[Comparative biochemical study of 2 natural inactive variants of the actinomycin C producer Actinomyces sp. 26-115 with varying sensitivity to actinomycin].

Two natural variants of the actinomycin C-producing organism Actinomyces sp-26-115, i.e. H1 and H2 differ in their sensitivity to exogenic actinomycin, colony morphology, growth dynamics on the synthetic medium and stability to ultrasound and lysozyme. Both variants synthesize no actinomycin. Variant H1 is sensitive to exogenic actinomycin, while variant H2 is resistant to it. Variants H1 and H2 have some similarity in the composition of membrane proteins. Still, they differ in the protein molecular masses, which are equal to 600000--500000, 220000, 130000. The active variant A and nonactive variant H2 have the most similar compositions of membrane proteins. These variants are also close in their growth dynamics, colony morphology, sensitivity to ultrasound and lysozyme. The membranes of all the variants studied contain phosphatidyl ethanol amide as the main phospholipid component. Insignificant differences are observed only with respect to the minor components. The content of teichoic acids in the cell walls of variant H2 is very high, slightly changes during the developmental stage and insignificantly increases on addition of actinomycin to the medium. The cell wall of variant H1 contains less amounts of teichoic acids. During the developmental stage they are liberated from the wall at a higher rate than peptidoglycan. The sensitivity to actinomycin does not increase with an increase in the culture age. It is probable that teichoic acid of the cell wall is one of the factors providing resistance to actinomycin in variant H2. It may be considered as a barrier preventing transport of exogenic actinomycin into the cell.

[Actinomyces israelii. Methods of isolation and identification. Apropos of 10 cases].

Actinomyces israelii was isolated from cervico-facial subacute or chronic infections (9 cases), and abdominal wall abscess (1 case). Two conditions were necessary for successful isolation of A. israelii: properly collected clinical specimen (pus aspirated from abscess or biopsy) and prolongated incubation of anaerobic cultures (at minimum 5 days). Two commercially available micro-method multitest systems (Api 20 anaerobes, Api Zym test) were compared with standard biochemical procedures. The three methods were evaluated as to the reproducibility and help in identifying anaerobic non spore-forming Gram-positive bacilli in a routine clinical laboratory.

Actinomyces hordeovulneris, a canine pathogen that produces L-phase variants spontaneously with coincident calcium deposition.

The spontaneous production of cell-wall-deficient filaments and protoplasts by a strain of Actinomyces hordeovulneris (UCD 81-332-9) in 10% sucrose L-form media is reported. Multiple mineral deposits were present within the variants at 48 hours. Electron microscopy revealed that these deposits were amorphous, dense, and at the inner face of the cytoplasmic membrane in wall-less protoplasts and also in filaments which had a thin wall of 10 nm. These cell-wall-deficient variants produced L-form colonies when cultured for an additional 48 hours on 10% sucrose-BYE L-form plates. The colonies were composed of only a few filaments and many vesicles which were negative with Dienes' stain. Silver substitution stains of UCD 81-332-9 cells that had been grown in L-form broth for 5 days revealed heavy calcification of all cells including protoplasts. Gram stains of L-form grown cells revealed the presence of long-beaded, infrequently branched gram-positive filaments similar to those observed in clinical specimens. The formation of cell wall-deficient variants with coincident mineralization is believed to be related to the phenomenon of sulfur granule formation in vivo.

[Actinomyces-like organisms in users of intrauterine contraception].

Actinomyces-like organisms (ALO) were found in 6.9% of cervical smears in 2133 IUD users. The chance of having co-infection with Trichomonas vaginalis was 2.7 times higher in women with ALO than in ALO negative women. The proportion of IUD users with ALO in cervical smears increased with the duration of the IUD use. A total of 108 IUD users with ALO were compared with the same number of IUD users without ALO in cervical smears. The women in the two groups were matched for the duration of the IUD use. During the IUD use the women with ALO had PID and vaginitis significantly more frequently than ALO negative women. No difference was found regarding the type of the IUD used in women with ALO in cervical smears compared to controls.

[Heterogeneity of L-valine pool in Actinomyces sp. 26-115 producing actinomycin C].

Free intracellular valine in Actinomyces sp. 26-115 producing actinomycin C its functionally heterogenous. There are at least 2 pools of free valine. One of them supplies valine for protein biosynthesis and the second for the antibiotic biosynthesis. The volume of the pools is estimated. Inadequate reaction of the pools to similar effects is indicative of differences in their properties. The pool participating in protein biosynthesis strives for preservation of its volume. The pool participating in the antibiotic biosynthesis is capable of enlarging its volume to various levels depending on the change in the volume of the common intracellular pool.

[Composition and properties of cholesterol oxidase from Actinomyces lavendulae].

The composition of cholesterol oxidase isolated by different methods from the Actinomyces lavendulae mycelium was studied by gel-chromatography. It was shown that depending on the isolation conditions the enzyme can be detected within the complexes with different molecular weights. The values of pH, pI and Km for the isolated components were determined. The thermal inactivation of the cholesterol oxidase components at different temperature intervals was investigated.

[Inhibition of glucose isomerase from Actinomyces olivocinereus 154 by polyols].

The inhibitory action of polyols, e. g. D-mannitol, D-glucitol, ribitol and xylitol, on the activity of glucose isomerase prepared from Actinomyces olivocinereus 154 was studied. All the polyols under study are purely reversible competitive inhibitors. The values of Ki for D-mannitol, D-sorbitol, D-arabitol, D-glucitol, ribitol and xylitol are 0.200, 0.140, 0.030, 0.024 and 0.020 M, respectively. The inhibition is to some extent decreased by increasing concentrations of the substrate and is completely abolished by increasing concentrations of Mg2+ and Co2+--up to 5.10(-2) and 5.10(-3) M, respectively.

[Amino acid composition and quaternary structure of glucosoisomerase (d-xylose-ketol-isomerase) from Actinomyces olivocinereus 154].

The amino acid composition of glucosoisomerase from Actinomyces olivocinereus 154 was investigated. The content of dicarboxylic acids--aspartic and glutamic--was found to be greater than that of basic acids--lysine, arginine and histidine. Hydrophobic acids were also detected to occur on appreciable quantities. No cysteine was seen in the enzyme. The experimental data on the effect of sodium dodecyl sulfate and urea suggests that the enzyme has a quaternary structure consisting of four nonidentical subunits.

[Chitinolytic activity of Actinomyces kurssanovii under periodic and flow cultivation].

Chitinolytic activity of Actinomyces kurssanovii cultivated on the medium containing highly purified crab chitine as carbon source was studied. Under periodic cultivation the first to be accumulated were the (CHI)--enzyme that degraded ground chitine and beta-N-acetylglucosaminidase (EC 3.2.1.30). Then by the 15--16th hour of the producer growth chitinase-poly-(1,4-beta-(2-acetamido-2-deoxy-D-glucoside)-glucanohydrolase (EC 3.2.1.14) and beta-N-acetylglucosaminidase splitting N-N-diacetyl chitobiase were intensively accumulated. It was demonstrated that a highly active chitinolytic preparation of Act. kurssanovii can be obtained under flow cultivation at a dilution coefficient of D = 0.03--0.12 hr-1. A correlation between the respiratory activity of the culture and components of the chitinase system was established.

[Endo-1,3-beta-glucanase from Actinomyces cellulosae].

Endo-1.3-beta-glucanase (EC 3.2.1.39) from Actinomyces cellulosae was purified 960-fold by ion-exchange chromatography on DEAE-cellulose and DEAE-Sephadex A-50 and by gel-filtration through Acrylex P-60. The enzyme was electrophoretically homogeneous and had molecular weight of about 30 000 as determined by polyacrylamide gel disc-electrophoresis in the presence of SDS and gel-filtration through Sephadex G-200. The isoelectric point lies at 4.0; the pH optimum for the non-purified enzyme preparation is 5.5, that for the purified one is 6.0. The enzyme is stable within the pH range of 7.0-8.0 and is rapidly inactivated in acid media. The enzyme does not hydrolyze laminaribiose, lichenane, barley glucan, pustulane, KM-cellulose, but splits laminarane to form high molecular weight oligosaccharides, a small number of low molecular weight oligosaccharides and a negligible amount of glucose. Studies with glucose oxidase showed that splitting of 1.3-beta-glucan results in a beta-form of glucose.

Actinomyces meyeri osteomyelitis: an unusual cause of chronic infection of the tibia.

A 50-year-old man had draining fistulas of the right tibula for 15 years. He had been previously treated with antibiotic therapy of an unknown nature and by 3 sequestrectomies spaced over the 15-year interval, without a cure having been achieved. Several anaerobic bacteria, including Actinomyces meyeri were isolated from the bone lesion. The involvement of an actinomycete in osteomyelitis of a long bone is a very rare occurrence. After an additional sequestrectomy and penicillin therapy of an appropriate nature, clinical remission was achieved for the first time in 15 years.

[Highly purified glucosoisomerase from Actinomyces olivocinereus 154].

The paper presents a method for producing highly purified glucosoisomerase from Actinomyces olivocinereus 154. The scheme of purification includes enzyme extraction from dry biomass, acetone fractionation, ammonium sulfate precicipation, and Sephadex G-200 chromatography. The highly purified enzyme is homogeneous as shown by polyacryl amide gel electrophoresis, analytical ultracentrifugation, and gel filtration. The highly purified enzyme has been prepared in a crystalline form. The molecular weight of the enzyme has been estimated by sedimentation equilibrium to be 162 000 and by gel filtration to be 158 000. The sedimentation constant of glucosoisomerase has been found to be 8.52 S.

[Isolation and characterization of endo-N-acetylmuramidase produced by Actinomyces levoris].

Precipitation by ammonium sulfate and a subsequent purification of the culture fluid of Actinomyces levoris by gel-filtration through Sephadex G-25, ion-exchange chromatography on DEAE-cellulose and CM-cellulose resulted in an enzyme which activley lyzes the cell walls of a hemolytic streptococcus of group A. The molecular weight (12,500), isoelectric point (pI 10,6) and amino acid composition of the enzyme were determined. The enzyme specificity was assayed using peptidoglycane isolated rom the cell walls of streptococcus of group A used as a substrate. An analysis of the hydrolysis products of peptidoglycane showed that the enzyme under study is an endo-beta-N-acetylmuramidase (EC 3.2.1.17).

Effect of actinomyces israelii on human peripheral lymphocytes.

Actinomycosis is rare and responds well to antibiotic therapy. The causative organism, Actinomyces israelii which is now considered to be a bacterium, has long been suspected of exerting an immunosuppressive effect. The authors provide further evidence of this in their study of three patients with actinomycosis and four healthy control subjects. Skin reactivity to various standard antigens was depressed in all seven patients. Peripheral lymphocytes from the patients and the control subjects were isolated. A. israelii was added to the cell cultures following challenge with several standard mitogens. Blast transformation in response to phytohemagglutinin was inhibited by A. israelii in all seven subjects, but was preserved in response to concanavalin A and pokeweed mitogen. A. israelii appears capable of inhibiting the subset of human peripheral lymphocytes responsive to phytohemagglutinin stimulation in vitro.

Actinomyces naeslundii, acute cholecystitis, and carcinoma of the gallbladder.

This report describes the first case of biliary actinomycosis associated with an adenocarcinoma of the gallbladder. Actinomyces naeslundii was encountered as a pure isolate after a precise and careful identification. Although, in diagnosis, cancer and actinomycosis are often confused, the simultaneous occurrence of actinomycosis in cancer lesions can happen. This case illustrates the diagnostic challenge of actinomycosis.

[Actinomyces viscosus in subdural empyema].

The authors describe the finding of Actinomyces viscosus in pus from a subdural empyema in a child patient. When searching for the probable primary source of this uncommon infection of the central nervous system the authors identified by cultivation the same bacterial strain in a periapical dental granuloma. Bacterial diagnosis and radical surgical and antimicrobial therapy led to normalization of a clinical condition with an initially unfavourable prognosis.

Actinomyces pyogenes: susceptibility of 103 clinical animal isolates to 22 antimicrobial agents.

Actinomyces pyogenes induces suppurative diseases in ruminants and many other animal species. Most of the earlier antimicrobial susceptibility data has been obtained by disk diffusion techniques. Minimal inhibitory concentrations (MIC) of 22 antibiotics for 103 strains of A pyogenes of animal origin were determined by agar dilution test (Mueller-Hinton agar supplemented with 5% sheep blood). All the strains were susceptible to penicillin G, amoxicillin, methicillin, cephalothin, cefoperazone, pristinamycin, kanamycin, gentamicin, spectinomycin, chloramphenicol, vancomycin, novobiocin and rifampin. Fifty-nine percent were resistant to streptomycin, 67% to tetracycline, doxycycline and minocycline, 12% to erythromycin, spiramycin and lincomycin. Most of the strains resistant to macrolides and lincosamides exhibited a constitutive MLS(B)-like phenotype. In the cultural conditions used, it was not possible to determine accurate MIC of fucidic acid and pefloxacin.

[Isolation of the membranes of Actinomyces sp. 26-115, a producer of actinomycin C].

A summation fraction of the membranes of Actinomyces sp. 26-115 was obtained as a result of lysis of its protoplasts in a hypotonic medium. The qualitative content of protein, lipids, phospholipids, nucleic acids, glucosamine and muramic acid was determined in the membranes at various stages of the organism development. Phosphatidylcholine is the main component of phospholipids in this organism. Intracellular actinomycin was found inside the protoplasts. Electrophoregrams of the microprotoplasts and membranes are presented. Actinomycin was also detected in the membranes. Still, it is not clear whether it is a component of the membrane or it is adsorbed on the membrane during the process of its isolation. The final conclusion on the relationship between the membrane and localization of actinomycin in the cell requires further investigation.
