Matrix compare analysis discriminates subtle structural differences in a family of novel antiproliferative agents, diaryl-3-hydroxy-2,3,3a,10a-tetrahydrobenzo[b]cycylopenta[e]azepine-4,10(1H,5H)-diones.

A diastereoselective synthesis of diaryl-3-hydroxy-2,3,3a,10a-tetrahydrobenzo[b]cycylopenta[e]azepine-4,10(1H,5H)-diones is described employing a tandem Michael-aldol addition as key step. The novel compounds exhibit antiproliferative activity in a panel of in vitro cultivated cancer cell lines. The bioinformatic tool COMPARE was able to discriminate between two closely related subgroups of the title compounds, namely 1,3- and 2,3-disubstituted derivatives.

The serological clostridium tumour test--application of discriminance analysis.

The microbiological tumour test is based on the detection of humoral antibodies to clostridial rods following intravenous injection of apathogenic clostridial spores. As serological methods we used the complement fixation test with a simultaneous short-time warm and long-time cold incubation as well as the passive haemagglutination test after absorption of clostridial group-specific antibodies. The evaluation of titre dynamics was rendered difficult by normal clostridial antibodies, by serological cross-reactions and, above all, by border-line reactions. The serological test results from 189 dogs investigated were evaluated in a multivariate analytical procedure. We determined the adequate combination of methods and antigens, thus improving the reliability of the test results and objectifying the predictive value of low sero-positive titres. On the basis of the computer-calculated individual weighting factors and the corresponding reference value it has become possible to evaluate and classify actual serological data emerging from the previous overall investigation.

Discriminative analysis of rat Sertoli and peritubular cells and their proliferation in vitro: evidence for follicle-stimulating hormone-mediated contact inhibition of Sertoli cell mitosis.

A new methodological approach using immunohistochemical markers for Sertoli cells (alpha inhibin), peritubular cells (alpha smooth muscle actin), and S-phase cells (bromodeoxyuridine; BrdU) is presented that allows an accurate and simultaneous analysis of morphogenetic and mitogenic changes occurring in vitro. Sertoli cells and peritubular cells were isolated by sequential enzymatic digestion from 7-day-old rats. Laminin, as a major component of the extracellular matrix of the seminiferous tubule, and FSH, as a hormone stimulating Sertoli cell proliferation, were tested for their ability to influence the morphology or mitotic activity of the cultured cells. After fixation, the coverslips were stained for these antigens with use of specific primary antibodies and horseradish peroxidase- or alkaline phosphatase-labeled secondary antibodies for visualization of the respective antigens with different-colored precipitates. This approach allowed us to distinguish the two cell populations, which could not be done unequivocally without the antibody staining. We scored striking changes in cell densities and cell ratios during the culture period. Peritubular cells showed a consistently higher BrdU-labeling index than Sertoli cells. While Sertoli cells were not labeled until Day 7, peritubular cells proliferated as soon as on Day 3, and their density doubled from Day 3 to Day 7. A linear negative correlation was established for Sertoli cell proliferation in response to their local density on the coverslip, indicating contact inhibition as a signal for cessation of mitosis. At high cell densities, this inhibition was partially overcome in the presence of FSH. The presence of laminin had striking effects on the morphogenetic response but only a minor influence on mitogenesis.

Exploring the dorsal surface of hominoid brain endocasts by stereoplotter and discriminant analysis.

One of the more vexing problems with hominoid endocasts has been to secure reliable information that goes beyond their volumes. One method is explored here, where a large number (N = 171) of radial distances from a homologous internal central point to the dorsal endocast surface are measured in a polar coordinate system. From two pilot studies, one with a hominoid sample of N = 64, and the other with an enlarged sample of N = 92, the following results can be mentioned tentatively: (1) there are residual data that differ taxonomically in different cortical regions once overall endocast size is corrected in allometric fashion; (2) the major cortical regions where these differences appear most strongly are in the lower parietal lobule, anterior occipital zone, and the dorsoanterior region of the frontal lobe; (3) the method shows excellent promise in objectively and quantitatively depicting taxa-specific shape differences in functionally understood cortical regions through multivariate statistical analyses.
