CO2Me)2] 2a with an organonitrile bearing an acceptor group proceeds under mild conditions in dry CH2Cl2 to give the trans-[PtCl2{NH=C(CH2CO2Me)ON=CR1R2}2] 3a-d isomers in moderate yield. The reaction of those ketoximes with trans-[PtCl2(NCCH2Cl)2] 2b under the same experimental conditions gives a 1 : 1 mixture of the isomers trans/cis-[PtCl2{NH=C(CH2Cl)ON=CR1R2}2] 3e-h and 4e-h in moderate to good yield. These reactions are greatly accelerated by microwave irradiation to give, with higher yields (ca. 75%), the same products which were characterized by IR and 1H, 13C and 195Pt NMR spectroscopies, FAB-MS, elemental analysis for the stable trans isomers, and X-ray diffraction analysis (3f). The diiminoester ligand in 3a was liberated upon reaction of the complex with a diphosphine.

Comparative evaluation on mouse nasal immunogenicity of arylmethane-, xanthene-, quinone-imine-, and acridine-dye-inactivated Sendai virus vaccines.

Twenty-seven kinds of organic dye-inactivated Sendai virus vaccines were prepared by treatment in dark at 23 C for 2 months or more, and selected with the high HA titers as a guide. Their nasal immunogenicities were examined in mice by contact infection and immunofluorescent method, and the relative merits of the dye-inactivants were determined. The strongest protection was elicited with acriflavine-, auramine O-, eosin Y-, neutral red-, night blue-, patent blue V-, thymol blue-, uranin-, and xylene cyanol FF-treated vaccines. Middling protective efficacy was induced by use of erio green B-, malachite green-, methyl green-, proflavine-, pyronin B-, and thionin-inactivated vaccines. Dye-inactivated vaccines that resulted in the weakest protection were Bindschedler's green-, bromothymol blue-, erythrosin B-, ethyl violet-, gallein-, light green SF yellowish-, methyl violet-, new methylene blue N-, phenol red-, rhodamine 6G-, spirit blue- and victoria blue B-treated ones. Serum HI titers developed by nasal vaccination were variable, and rose still more in most vaccinated groups postexposure. Elicitation of the most effective nasal immunogenicity in dye-inactivated vaccines appeared to depend on selective modification of capsid protein or ribose in viral core with dyes possessing definite functions, despite the different molecular structures.

Solubilization of spermidine-sensitive (+)-[3H]5-methyl-10,11-dihydro-5H-dibenzo[a,d]cyclohepten-5,10-imine ([3H]MK-801) binding activity from rat brain.

The receptor-ionophore complex of the N-methyl-D-aspartate (NMDA)-sensitive receptor was solubilized by deoxycholic acid from rat brain using (+)-[3H]5-methyl-10,11-dihydro-5H-dibenzo[a,d]cyclohepten-5,10-imi ne ([3H]MK-801) binding as a marker for the receptor. Gel filtration of the solubilized preparations on a Sephadex G-25 column revealed significant [3H]MK-801 binding sensitive to potentiation by glutamate and glutamate/glycine, which was prevented by competitive antagonists for the NMDA and strychnine-insensitive glycine (GlyB) sites. In contrast to NMDA and glycine, spermidine markedly potentiated the amount of [3H]MK-801 binding in solubilized preparations by increasing the apparent affinity of the ligand. In the presence of all three stimulants, the solubilized preparations exhibited pharmacological profiles similar to those in the membrane preparations. These results clearly indicate that the whole macromolecular NMDA receptor-ionophore complex is solubilized under the experimental conditions used.
