Prediction of human drug metabolizing enzyme induction.

New chemical entities are routinely screened in vitro and in vivo for their ability to induce cytochrome P450s (CYP), other drug-metabolizing enzymes and possibly transporters in an attempt to more accurately predict clinical parameters such as drug-drug interactions and clearance in humans. Some of these potential therapeutic agents can cause induction of the metabolism of another molecule or auto-induction thereby increasing their own metabolism and elimination, as well as potentially any molecules metabolized by the same enzyme(s). Key CYPs in the 1A, 2B, 2C, and 3A families have all been shown to be inducible. It would be clearly advantageous to know the potential for a compound to induce drug metabolizing enzymes or transporters prior to clinical development, and many in vitro systems have been developed for this purpose. Newer computational technologies are also being applied in order to attempt to predict induction from the molecular structure alone before a molecule is even synthesized or tested. This review will cover the various in vitro and in silico methods developed for prediction of key inducers of CYPs and other proteins, as well as the limitations of such technologies and applications in the future.

Comparative effects of mono-, di-, tri-, and tetrasulfides derived from plants of the Allium family: redox cycling in vitro and hemolytic activity and Phase 2 enzyme induction in vivo.

Epidemiological evidence indicates that a high dietary intake of plants of the Allium family, such as garlic and onions, decreases the risk of cancer in humans. It has been suggested that this effect is due to the ability of the aliphatic mono-, di-, tri-, and tetrasulfides derived from these vegetables to increase tissue activities of Phase 2 detoxification enzymes. In contrast, toxic effects have been recorded in domestic and farm animals after the consumption of garlic or onions, involving oxidative damage to erythrocytes and consequent hemolytic anemia. This effect again has been attributed to the aliphatic sulfides. In the present study, the ability of sulfides derived from garlic and onions to generate "active oxygen" species and cause oxidative damage to erythrocytes in vitro has been compared, together with their ability to cause hemolytic anemia and increase the activity of the Phase 2 enzymes quinone reductase (QR) and glutathione S-transferase (GST) in rats. Monosulfides were without significant effect on any parameter. Di-, tri-, and tetrasulfides generated hydrogen peroxide in the presence of GSH and hemoglobin and caused oxidative damage to erythrocytes in vitro. The activity decreased in the order of tetra- > tri- > disulfide, with the allyl compounds being more potent than the propyl. In vivo, both allyl and propyl tri- and tetrasulfides were powerful hemolytic agents. In contrast, only the allyl sulfides increased the activities of QR and GST; the propyl derivatives were completely without effect. Allyl and propyl tri- and tetrasulfides, thus, may contribute to the toxic effects of Allium vegetables, while only the allyl derivatives are effective in increasing tissue activities of cancer-protective enzymes.

cis-terpin hydrate metabolism by a Brevibacterium: patterns of enzyme induction, and accumulation of -terpineol in growth.

A brevibacterium, strain TH-4, previously isolated by aerobic enrichment on the monocyclic monoterpenoid cis-terpin hydrate as a sole carbon and energy source, was found to grow on alpha-terpineol and on a number of common sugars and organic acids. Oxidation of these terpenoids was shown to occur via an induced enzyme system, as measured manometrically by oxygen uptake and prevention of protein synthesis with chloramphenicol or puromycin. Oxidation of terpin hydrate by cell suspensions appeared to be coincidentally induced by growth on alpha-terpineol, and oxidation of alpha-terpineol similarly appeared to be induced by growth on terpin hydrate. Culture fluids in which the TH-4 organism was grown at the expense of cis-terpin hydrate were found to contain (-)-alpha-terpineol in combined butanol-ether extracts. The isolated compound was shown to be chromatographically and spectrophotometrically identical to an authentic sample of alpha-terpineol. The stereospecificity of an enzymatic dehydration of terpin hydrate to alpha-terpineol is considered.

Rat lung antioxidant enzyme induction by ozone.

We exposed rats of different ages (weights approximately 45-300 g) to 0.7 ppm O3 for 1-5 days. At 5 days lungs of O3-exposed rats had higher activity of Cu,Zn superoxide dismutase (SOD), Mn SOD, catalase, and glutathione peroxidase than air-breathing rats; this greater activity was not due to blood-associated enzyme activity. The greater enzyme activity occurred with a higher concentration of the mRNA for each enzyme (Mn SOD not measured) without altered stability of these mRNAs. In adult rats the concentrations of these mRNAs were measured after 1, 3, and 5 days exposure to O3 and were elevated by day 3. The intergroup differences (air vs. O3) among antioxidant enzymes (AOEs) were unequal, and the intergroup differences in concentration of the specific AOE mRNA were greater than the differences in activity of their AOE. We conclude exposure to O3 led to greater expression of AOE genes; the increased expression was mediated pretranslationally probably at the level of transcription.

Amelioration of experimental allergic encephalitis (EAE) through phase 2 enzyme induction.

The multiple sclerosis (MS) lesion is characterized by an inflammatory cell mediated attack on white matter. Oxidative stress appears to play a role in the onset and progression of MS. We reasoned that decreasing oxidative stress might ameliorate MS. One way of decreasing oxidative stress is to induce phase 2 enzymes. The model chosen to test this hypothesis was experimental allergenic encephalomyelitis (EAE) induced in the Lewis rat. The 26 animals were placed into two groups: 1) those on normal rat chow, 2) those on rat chow containing 250 mumoles t-butylhydroxyanisole (BHA)/kg. After 2 weeks, animals were administered 100 micrograms guinea pig myelin basic protein and examined daily in a blinded fashion. Twenty-nine days later, animals were sacrificed, blood collected for glutathione (GSH) measurements and tissues collected for histology. Six of the 13 control chow animals developed hindlimb weakness or paralysis while 5 developed tail weakness only. Only 1 BHA fed animal exhibited symptoms--hindlimb weakness. Clinical symptoms correlated well with the extent of perivascular lymphocyte infiltration. Animals with BHA in the diet had 20% higher red cell GSH indicting induction of phase 2 enzymes. We conclude that dietary phase 2 enzyme inducers should be examined for their ability to ameliorate MS.

Effect of dimethylnitrosamine on enzyme induction in rat liver.

1. The effects of various doses of dimethylnitrosamine on the hydrocortisone induction of tryptophan pyrrolase were studied. A single LD(50) dose of dimethylnitrosamine inhibits the synthesis of the enzyme if given within the first 5hr. of the induction. A quarter of this dose also inhibits the synthesis of the enzyme, but, in addition, retards the rate of decay of the enzyme. 2. A single small dose of dimethylnitrosamine significantly inhibits the hydrocortisone induction of tryptophan pyrrolase for at least 14 days without causing widespread damage to liver substructure. 3. The inhibition by dimethylnitrosamine of induced tryptophan pyrrolase synthesis is probably independent of any action of the toxin on the synthesis of cofactors necessary for full expression of the enzyme's activity. Dimethylnitrosamine appears to act directly on the synthesis of the enzyme protein. 4. The synthesis of that form of the enzyme which is most sensitive to hydrocortisone is apparently also most susceptible to the action of dimethylnitrosamine. 5. It is suggested that the inhibition of protein synthesis by dimethylnitrosamine may be a result of methylation of messenger RNA, which then is unable to code effectively for amino acid polymerization.

[Enzyme induction in Streptomyces hydrogenans. IV. (1) Qualitative and quantitative changes in RNA content and RNA synthesis during induction].

For isolation of RNA, freeze-dried cells of Streptomyces hydrogenans were disrupted by grinding with kieselguhr. Application of diethylpyrocarbonate (diethyl oxydiformate) in an extraction procedure yielded undegraded nucleic acids of high purity. Best separation of extracted nucleic acids was achieved by electrophoresis on 2% mixed agarose-acrylamide gels. After application of 11beta, 21-dihydroxy-4,14 (20)-pregnadien-3-one to the culture medium, the amount of acid-precipitable RNA in the cells decreased to 50% within 20 min. Concimitantly, the rate of incorporation of precursors into RNA is much slower immediately after addition of the inducer but increases during the next 2 h. 3-4 h after induction there is no difference in the RNA content of induced and control cells. Degradation of stable as well as unstable RNA was observed. Simultaneous addition of inducer and rifamycin inhibits the production of 20beta-hydroxysteroid dehydrogenase, suggesting the synthesis of special mRNA for this enzyme. Based on experiments with antibiotics, the half-life of total mRNA was calculated to be 3 min in control cells, and about 4 min in induced cells. Using a double isotope labelling technique, we established the existence of specific mRNA in the induced cells. Together with the longer half-life of mRNA in the induced cells, the increased transcription may allow the 40-fold stimulation of the synthesis of 20beta-hydroxysteroid dehydrogenase.

The effect of repeated administration of diftalone on enzyme induction.

Plasma concentrations of diftalone have been examined in normal volunteers after a single dose (500 mg) and after 500 mg doses given twice daily for one week. An increase in post dosing urinary excretion of D-glucaric acid showed a correlation with the ratio of calculated to observed areas under the plasma concentration, time curve following the final dose in the multiple dosing studies, indicating that hepatic microsomal enzymes are induced after repeated administration of the drug. Single dose studies in the presence of aluminium hydroxide and sodium bicarbonate showed that the antacids had no significant effect on the absorption of diftalone.

Endotoxin inhibition of glucocorticoid enzyme induction and in vivo 3H-dexamethasone labelling of rat liver nuclei.

1. The effect of endotoxin on glucocorticoid (GC) induction of liver TO and TAT was investigated. 2. It was found that endotoxin inhibited not only TO GC induction, but also that of TAT, though to a lesser extent (17.41%). 3. Endotoxin did not influence the binding capacity of liver cytosol for 3H-dexamethasone at the second hour after the toxin administration. 4. In in vivo experiments endotoxin inhibited with 57.2% the binding of 3H-dexamethasone to hepatic nuclei. 5. It is suggested that the lower extent of endotoxin inhibition of GC induction of TAT may be due to the counteracting action of some inductor(s) for TAT only.

Effect of pesticide exposure on human microsomal enzyme induction.

1 The antipyrine half-life, metabolic clearance rate and the apparent volume of distribution were measured in six subjects handling malathion. These results were compared with six appropriate controls. 2 Occupational exposure of malathion in subjects reduced the antipyrine half-life from 9.18 +/- 2.16 h in controls to 4.73 +/- 1.65 h. 3 It is concluded that malathion exposure increases the rate of antipyrine elimination, possibly by inducing the hepatic microsomal enzymes.
