In vitro fermentation of cellulose, beet pulp, citrus pulp, and citrus pectin using fecal inoculum from cats, dogs, horses, humans, and pigs and ruminal fluid from cattle.

We evaluated the influence of gastrointestinal tract microflora from several species on fiber fermentation characteristics in vitro. Selected fibrous substrates (cellulose, beet pulp, citrus pulp, and citrus pectin) were incubated for 6, 12, 24, and 48 h with ruminal fluid from cattle or feces from dogs, cats, pigs, horses, or humans. When data were pooled across all substrates and fermentation times, OM disappearance (29.4%) and acetate, propionate, butyrate, and total short-chain fatty acid (SCFA) production (1.09, .41, .12, and 1.61 mmol/g of OM, respectively) were lowest (P < .05), and lactate production (.23 mmol/g of OM) was greatest (P < .05) for horse fecal microflora compared with samples from the other species. The greatest (P < .05) acetate production resulted when substrates were fermented by cat fecal microflora (2.38 mmol/g of OM). The greatest (P < .05) propionate productions resulted from pig fecal and cattle ruminal microflora (.88 and .83 mmol/g of OM, respectively), and the greatest (P < .05) butyrate productions resulted from human and pig fecal microflora (.39 and .40 mmol/g of OM, respectively). Total SCFA production was greatest (P < .05) for cat fecal microflora (3.38 mmol/g of OM). When data were pooled across the species, substrate OM disappearance and SCFA production ranked from least to greatest in the following order: cellulose < beet pulp < citrus pulp < citrus pectin. The fermentability of different fibrous substrates by fecal or ruminal microflora from various species seems to be dependent not only on the fermentative activity of the microbial population but on other factors as well, perhaps lag time and rate of digesta passage.

An investigation into the floating behaviour of a pectin-containing anti-reflux formulation (FF5005) by means of gamma scintigraphy.

The gastric distribution and residence time of a new pectin-containing formulation, FF5005 (Farma Food A/S, Denmark), was investigated by using the technique of gamma scintigraphy in six healthy volunteers after administration with a radiolabelled meal. The formulation and test meal were radiolabelled with indium-113m and technetium-99m, respectively, and the formulation was administered to the subjects 30 min after the labelled meal. FF5005 was shown to float and form a discrete phase on top of the stomach contents and emptied from the stomach more slowly than the food (p less than 0.05, Wilcoxon signed rank test). The times taken for the formulation and test meal to half-empty from the stomach (T50) were 4.13 +/- 0.69 h (mean +/- SD) and 2.17 +/- 0.15 h (mean +/- SD), respectively. Greater than 50% of the formulation remained in the fundal region of the stomach for 3 h. FF5005 produced in vivo behaviour similar to that of established alginate-containing anti-reflux agents, and the pectin content of the formulation was shown to decrease the rate of emptying of the meal.

Effects of different levels of gum arabic, low methylated pectin and xylan on in vitro digestibility of beta-lactoglobulin.

Plant hydrocolloids used in the food industry to improve texture and stability of food, such as dairy products, can reduce protein digestibility and, consequently, modify the bioavailability of amino acids. We studied the in vitro hydrolysis at 37 degrees C of beta-lactoglobulin (beta-lg) in mixed dispersions containing either gum arabic or low-methylated pectin or xylan at levels of 0, 1, 10, 20, 30, and 50% weight. Proteolysis used either pepsin alone by progressive reduction of pH during proteolysis or pepsin followed by trypsin and chymotrypsin in two different dialysis bags with a molecular weight (MW) cutoff of 1000 or 8000 Da. Results showed that beta-lg was almost resistant to pepsin digestion and that the three plant hydrocolloids inhibited significantly beta-lg digestibility as determined using dialysis bag with a 1000-Da MW cutoff. Among the three polysaccharides used, xylan showed a digestibility decrease greater than that obtained with gum arabic and low-methylated pectin. On the other hand, no significant effect of polysaccharides on the in vitro beta-lg digestibility was detected using the dialysis bag with an 8000 Da MW cutoff. This mainly suggests that peptides with MW in the range 1000 to 8000 Da may interact with polysaccharides more than peptides and proteins with a greater molecular weight to decrease the protein digestibility, and that the nature of the polysaccharides plays a role in the interaction.

Beta-glucan and pectin derivatives stimulate prolactin secretion from hypophysis in vitro.

Previous work has shown that various plant extracts administered to animals stimulate milk protein synthesis through the secretion of prolactin. It has also been shown that beta-glucan and pectin are the active molecules capable of stimulating prolactin release in vivo after intravenous injections. In this work, it is shown that beta-glucan and several pectin derivatives are able to stimulate prolactin secretion from hypophysis fragments incubated for 2 hr in a synthetic medium.

Relationship between caesium (137Cs) load, cardiovascular symptoms, and source of food in 'Chernobyl' children -- preliminary observations after intake of oral apple pectin.

Seventeen years after the nuclear power accident at Chernobyl, most of the radio-contamination among the population of Southern Belarus is caused by incorporation of long-lived radioisotopes. The varying levels of 137Cs observed among children in this area are explained by the source of their food, especially by the consumption of contaminated milk produced privately. We stratified children from rural areas of Belarus (caesium [137Cs] contamination >5 Ci/km(2)) by their 137Cs loads into three distinct groups (group 1, <5 Bq/kg body weight [BW]; group 2, 38.4 +/- 2.4 Bq/kg BW; group 3, 122 +/- 18.5 Bq/kg BW). We determined the relationship between the 137Cs load and the children's main source of food and recorded their cardiovascular symptoms. Cardiovascular symptoms, ECG alterations, and arterial hypertension were significantly more frequent in children with high 137Cs burden than in children with very low 137Cs burden. Children with moderate and high 137Cs loads (groups 2 and 3) received apple pectin, a food additive, for 16 days. Apple pectin significantly decreased 137Cs loads in these groups (39% and 28%, respectively). ECG alterations improved, while cardiovascular symptoms and hypertension did not change in any group.

Effects of daily oral administration of quercetin chalcone and modified citrus pectin on implanted colon-25 tumor growth in Balb-c mice.

The health benefits of fruits and vegetables have been the subject of numerous investigations over many years. Two natural substances, quercetin (a flavonoid) and citrus pectin (a polysaccharide found in the cell wall of plants) are of particular interest to cancer researchers. Two modified versions of these substances - quercetin chalcone (QC) and a pH-modified citrus pectin (MCP) - are the focus of this study. Previous research has confirmed that quercetin exhibits antitumor properties, likely due to immune stimulation, free radical scavenging, alteration of the mitotic cycle in tumor cells, gene expression modification, anti-angiogenesis activity, or apoptosis induction, or a combination of these effects. MCP has inhibited metastases in animal studies of prostate cancer and melanoma. To date, no study has demonstrated a reduction in solid tumor growth with MCP, and there is no research into the antitumor effect of QC. This study examines the effects of MCP and QC on the size and weight of colon-25 tumors implanted in balb-c mice. Fifty mice were orally administered either 1 ml distilled water (controls), low-dose QC (0.8 mg/ml), high-dose QC (1.6 mg/ml), low-dose MCP (0. 8 mg/ml) or high-dose MCP (1.6 mg/ml) on a daily basis, beginning the first day of tumor palpation (usually eight days post-implantation). A significant reduction in tumor size was noted at day 20 in all groups compared to controls. The groups given low-dose QC and MCP had a 29-percent (NS) and 38-percent (p<0.02) decrease in size, respectively. The high-dose groups had an even more impressive reduction in size; 65 percent in the QC group and 70 percent in the mice given MCP (both p<0.001). This is the first evidence that MCP can reduce the growth of solid primary tumors, and the first research showing QC has antitumor activity. Additional research on these substances and their effect on human cancers is warranted.

[Enzymatic decomposition of pectin in the presence of amino acids and proteins].

The influence of amino acids--glycine, alpha-alanine, lysine, phenylalanine, serine, methionine, cysteine, tryptophan, aspartic acid and asparagine--and proteins--chick egg albumin and autolysate of Aspergillus niger biomass--on the disintegration of beet pectine by the Asp. niger enzymic preparation was investigated. This process was shown to depend on the chemical composition and concentration of amino acids and the protein nature. All the tested amino acids, except tryptophan, at doses of 0.0001 to 0.5% stimulated the activity of pectolytic enzymes. The effect of proteins was different: Asp. niger autolysate showed no essential influence and egg albumin inhibited pectine disintegration.

Comparison of the moisturization efficacy of two vaginal moisturizers: Pectin versus polycarbophil technologies.

This study was designed to compare the vaginal deposition and moisturization of two vaginal moisturizers, Summer's Eve (SE), based on pectin, and Replens (Rp), based on polycarbophil, in a double-blind crossover study design. Fifty-one female patients were each randomly assigned to one of two treatment groups. After a one-week washout period, the products were used for two weeks. After another one-week washout period, product assignments were switched. Colposcopy examinations were performed at the beginning and at the end of each product use. Of the forty-seven patients completing the study, 41 (87%) were found to have no vaginal residue after SE vaginal moisturizer, while only 25 (53%) were found to have no vaginal residue after using Rp vaginal moisturizer. No difference in relief of vaginal dryness or in product acceptance was found between the two products. This study shows that the use of SE vaginal moisturizer, based on pectin, resulted in significantly less vaginal residue compared to Rp vaginal moisturizer, based on polycarbophil, and in comparable relief of vaginal dryness. These results strongly suggest that bioadhesion is not important in vaginal moisturizers.

[Topical pectins for the treatment of burn wounds].

The results of the burned wounds treatment with 1-2% solution of apple or beet pectins were analysed. The control results were examined at the same patients with symmetrical wounds treated by chlorhexidine, levomecol, furacillin liniment. Clinical evaluation demonstrated good tolerability of the treatment regime, absence of side effects and complications, inhibition of inflammation, acceleration of epithelisation of the burns II-IIIA class, allowed to diminish the preliminary period before autodermoplastic operation (burns of IIIB class), diminished microbial dissemination and dissemination with Pseudomonas aeruginosa. The best results were demonstrated when the pectins application was performed since the first day after the burned injury.

[The use of pectin-containing enterosorbents in exposure to radionuclides and heavy metals].

The pectin-vitamin composition (PVC) and its complex with active carbon (AC) was investigated for use as an oral sorbent to protect radionuclide (Sr-85, Cs-137) accumulation. PVC is composed of the natural mixture of cellulose, pectin (Ca-salt), flavonoides, vitamins of the B-group, PP and C. Investigation of PVC radioprotective action was carried out on the two groups of rats (n = 30) which received 1306 Bk/animal of Sr-85 or 343 Bk/animal of Cs-137 daily. One of the group of rats received every day also 300 mg of PVC. After 30 days level of Sr-85 was less by 56.6%, level of Cs = 137 was less by 27.6% in PVC-treated than in the control group (P < 0.05). Composite sorbent consisting of PVC and AC was slightly less effective in removing of Sr and Cs.

[Role of pectins in the complex treatment in patients in neurointensive care units].

Clinical and laboratory estimation of the efficacy of pectins in complex treatment of patients with intracranial hematoma was performed. It was shown that in the group of the patients treated with pectins vs the control group development of pyo-inflammatory infections was less frequent, the indices of the immunity status improved and a more rapid decrease of the intoxication and a more rapid normalization of the composition of various biotopes in the patients were observed.

Adhesion of human neutrophils to fibronectin is inhibited by comaruman, pectin of marsh cinquefoil Comarum palustre L., and by its fragments.

Earlier, we detected antiinflammatory action of comaruman, pectin of the marsh cinquefoil Comarum palustre L. This effect can be explained by new data concerning inhibition of adhesion of human neutrophils to fibronectin by comaruman and its fragments. The galacturonan backbone fragment of molecular mass >10 kD appears to be the active region of the comaruman macromolecule. Comaruman CP (50-200 microg/ml) was found to decrease adhesion of neutrophils stimulated by phorbol 12-myristate 13-acetate (PMA, 1.625 microM) and by dithiothreitol (DTT, 0.5 mM). The fragments of comaruman CP-H (100 kD), CP-H1 (10-50 kD), and CP-H2 (100 kD) obtained by acidic hydrolysis and representing regions of linear polygalacturonan are shown to inhibit neutrophil adhesion more than the crude pectin. A fragment CP-E (<10 kD) obtained using pectinolysis and representing a branched region of the comaruman macromolecule failed to influence cell adhesion. The parent comaruman CP as well as fragments of its polygalacturonan backbone diminish PMA-initiated generation of oxygen radicals in neutrophils.

[Enterosorption with pectin-containing medication in the treatment of peritonitis].

UNLABELLED: One hundred and fifty patients aged 19 to 86 years with general peritonitis mainly in a toxic (85.3%) stage were examined. The leading causes of peritonitis were acute appendicitis (24.0%) and perforation of gastroduodenal ulcers (21.3%). Three groups of patients were formed with a pair samples method for a comparative analysis of treatment RESULTS: The control group of patients was treated conventionally. Entero-sorption was performed through naso-intestinal tube with well-known carbonic sorbent UAO-A and pectin-containing medication made of red beet. The results were compared. A 1.3-fold decrease in entero-paresis naso-intestinal intubation duration, a relief in severity of the patients' condition by SAPS scale as early as on day 2, improvement of immunity indices, faster reduction of toxicity of blood plasma and leukocytic index of intoxication, a 12% decrease in postoperative pneumonia morbidity and postoperative lethality from 14 to 6.6% demonstrated advantages of entero-sorption with pectin-containing medication.

Natural hypocholesterolemic agent: pectin plus ascorbic acid.

An addition of 5% citrus pectin and 0.5% ascorbic acid to high-fat diet of guinea pigs prevented total cholesterol accumulation in blood serum and the liver. Two groups of persons were given a preparation containing a daily dose of 15 g pectin and 450 mg ascorbic acid for 6 weeks. In 21 healthy persons with mild hypercholesterolemia total serum cholesterol dropped significantly by 24 mg/100 ml (8.6%), while the concentration of high density lipoprotein cholesterol remained unchanged. In 11 hyperlipemic outpatients (type IIa, IIb and IV) total serum cholesterol dropped by 68 mg/100 ml (18.7%). The changes in triglyceridemia proved inconsistent.

Effect of pectin on the function of the rat exocrine pancreas.

The author studied the effect of three weeks' administration of a 5% pectin concentration in the standard (Larsen) diet on basal and cholecystokinin octapeptide-(CCK8-) stimulated pancreatic exosecretion in rats. Protein, amylase and trypsinogen output determined in pancreatic juice obtained under basal conditions and after CCK8 stimulation of the exocrine pancreas showed no statistically significant differences between rats fed on the standard and the pectin-enriched Larsen diet. Our observations and studies by other authors show that the direct effect of dietary fibre on the exocrine function of the pancreas is an open question as far as further research is concerned.

Modulation of postprandial glycaemia and insulinaemia by pectin in mixed nutrient combinations.

The present study was designed to examine the effect of pectin (P) on postprandial glycaemia and insulinaemia when ingested with glucose (G), casein (Cs) and corn oil (Co) in various combinations. The study was conducted on five healthy male volunteers, on each of whom five meal tolerance tests were performed. The meals were isocaloric and consisted of G; G and P; G, Cs and P; G, Co and P; and G, Cs, Co and P. The meals were administered after an overnight fast. In addition to a fasting blood sample, blood was collected 0.5, 1.0, 1.5 and 2.0 h after ingestion for measurement of serum glucose and insulin levels. The glycaemic and insulinaemic response to GP did not differ significantly from that to G. All the other meals, viz. GCsP, GCoP and GCsCoP, gave a significant reduction in postprandial glycaemia as compared to G. The corn oil containing meals, viz. GCoP and GCsCoP, in addition, gave a significant reduction in postprandial insulinaemia as compared to G. Pectin alone is not a dependable dietary constituent for reducing postprandial glycaemia. Its combination with protein and fat significantly lowers the postprandial glycaemic as well as insulinaemic response to orally administered glucose.

[Electron microscopic study of pectin solutions].

Electron microscopic study was carried out of the structure of pectin 0.01% aqueous solutions according to esterification degree at different pH medium. Preparations were prepared by freezing-drying method of solution drop on the formvar film and by the negative contrasting method. Microfibrils of 60-180 A diameter were found. Their aggregation increased with a decrease of the charge of pectin macromolecule (increase of esterification, decrease of pH).

[Immobilization of proteolytic complexes on pectin-containing carriers].

A number of sorbents were synthesized on the basis of pectin and then used for immobilization of proteolytic complexes--pancreatin and protosubtilin. The best properties were shown by the enzyme preparations based on pectin, formaldehyde and melamin (PFM). Thus immobilization of pancreatin on PFM through Fe(III) ions gave a preparation with the activity of 79 000 mumole/g X h with respect to methyl ester of L-tryptophane (the activity yield is 91%). The pH optimum for all immobilized preparations was shifted towards the alkaline region. The thermostable fraction of the immobilized preparations retains the activity at 60 degrees for a long time.
