Quantitative investigations of the adhesiveness of circulating polymorphonuclear leucocytes to blood vessel walls.

1. A new simple method is described for quantitating the adhesiveness of circulating polymorphonuclear leucocytes, or granulocytes, to the walls of blood vessels. The cheek pouch of anaesthetized hamsters or a small part of the mesentery of anaesthetized mice were prepared for continuous microscopic observation of selected venules. Those granulocytes which moved sufficiently slowly to be individually visible were counted for 1 or 2 min periods as they rolled past a selected point on one side of a vessel. The velocity distribution of these cells was determined by analysing films. Films were used also to measure mean blood flow velocity in the venules by observing embolizing platelet thrombi induced by the iontophoretic application of adenosine diphosphate. Emigration of granulocytes into the tissues was quantitated by enumerating them in standard areas of stained histological sections.2. In control experiments with hamster cheek pouch venules, the rolling granulocyte count usually passed through a maximum shortly after the preparation was set up and then fell to a low constant value. In mouse mesentery venules the count remained at a low approximately constant value from the beginning for at least 3 hr.3. The mean velocity of blood flow in the venules was between 900 and 200 mu/sec. All rolling granulocytes moved much more slowly; in hamster cheek pouch venules the mean velocity was about 20 mu/sec and in mouse mesentery venules about 10 mu/sec. Around these means the velocity distribution of individual cells was narrow.4. Rolling of granulocytes was abolished by superfusing ethylenediamine tetra-acetate (EDTA, 0.1 M) suggesting that the phenomenon depends on calcium or magnesium ions.5. Agents were applied locally to the observed venules. Human serum albumin, trypsin or histamine in high concentrations did not affect the rolling granulocyte count.6. The rolling granulocyte count was increased during the application of Hammarsten casein or Escherichia coli culture filtrate which are chemotactic to granulocytes in vitro. These agents did not cause alterations in mean blood flow velocity in the observed venules which might have accounted for the effect on the rolling granulocyte counts. When E. coli culture filtrate was perfused through mouse intestine the increase in rolling granulocyte count in the draining venous blood was proportional to the logarithm of the concentration of filtrate.7. The rolling granulocyte count was also increased by the local application of plasma globulin permeability factor or lymph node permeability factor.8. Granulocyte counts in standard histological sections showed no significant increases in control preparations but considerable increases following the application of Hammarsten casein.

Hematopoietic and hematologic properties of bestatin in normal and cyclophosphamide myelosuppressed mice.

Bestatin is a potent inhibitor of aminopeptidase, an enzyme found in abundance in the membrane of monocytes and macrophages. Following binding of bestatin to cells in the histiocytic lineage, the production of colony stimulating activity is upregulated (both in vitro and in vivo) with subsequent increases in hematopoietic and hematologic parameters. We report that the frequency and absolute numbers of CFU-GM as well as entry of CFU-GM into S phase (a measure of progenitor cell activity) is upregulated by treatment of animals with bestatin. This results in an increase in bone marrow cellularity in cyclophosphamide suppressed mice and an increase in the absolute neutrophil count in normal and suppressed mice. The therapeutic application of this hematopoietic modulator has been demonstrated in combination cyclophosphamide and bestatin therapeutic protocols. Because this indication for bestatin has only recently been recognized, few clinical studies have been undertaken utilizing appropriate surrogates of hematopoietic activity. However, preliminary clinical evidence of hematopoietic activity by this non-toxic dipeptide, as reviewed here, suggests that this may be an appropriate strategy for the treatment of myelosuppressed patients.

Efficacy of broadcast and perimeter applications of S-methoprene bait on the red imported fire ant in grazed pastures.

The red imported fire ant, Solenopsis invicta (Buren), is a major pest in the United States because of its painful sting. Toxic bait has been an important management tool against fire ants, but site registrations prohibit applications of most baits on grazed pastures. Extinguish, containing the insect growth regulator methoprene, was selected for this study because it has a broad site registration that includes grazed pastures. The primary objective of this research was to evaluate the efficacy for control of red imported fire ants by using broadcast applications of methoprene bait at a label rate of 1,121 g/ha versus applications around the perimeter of a target area at the reduced rate of 280 g/ha. Grazed pastures in Lee County, Alabama, and Chambers County, Alabama, were selected for this study, with broadcast treatments, perimeter treatments, and controls replicated three times at each site. All mounds were counted and rated using the USDA population index before applications and then at 8 and 16 wk posttreatment. Perimeter applications did not significantly reduce S. invicta mound abundance, but bait treatments significantly reduced mound abundance at 16 wk posttreatment at site 1 where applications were conducted in early evening. However, broadcast applications were not effective at site 2 where treatments were conducted in early morning with warmer temperatures. Emergence of winged alates was observed at 12 wk posttreatment, followed by a high density of incipient mounds that may have masked the full treatment effect of methoprene applications at site 2. Methoprene bait was effective in reducing abundance of S. invicta only when full label rates were applied.

A microtiter plate assay using cellulose acetate filters for measuring cellular [3H]serotonin release.

An assay system for measuring the release of tritium-labeled serotonin that accompanies cellular degranulation is described. The assay is carried out with [3H]serotonin loaded rat basophilic leukemia (RBL) cells that have been sensitized with IgE, and aliquots of these are combined with dilutions of specific antigen in the wells of a microtiter plate together with appropriate control samples. The released activity in all of the wells is harvested simultaneously and quickly using commercial cellulose acetate filters which are counted following addition of scintillation fluor. In many applications this plate assay is faster and more convenient than the conventional test tube assay, and the results obtained are shown to be comparable. For the plate assay, the effect of cell concentration and some other conditions are shown. In general it is found that greatly improved results can be obtained if the cells are cultured overnight during the [3H]serotonin incorporation. With this assay it is shown that RBL cells also release incorporated 51Cr specifically during degranulation.

[The course of pregnancy, delivery and puerperium in women with varices and thrombophlebitis of lower extremities, after application of low molecular weight heparins].

OBJECTIVES: Estimation of the long term prophylactic or therapeutic application of low molecular weight heparin (LMWH) on the platelets count, and incidence bleedings during pregnancy, delivery and puerperium in the women with varices of lower extremities and past thrombophlebitis of lower extremities. MATERIAL AND METHODS: 5212 pregnant, women in labour and in puerperium divided into 4 groups.; 142 women with varices and thrombophlebitis of lower extremities (group I); 10 with past thrombophlebitis of lower extremities (group II); 15 with thrombophlebitis in current pregnancy; 5045 without vascular complications (group IV--control). In group I during pregnancy compression therapy was applied (stockings) and low molecular weight heparins (LMWH) in course of puerperium. In group II during the 1st trimester of pregnancy and in labour the same heparin doses were administered, while the doses were increased in the 2nd and 3rd trimester. In group III, when thrombophlebitis was stated non-steroid anti-inflammatory drugs and LMWH were administered. In all cases treated with heparin both number of platelets and incidents of bleedings from genitourinary tract were observed. Presence of embolic complications was also noted. RESULTS: No cases of decrease platelets number or bleedings from genitourinary tract were observed in group I-III during administering of LMWH. In women in group II where prophylactic with LMWH was applied no incidences of recurrent thrombophlebitis during pregnancy and puerperium were observed. In group I-III all newborns were born in good condition and no complications were observed. Average blood loss during both labour and cesarean section, among women in group I-III was not significantly different comparing with control group. No incidences of pulmonary artery embolism or decrease number of platelets were observed. CONCLUSIONS: 1. The long term prophylactic or therapeutical administration of LMWH in the women with varices of lower extremities or thrombophlebitis has no influence on the platelets count and incidence of bleedings from genitourinary tract during pregnancy or increase of blood loss during labour and puerperium. 2. In the women with past thrombophlebitis of the lower extremities after application of LMWH during pregnancy there were no recurrence observed.

The relationship between lumbar segmental motion and pain response produced by a posterior-to-anterior force in persons with nonspecific low back pain.

STUDY DESIGN: Cross-sectional. OBJECTIVE: To investigate the association between lumbar segmental motion and pain response during the application of a posterior-to-anterior (PA) force to the lumbar spinous processes in persons with nonspecific low back pain. BACKGROUND: Although low back pain is believed to be associated with altered segmental motion of the lumbar spine, the relationship between subjective reports of pain and objective measurements of segmental motion has not been established. METHODS AND MEASURES: Thirty-five individuals between 18 and 45 years of age with nonspecific low back pain (less than 3 months' duration) participated. All subjects participated in 2 separate procedures: (1) segmental motion assessment during a PA force application over the lumbar spinous processes using dynamic magnetic resonance imaging (MRI), and (2) pain assessment during a PA force application procedure outside of the MRI environment. Frequency counts were used to determine the lumbar segments that were most painful, and which functional spinal units had the most and least motion. Fisher exact tests were performed to determine if an association existed between the most painful segment and the functional spinal unit with the most or least motion. RESULTS: L5 was deemed the most painful segment in nearly half of the participants (48.1%). The L1-2 and L3-4 functional spinal units most frequently had the most motion (25.9% each) and the L4-5 functional spinal units most frequently had the least motion (29.6%). No association was found between the most painful segment and the functional spinal units with either the most or least motion. CONCLUSION: The results of this study indicate that an assumption regarding segmental motion cannot be inferred from the pain response when using a PA force application procedure.

Seeing through the stratum corneum.

The stratum corneum (SC) provides a vital barrier membrane between the external environment and the vulnerable internal tissues of the skin. It impedes the flow of water, the penetration of xenobiotics, and invasion of pathogenic micro-organisms. It also has protective capacity against ultraviolet radiation and thermal injury. As routine histopathology provides a misleading picture of a disorganized and shadowy SC, we would recommend the skin surface biopsy technique. This painless technique is easy and reliable in obtaining information from the SC. It demonstrates the geometric patterns of the surface, the openings of the eccrine ducts and hair follicles. The skin surface biopsy technique is also ideal for the investigation of the in situ microbiology of skin. Staining with periodic acid Schiff reagent makes it possible to see ringworm fungi, pityriasis versicolor, candida species, or erythrasma micro-organisms. Scanning electron microscopy can be employed when the higher magnification is needed. Histochemical applications include silver staining for melanin particle, potassium ferricyanide staining for blood pigments and lipid staining with Sudan red, for sebum. The rate of movement of topically applied drugs into the skin can be measured using the skin surface biopsy technique. The concentration of radiolabelled drugs can be counted and compared. Comedogenicity and DNA analysis are other applications of this non-invasive technique.

A new assay for leukocyte chemotaxis using cell retrieval, electronic particle counting and flow cytometry.

A new micropore membrane assay for leukocyte migration has been devised. It permits the complete retrieval in monodisperse suspension of functionally intact cells that have traversed the membrane, thus allowing the application of precise, automated techniques, including flow cytometry and electronic particle counting. Hemocytometers may also be used. Direct comparison with 2 different conventional membrane methods showed that the new method performed superiorly. It was also much more economical with regard to time and labor. This technique permitted detection of functional differences between leukocytes isolated from blood in different ways. Data on the duration of concentration gradients in chemotaxis chambers are also presented.

An application of a two-level non-Gaussian state-space model in the analysis of longitudinal papilloma count data.

In this study, a dynamic Bayesian two-level non-Gaussian state-space model is applied in the statistical analysis of longitudinal detectable papilloma count data. This two-level model is established on the basis of the state and hyper-state parameters that depend on the model parameters of biological significance, namely, the initiation rate of normal cells and the birth- and death-rates of initiated cells. As the time-dependent model parameters fluctuate dynamically and stochastically over time, so will the state and hyper-state parameters; thus, smoothness priors that allow a wide range of shapes with no specific forms required on the state/hyper-state parameters can be employed in this model. Gibbs sampler, a Markov chain Monte Carlo approach, is used to implement the Bayesian inferences on the state as well as the hyper-state parameters; and the estimates of the model parameters can be obtained thereon. Illustrations of the Bayesian inference procedure are given by using the datasets from a simulation study as well as a laboratory experiment by Brook et al. [E.A. Brooks, C.M. Kohn, P.J.M. van Birgelen, G.W. Lucier, C.J. Portier, Stochastic models for papilloma formation following exposure to TCDD, Organohalogen Compd. 41 (1999) 521]. Comparing to the parametric approach that assigns specific forms on the time-dependent model parameters, different conclusions are drawn by the two-level state-space modeling approach considered in this study.

Effect of hyperthermic water bath on parameters of cellular immunity.

Effects of hyperthermic water bath on selected immune parameters (lymphocyte subpopulations, natural killer (NK) cell counts and their activity) were studied in a group of 10 volunteers. Application of hyperthermic water bath (both topical and whole-body) was followed by a significant reduction of relative B lymphocyte counts. Whole-body hyperthermic water bath reduced relative total T lymphocyte counts, increased relative CD8+ T lymphocyte and NK cell counts and increased NK activity. Whole-body hyperthermic bath increased somatotropic hormone (STH) activity in eight out of 10 volunteers; higher relative counts of CD8+ lymphocytes and NK cells were observed compared with the group of volunteers not responding to hyperthermic water bath by STH secretion. In five volunteers STH was released in response to local hyperthermic water bath and the NK activity of lymphocytes also increased but their relative counts did not. The results suggest that these increases in CD8+ lymphocyte and NK cell counts are probably dependent on increased STH production.

Purine loading, stem-loops and Chargaff's second parity rule: a discussion of the application of elementary principles to early chemical observations.

DNA base compositions were determined chemically long before sequencing technologies permitted the direct counting of bases. Some recent observations made using modern sequencing technologies could have been deduced by application of elementary principles to early chemical observations. This paper draws attention to the fact that the potential for significant stem-loop structure is a general property of single-stranded DNA (genic and non-genic) and hence for any corresponding transcripts, whether they function by virtue of their structure (eg rRNA) or as mRNA. Furthermore, there is Chargaff's second parity rule: in single strands, the percentage of purines approximately equals the percentage of pyrimidines. Since, in stems, purines match pyrimidines, Szybalski's rule that transcripts violate the second parity rule in favour of purines, must apply to loops. Since purine loading occurs in both mesophilic and thermophilic species, genes with transcripts that need stable secondary structures for functioning at high temperatures must achieve this by selectively increasing the GC percentage (GC%) of stems, while retaining purine loading of loops. Arguments that purine loading is specific for the loops of RNA-synonymous strands of genes whose transcripts function by virtue of their secondary structure (ie rRNAs, not mRNAs) need to take into account, as controls, the loop regions of mRNA-synonymous strands. Entire genes, or entire genomes where gene orientation is not considered, are not appropriate controls.

[Quantitative analysis in nuclear nephrourology: clearance, uptake and transit time].

Radionuclide renal study is only a method that can provide anatomical as well as functional information on the kidney. There have been many reports on quantification of the renal function using radionuclides. They are summarized to two groups: blood and/or urine sampling method and external counting method. The former is accurate and reproducible in quantification of the renal function, but is rarely applicable in a routine practice due to technical complexity. On the contrary, the external counting method by means of a gamma camera and dedicated computer system is simple and practical in a routine use, but is less in reliability of quantitated renal function than sampling method. In this paper, technical and diagnostic characteristics of the quantitative methods on renal function using the radionuclide are reviewed.

A calculation and number processing battery for clinical application in illiterates and semi-literates.

Ten simple tasks assessing counting, number processing, elementary calculation and quantity estimation were proposed to 122 normal Brazilian adults aged between 18 and 58 years with 0, 1, 2, 3 or 4 years of education. Tasks such as counting the number of elements in small sets were almost perfectly mastered by these illiterate or semi-literate normal subjects; however in other tasks (e.g. those assessing knowledge of the correspondence between numbers and banknotes) a sizeable proportion of the sample showed errors. The pattern of errors was analysed to identify difficulty factors. A strong gender effect with better performance in men than women was observed, which was even greater than the expected effect of educational level. Results in normals allowed to propose cut-off scores for neuropsychological assessment in brain-damaged patients with very low levels of education, which were tested in a small sample of illiterate or semi-literate patients with cerebrovascular accident. It is argued that the relatively neglected area of neuropsychological assessment in illiterates is of great practical and theoretical interest.

Detection of platelet-derived microparticles using flow cytometry and its clinical application.

OBJECTIVE: To establish a flow cytometric internal standard method for counting platelet-derived microparticles (PMPs) and to study its clinical significance. METHODS: PMPs suspension (platelet poor plasma, PPP) was extracted by gradual centrifugation. According to the size of PMPs, 3 microm and 0.8 microm latex beads were used as internal standards for the quantitation. PMPs were counted by adjusting flow cytometric discrimination and voltage of forward scatter and side scatter. RESULTS: In 30 healthy donors, the average concentration of resting PMPs was (1.2 x 10(5) +/- 5.7 x 10(4))/ml and that of activated PMPs was (1.6 x 10(6) +/- 9.1 x 10(5))/ml. Compared with healthy donors, PMPs mean value was significantly higher (P < 0.001) in 18 patients with coronary artery disease, 12 with acute cerebral infraction and 23 with chronic renal failure [the average PMPs concentration, (6.1 x 10(5) +/- 2.5 x 10(5))/ml, (6.8 x 10(5) +/- 3.4 x 10(5))/ml and (5.9 x 10(5) +/- 3.1 x 10(5))/ml respectively]. However, no significant difference in PMPs concentration was observed in 25 patients with acute leukemia and severe thrombocytopenia during the aplastic phase after chemotherapy [(1.3 x 10(5) +/- 6.1 x 10(4))/ml, (P > 0.05)]. CONCLUSIONS: PMPs is a useful indicator in monitoring platelet activation, and plays an important role in thrombotic disease. By flow cytometric internal standard method, PMPs can be counted rapidly and accurately, which may be very helpful in interlaboratory comparative studies.

Multi-level zero-inflated poisson regression modelling of correlated count data with excess zeros.

Count data with excess zeros relative to a Poisson distribution are common in many biomedical applications. A popular approach to the analysis of such data is to use a zero-inflated Poisson (ZIP) regression model. Often, because of the hierarchical study design or the data collection procedure, zero-inflation and lack of independence may occur simultaneously, which render the standard ZIP model inadequate. To account for the preponderance of zero counts and the inherent correlation of observations, a class of multi-level ZIP regression model with random effects is presented. Model fitting is facilitated using an expectation-maximization algorithm, whereas variance components are estimated via residual maximum likelihood estimating equations. A score test for zero-inflation is also presented. The multi-level ZIP model is then generalized to cope with a more complex correlation structure. Application to the analysis of correlated count data from a longitudinal infant feeding study illustrates the usefulness of the approach.

The effect of a probiotic Enterococcus faecium product in diets of healthy dogs on bacteriological counts of Salmonella spp., Campylobacter spp. and Clostridium spp. in faeces.

An experiment was conducted to determine the effects of a probiotic on selected faecal bacteria of healthy dogs under different feed and environmental conditions. For the study 12 dogs kept in households were used for an 18-day supplementation with a patented commercially available strain of E. faecium NCIB 10415 (Enteroferm). In order to minimize losses the probiotic product was orally applicated once a day before meals at a dose of 2 g per dog (9.2 x 10(9) CFU). The faeces were collected before the beginning of the supplementation and at the end of the 18-day application period. In order to exclude contamination, all faeces were taken rectally. Before and at the end of the experimental period total Salmonella spp., Campylobacter spp. and Clostridium spp. counts were determined in fresh faeces using selective media. It was demonstrated that the 18-day application of the probiotic E. faecium product induced modifications on the gastrointestinal microflora in all dogs. While Salmonella spp. and Campylobacter spp. counts were in majority of the dogs higher than before the application. Clostridium spp. counts were significantly reduced in 10 of 12 dogs. According to the guidelines for the evaluation of the efficiency of microorganisms in dogs a relevant efficacy effect was supported by this data. However, a beneficial effect of the probiotic product on healthy dogs remains questionable.

Using the Microcyte flow cytometer to monitor cell number, viability, and apoptosis in mammalian cell culture.

The Microcyte is a novel, portable flow cytometer based on diode laser technology whose use has been established for yeast and bacterial analysis. We present data that demonstrate its suitability for routine mammalian cell counting and viability determination. To extend its range of applications in the field of animal cell culture biotechnology, a test to determine the number of apoptotic cells present has been developed for use with the instrument. Apoptosis was induced in hybridoma cell cultures by treatment with camptothecin. Apoptotic cells were labeled with biotinylated Annexin V and then visualized using a streptavidin-allophycocyanin conjugate. Their numbers were counted, and the cell size of the apoptotic cell population was determined using the Microcyte.

Patient-orientated metered capillary blood collection.

A small collector bottle, with a precision-moulded top, allows capillary blood from a finger-prick to be automatically metered into a diluent for subsequent laboratory analysis. This device, which has been successfully used by patients to obtain full blood counts, glycosylated haemoglobin values and blood glucose measurements, may have wide application.
