LC-PDA and LC-ESI-MS separation and determination of process-related substances arising from stilbene-type fluorescent whitening agents. Application to monitoring of their photodegradation products in industrial effluents and aqueous environmental systems.

A simple and rapid gradient elution high-performance liquid chromatographic method using photodiode array and electrospray ionization mass spectrometric detectors was developed for separation and determination of the process-related substances and photodegradation products of stilbenesulfonic acids, viz. 4,4'-dinitrostilbene-2,2'-disulfonic acid (DNSDA), 4-amino-4'-nitrostilbene-2,2'-disulfonic acid (ANSDA), and 4,4'-diaminostilbene-2,2'-disulfonic acid (DASDA) in industrial waste waters. Gradient elution was carried out using ammonium acetate and acetonitrile as mobile phase and an Inertsil-ODS 3V column for separation. The negative-ion electrospray ionization mass spectra containing [M-H]- ions of sulfonic acids allowed molecular mass determination of unknowns and the structures were proposed on the basis of the fragment ions in the MS/MS spectra.

[Enzyme immunoassay (EIA) for the estrogenic stilbene derivative diethylstilbestrol (DES) (author's transl)].

An enzyme immunoassay (EIA) for diethylstilbestrol (DES) is described, which is based on the competitive reaction between DES and peroxidase-labelled DES for an antibody, which has been bond to an insoluble support. The preparation of the enzyme-labelled DES necessitates the use of highly purified 4-O-(carboxypropyl)-diethylstilbestrol (CP-DES) to obtain a highly immunoreactive conjugate. CP-DES was synthesized from DES and ethyl-4-bromobutylate and was purified by a novel isolation procedure involving a simple solvent partition followed by hydrolysis of the ester. It was characterized by it's chemical and biological properties. The assay covers the range from 0.2 to 20 pmol/test (approximately equal to 0.05-5 ng/test). It has sufficient sensitivity and specificity compared with radio immuno assay to make it a potential method for controlling the misuse of this estrogenic substance in animal production.

The disulfonic stilbene DIDS and the marine poison maitotoxin activate the same two types of endogenous cation conductance in the cell membrane of Xenopus laevis oocytes.

In the present experiments we exposed the intra- or extracellular surface of excised giant membrane patches of Xenopus laevis oocytes bathed in 140 mmol/l Na-aspartate solution to the anion transport inhibitor 4,4'-diisothiocyanatostilbene-2,2'-disulfonate (DIDS, 250 micromol/l). We observed that DIDS activated at least two cation conductances: (1) a non-selective cation (NSC) conductance that was mediated by channels of approximately 27 pS and resembled the stretch-activated cation conductance that has been observed in the oocyte cell membrane previously, and (2) a Na+-selective conductance, the single-channel events of which could not be resolved and which resembled the depolarization-induced Na+ conductance that has also been observed in the oocyte cell membrane previously. Both conductances were blocked by 1 mmol/l amiloride from the intra- and extracellular surfaces but inhibition of the NSC conductance by extracellular amiloride was less pronounced. Both conductances activated only slowly with a delay of 15-60 s after application of DIDS and remained active even after DIDS was washed off. This suggests that DIDS caused the exocytosis of preformed channels and this interpretation was supported by our additional observation that extracellular application of maitotoxin (MTX) mimicked the effects of DIDS. MTX is a marine toxin that has recently been reported to induce exocytosis in Xenopus laevis oocytes. The fact that DIDS and MTX each carry two sulfonyl groups suggests that they act on the same positively charged binding sites of an exocytosis-inducing protein. Our observations demonstrate that using DIDS to inhibit heterologously expressed anion transporters in the cell membrane of Xenopus laevis oocytes may compromise proper determination of the transporter currents. This effect can be prevented if the DIDS-activated endogenous cation conductances are suppressed by application of amiloride to the cytoplasmic surface of the cell membrane.

Catalysis of the oxidation of steroid and stilbene estrogens to estrogen quinone metabolites by the beta-naphthoflavone-inducible cytochrome P450 IA family.

Diethylstilbestrol (DES) or catecholestrogens are metabolized by microsomal enzymes to quinones, DES Q or catecholestrogen quinones, respectively, which have been shown to bind covalently to DNA and to undergo redox cycling. The isoforms of cytochrome P450 catalyzing this oxidation of estrogens to genotoxic intermediates were not known and have been identified in this study by (a) using microsomes of rats treated with various inducers of cytochrome P450; (b) using purified cytochrome P450 isoforms; and (c) examining the peroxide cofactor concentrations necessary for this oxidation by microsomes or pure isoenzymes. The highest rate of oxidation of DES to DES Q was obtained using beta-naphthoflavone-induced microsomes (14.0 nmol DES Q/mg protein/min) or cytochrome P450 IA1 (6.4 pmol DES Q/min/pmol P450). Isosafrole-induced microsomes or cytochrome P450 IA2 oxidized DES to quinone at one-third or one-fifth of that rate, respectively. Low or negligible rates of oxidation were measured when oxidations were catalyzed by microsomal rat liver enzymes induced by phenobarbital, ethanol, or pregnenolone-16 alpha-carbonitrile or by pure cytochromes P450 IIB1, IIB4, IIC3, IIC6, IIE1, IIE2, IIG1, or IIIA6. Cytochrome P450 IA1 also catalyzed the oxidation of 2- or 4-hydroxyestradiol to their corresponding quinones. The beta-naphthoflavone-induced microsomes and cytochrome P450 IA1 had the highest "affinity" for cumene hydroperoxide cofactor (Km = 77 microM). Cofactor concentrations above 250 microM resulted in decreased rates of oxidation. The other cytochrome P450 isoforms required much higher cofactor concentrations and were not inactivated at high cofactor concentrations. The data demonstrate that beta-naphthoflavone-inducible cytochrome P450 IA family enzymes catalyze most efficiently the oxidation of estrogenic hydroquinones to corresponding quinones. This oxidation may represent a detoxification pathway to keep organic hydroperoxides at minimal concentrations. The resulting quinone metabolites may be detoxified by other pathways. However, in cells with decreased detoxifying enzyme activities, quinones metabolites may accumulate and initiate carcinogenesis or cell death by covalent arylation of DNA or proteins.

Stable derivatives for the gas chromatographic determination of synthetic anabolic stilbene residues (diethylstilbestrol, dienestrol and hexestrol) in meat and organs of treated cattle in the sub-parts per billion (10(9)) level.

A method for the determination of diethylstilbestrol and the related compounds dienestrol and hexestrol residues in meat and organs of treated cattle is described. After extraction and clean-up, these synthetic estrogens are subjected to reaction with pentafluorobenzoyl chloride, which gives very stable perfluoro esters that are suitable for gas chromatographic determination using an electron-capture detector. With the careful clean-up and the very sensitive response of these derivatives, it is possible to reach a limit of detection in the sub-parts per billion (10(9)) range starting with only 5 g of sample.

Effects of DIDS, a disulfonic stilbene derivative, on chloride movements in toad skeletal muscles.

In order to investigate the characteristics of the movement of Cl- ions in toad skeletal muscles we decided to study the relative membrane permeabilities of chloride and nitrate and the effects of DIDS (4,4'-diisothyocyanatostilbene-2,2'-disulphonate) upon the hyperpolarizations produced in muscle fibers when chloride or nitrate ions rapidly replace impermeant sulphate ions in the external solution. For experiments where membrane potential changes were recorded in response to sudden changes in extracellular solutions, small bundles from the semitendinosus muscles were used. We showed that DIDS reduced in a reversible manner the Cl- permeability (pCl) in toad skeletal muscle fibers. The results supporting this conclusion were the following. First, a diminished hyperpolarization in response to a sudden exposure of the fibers to a solution containing Cl-. In these experiments DIDS reduced the pCl/pK ratio to 5.5 from a control value of 12. Second, a smaller transient of the resting potential when [Cl]o was changed from 120 to 30 mM and vice versa.

Disulfonic stilbenes prevent beta-amyloid (25-35) neuronal toxicity in rat cortical cultures.

Anion exchange proteins were recently identified among some of the proteins found clustered together in the hallmark plaques and tangles of Alzheimer's patient's brains. Anion exchange proteins underlie chloride/bicarbonate exchange, cell shape regulation and participate in removal of aged cells by the immune system. In this study we compared the neuroprotective efficacy of an anion exchanger inhibitor, 4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid (DIDS), against beta-amyloid((25-35)) neurotoxicity, staurosporine-induced apoptosis and glutamate-induced necrosis in primary cortical cultures. We demonstrate potent neuroprotective efficacy with DIDS against beta-amyloid((25-35)) and staurosporine, but not against glutamate. Our results suggest that anion exchange proteins may play an important role in beta-amyloid toxicity and that DIDS may represent a viable therapeutic agent for Alzheimer's disease.

Retinobenzoic acids. 3. Structure-activity relationships of retinoidal azobenzene-4-carboxylic acids and stilbene-4-carboxylic acids.

Alkyl-substituted azobenzene-4-carboxylic acids are potent differentiation inducers of human promyelocytic leukemia cell line HL-60 to mature granulocytes. Their structure-activity relationships are very similar to those of other retinoidal benzoic acids which are generally represented by 4 and named retinobenzoic acids. The structure-activity relationships of azobenzenecarboxylic acids can also be applied to the known retinoid TTNPB (3). Thus, (E)-4-[2-(3,4-diisopropylphenyl)-1-propenyl]benzoic acid (St30 (28] and (E)-4-[2-(3-tert-butylphenyl)ethenyl]benzoic acid (St40 (29], the acyclic alkyl analogues of TTNPB, are nearly as active as retinoic acid. Among the oxidatively derived compounds (Az90, Ep series and Ox series) of azobenzene- or stilbenecarboxylic acids, Az90 (71) and Ep80 (61) have strong activities. However, all the bishydroxylated derivatives of TTNPB are inactive, while a diketo analogue Ox580 (69) has only weak potency. The activities of conformationally restricted compounds of TTNPB offer some information on the stereochemistry of the active form of these retinoidal compounds.

A stable cis-stilbene derivative encapsulated in cucurbit[7]uril.

cis-Diaminostilbene dihydrochloride encapsulated in cucurbit[7]uril does not spontaneously isomerize to the trans isomer at room temperature as a result of the strong host-guest interactions including strong hydrogen bonds between the two protonated amine termini of the C-shaped guest and the portal oxygen atoms of the host.

Cyclodextrin complexation of a stilbene and the self-assembly of a simple molecular device.

(E)-4-tert-Butyl-4'-oxystilbene, 1(-), is thermally stable as the (E)-1(-) isomer but may be photoisomerized to the (Z)-1(-) isomer as shown by UV-vis and (1)H NMR studies in aqueous solution. When (E)-1(-) is complexed by alphaCD two inclusion isomers (includomers) form in which alphaCD assumes either of the two possible orientations about the axis of (E)-1(-) in alphaCD.(E)-1(-) for which (1)H NMR studies yield the parameters: k(1)(298 K)= 12.3 +/- 0.6 s(-1), DeltaH(1)(++)= 94.3 +/- 4.7 kJ mol(-1), DeltaS1(++)= 92.0 +/- 5.0 J K(-1) mol(-1), and k(2)(298 K)= 10.7 +/- 0.5 s(-1), DeltaH(2)(++)= 93.1 +/- 4.7 kJ mol(-1), DeltaS2(++)= 87.3 +/- 5.0 J K(-1) mol(-1) for the minor and major includomers, respectively. The betaCD.(E)-1(-) complex either forms a single includomer or its includomers interchange at the fast exchange limit of the (1)H NMR timescale. Complexation of 1(-) by N-(6(A)-deoxy- alpha-cyclodextrin-6(A)-yl)-N'-(6(A)-deoxy- beta-cyclodextrin-6(A)-yl)urea, results in the binary complexes 2.(E)-1(-) in which both CD component annuli are occupied by (E)-1(-) and which exists exclusively in darkness and 2.(Z)-1(-) in which only one CD component is occupied by (Z)-1(-) and exists exclusively in daylight at lambda > or = 300 nm. Irradiation of solutions of the binary complexes at 300 and 355 nm results in photostationary states dominated by 2.(E)-1(-) and 2.(Z)-1(-), respectively. In the presence of 4-methylbenzoate, 4(-), 2.(Z)-1(-) forms the ternary complex 2.(Z)-1(-).4(-) where 4(-) occupies the second CD annulus. Interconversion occurs between 2.(Z)-1(-).4(-) and 2.(E)-1(-)+4(-) under the same conditions as for the binary complexes alone. Similar interactions occur in the presence of 4-methylphenolate and 4-methylphenylsulfonate. The two isomers of each of these systems represent different states of a molecular device, as do the analogous binary complexes of N,N-bis(6(A)-deoxy- beta-cyclodextrin-6(A)-yl)urea, 3, [3.(E)-1(-) and 3.(Z)-1(-), where the latter also forms a ternary complex with 4(-).

Pressure control of diastereodifferentiating [2 + 2] photocycloaddition of (E)-stilbene to chiral fumarate upon direct and charge-transfer excitation.

The geometry (donor-acceptor distance) and association constant of the ground-state CT complex were considerably affected by applied pressure, thus enhancing the reaction rate and the adduct yield particularly upon CT excitation, but the diastereoselectivity did not show any substantial change at elevated pressures in both excitation modes.
