[Effects of genistein on the expressions of cell cycle proteins in salivary adenoid cystic carcinoma cell line SACC-83].

PURPOSE: To investigate the molecular mechanism of cell cycle arrest induced by tyrosine protein kinase inhibitor, genistein, in human salivary adenoid cystic carcinoma cell line SACC-83. METHODS: SACC-83 cells cultured in vitro were treated with genistein, the expressions of CyclinB1, Cdk1, CyclinD1 and Cdk4 proteins were detected with Western blotting, and the results were quantitatively analyzed by FluorChem V2.0 software, statistical analysis was performed with analysis of variance using SPSS11.5 software. RESULTS: With the increase of concentration of genistein and the elongation of time, the expression of CyclinB1, Cdk1, CyclinD1 and Cdk4 proteins was significantly decreased. Treated with 220 micromol/L of genistein for 3 days, the expression level of CyclinB1, Cdk1, CyclinD1 and Cdk4 proteins was 58%, 64%, 46% and 43% of the control group, respectively (P<0.01). CONCLUSION: The cell cycle G2/M arrest induced by genistein in human salivary adenoid cystic carcinoma cell line SACC-83 may be associated with the downregulations of CyclinB1, Cdk1, CyclinD1 and Cdk4 protein expressions.

Primary adenoid cystic carcinoma of the esophagus. Report of a case and review of the literature.

A case of primary adenoid cystic carcinoma of the esophagus is reported and a total of 15 cases was reviewed. There were 9 men, although earlier reports indicated the disease was more common in women. The lesion was located in the middle third of the esophagus in 10 cases. The possibility of tumor development from embryonal rests of the tracheobronchial tree in the esophagus is discussed. The prognosis of adenoid cystic carcinomas is not well known, but the treatment and prognosis appear similar to those of epidermoid carcinomas of the esophagus.

[A clinical study of oral adenoid cystic carcinoma].

Author experienced a case of tubular type, and a case of solid & basaloid type in adenoid cystic carcinoma and presented these cases through the review of literature. The paralysis was found. Both cases showed perineural, muscular tissue and bony invasion. Respectively hemimaxillectomy, and subtotal maxillectomy were operated. The origin of adenoid cystic carcinoma was considered as myoepithelial cell combined with intercalated duct reserve cell.

[Long-term prognosis of adenoid cystic carcinoma of the trachea].

A study was made on the prognosis of 26 tracheal adenoid cystic carcinoma surgery cases reported in Japan between January 1978 and January 1984. The study was based on a questionnaire sent to doctors who reported prognosis as of May 1991. Twelve cases out of the 26 cases underwent postoperative irradiation. Seven out of the 12 cases survived 9 to 13 years after surgery without any recurrence. Two out of the 12 cases survived 8 and 10 years after surgery with multiple pulmonary metastases. Three out of the 12 cases died of a local recurrence 3, 3 and 9 years after surgery. Four cases received irradiation after a local recurrence was revealed postoperatively. Three out of the 4 cases survived 9, 10 and 11 years after surgery, but 1 out of the cases died 6 years after surgery. Four cases received no irradiation not only after surgery but also after detection of a local recurrence. All of them died within 9 years after surgery. There were six cases who underwent no postoperative irradiation and showed no local recurrence. One out of the 6 cases died of pancreas cancer 7 years after surgery, but the other cases survived 8 to 11 years after surgery without any recurrence. A study was also made on the prognosis of 9 cases with adenoid cystic carcinoma of the trachea who underwent surgery at Keio University and Tokai University until April 1982 (4 cases overlap with the above-mentioned cases). Six out of 9 cases received postoperative irradiation. Four cases survived 9 to 12 years after surgery without any recurrence.(ABSTRACT TRUNCATED AT 250 WORDS)

[Inhibitory effect of p53 gene on telomerase activity and proliferative activity in salivary adenoid cystic carcinoma cells].

OBJECTIVE: To evaluate the inhibitory effect of p53 gene on salivary adenoid cystic carcinoma (SACC) cells. METHODS: Adenoviral vector pDeltaE1-p53 was constructed and transfected into SACC-83 cells. The enhanced p53 expression was measured by reverse transcription polymerase chain reaction (RT-PCR), and the effects of transfected p53 on SACC-83 cells were analyzed by TRAP-PCR-ELISA, luciferase reporter, flow cytometry (FCM), soft agar assay and tumorigenicity test. RESULTS: The expression of p53 gene in SACC-83 cells was increased after introduction of pDeltaE1-p53. The telomerase activity and the transcriptional activity of hTERT promoter were inhibited. The cells cycles of transfected SACC-83 were arrested in G(1) phase and the rate of colony-formation was decreased, and similarly the tumorigenicity in nude mice was also decreased. CONCLUSIONS: The introduction of wild-type p53 by adenoviral vector could suppress the telomerase activity and malignant phenotypes of salivary adenoid cystic carcinoma cells.

[Adenoid cystic carcinoma and basal cell adenoma of the parotid gland: a compromised differential diagnosis].

The adenoid cystic carcinoma of the major and minor salivary glands grows slowly but spreads relentlessly into the surrounding tissues, in particular along the perineural spaces. It may appear clinically to be circumscribed, true encapsulation is lacking and the insidious invasion is demonstrated histologically. Another characteristic of the tumor is a high rate of metastases to distant organs such as lung, bobe or liver. The prognosis relatively poor despite surgical excision. The monomorphic basal cell adenoma of salivary glands is a rare tumor. Its diagnosis is, occasionally, difficult with adenoid cystic carcinoma on the clinical and pathology basis. The authors shown a method for this diagnosis consistent in a morphometric analysis of the neoplasic cells.

[Assessment of adenoid size in children by flexible fibrescope examination].

OBJECTIVE: To assess the adenoid size in children by flexible fibrescope examination and outline an effective therapeutic program according to the examination. METHOD: In this study, the flexible fibrescope examination of the nasal cavity and nasopharynx and a lateral skull radiograph examination were performed in 130 children with snoring and the same examinations were performed in another 30 children without snoring. RESULT: The size of adenoid was negatively associated with the degree of snoring. (P < 0.01). There is a high correlation between the flexible fibrescope examination and the lateral skull radiograph examination (r = 0.435, P < 0.01). CONCLUSION: The application of the flexible fibrescope examination of the nasal cavity and nasopharynx has proved to be of great clinical value, especially in the selection of children for adenoidectomy. It is an examination with minor invasion which can be well tolerated by most children, giving more information than a lateral skull radiograph and avoiding unnecessary radiation.

Adenoid cystic carcinoma manifesting as maxillary jaw pain refractory to conventional treatment: a case report.

Adenoid cystic carcinoma is an indolent, slow-growing tumor that may first cause low-grade pain in the affected region. This article describes a case involving adenoid cystic carcinoma of the maxilla that was present for approximately nine years. Prior to diagnosis, five dentists reported that the patient had anisocoria, migraine headaches, and low-to-moderate upper jaw pain that was refractory to conventional therapy. A surgical resection was performed; after a period of soft tissue healing, radiation therapy was initiated. The surgical defect was obturated using an interim removable prosthesis while awaiting final reconstruction by a maxillofacial prosthodontist. This article examines possible reasons why this lesion was not diagnosed sooner and discusses how this case should raise the general dentist's awareness of such lesions.

[An immunohistochemical study of keratin in adenoid cystic carcinoma of the salivary gland].

An immunohistochemical study of keratin was performed in forty-five cases of adenoid cystic carcinoma of salivary gland, the results were as follows: keratin was distributed in the duct system of normal salivary gland, but the acini were negative. The distribution of keratin were varied in different patterns of adenoid cystic carcinoma. The amount of synthesis of keratin was many in tubular type of high differentiated carcinoma, but, in basaloid solid type of low differentiated carcinoma, the synthesis of keratin was less; the content of keratin in cribriform and trabecular type was between the tubular type and basaloid solid type, and the former had more keratin than the latter.

Heterogeneity and co-expression of intermediate filament proteins in adenoid cystic carcinoma of salivary glands.

Among a series of 76 adenoid cystic carcinomas (ACC), 51 cases with cibriform or trabecular patterns were selected for an immunohistochemical study. These tumors were composed of three types of cells: basaloid, myoepithelial and ductal luminal acinous or tubular cells with a variable amount of each cell type from one case to another. Monoclonal antibodies (MoAb) against keratins (PKK1, KL1, K8.12, K8.13, K4.62 anti-cytokeratins antibodies) and against vimentin were used. Tubular cells were characteristically marked by anti-cytokeratins MoAb, but with a great heterogeneity of keratin distribution. In myoepithelial cells, keratin was absent or slightly positive and vimentin was present, with co-expression of the two types of filaments in some cells. Like myoepithelial cells, basaloid cells were positive to anti-vimentin antibody and negative or slightly positive to anti-keratin antibodies, with sometimes a co-expression of vimentin and keratin filaments in the same cell. Histogenesis of adenoid cystic carcinomas was discussed. Progenitor intercalated duct reserve cells may change into ductal luminal and myoepithelial tumor cells. Otherwise, basaloid cells, arising also from intercalated duct reserve cells, are able to acquire some secretory organites.

[Tracheal adenoid cystic carcinoma presenting as a thyroid tumor].

Primary tumors of the trachea are rare. Adenoid cystic carcinoma is the second most common tracheal malignancy and its symptoms are nonspecific. A 66-year-old woman presented with complaints of progressive dyspnea and cough of a two-week duration. She had received asthmatic treatment many times for the past four years. Physical examination revealed a large, firm, nontender, multilobular mass in the thyroid gland. Indirect laryngoscopic examination showed subglottic stenosis. On a plain cervical radiograph, there was narrowing of the tracheal lumen. Emergent tracheotomy was performed and a biopsy was taken from the mass. Histopathologic examination revealed adenoid cystic carcinoma, suggesting a thyroid mass with a tracheal origin. Despite medical treatment, the patient died in the fourth month.

[Adenoid cystic carcinoma of ENT associated with hard lump: a report of 9 cases].

OBJECTIVE: To assess the clinical and pathological features of the patients with ENT adenoid cystic carcinoma (ENTACC) who presented with hard lump, and to detect its prognosis. METHOD: A retrospective analysis was performed on 9 patients with ENTACC by their clinical data. RESULT: The period from the occurrence of primary symptoms to definite diagnosis was 4 years and 3 months on average. All patients were followed up from 6 months to 15 years. All of the cases relapsed after operation and radiotherapy. Five patients died, among which 3 died with lung metastases, 2 with local lymphatic metastases, 1 with bone metastases and 1 with encephalic diffusion. The other 4 cases still survive. The longest survival period with tumor was 6 years and 3 months. CONCLUSION: The pathological result of these 9 cases were all salivary gland derived adenoid cystic carcinoma of solid type. It was mainly characterized by the development of ENT hard lump. All of the patients should be diagnosed in time. Extensive excision and postoperative radiotherapy are needed at the first time. The tumor of solid histological pattern appears with much poorer prognosis compared with that of other histological pattern.

Cytologic characteristics of adenoid cystic carcinoma of the cervix uteri.

Cytologic characteristics of three cases of adenoid cystic carcinoma of the cervix uteri are reviewed. The clusters of malignant cells tend to form large acini and mimic the pattern seen in histologic sections. The resemblance to cells of adenocarcinoma of the endometrium is noted. When the acinar pattern is recognized in cervical smears, adenoid cystic carcinoma of the cervix may be suspected.

[Adenoid cystic carcinoma extended to Gasserian ganglion-report of two cases (author's transl)].

Adenoid cystic carcinoma is an uncommon tumor, especially in cases of its extension into the cranial cavity. Two cases of this tumor involving Gasserian ganglion were reported. The first case was a 47 year old female who developed paresis of the right side fifth cranial nerve. The plain skull X-ray of the axial view demonstrated enlargement of the right sided foramen ovale. The tumor was removed exploring the middle fossa extradurally, however its extention through the foramen ovale remained behind. A small induration was found at the base of the oral cavity postoperatively, and the biopsy revealed characteristic findings of adenoid cystic carcinoma histologically. Radiation therapy was followed by considerable clinical improvement.

Adenoid cystic carcinoma of the breast. Light and electron microscopy and a brief review of the literature.

The case of two patients with adenoid cystic carcinoma of the breast are reported. In case 1, the predominant pattern was that of a solid, basaloid, cylindromatous tumor, whereas case 2 showed the typical adenoid cystic appearance. Results of electron microscopical study confirmed that many of the cystic spaces seen by light microscopy were extracellular compartments enclosed by tumor cells. Many of the tumor cells contained densely packed fibrils, which support the myoepithelial origin of these tumors. Approximately 92 well-documented reports of this lesion are briefly reviewed.

[Neutron therapy for adenoid cystic carcinoma].

In the treatment of adenoid cystic carcinomas, certain situations such as inoperable tumors, macroscopic tumor residues following surgery and recurrences occurring with the previous therapeutic strategies (surgery and/or photon radiotherapy) are afflicted with frequent local recurrences. In these situations the use of radiotherapy with fast neutrons (densely ionizing radiation with high energy transfer) results in absence of recurrences in approx. 70% of the cases, according to data in the literature. The advantage of using neutrons lies in their greater biological effect on slowly growing tumors. In the Hospital for Radiotherapy of the Münster University neutron therapy has been applied since 1986 for these indications using a DT neutron generator (14 MeV). Based on the results obtained in the treatment of 8 patients and a review of the literature, indications for, treatment planning and implementing of neutron therapy is illustrated. Particular consideration is given to CT and MRT. Neutron therapy offers an alternative to previously existing treatment methods for locally advanced adenoid cystic carcinomas or highly differentiated malignomas of the salivary glands. Its effectiveness should be verified by studies involving larger numbers of patients and longer observation periods.

[Adenoid hypertrophy as initial symptom of an infection with the human immunodeficiency virus].

A woman aged 25 yr consulted the Department of Otorhinolaryngology with complaints of persistent nasal obstruction caused by adenoid hypertrophy recurring shortly after recent adenoidectomy. Previous elaborate investigations did not reveal the cause of this unexplained phenomenon. Testing for human immunodeficiency virus (HIV) was positive. In case of an unexplained recurrent adenoid hypertrophy, an infection with the human immunodeficiency virus should be considered in the differential diagnosis.
