Immunolocalization of acidic and basic fibroblast growth factors during mouse odontogenesis.

Acidic and basic fibroblast growth factors (aFGF and bFGF), are both known to bind to extracellular matrix components, particularly proteoheparin sulfates, and to regulate in vitro proliferation, differentiation and morphology of cells of neuroectodermal and mesodermal origins. Their patterns of distribution were studied during mouse odontogenesis by means of indirect immunofluorescence and immunoperoxidase histochemistry on frozen fixed sections and after Bouin's fixative and paraffin embedding. Localization of aFGF on frozen fixed sections was observed in the oral epithelium, dental lamina and oral mesenchyme (day-12 of gestation), the stellate reticulum and oral epithelium (day-14), the stratum intermedium and at the basal and apical poles of preameloblasts at bell stage. After birth aFGF epitopes were localized within the predentin-dentin area, the stratum intermedium and at the secretory pole of ameloblasts. There was no staining with anti-aFGF antibodies after Bouin's fixative and paraffin embedding. In contrast, using this protocol, intense stainings were found with anti-bFGF antibodies predominantly within dental and peridental basement membranes and mesenchyme: staining of the dental basement membranes was transient (bud and cap stage) and discontinuous; a preferential concentration of bFGF epitopes in the condensed dental mesenchyme of incisors (cap stage) and the dental papillae mesenchymal cells of molars (bell stage) was observed in the posterior and the cervical part of tooth germs. An intense immunostaining of the stellate reticulum with anti-bFGF antibodies was also found on paraffin sections from bud to bell stage.(ABSTRACT TRUNCATED AT 250 WORDS)

[Allotransplantation of meniscus in dog's knee. An experimental study].

This experimental study was to show the difference and outcome of allotransplantation and autotransplantation of meniscus in dogs. In the experimental group (9 dogs), the lateral meniscus was removed and replaced by one preserved for 2 weeks in tissue culture. In the control group (9 dogs), the lateral meniscus was excised but was sutured back to the place immediately. The specimens in each group were taken at 3, 6, 12, and 24 weeks after operation, and were examined microangiographically and histologically under light microscope and electron microscope. The results in both groups showed that the meniscus united to the capsule, chondrocytes in the menisci were intact and the collagen fiber orientation was regular. The articular cartilage of the uncovered area of the tibial plateaus showed normal appearance, and vascular neogenesis in the meniscal periphery was found 12 weeks after operation. These observations suggest that allogenic meniscus maintains its original histologic pattern and functional behavior after its transplantation. It makes possible to protect the knee joint from osteoarthritis.

[Experimental study of preventing postoperative stenosis by modifying artificial esophagus in dogs].

OBJECTIVE: To observe the effect of the modified artificial esophagus on postoperative stenosis in dogs. METHODS: The models of defected esophagus were established in dogs. The double-layered membrane tube (modifying type) was implanted in the test group (n = 10) and the esophageal stent was further inserted when the stenosis occurred. The single pattern tube (original type) was transplanted to the control group (n = 30). The dilation treatment was performed to relieve the postoperative stenosis; alternatively, the esophageal stent was implanted in the unsuccessful dogs. RESULTS: The average artificial esophagus removal time was 19.10 days in the test group, which was significantly lower than 39.07 days in the control group (t = 15.6, P = 0.000). No obstruction after removal was observed in the experimental group. The incidence of postoperative stenosis had no significant difference between these two groups. CONCLUSION: The double-layered membrane tube can make the tube removal safer by shortening the removal time.

Ventricular extrasystoles masquerading as aberrantly conducted atrial extrasystoles because of postectopic T wave change.

This presentation reflects a case where broad and bizarre premature QRS complexes are preceded by sinus beats whose T wave is "abnormal," and seems to contain a premature P wave. A diagnosis of atrial extrasystoles with aberrancy thus could be entertained. The extrasystoles, however, are ventricular in origin. The pattern is explained on the basis of postectopic T wave change, that is, the change in configuration of the T wave that occurs in the sinus beat after an extrasystole.

Phylogenetic analysis of Alloglossidium Simer, 1929 (Digenea: Plagiorchiiformes: Macroderoididae) with discussion of the origin of truncated life cycle patterns in the genus.

Alloglossidium comprises 9 species of North American plagiorchiiform digeneans using ictalurid catfish, freshwater crustacea, and hirudinid leeches as definitive hosts. Two hypotheses about the evolution of this array of definitive hosts were examined using phylogenetic systematic analysis. Two most parsimonious trees, based on 15 homologous series derived from morphological data, each indicated the 2 species utilizing ictalurid catfish definitive hosts are basal members of the group, whereas the 2 species using freshwater crayfish definitive hosts and the 5 utilizing leech definitive hosts each comprise relatively derived monophyletic sister groups. The results suggest that species using crustaceans as definitive hosts are derived by life cycle truncation, whereas those using leeches as definitive hosts appear to be derived through a switch from crustaceans to leeches.

[Contribution of electrophysiology to the study of tianeptine and other antidepressive agents].

Intravenous perfusion of tianeptine reduces the frequency of coeruleus locus neural discharge. It does not affect the rate of neural discharge in the dorsal raphe, increase the rate of neural discharge in the ventral tegument air and the rate of discharge in the pyramidal cells of the hippocampus (CA1). In comparison, intravenous perfusion of clomipramine decreases the rate of neural discharge in the coeruleus locus, the dorsal raphe, the ventral tegument air (temporarily) and the pyramidal cells of the hippocampus. Iontophoretic application of tianeptine does not affect the response of pyramidal cells in the hippocampus to application of serotonin or gamma amino-butyric acid (GABA), but lowers recovery time after iontophoretic administration of serotonin or GABA. Iontophoretic application of clomipramine increases pyramidal cell response to serotonin but not to GABA and lengthens the recovery time after serotonin and GABA. The results are in agreement with the fact that tianeptine, unlike clomipramine, increases serotonin capture. Tianeptine leads to an original electrophysiologic pattern distinct from classic antidepressants and clomipramine.

Systemic circulation with high frequency blood flow--analysis from neurophysiological point of view.

The vibrating electromagnetically driven univalved artificial heart, which provided more than 10 1/min at 10 Hz/sec. vibration, was developed in our team. This vibrating electromagnetic pump (VEMP) produced the original flow pattern compared with the flow pattern of the natural heart, so that circulatory regulatory system during maintain the circulation with VEMP must be investigated before the clinical application. In order to assess the effect of high frequency blood flow produced by the VEMP from the neurophysiological point of view, power spectral analysis of the hemodynamics was performed. VEMP was implanted as the total left heart bypass in adult goats and right heart circulation was maintained by natural hearts. In arterial blood pressure during maintain circulation by the high frequency blood flow, Mayer waves (0.01-0.15 Hz) were significantly decreased, though respiratory waves (0.25-0.40 Hz) were not significantly changed. These results suggest that circulatory regulatory system was influenced by the high frequency blood flow produced by the VEMP.

[Enzyme-and immuno-histochemistry of muscle biopsies in severe infantile spinal muscular atrophy].

The basic lesions in the muscle biopsies of 11 cases of severe infantile spinal muscular atrophy showed typical neurogenic fascicular muscular atrophy. It was found through histochemistry staining that both types of muscle fibres were involved which were characterised by the presence of type grouping of the damaged fibers, anyhow, the original mosaic pattern of both types were still partially reserved. With anti-laminin immunofluorescence staining, it was demonstrated that the basal membrane of atrophied small muscle fibre was still intact. It is considered that the main manifestation of damage on spinal anterior horn neuron is denervation of the skeletal muscle; meanwhile, the function of the muscle fibre in regulations carbohydrate metabolism is not apparently obstructs. The pathologic process of this disease develops from denervation to renervation and then back to denervation again. Therefore the clinical feature of muscle atrophy in this disease is progressive and developmental.

Patterns of failure in the original Besser bipolar prosthesis.

Fourteen original Besser endoprostheses inserted in young patients with osteonecrosis of the femoral head were reviewed. Two cups dislocated early and 1 other required replacement due to acetabular degeneration. Component disassembly occurred in 4 hips, which had exhibited persistently vertical cups. Their stems revealed notching medially and the cups showed gross erosion and failure. The remaining 5 hips were mobile and pain free, but in 3 of these the cups exhibited a persistently vertical orientation and, as such, are predisposed to failure. Only one article reporting early good results using this prosthesis has been published. The failure rate of 50% in this series is disappointing.

Trichoepithelioma of the vulva. A report of two cases.

Trichoepithelioma usually presents clinically as single or multiple nodules on the face, scalp, neck and trunk, and no cases of it on the vulva have been described before. Vulvar trichoepithelioma has complex histologic patterns and originates in appendages that simulate malignancy.

Differentiation of rennet from other milk-clotting enzymes by polyacrylamide gel electrophoresis.

Polyacrylamide gel electrophoresis was used to differentiate animal rennet and other milk-clotting enzymes. After electrophoresis, the separated components were visualized by staining with aniline blue-black. Two prominent proteins were found in calf and bovine rennet, while only 1 major protein was observed in pepsin and enzymes of microbial origin. These patterns provided a basis for distinguishing animal rennet and the other enzymes as well as a means of identifying each type of enzyme by the characteristic pattern shown.

Recombinant interferon alpha-2 modulates hairy cell phenotype "in vitro".

The "in vitro" effect of IFN-alpha on the phenotypic profile of atypical cells from 5 hairy cell leukemia patients was investigated in a 72 hr culture assay. Cytochemical investigations revealed a dramatic decrease in the cytoplasmic content of acid phosphatase and tartrate resistant acid phosphatase in the absence of any apparent morphological modification. Flow cytometry showed that IFN-alpha markedly reduced the density of surface Ig without modifying the original isotype pattern. The expression of the receptor for the Fc fragment of IgG was also reduced. The class II MHC antigen recognized by the monoclonal antibody 12 remained essentially unchanged. Hairy cells were negative for OKT10 and PCA-1 and remained so after IFN-alpha incubation. Present data indicate that IFN-alpha is able to consistently and selectively affect membrane and cytoplasmic features of hairy cells in a short term period. The possibility is envisaged that these changes may be related to the therapeutic efficacy of IFN-alpha.

Cellular migration and morphology in corneal endothelial wound repair.

After a mechanical denudation of rabbit corneal endothelial cells, the healing process was followed with wide-field specular microscopy. Individual cell migration and morphologic changes were analyzed by computer-assisted morphometry. The cells surrounding the wound migrated to cover the defect without producing intercellular gaps. The greatest cellular migration and morphologic alterations occurred close to the wound edge. As the cells migrated toward the wound, they elongated and increased their surface area in the direction of the migration. As the healing proceeded, the cells lost their original hexagonal pattern, which returned after coverage was complete. The wound was covered completely by large, irregularly shaped cells showing mitotic figures between 24 and 48 hr. During this period, cellular migration decreased and normal cellular morphology began to recover. When mitosis decreased, the normal cellular pattern rearranged towards a more hexagonal shape. During the healing process, the degree and direction of cellular migration varied from cell to cell. Additionally, changes in cell-to-cell contact (positional changes of neighboring cells) occurred in one-third of migrating cells. Such cellular migration can account for monolayered cells sliding without producing gaps between individual cells.

Variations of endogenous chicken proviruses: characterization of new loci of endogenous proviruses in the genome of Italian partridge chickens.

The composition and structure of endogenous proviruses present in the genome of Italian Partridge chickens were studied by the method of blot hybridization using RAV-2 [32P]DNA or LTR of RSV as hybridization probes. The genomes of 5 out of 39 chickens analyzed did not contain endogenous proviruses related to RAV-2. Different sets of five so far undescribed endogenous proviruses, differing in the structure and location, were detected in the DNA of other IP chickens. None of them is identical in its structure to the DNA of the endogenous chicken virus RAV-0, all five loci of endogenous proviruses of IP chickens were defective. The origin, the patterns of genetic variation and the function of endogenous proviruses are discussed.

Fetal gastric and small intestine pattern of intestinal mucus antigens in human gastric carcinomas.

The present immunohistological study was performed to investigate the expression of intestinal mucus-associated antigens in the different histological types of gastric carcinoma according to the classification of Laurèn and the WHO classification. We used the following antigenic markers: M3, present in all the goblet cells of the whole small and large intestine; M3SI, expressed in all the goblet cells of the small intestine, but only in some of them of the large intestine; M3D, mainly produced in the upper small intestine; and M3C, specific for colonic mucus cells. All these antigens were found to be similarly expressed in both Laurèn's intestinal and diffuse types. Nevertheless, M3 and M3C appeared to be more largely produced in carcinomas showing well-differentiated cells (tubulopapillary, mucinous, and signet ring cells according to the WHO classification). Our results evidenced the occurrence of two main fetal antigenic patterns in gastric carcinomas, one of the gastric type (M3SI produced to a larger extent than M3 and M3C) and the other one of the small intestinal type (M3 and M3SI more largely expressed than M3C). On the basis of their similar antigenic pattern, the histogenesis of carcinomas showing the fetal small intestinal antigenic profile may be associated with intestinal metaplasia. On the other hand, carcinomas with the fetal gastric pattern may originate from undifferentiated stem cells of the gastric mucosa. Thus, such immunohistological studies could lead to a better understanding of the histogenesis of gastric carcinomas.

Primary mediastinal seminoma.

Three new cases of primary mediastinal seminoma are presented, and 124 other cases in the literature are reviewed. These tumors primarily affect white males in the third decade. Chest pain is the most common symptom, but 20 per cent of the patients can be asymptomatic. Testicular atrophy occurred in only 7 patients. In one of these a testicular seminoma developed seven years after treatment of a primary mediastinal tumor. Supraclavicular adenopathies were found in 10 patients. Chest x-ray films typically show an anterior mediastinal noncalcified mass with smooth lobulated borders. Twenty-eight of 74 surgical resections attempted were successful; 13 of these received adjuvant irradiation. Of 64 cases in which radiation therapy was the primary mode of treatment, local failures occurred only in 7 cases. These tumors spread primarily by distant metastases. Theories of tumoral origin and patterns of spread are discussed. A dose of 3,000 rad in three weeks and 4,500 rad in four and one-half weeks to the mediastinum and supraclavicular areas is recommended for minimal and bulky disease. Primary mediastinal seminoma is as radiosensitive as its testicular counterpart.

The radiology of chronic neck pain: sequelae of occult traumatic lesions.

Occult traumatic lesions of the cervical spine usually involve the smaller, posterior elements of the vertebrae. Chronic neck pain associated with these lesions is intermittent and of variable severity, but with patterns that are fairly characteristic of the osseous lesions. Similarly, the long-term radiographic sequelae occur in fairly predictable patterns. The original osseous injury characteristically involves a fracture deformity and frequently some instability. There is a closed five point suspension system at each of the levels of the lower cervical spine, so that each of these five points is stressed whenever any one of them is deformed. Thus there follows bilateral hypertrophic changes and disc deterioration. Satellite changes also occur. The original deformity usually leads to rotation at the level of the lesion, cervical malalignment, and muscle imbalance. Therefrom the osseous and disc changes associated with chronic torticollis may occur, as well as distant instabilities.

Hemolytic uremic syndrome and crescentic glomerulonephritis complicating childhood nephrosis.

Three children with nephrotic syndrome and focal segmental glomerulosclerosis and/or mesangial proliferation on renal biopsy developed the sudden onset of renal failure, microangiopathic hemolytic anemia, and thrombocytopenia. Two of the three children developed crescentic glomerulonephritis and never regained renal function while the third showed no change from his original histologic pattern and also developed chronic renal failure. These cases suggest an association between the lipoid nephrosis-focal segmental glomerulosclerosis group of glomerular diseases and crescentic glomerulonephritis, and may represent an unusual pathway in the evolution of childhood nephrosis.
