Reliability of transfer function estimates in cardiovascular variability analysis.

Transfer function (TF) analysis is a widely diffused technique in the assessment of the relationship between short-term cardiovascular variability signals, particularly blood pressure, heart rate and respiration. To guarantee the reliability of the estimates, a conventional threshold of 0.5 on the magnitude squared coherence (MSC) is commonly used, although (i) other analysis parameters play a role and (ii) lower values of MSC are frequently unavoidable in physiological systems. In this study, computer simulations are performed to assess the dependency of the bias and standard deviation (SD) of TF estimates on record length (RL), spectral window bandwidth (Bw) and MSC; to evaluate the accuracy of theoretical expressions for the computation of the confidence interval (CI) of the estimates; and to assess, in some representative situations, how faithfully observed TF shapes reproduce the underlying true functions in conditions of very low MSC. The accuracy of TF estimates increases non-linearly with increasing RL, and the benefit over 7 min is small. Using this RL, the relative bias for the TF modulus is < 10% for MSC > 0.2. Estimates of TF phase are unbiased. The SD of both the modulus and phase increases linearly as the MSC decrease to 0.4 and then, for lower MSC, increases markedly with nonlinear behaviour. Bw= 0.03Hz appears to be most suitable to reduce the error, preserving spectral resolution. CIs for the TF phase are highly reliable, whereas those for the modulus tend to be slightly narrower than the nominal value at high coherence values. Major features of the TF shape appear to be preserved in simulations with very low MSC. The major problem in TF estimation is the sharp increase in the variability of the measurements as the coherence decreases towards the lowest values. The combination of RL > or = 420s and Bw= 0.03Hz should be suggested in short-term cardiovascular variability studies. Although basic features of the true TF can be recovered even when the MSC is < 0.5, much greater values can be necessary when accurate point estimates are needed. Theoretical expressions for the computation of confidence intervals of the TF are adequate for practical purposes.

Vowel and consonant recognition of cochlear implant patients using formant-estimating speech processors.

Vowel and consonant confusion matrices were collected in the hearing alone (H), lipreading alone (L), and hearing plus lipreading (HL) conditions for 28 patients participating in the clinical trial of the multiple-channel cochlear implant. All patients were profound-to-totally deaf and "hearing" refers to the presentation of auditory information via the implant. The average scores were 49% for vowels and 37% for consonants in the H condition and the HL scores were significantly higher than the L scores. Information transmission and multidimensional scaling analyses showed that different speech features were conveyed at different levels in the H and L conditions. In the HL condition, the visual and auditory signals provided independent information sources for each feature. For vowels, the auditory signal was the major source of duration information, while the visual signal was the major source of first and second formant frequency information. The implant provided information about the amplitude envelope of the speech and the estimated frequency of the main spectral peak between 800 and 4000 Hz, which was useful for consonant recognition. A speech processor that coded the estimated frequency and amplitude of an additional peak between 300 and 1000 Hz was shown to increase the vowel and consonant recognition in the H condition by improving the transmission of first formant and voicing information.

Rapid fat/water assessment in knee bone marrow with inner-volume RARE spectroscopic imaging.

An inner-volume modification of a spectroscopic imaging technique based on RARE (rapid acquisition with relaxation enhancement) sequences is shown to provide, in less than 1 minute, spectra suitable for fat/water peak-area integration from selected imaging columns containing more than 1,000 5 x 5 x 1-mm voxels. A limitation is the unavoidable T2 weighting of the spectral peaks, which influences estimations of true fat/water composition from the spectra. Studies obtained in the knees of healthy volunteers demonstrate the ability of the technique to enable characterization of the two major proton peaks in bone marrow. Data collected at two or more effective TEs are suitable for extrapolation of fat/water estimates to zero TE.

NMR spectral quantitation by principal-component analysis. II. Determination of frequency and phase shifts.

This paper extends the use of principal-component analysis in spectral quantification to the estimation of frequency and phase shifts in a single resonant peak across a series of spectra. The estimated parameters can be used to correct the spectra accordingly, resulting in more accurate peak-area estimation. Further, the removal of the variations in phase and frequency cause by instrumental and experimental fluctuations makes it possible to determine more accurately the remaining variations, which bear biological significance. The procedure is demonstrated on simulated data, a 3D chemical-shift-imaging dataset acquired from a cylinder of inorganic phosphate (Pi), and a set of 736 31P NMR in vivo spectra taken from a kinetic study of rate muscle energetics. In all cases, the procedure rapidly and automatically identifies the frequency and phase shifts present in the individual spectra. In the kinetic study, the procedure is used twice, first to adjust the phase and frequency of a reference peak (phosphocreatine) and then to determine the individual frequencies of the Pi peak in each of the spectra which further can be used for estimation of pH changes during the experiment.

A new method of the description of the information flow in the brain structures.

The paper describes the method of determining direction and frequency content of the brain activity flow. The method was formulated in the framework of the AR model. The transfer function matrix was found for multichannel EEG process. Elements of this matrix, properly normalized, appeared to be good estimators of the propagation direction and spectral properties of the investigated signals. Simulation experiments have shown that the estimator proposed by us unequivocally reveals the direction of the signal flow and is able to distinguish between direct and indirect transfer of information. The method was applied to the signals recorded in the brain structures of the experimental animals and also to the human normal and epileptic EEG. The sensitivity of the method and its usefulness in the neurological and clinical applications was demonstrated.

Persistent effects of high frequency repetitive TMS on the coupling between motor areas in the human.

Repetitive transcranial magnetic stimulation (rTMS) shows promise as a treatment for various movement and psychiatric disorders. How rTMS may have persistent effects on cortical function remains unclear. We hypothesised that it may act by modulating cortico-cortical connectivity. To this end we assessed cortico-cortical coherence before and after high frequency rTMS of the motor cortex. Sixteen healthy subjects received a single train (5 Hz, active motor threshold, 50 stimuli) of rTMS to the left motor hand area. Spectral power and coherence estimates were calculated between different EEG signals at rest and while muscles of the distal upper limb were tonically contracted. Repetitive TMS over the left motor hand area caused a significant decrease in the intrahemispheric EEG-EEG coherence between motor and premotor cortex in the 10.7-13.6 Hz (upper alpha band) lasting a few minutes after stimulation. There was no significant change in interhemispheric EEG-EEG coherence between motor areas. Thus, high frequency rTMS of the motor cortex decreases ipsilateral cortico-cortical intrahemispheric in the upper alpha band.

Status and progress in on-line spectrometric monitoring and control of plant nutrient solutions.

Biotronics has been involved, under NASA sponsorship, in a wide ranging research and development program for instrumentation used in the monitoring and control of controlled environment agriculture. This program has embraced both chemical monitoring of plant nutrient solutions as well as microbiological monitoring of bacteria and fungi in these same solutions. This paper emphasizes the microbiological monitoring aspects of this program. In contrast to traditional methods of microbiological analysis based on culturing, staining and microscopic observation, the development described here is based on spectroscopic measurements, more specifically spectral fluorometry. The rationale, objectives, analytical methods and new instrumentation employed in the development of an on-line microbiological analyzer (MBA) are presented in some detail. Finally, the signal processing/pattern recognition methods used to evaluate the spectral data and produce estimates of microbial populations are described along with experimental test results to conclude the paper.

Circular dichroism and magnetic circular dichroism spectra of chlorophylls a and b in nematic liquid crystals. II. Magnetic circular dichroism spectra.

Absorption and magnetic circular dichroism (MCD) spectra are reported for chlorophyll (Chl) a and Chl b dissolved in nematic liquid crystal solvents. The spectra were measured with the dye molecules oriented uniaxially along the direction of. the magnetic field and measuring light beam. It is significant that under such conditions the MCD spectra recorded in the wavelength region of the Q and Soret bands of the chlorophyll are essentially unchanged with respect to rotation of the sample cell around this axis, even though there is almost complete orientation of the chlorophyll molecules by the liquid crystals. The MCD spectra of Chl a and b in the nematic liquid crystal solvents used in this study are surprisingly similar to the spectra obtained under isotropic conditions. These results illustrate an important technique with which to examine the optical spectra of dyes oriented in liquid crystal matrices in which the anisotropic effects can be reduced the negligible proportions by the application of a strong magnetic field parallel to the direction of the measuring light beam. The first deconvolution calculations are reported that describe the deconvolution of pairs of absorption and MCD spectra, in the Q and B band regions, for both Chl a and b. The spectral analysis to obtain quantitative estimates of transition energies was accomplished by carrying out detailed deconvolution calculations in which the both the absorption and MCD spectral envelopes were fitted with the same number of components; each pair of components had the same hand centres and bandwidth values. This procedure resulted in an assignment of each of the main transitions in the absorption spectra of both Chl a and b. Chl a is clearly monomeric, with Qy, Qx, By and Bx located at 671, 582, 439 and 431 nm, respectively. Analysis of the spectral data for Chl b located Qy, By and Bx, at 662, 476 and 464 nm, respectively.

Quality control of herbal medicines.

Different chromatographic and electrophoretic techniques commonly used in the instrumental inspection of herbal medicines (HM) are first comprehensively reviewed. Chemical fingerprints obtained by chromatographic and electrophoretic techniques, especially by hyphenated chromatographies, are strongly recommended for the purpose of quality control of herbal medicines, since they might represent appropriately the "chemical integrities" of the herbal medicines and therefore be used for authentication and identification of the herbal products. Based on the conception of phytoequivalence, the chromatographic fingerprints of herbal medicines could be utilized for addressing the problem of quality control of herbal medicines. Several novel chemometric methods for evaluating the fingerprints of herbal products, such as the method based on information theory, similarity estimation, chemical pattern recognition, spectral correlative chromatogram (SCC), multivariate resolution, etc. are discussed in detail with examples, which showed that the combination of chromatographic fingerprints of herbal medicines and the chemometric evaluation might be a powerful tool for quality control of herbal products.

Spatial dependency of trapezius muscle activity during repetitive shoulder flexion.

The purpose of this study was to explore changes in spatial muscle activation within the three divisions of the trapezius muscle during a dynamic, cyclic task of the upper limb. Surface EMG signals were detected from thirteen healthy subjects from the upper, middle and lower divisions of the trapezius muscle at multiple electrode sites in the cephalad-caudal direction during a repetitive shoulder flexion task. Initial values and rate of change of average rectified value (ARV) and of instantaneous mean power spectral frequency (iMNF) were estimated at 45 degrees , 90 degrees and 120 degrees of shoulder flexion throughout the 5-min task. The location of the electrodes had a significant effect on initial EMG ARV for both the upper and middle division of the trapezius muscle (P<0.05). Both the rate of change and normalized rate of change of ARV were greatest for the most cranial muscle fibers of the upper division (P<0.05). Initial values and rates of change of iMNF were also affected by electrode location for the upper and lower divisions of the trapezius muscle (P<0.05). These results demonstrate that muscle activity and its changes over time depend on position within the three divisions of the trapezius muscle during a dynamic, cyclic task of the upper limb. This suggests non-uniform muscle fiber distribution and/or recruitment. The results also highlight the importance of multiple recording sites when investigating trapezius muscle function in dynamic tasks.

The single basis filter diagonalization method: a rapid multidimensional data processing scheme.

A new way to apply the filter diagonalization method (FDM) that results in a large increase in the speed of calculation of multidimensional NMR spectra is presented. The speed increase is accompanied by slight differences in spectral lineshapes, although frequency estimates remain essentially identical. For contoured spectra, the method does not result in appreciable differences from the full FDM calculation. Optimal parameter sets for an FDM calculation can be estimated far more rapidly, which makes the FDM more straightforward to employ in practice. The performance of the method versus the full FDM was investigated for both model and experimental signals. The effect of noise on the method was also studied.

Spectrophotometric determination of acidity constants of alizarine red S in water, water-Brij-35 and water-SDS micellar media solutions.

The acidity constants of Alizarine Red S in water, water-Brij-35 and water-SDS micellar media solutions at 25 degrees C and an ionic strength of 0.1 M have been determined spectrophotometrically. To evaluate the pH-absorbance data, a resolution method based on the combination of soft- and hard-modeling is applied. The acidity constants of all related equilibria are estimated using the whole spectral fitting of the collected data to an established factor analysis model. DATAN program applied for determination of acidity constants. Results show that the pKa values of Alizarine Red S are influenced as the percentages of a neutral and an anionic surfactant such as Brij-35 and SDS, respectively, added to the solution of this reagent. Effect of surfactant on acidity constants and pure spectrum of each component are also discussed.

Monitoring imaging of lesions induced by high intensity focused ultrasound based on differential ultrasonic attenuation and integrated backscatter estimation.

We investigated the feasibility of two monitoring imaging methods to visualize and evaluate the high intensity focused ultrasound (HIFU) induced lesions in vitro during and after their formation, which were based on differential ultrasonic parameter estimation. Firstly, ultrasonic attenuation slope of tissue sample was estimated based on the spectral analysis of ultrasound RF backscattered signals. The differential attenuation slope maps were acquired, which were interpreted as the differences between the pretreatment image and those obtained in different stages during HIFU therapy. Secondly, ultrasonic integrated backscatter (IBS), defined as the frequency average of the backscatter transfer function over the useful bandwidth, was proposed quantitatively to evaluate the extent of lesions with the same RF signals as the first method. Differential IBS maps were also acquired to visualize temporal evolution of lesion formation. It was found in pig liver in vitro that more precise definition of the treated area was obtained from the differential IBS images than from differential attenuation slope images. Dramatic increase in both attenuation and IBS value was observed during the therapy, which may be related to dramatic enhancement of cavitation due to boiling and accompanying tissue damage. Two methods to obtain one differential image were compared and the cumulative differential image was found to be able to eliminate noises and artifacts to some extent, which was the cumulation of a series of differential images acquired from the differences between the temporally adjacent RF data frames. Moreover, we presented a bidirectional color code for identification of the artifacts due to tissue movements caused by HIFU radiation force. We conclude that cumulative differential IBS images have the potential to monitor the formation of HIFU-induced lesions.

A Fourier method for the analysis of exponential decay curves.

A method based on the Fourier convolution theorem is developed for the analysis of data composed of random noise, plus an unknown constant "base line," plus a sum of (or an integral over a continuous spectrum of) exponential decay functions. The Fourier method's usual serious practical limitation of needing high accuracy data over a very wide range is eliminated by the introduction of convergence parameters and a Gaussian taper window. A computer program is described for the analysis of discrete spectra, where the data involves only a sum of exponentials. The program is completely automatic in that the only necessary inputs are the raw data (not necessarily in equal intervals of time); no potentially biased initial guesses concerning either the number or the values of the components are needed. The outputs include the number of components, the amplitudes and time constants together with their estimated errors, and a spectral plot of the solution. The limiting resolving power of the method is studied by analyzing a wide range of simulated two-, three-, and four-component data. The results seem to indicate that the method is applicable over a considerably wider range of conditions than nonlinear least squares or the method of moments.

Measurement of ataxic symptoms with a graphic tablet: standard values in controls and validity in Multiple Sclerosis patients.

Aim of our study was to find a specific measure for the intensity of upper limb tremor and other ataxic symptoms in Multiple Sclerosis (MS) patients, and to establish standard values and test quality parameters. Three hundred and forty-two consecutive patients with different symptoms in the upper limbs (upper motor neuron symptoms, cerebellar upper limb ataxia, and/or sensory deficits in the upper limbs) and 140 healthy controls took part in the study. All patients and controls had to trace over a 25 cm high figure '8' on a graphic tablet, to tap with the stylus on the tablet and to perform the nine-hole-peg test (9HPT). Patients were additionally examined using clinical standard scales to classify motor dysfunctions of the upper limbs. One hundred and eighty-nine patients and 27 controls were tested twice to investigate the test reliability. Kinematic analysis of the tablet data was performed by kernel estimators, oscillatory activity by spectral analysis. Total power in the 2--10 Hz band was very specific for ataxia versus other motor symptoms. Tapping and 9HPT could well distinguish patients from controls, and patients with predominant motor neuron or cerebellar symptoms from patients with predominant sensory dysfunctions. Mean drawing error did not differ between motor and sensory dysfunctions. The test--retest reliability was similarly high for both spectral analysis and 9HPT.

A simple measure of correlation across time, frequency and space between continuous brain signals.

This paper introduces a simple but systematic method to estimate correlations between the spectral energy of two continuous electrophysiological signals in such a way that it can detect relationships between different frequencies and different latencies. From two series of signals (e.g. electroencephalogram, magnetoencephalogram or local field potentials) recorded from two sites in response to repeated sensory stimulations, the method computes the time-frequency energy of each signal. Then, it computes the Spearman rank order correlation coefficient across all the trials between the energy of the first signal series in one time-frequency region and the energy of the second signal series in a second time-frequency region. The method was designed to analyze interactions between frequency bands, in an effort to describe how the main brain rhythms interact with each other across time and space. It was applied to two simulations and to intracranial electro-encephalogram (EEG) recordings obtained from an epileptic patient performing two verbal discrimination tests (a phonological and a semantic task). It led to the identification of different correlations patterns in the gamma band depending on the level of semantic analysis performed by the patient.

Human anterior and frontal midline theta and lower alpha reflect emotionally positive state and internalized attention: high-resolution EEG investigation of meditation.

EEG spectral power and coherence estimates in the individually defined delta, theta, alpha-1, alpha-2, and alpha-3 bands were used to identify and characterize brain regions involved in meditative states, in which focused internalized attention gives rise to emotionally positive "blissful" experience. Blissful state was accompanied by increased anterior frontal and midline theta synchronization as well as enhanced theta long-distant connectivity between prefrontal and posterior association cortex with distinct "center of gravity" in the left prefrontal region (AF3 site). Subjective scores of emotional experience significantly correlated with theta, whereas scores of internalized attention with both theta and alpha lower synchronization. Our results propose selective associations of theta and alpha oscillating networks activity with states of internalized attention and positive emotional experience.

The effects of subthreshold 1 Hz repetitive TMS on cortico-cortical and interhemispheric coherence.

OBJECTIVES: Repetitive transcranial magnetic stimulation (rTMS) shows promise as a treatment for various movement and psychiatric disorders. Just how rTMS may have persistent effects on cortical function remains unclear. We hypothesised that it may act by modulating cortico-cortical and interhemispheric connectivity. To this end we assessed cortico-cortical and interhemispheric coherence before and after low frequency, subthreshold rTMS of the left motor cortex. METHODS: Fifteen healthy subjects received one train (1Hz, 90% of active motor threshold, 1500 stimuli) of rTMS to the left motor hand area. Spectral power and coherence estimates were calculated between different electroencephalogram (EEG) signals at rest and while muscles of the distal upper limb were tonically contracted. RESULTS: rTMS over the left motor hand area caused a significant increase in ipsilateral EEG-EEG coherence and in the interhemispheric coherence between motor areas in the alpha band. The effects of rTMS lasted up to 25 min post-stimulation. There was no significant change in EEG-EEG coherence over the hemisphere contralateral to stimulation. CONCLUSIONS: Low frequency, subthreshold rTMS of the motor cortex increases ipsilateral cortico-cortical and interhemispheric coherence in the alpha band. This may, in part, mediate the inhibitory effects of low frequency rTMS.
