[Spatial pattern of Tetranychus urticae population in apple tree garden].

Spatial pattern of insect population is an important aspect of insect ecology. It reveals the spatial structure characteristics of populations, and provides a basis for relevant sampling techniques and data transformation. Tetranychus urticae Koch is a serious pest of apple tree. Its spatial pattern and time-series dynamics were analyzed by calculating four indices of aggregation, and parameters of Iwao model. The results showed that the spatial pattern of T. urticae in apple tree garden belonged to an aggregated distribution pattern. The aggregation intensity was the highest in upper and southern part of crown, and the lowest in its lower and inner part. The aggregation was due to the biological habits of T. urticae and environmental heterogeneity. No matter T. urticae was in the upper, middle, or lower crown of apple tree, its aggregation intensity was higher before first ten days of June, and then gradually decreased after the middle ten days of June.

Evaluation of the immunomodulatory activity of Aronia in combination with apple pectin in patients with breast cancer undergoing postoperative radiation therapy.

The aim of the present study was to evaluate the immunomodulatory activity of Aronia in combination with apple pectin in patients with breast cancer in the course of postoperative radiation therapy. Monoclonal antibodies were used to assay specific T cell subsets. Tests were performed prior to and after 26 and 50 Gy of irradiation. The study comprised 42 women (19 to 65 years of age) receiving 15 g of apple pectin in combination with 20 ml of Aronia concentrate (Bioactive Substance Laboratory--Plovdiv) twice daily during postoperative irradiation. Irradiation was performed by a 60Co-Rokus according to individualized treatment schedules. The following T lymphocyte populations were tested--CD3 total T lymphocytes, CD4 helper and inducer T cells, CD8 suppressor and cytotoxic T cells and NK cells. The levels of the polypeptide tissue antigen (TPA), an oncofetal protein, were tested in parallel. The TPA was used to assess treatment outcome in our patients. A group of 25 age-matched women with breast cancer served as controls. Immune status analysis of controls was performed prior to and following postoperative radiation. A total of 880 serum samples were tested. Assays of immunity parameters in the patients receiving Aronia in combination with apple pectin showed that CD4 and CD8 T cell counts increased significantly (P < 0.01 and P < 0.05 respectively). In control patients CD3 T cell levels lowered, the other T cell levels remained unchanged. Initially the number of NK cells was increased in both groups of patients. It remained constant throughout the course of the study. The normal levels of TPA in both groups of patients indicated a good treatment outcome due to the adequacy of surgery and in combination with radiation therapy.

Antioxidant properties of apple juice and its protection against Cr(VI)-induced cellular injury.

Reactive oxygen species (ROS) have been shown to cause a broad spectrum of damage to the biological system. ROS-mediated reactions are believed to play a key role in Cr(VI)-induced carcinogenesis. Using electron spin resonance (ESR) spin trapping, the present study shows that apple juice efficiently scavenged hydroxyl radicals generated by Cr(VI) reduction catalyzed byglutathione reductase/NADPH. Apple juice reduced Cr(VI)-induced lipid peroxidation, DNA damage, cell apoptosis, and NF-kappaB activation in human lung epithelial A549 cells. The lipid peroxidation was measured by a colorimetric assay, DNA damage by single cell gel electrophoresis assay, induction of cell apoptosis by flow cytometry, and activation of NF-kappaB by luciferase assay. The results show that through its antioxidant properties, apple juice can protect Cr(VI)-induced cellular injury and may help reduce its carcinogenic potentiaL

[Cystogenous nematode, Heterodera (Globodera) mali sp. n., an apple parasite in Kazakhstan].

There has been no information on cystogenous nematodes of the genus Heterodera injuring apple trees. The paper checks the first finding of these nematodes on the roots of a cultivated apple tree. The nematodes proved to be a new species, Heterodera mali sp. n. The apple cystogenous nematode resembles the potato cystogenous nematode H. rostochiensis Woll. in having a round body, small cleft of the vulva and similar sizes of eggs. It differs from H. rostochiensis in smaller sizes (on the average 5 against 9) very transparent and thin cuticle, a small distance between vulva and anus (on the average 43 against 86), a smaller value of the Granek's index (on the average 1.3 against 4.6), a smaller number of cuticular folds between vulva and anus (on the average 7 against 21) and different plant-hosts. A detailed description of H. mali sp. n. is given.

[Desiccation and nitrate accumulation of apple orchard soil on the Weibei dryland].

The study of apple orchard soil on the Weibei dryland showed that high input and high yield not only resulted in soil desiccation, but also induced nitrate accumulation in the soil profile. Generally, the soil layers with accumulated nitrate located in 40 - 260 cm depths, and the maximum nitrate content was 403.4 mg x kg(-1). Owing to the continuous application of nitrogen fertilizer, the accumulated nitrate could hardly be reused in the future, which might lead to environmental pollution. Because the soil storage water could not adjust the water utilization of apple trees, the yield of orchard was affected by precipitation. Therefore, the nitrogen application rate should be reduced, and actions should be taken to improve soil moisture in apple orchard.

[Changes in antioxidative system and cell ultrastructure in the fruit peels of apple during sunburn development].

Fruits from 6-year-old apple trees (Malus domestica Borkh cv. Fuji) were used as materials to test the changes of antioxidative system and cell ultrastructure of fruit peels during sunburn development. Fruit sunburn appeared in mid July, 2002. The process of apple fruit sunburn was divided into three phases (0 degrees : control): 1 degrees : bleaching, 2 degrees : brownness, and 3 degrees : necrosis. Fuji apple fruits in different sunburn state were picked. Phenolic compounds, membrane protective enzymes (SOD, POD, PPO, CAT), cell membrane lipid peroxidation and cell ultrastructure in fruit peel were studied. The result showed that the degree of cell membrane lipid peroxidation enhanced along with development of sunburn. The activity of membrane protective enzymes also increased remarkably. However, the cell structure kept its integrity, except some organelles which partly disassembled, and cytoplasm and vacuoles became enriched with electron-dense substances while fruit peels became pale. As peel became brown, chlorogenic acid, quercetin, rutin and myricetin accumulated, and cells of outer layers of the epidermis collapsed correspondingly, cell wall became thicker. It is suggested that changes in both cell ultrastructure and antioxidative system confirm that physiological state of fruit peels becomes disordered during sunburn development.

[Effects of NaCl stress on Ca, Mg, Fe and Zn contents of different apple organs].

The study with two-year-old potted apple tree (Fuji variety with M. hupehensis Reld stock) showed that under NaCl stress, the average contents of Ca, Mg, Fe and Zn in different apple organs sampled at 4 periods had no significant changes, while the content ratios of test elements to Na decreased significantly, especially under high NaCl (3 per thousand) stress, which would result in the unbalance of mineral nutrients in the tree. The Ca content of different apple organs under and without NaCl stress was in order of trunk phloem > leaf and vegetative shoot > root > trunk xylem; Mg content was vegetative shoot and root > trunk xylem and phloem, and leaf; Fe content was root > leaf > trunk phloem and vegetative shoot > trunk xylem; Zn content was vegetative shoot > leaf > root and trunk phloem > trunk xylem. The element contents under NaCl stress showed different degrees of fluctuation, comparing with the control.

[Mapping of apple Co gene using SSR markers].

Columnar apple is an important genetic resource for tree form breeding of apple. In this study, 106 individuals of the F1 population derived from 'Spur Fuji' (coco)x 'Telamon'(Coco) were used as plant materials for screening SSR markers linked to gene. By bulked segregating analysis (BSA), eight SSR markers from the tenth linkage group of apple genome were tested. Finally, three of them, COL, CH02a10 and CH03d11, were identified to be SSR markers of Co gene. Linkage analysis showed that the genetic distance of COL, CH02a10 and CH03d11 to Co locus was 15.3cM, 22.2cM and 3.9 cM, respectively. On the linkage map of these markers, Co gene was located between COL and CH03d11.

[Diminution of blood and hepatic cholesterol induced by an apple-supplemented diet in the hamster. Trials in man].

The effects of an apple-supplemented diet on plasma and liver cholesterol were investigated in two strains of Hamster, a normal one and another which exhibits a spontaneous accumulation of cholesteryl esters in the liver (FEC animals). Adding of apples to the standard diet promoted a 20 % decrease of cholesterolemia in normal animals and normalized the high level of plasma cholesterol in the FEC animals. Similarly, the cholesterol content of the liver was diminished. The levels of cholesteryl esters were strongly lowered, threefold and tenfold respectively in normal and in FEC animals. Incorporation of labelled mevalonate into cholesterol was increased, but esterification of the neosynthesized sterol was markedly reduced in the liver from FEC animals. In human daily ingestion of apples caused a significative reduction (16 %) of cholesterolemia.

[Changes in amino acid and fatty acid contents as well as activity of some related enzymes in apple fruit during aroma production].

Aroma volatiles from apple (Malus domestica Borkh. var. Starkrimson) fruit at different stages of maturity were collected by solid adsorbent-Tenax-GC and determined by thermodesorption and GC-MS. Production of propyl acetate, butyl acetate, ethyl 2-methyl-butanoate and total ester volatiles and changes in concentration of the precursors of aroma biosynthsis--free amino acids and fatty acids and activities of lipoxygenases (LOX) and alcohol acetyltransferase (AAT) in apple fruits during ripening were studied. The results showed that propyl acetate and total esters were very low when the endogenous ethylene formation of the fruit was very low. At the stage of the increase in ethylene production, the rate of formation of propyl acetate and total esters increased. Butyl acetate appeared at the beginning of ethylene rise and increased thereafter. Ethyl 2-methyl-butanoate was produced at the beginning of climacteric stage and then increased sharply (Figs.1). These facts suggest that the aroma production is closely related to ethylene production. Among the 14 free amino acids detected in fruit, isoleucine which is considered to be the biosynthetic precursor of some branched chain esters showed a great increase during fruit ripening while the others decreased or remained stable (Table 1). The accumulation of isoleucine suggested that isoleucine supply in fruit may not limit the biosynthesis of esters with branched chain alkyl groups. Concentrations of free fatty acids such as palmitic, linolenic, oleic, linoleic, stearic acids increased before the increase of aroma production, decreased with the increase of aroma production and showed an increase at postclimacteric stages (Fig.2). LOX activity increased at climacteric stages and declined rapidly thereafter. AAT activity increased sharply at the early stage of fruit maturity when the aroma was very low and remained at a stable high level during fruit ripening (Fig.3) indicating that the AAT activity is not the limiting factor for aroma formation in apple fruit.

Latex-vegetable syndrome due to custard apple and aubergine: new variations of the hevein symphony.

An increasing number of vegetables with crossreactions to latex are being described in patients with latex-vegetable syndrome. We present two of these vegetables, custard apple linked in two previous cases with latex sensitisation, and aubergine, that had not been described up to now in patients with latex sensitisation. The diagnosis of both cases was based on the clinical history, positive skin prick test (SPT) and specific IgE to the offending vegetables, as well as to positive SPT and specific IgE levels to latex and the major fruits involved in the latex-fruit syndrome (avocado, banana, and chestnut). Further, crude extracts from latex, custard apple and aubergine, as well as the purified allergens Hev b 6.02 and Prs a 1 were used in in vitro and in vivo assays: IgE immunodetection, histamine release (HRT) and basophil activation (BAT) tests and skin prick tests. In case 1, both purified Hev b 6.02 and Prs a 1 induced positive responses in skin prick tests, high levels of basophil activation and histamine release. Specific IgE immunodetection uncovered a reactive band of 45 kd in the crude custard apple extract, which was also recognized by anti-chitinase monospecific antibodies. The serum from patient 1 also detected Prs a 1 in immunodetection. Hev b 6.02 produced positive skin responses and showed high biological activity in HRT and BAT in the case of patient 2. However, Prs a 1 was reactive neither in SPT nor in IgE immunodetection. In fact, no band was detected using the serum of patient 2 in avocado or aubergine extracts. By contrast, Prs a 1 reached high values of basophil activation and over 10% of histamine release in case 2.

[Study on predicting sugar content and valid acidity of apples by near infrared diffuse reflectance technique].

The nondestructive method for quantifying sugar content (SC) and available acid (VA) of intact apples using diffuse near infrared reflectance and optical fiber sensing techniques were explored in the present research. The standard sample sets and prediction models were established by partial least squares analysis (PLS). A total of 120 Shandong Fuji apples were tested in the wave number of 12,500 - 4000 cm(-1) using Fourier transform near infrared spectroscopy. The results of the research indicated that the nondestructive quantification of SC and VA, gave a high correlation coefficient 0.970 and 0.906, a low root mean square error of prediction (RMSEP) 0.272 and 0.056 2, a low root mean square error of calibration (RMSEC) 0.261 and 0.0677, and a small difference between RMSEP and RMSEC 0.011 a nd 0.0115. It was suggested that the diffuse nearinfrared reflectance technique be feasible for nondestructive determination of apple sugar content in the wave number range of 10,341 - 5461 cm(-1) and for available acid in the wave number range of 10,341 - 3818 cm(-1).

Determination of patulin in apple juice by liquid chromatography.

A method was developed and validated in-house for the determination of patulin (PAT), a toxic mold metabolite, in apple juice. The sample was extracted with ethyl acetate-hexane and analyzed by liquid chromatography equipped with a C18 column and diode array detector. The mobile phase used for the quantification was water-ethanol, at a flow rate of 0.5 mL/min. The method showed a mean recovery of 84.8%, the relative standard deviation obtained in the precision study was <7.7%, the quantification and detection limits were 7 and 3 microg/L, respectively, and the linear range for PAT in apple juice was 2.6-650 microg/L. The ruggedness was evaluated by an intralaboratory experiment, in which 5 factors were studied, and only one was found to influence the observed results. The developed method is fast, practical, and simple; the solvents (except hexane) and reagents used were nontoxic. The results of the validation confirmed the efficiency of the method, which is sensitive enough to be used in studies required to quantify PAT in apple juice.

Determination of daminozide in apples by gas chromatography/chemical ionization mass spectrometry.

A method using gas chromatography/chemical ionization mass spectrometry (GC/CIMS) for the determination of daminozide residues in apples has been developed. Daminozide was separated from the sample matrix by water extraction and cation exchange, converted to the methyl ester by treatment with HCl-methanol, and determined by GC/CIMS using succinonitrile as an internal standard. The detection level was 0.05 ppm. Recoveries were 92-104% from apples spiked at the 0.05-0.5 ppm levels. Of the 25 apple samples analyzed, only 2 were positive for daminozide (1.04 and 0.32 ppm).

Familial apple peel jejunal atresia: surgical, genetic, and radiographic aspects.

Fifty-seven cases of apple peel jejunal atresia have been reported in the English literature. Patients with this anomaly have a high incidence of prematurity (70%), malrotation (54%), short gut syndrome (74%), multiple atresias (15%), complications (63%), and mortality (54%). Mortality has decreased from 63% to 47% since 1970. We report three new cases of apple peel jejunal atresia, including two from one family. Radiographic evidence of high small bowel obstruction and a malrotated microcolon on preoperative roentgenogram with barium enema should suggest this diagnosis. Five families, including ours, have been reported in which more than one child was affected, and it has been suggested that this disorder is transmitted by an autosomal recessive gene. However, the occurrence of conventional intestinal atresia in other siblings, the association with multiple atresias, and discordance in a set of apparently monozygotic twins indicate that there may be a more complex spectrum of genetic transmission. Subsequent siblings are at increased risk for apple peel atresia or related malformations. Prenatal ultrasound can facilitate early diagnosis and treatment.

Gas-liquid chromatographic determination of fenvalerate insecticide residues in processed apple products and by-products.

Apples from trees treated in the field at 2-week intervals (9 foliar applications) with the synthetic pyrethroid insecticide fenvalerate (cyano(3-phenoxyphenyl) methyl 4-chloro-alpha-(1-methylethyl)-benzeneacetate) were processed into apple sauce, juice, pomace, and peels plus cores. Gas-liquid chromatographic analysis of the commodities for fenvalerate showed the sauce and juice to be essentially residue-free, the whole apples to contain about 0.4 ppm, and the pomace and peels plus cores to contain about 2 and 1.5 ppm, respectively. Agreement among 5 laboratories using modifications of the same basic method was good.

Liquid chromatographic method for rapid determination of total avermectin B1 and 8,9-Z-avermectin B1 residues in apples.

A liquid chromatographic method has been developed and validated for the rapid determination of avermectin B1 and 8,9-Z-avermectin B1 residues in apples. The avermectins are extracted from the crop matrix with an acetonitrile-water-hexane mixture; the extract is cleaned up on an aminopropyl solid-phase extraction column. The avermectins are derivatized with trifluoroacetic anhydride and analyzed by reversed-phase liquid chromatography with fluorescence detection. Recoveries of avermectins from apples fortified with about 2-77 ppb avermectin B1a or 2-27 ppb 8,9-Z-avermectin B1a averaged 85%. The limit of quantitation is 2 ppb (signal-to-noise [S/N] ratio, 12) and the limit of detection is 1 ppb (S/N ratio, 6) for each analyte. The assay is a simple, rapid, and sensitive method for monitoring the total amount of avermectin residues in apples.

Comparative fermentation behaviour and chemical characteristics of Saccharomyces and Zymomonas fermented culled apple juice.

Ethanol production from culled apple juice showed that fermentability of the juice could be enhanced by addition of DAHP or ammonium sulphate in Saccharomyces and DAHP in Zymomonas fermentation. Addition of trace elements inhibited both the fermentations and ethanol, consequently. With respect to by-products of fermentation, no clear advantage of Zymomnas fermentation of culled apple juice could be observed. Differences in physico-chemical characteristics of the fermented apple juice were also noted. Saccharomyces cerevisiae proved to be better than Zymomonas in most of the parameters and is preferrable from handling and spoilage point of view.
