Trace elements and superoxide dismutase in benign and malignant breast diseases.

This study was planned to determine the probable changes in trace element levels and superoxide dismutase (SOD) activity in women with neoplastic breast diseases. Measurements were performed in three different groups. The first group consisted of 20 healthy women, control group, the second group contained 16 patients with benign breast disease and the third group contained 39 patients with malignant breast disease. The trace element concentrations were determined by using atomic absorption spectrophotometry and SOD activity by using spectrophotometry. When compared with the control values, the plasma copper levels were slightly increased in the second group and significantly in the third group (p < 0.001). The difference between the benign and malignant groups was also significant (p < 0.001). The red cell copper values showed a marked decrease in both groups (p < 0 001). Although there were increases in the plasma zinc levels of both patient groups, the differences were not significant statistically. But, the red cell zinc values showed an significant increase in benign and malignant patients compared to the control group (p < 0.001) (p < 0.001). The plasma magnesium and red cell magnesium values did not show significant differences. The red cell SOD activity showed an significant increase in the benign and malignant patient groups (p < 0.001). The results of this study suggested that reactive oxygen metabolites may play a pathogenetic role in the both benign and malignant tumor development, which is reflected by the change in SOD activity, and in trace element concentrations.

Trace element signature of subduction-zone fluids, melts and supercritical liquids at 120-180 km depth.

Fluids and melts liberated from subducting oceanic crust recycle lithophile elements back into the mantle wedge, facilitate melting and ultimately lead to prolific subduction-zone arc volcanism. The nature and composition of the mobile phases generated in the subducting slab at high pressures have, however, remained largely unknown. Here we report direct LA-ICPMS measurements of the composition of fluids and melts equilibrated with a basaltic eclogite at pressures equivalent to depths in the Earth of 120-180 km and temperatures of 700-1,200 degrees C. The resultant liquid/mineral partition coefficients constrain the recycling rates of key elements. The dichotomy of dehydration versus melting at 120 km depth is expressed through contrasting behaviour of many trace elements (U/Th, Sr, Ba, Be and the light rare-earth elements). At pressures equivalent to 180 km depth, however, a supercritical liquid with melt-like solubilities for the investigated trace elements is observed, even at low temperatures. This mobilizes most of the key trace elements (except the heavy rare-earth elements, Y and Sc) and thus limits fluid-phase transfer of geochemical signatures in subduction zones to pressures less than 6 GPa.

Lipids, lipoproteins, apolipoproteins, selected trace elements and minerals in the serum of children on valproic acid monotherapy.

We evaluated the serum levels of lipids, lipoproteins, apolipoproteins, along with a number of minerals and trace elements such as Ca, Mg, Cu and Zn in a group of children after 6 months of valproic acid monotherapy. Thirty patients with seizures, mean age, 9.8+/-2.6 years and 79 healthy children (controls), mean age, 10.9+/-3.2 years, formed the two styd groups. The patient group was treated with valproic acid (27.9+/-14.8 mg/kg/24 hr). Patients underwent clinical and laboratory evaluations including liver function tests, NH3, lipid, mineral and selected trace element levels before and after six months on valproic acid treatment, whereas controls only one evaluation. Liver function data and NH3 levels were found to be elevated in the group of patients, whereas albumin level was reduced. Triglycerides, total cholesterol, HDL-C, apolipoprotein (ApoA)-1, Apo B and Ca concentrations were found relative to control values, LDL-C, VLDL-C, Mg, Cu, Zn, were measured significantly altered (P<0.0001) compared to controls. The ratios ApoA-1/ApoB, HDL-C/ApoA-1, LDL-C/Apo B, which were closely related to the size of LDL particles, where correlated with Zn/Cu (P<0.001). Serum lipid profile, especially LDL size, indirectly evaluated for the first time and metal levels were found to be significantly changed, after six months on valproic acid monotherapy, suggesting a possible risk of developing coronary heart disease. Since valproic acid is a long-term treatment, it could be recommended that the incorporation of measurements of lipids, lipoproteins, apolipoproteins and trace elements in the "follow up" laboratory testing could be a preventive measure.

Elevated trace element concentrations in southern toads, Bufo terrestris, exposed to coal combustion waste.

A number of recent studies have linked developmental, physiological, and behavioral abnormalities in amphibians to coal combustion wastes (coal ash). Few studies, however, have determined trace element concentrations in amphibians exposed to coal ash. In the current study we compare total body concentrations of 20 trace elements in adult southern toads, Bufo terrestris, inhabiting coal ash settling basins with toads that were not exposed to the combustion wastes (reference). In addition, we document the accumulation of trace elements in toads transplanted from reference sites to field enclosures in an ash settling basin for 7 and 12 weeks. Arsenic, selenium, and vanadium levels were significantly elevated in toads captured at the ash-contaminated site in comparison to toads from the reference site. All three of these elements were also significantly elevated in toads exposed to the contaminated habitat for only 7 weeks. Our study suggests that adult anurans can bioaccumulate particularly high levels of selenium and may be useful bioindicators in agricultural and coal ash-impacted habitats.

Changes in soil and leaf trace element concentrations: a study in Naples city centre.

The spatial and temporal changes of trace element concentrations in leaves and soils of an urban area were investigated simultaneously. For this purpose, in May and September 2001 and in January and May 2002, samples of Quercus ilex leaves and surrounding soils were taken at 5 sites in central Naples and on Mt. Vesuvius, a remote area, as control. The contents of Cd, Cr, Cu, Fe, Mn, Pb, V and Zn were analysed in both matrices by atomic absorption spectrometry. The urban sites in Naples showed different spatial trends of contamination depending on each element. The highest leaf concentrations of Cr, Cu, Fe, Pb, V and Zn were measured at the sites affected by a high traffic flow. The highest Cd, Cu, Pb and Zn soil concentrations were measured at different urban sites. The remote soil showed the highest values of Cr and V. The significant correlations between Pb and Zn in each matrix and between leaf and soil Pb and Zn concentrations suggest a common origin and an influence of wet and dry airborne deposition upon foliage and soils. Cd, Pb and Zn concentrations decreased and Cr increased from May 2001 to May 2002 in both matrices. The season and sampling period do not seem to affect trace element concentrations of Q. ilex mature leaves.

Automated in situ trace element analysis of silicate materials by laser ablation inductively coupled plasma mass spectrometry.

This paper describes the automated in situ trace element analysis of solid materials by laser ablation (LA) inductively coupled plasma mass spectrometry (ICP-MS). A compact computer-controlled solid state Nd:YAG Merchantek EO UV laser ablation (LA) system has been coupled with the high sensitivity VG PQII S ICP-MS. A two-directional communication was interfaced in-house between the ICP-MS and the LA via serial RS-232 port. Each LA-ICP-MS analysis at a defined point includes a 60 s pre-ablation delay, a 60 s ablation, and a 90 s flush delay. The execution of each defined time setting by LA was corresponding to the ICP-MS data acquisition allowing samples to be run in automated cycle sequences like solution auto-sampler ICP-MS analysis. Each analytical cycle consists of four standards, one control reference material, and 15 samples, and requires about 70 min. Data produced by Time Resolved Analysis (TRA) from ICP-MS were later reduced off-line by in-house written software. Twenty-two trace elements from four reference materials (NIST SRM 613, and fused glass chips of BCR-2, SY-4, and G-2) were determined by the automated LA-ICP-MS method. NIST SRM 610 or NIST SRM 613 was used as an external calibration standard, and Ca as an internal standard to correct for drift, differences in transport efficiency and sampling yield. Except for Zr and Hf in G-2, relative standard deviations for all other elements are less than 10%. Results compare well with the data reported from literature with average limits of detection from 1 ng x g(-1) to 455 ng x g(-1) and less than 100 ng x g(-1) for most trace elements.

[A case control study on the relationship between trace elements and human neural tube defects].

OBJECTIVE: To explore the relationship between multi-trace elements levels in hair and human neural tube defects as well as other risk factors. METHODS: Using 88 paired cases and controls, an 1:1 matched case control study was carried out. The study subjects were collected from the China-U. S. Collaborative Project on Neural Tube Defects Prevention and Birth Defects Surveillance System. Risk factors were obtained by field investigation with standardized questionnaires and hair trace elements levels were determined by AAS and ICP-MS methods. Microwave digestion was used to digest hair samples. The detected elements would include three groups, namely nutritional elements: Cr, Mn, Cu, Zn, Co, Mo; toxic elements: Pb, As, Cd, Hg; and Lanthanons: Y, La, Pr, Nd. Cox Proportional Hazard Regression Model was used to perform risk factors analysis. RESULTS: Pregnancy fever appeared to be a risk factor of neural tube defects (OR = 6.525, P = 0.034) while hair zinc level (OR = 0.541 microg/100 g, P = 0.02) and times of prenatal physical examination (OR = 0.634, P < 0.001) served as two protective factors appeared in the last model. CONCLUSION: Zinc deficiency might serve as a risk factor for human neural tube defects, suggesting that the avoidance of pregnancy infection together with more periodical prenatal physical examination might reduce the incidence of neural tube defects.

Average mineral and trace element content in daily adjusted menus (DAM) of French adults.

This article is based on data on food consumption gathered from 10,000 households. The daily energetic intake of 2,000 to 2,500 kcal (8,360 to 10,450 kJ) (beverages excluded) provides adults with less than 80% of the recommended amount of several essential minerals and trace elements. For women, beverages contribute only slightly to the total amount of mineral and trace element intake, because the major beverage sources of minerals and trace elements are alcoholic drinks (wine, beer). For men, alcoholic drinks could provide an additional amount of these elements. Consequently, it seems the risk of deficiency appears in adults, with women particularly concerned by this problem. The present study demonstrates that the intake of the following elements might be insufficient in women: fluorine, manganese, iron, copper, zinc and magnesium.

Geology, trace elements and health.

Both animal and vegetable life depend for their existence on appropriate amounts of various trace elements, albeit in very small amounts. This paper lists some of these trace elements and the ailments in which they play an important role. The elements discussed are gold, platinum, copper, lead, zinc, aluminium, silica, mercury, cadmium, selenium, arsenic, and iodine. The diseases involved range from multiple sclerosis, various cancers, arthritis, goitre, Down's Syndrome, and mental retardation. Less well known are Keshan, Alzheimer's, Itai-Itai, and Minamata diseases. Of particular interest in the latter part of the twentieth century is the discovery that serious deficiencies of either copper or zinc in the diet of animals may break down their immune defence mechanisms. The ability of certain plants selectively to concentrate particular heavy metals in their tissues and pollen is discussed.

Dephytinisation of soyabean protein isolate with low native phytic acid content has limited impact on mineral and trace element absorption in healthy infants.

Infant formulas based on soyabean protein isolate are often used as an alternative to cows'-based formulas. However, the presence of phytic acid in soya formulas has raised concern about the absorption of trace elements and minerals from these products. The aim of the present study was to evaluate mineral and trace element absorption from regular and dephytinised soya formula in healthy infants. Soyabean protein isolate with a relatively low native content of phytic acid was used for production of a regular soya formula (300 mg phytic acid/kg liquid formula) and an experimental formula was based on dephytinised soya protein isolate (<6 mg phytic acid/kg liquid formula). Using a crossover study design, apparent mineral and trace element absorptions were measured by a stable isotope technique based on 72 h faecal excretion of non-absorbed stable isotopes (Zn, Fe, Cu and Ca) and by the chemical balance technique (Mn, Zn, Cu and Ca) in nine infants (69-191 d old). Fe absorption was also measured by erythrocyte incorporation 14 d after intake. The results from the present study demonstrated that Zn absorption, measured by a stable isotope technique, was significantly greater after dephytinisation (mean value 16.7 v. 22.6 %; P=0.03). No other statistically significant differences between the two formulas were observed. The nutritional benefit of dephytinisation was marginal in the present study. Based on these results, the use of soyabean protein isolate with low native content of phytic acid should be promoted for production of soya formulas and adequate addition of ascorbic acid to enhance Fe absorption should be ensured in the products.

Effect of pectin on some electrolytes and trace elements in patients with hyperlipoproteinemia.

BACKGROUND: Water-soluble dietary fibres are widely used in preventive and curative nutrition, especially in cases of hyperlipoproteinemia. Occurrence of disturbances of the electrolyte and trace element metabolism causing adverse effects in long-term treatment with pectin can be theoretically substantiated. METHODS: Seventy three patients with hyperlipoproteinemia, aged from 40 to 69 years, were included in the study. They were treated with a natural product, based on high-esterified pectin granulated with sorbitol. The patients were examined prior to the treatment and once monthly during the three-month course of therapy. RESULTS: The results of the comparative assessment of the electrolytes (sodium, potassium, chloride, ionised calcium, total and ionised magnesium) failed to reach statistical significance during the administration of the pectin product. Neither did the serum level of trace elements (iron and copper) change significantly during the observed period. CONCLUSIONS: The biochemical analysis of the serum level of certain electrolytes and trace elements indicate that daily administration of 15 g of high-esterified apple pectin for a three-month period has no adverse effects. Therefore pectin products can be included in the therapeutic schedule in the treatment of hyperlipoproteinemia.

Concentrations and modes of occurrence of trace elements in the Late Permian coals from the Puan Coalfield, southwestern Guizhou, China.

The concentration, mode of occurrence, and origin of trace elements in the Late Permian coals from the Puan Coalfield, southwestern Guizhou, China, were examined using inductively coupled plasma-mass spectrometry (ICP-MS), X-ray fluorescence (XRF), cold-vapor atomic absorption spectrometry (CV-AAS), ion-selective electrode method (ISE), sequential chemical extraction procedure (SCEP), scanning electron microscopy equipped with energy-dispersive X-ray (SEM-EDX), and optical microscope. Results show that minerals in the No. 2 Coal from the Puan Coalfield were mainly made up of epigenetic pyrite of low-temperature hydrothermal fluid origin and kaolinite of detrital terrigenous origin. Elements including As (36.9 microg/g), Cd (10.2 microg/g), Cr (167.3 microg/g), Cu (365.4 microg/g), Hg (2.82 microg/g), Mo (92.6 microg/g), Ni (82.6 microg/g), Pb (184.6 microg/g), Se (6.23 microg/g), Zn (242.3 microg/g), and U (132.7 microg/g) are significantly enriched in the No. 2 Coal from the Puan Coalfield. However, concentrations of trace elements in the other four coals, the No. 1, No. 8, No. 11, and No. 18 Coals, were close to the usual ranges found for Guizhou of China, China, and USA. Results of SEM-EDX and SCEP showed that As, Cd, Hg, Mo, Ni, Pb, and Zn occur mainly in veined pyrite, while Cr, Cu, and U distribute mainly in kaolinite, indicating that the low-temperature hydrothermal fluid and detrital materials of terrigenous origin are the main contributors to the enrichment of these trace elements in the No. 2 Coal.

Concentration and subcellular distribution of trace elements in liver of small cetaceans incidentally caught along the Brazilian coast.

Concentrations of trace elements (V, Cr, Mn, Fe, Co, Cu, Zn, Ga, As, Se, Rb, Sr, Mo, Ag, Cd, Sb, Cs, Ba, T-Hg, Org-Hg, Tl and Pb) were determined in liver samples of estuarine dolphin (Sotalia guianensis; n = 20), Franciscana dolphin (Pontoporia blainvillei; n = 23), Atlantic spotted dolphin (Stenella frontalis; n = 2), common dolphin (Delphinus capensis; n = 1) and striped dolphin (Stenella coeruleoalba; n = 1) incidentally caught along the coast of Sao Paulo State and Parana State, Brazil, from 1997 to 1999. The hepatic concentrations of trace elements in the Brazilian cetaceans were comparable to the data available in literature on marine mammals from Northern Hemisphere. Concentrations of V, Se, Mo, Cd, T-Hg and Org-Hg increased with increasing age in liver of both estuarine and Franciscana dolphins. Very high concentrations of Cu (range, 262-1970 microg/g dry wt.) and Zn (range, 242-369 microg/g dry wt.) were observed in liver of sucklings of estuarine dolphin. Hepatic concentrations of V, Se, T-Hg, Org-Hg and Pb were significantly higher in estuarine dolphin, whereas Franciscana dolphin showed higher concentrations of Mn, Co, As and Rb. Ratio of Org-Hg to T-Hg in liver was significantly higher in Franciscana dolphin than estuarine dolphin, suggesting that demethylation ability of methyl Hg might be lower in liver of Franciscana than estuarine dolphins. High hepatic concentrations of Ag were found in some specimens of Franciscana dolphin (maximum, 20 microg/g dry wt.), and 17% of Franciscana showed higher concentrations of Ag than Hg. These samples with high Ag concentration also exhibited elevated hepatic Se concentration, implying that Ag might be detoxified by Se in the liver. Higher correlation coefficient between (Hg+0.5 Ag) and Se than between Hg and Se and the large distribution of Ag in non-soluble fraction in nuclear and mitochondrial fraction of the liver also suggests that Ag might be detoxified by Se via formation of Ag2Se in the liver of Franciscana dolphin.

High resolution inductively coupled plasma mass spectrometry (HR-ICPMS) determination and multivariate evaluation of 10 trace elements in mussels from 7 sites in Limfjorden, Denmark.

The contents of V, Cr, Co, Ni, Cu, Ga, Rb, Cd, Ba, and Pb in the soft tissue of blue mussel (Mytilus edulis) were determined by a high resolution inductively coupled plasma mass spectrometry (HR-ICPMS) method. Sample digestions were performed in closed microwave vessels using nitric acid and hydrogen peroxide. Using HR-ICPMS it is possible to resolve the analytical peaks from otherwise interfering polyatomic ions with a mass resolution setting of 4,000 (Cr, Ni, Cu). The proposed method was validated using a mussel tissue reference material (NIST SRM 2974). The proposed method was applied to real samples of blue mussel from seven sites in the inlet "Limfjorden", Denmark, and the levels of trace elements found were compared with the levels found in an earlier study. For the mussel samples large inter-regional differences in trace element concentrations in the tissues were recorded. The mussels from the different sites could be separated using principal component analysis (PCA). Comparison with the levels of trace elements in mussels found in 1982 showed that the trace elemental contamination has increased during the last 15 years. From the data obtained, mussel tissue appears to be good bio-indicator for identification of coastal areas exposed to metallic contaminants.
